Abstract
Description
[0001] The present invention relates to the use of selected ultraviolet (UV) filters selected from the group consisting of octocrylene, ethylhexyltriazone and butyl methoxydibenzoylmethane for protecting skin bacteria of the genus Lactobacillus, such as L. crispatus, preferably in the simultaneous presence of Staphylococcus epidermidis, from UV radiation to promote the maintenance of a healthy skin microbiome after UV irradiation.
[0002] The skin is the outermost protective layer of an organism and is the largest organ in the human body. It acts as a barrier and protects the body from external factors. It is also well known that the skin surface is inhabited by a variety of microorganisms that form the skin microbiome (often also referred to as the skin microbiome). Most of these microorganisms are symbiotic organisms that live in a mutually beneficial symbiotic relationship with the skin's immune system. Dysbiosis of the skin microbiome is associated with many skin pathologies, such as acne, atopic dermatitis, and psoriasis.
[0003] Endogenous inhabitants of healthy skin are lactobacilli (Lactobacillus family), such as Lactobacillus crispatus. They have been described as playing a beneficial role in the skin habitat, where they play a multifactorial local mechanism of action against pathogens and inflammation. On the one hand, lactobacilli have been shown to compete directly with skin pathogens by inhibiting adhesion, producing antimicrobial metabolites, and by affecting pathogen metabolism. The competitive antipathogenic effects of lactobacilli have been mechanistically described for common different skin pathogens, such as Staphylococcus aureus, Propionibacterium acnes, and Candida albicans. On the other hand, lactobacilli also have the immunomodulatory ability associated with reducing excessive skin inflammation. Their effects on the immune system are mediated by bacterial metabolites and cell wall-related or excretion microorganism-associated molecular patterns (MAMPs). In addition, lactobacilli can also enhance skin barrier function, such as when the skin barrier function is destroyed due to infection or inflammatory skin diseases.
[0004] It is well known that UV light destroys microorganisms such as bacteria. Therefore, UV radiation may induce significant changes in the skin microbiome and thereby may alter the immune response and / or lead to a compromised skin barrier.
[0005] Therefore, there is a continuing need to selectively protect beneficial bacteria, such as skin bacteria, in particular of the genus Lactobacillus, from UV radiation. Furthermore, it is also desirable to simultaneously protect another beneficial skin commensal, Staphylococcus epidermidis.
[0006] Therefore, the inventors have developed a targeted in vitro liquid model in a 96-well microplate format to screen for UV protection active substances ("UV filters") that enable bacterial survival after UV irradiation. The effects of selected UV irradiation conditions tested on the microbial populations Lactobacillus crispatus and Staphylococcus epidermidis in the presence of UV filter samples and combinations thereof were evaluated by the evolution of the residual microbial population after 4 h. The bacterial growth inhibition as a result of UV irradiation was compared with that of the same strains without UV irradiation and expressed as a percentage (%) of survival. In addition, a clinical single-blind placebo-controlled trial specifically evaluated the effects of UV irradiation on the composition of the skin microbiome of human skin and the protection of the skin microbiome by a representative SFP20 sunscreen versus a placebo formulation.
[0007] Surprisingly, it has now been found that certain UV filter substances are particularly suitable for protecting skin bacteria of the genus Lactobacillus, such as in particular Lactobacillus crispatus, more preferably in the simultaneous presence of Staphylococcus epidermidis, from the harmful effects of UV radiation by significantly and selectively increasing the survival rate of skin bacteria of the genus Lactobacillus, such as in particular Lactobacillus crispatus, more preferably in the simultaneous presence of Staphylococcus epidermidis after irradiation with UV light.
[0008] Thus, in a first embodiment, the present invention relates to a method for protecting bacteria of the genus Lactobacillus, preferably in the simultaneous presence of Staphylococcus epidermidis, from the harmful effects of UV radiation, said method comprising the step of applying to an external surface of a human body having a skin microbiota comprising said bacteria, before exposing said external surface to UV radiation, an effective amount of a sunscreen composition comprising one or more UV filters selected from the group consisting of octocrylene, butyl methoxydibenzoylmethane and ethylhexyl triazone.
[0009] In another embodiment, the present invention relates to the use of one or more UV filters selected from the group consisting of octocrylene, butyl methoxydibenzoylmethane and ethylhexyl triazone for protecting bacteria of the genus Lactobacillus from the harmful effects of UV radiation, preferably in the simultaneous presence of Staphylococcus epidermidis.
[0010] In an additional embodiment, the present invention is directed to a method for increasing the differential abundance of bacteria of the genus Lactobacillus in the skin microbiome of an individual after exposure to UV radiation, preferably in the simultaneous presence of Staphylococcus epidermidis, the method comprising the step of topically applying to an external surface of a body in need thereof, prior to exposing the external surface to UV radiation, an effective amount of a sunscreen composition comprising one or more UV filters selected from the group consisting of octocrylene, butyl methoxydibenzoylmethane and ethylhexyl triazone.
[0011] In another embodiment, the present invention also provides a method for protecting the external surfaces of the human body by selectively increasing the differential abundance of skin bacteria of the genus Lactobacillus after exposure to UV radiation, preferably in the presence of Staphylococcus epidermidis in the skin microbiome, the method comprising the step of topically applying to the external surface a sunscreen composition comprising one or more UV filters selected from the group consisting of octocrylene, butyl methoxydibenzoylmethane and ethylhexyl triazone before exposure to UV radiation.
[0012] In another embodiment, the present invention relates to the use of one or more UV filters selected from the group consisting of octocrylene, butyl methoxydibenzoylmethane and ethylhexyl triazone, or a sunscreen composition comprising one or more of said UV filters, for selectively increasing the differential abundance of skin bacteria of the genus Lactobacillus after exposure to UV radiation, preferably in the presence of Staphylococcus epidermidis in the skin microbiome of an individual in need thereof.
[0013] In another embodiment, the invention relates to a composition comprising one or more UV filters selected from the group consisting of octocrylene, butyl methoxydibenzoylmethane and ethylhexyl triazone for use in the treatment or prevention of dysbiosis and / or for strengthening the surface barrier of the skin after UV radiation.
[0014] All methods and uses disclosed herein are particularly suitable for treating or preventing dysbiosis and / or strengthening the surface barrier of the skin after UV radiation. The compositions, methods and uses as disclosed herein are suitable for maintaining a healthy and well-balanced skin microbiome after UV radiation.
[0015] Since the number of bacteria of the genus Lactobacillus increases, ie the abundance increases, after UV irradiation, the metabolites secreted by said bacteria may also increase.
[0016] It is well known to those skilled in the art that compositions comprising UV filters may also be referred to as sunscreen compositions.
[0017] Without wishing to be bound by theory, these metabolites are believed to contribute to the body's natural defence system. Thus, in another aspect, the invention relates to the use of a sunscreen composition comprising one or more UV filters selected from the group consisting of octocrylene, butyl methoxydibenzoylmethane and ethylhexyl triazone for improving the natural defence system of a surface of the human body by the balance of the microbiota after exposure of the surface to UV radiation.
[0018] In a further aspect, the present invention also relates to a method for improving the natural defence system of a surface of the human body by balancing the microbiota, said method comprising the steps of applying to said surface a sunscreen composition comprising one or more UV filters selected from the group consisting of octocrylene, butyl methoxydibenzoylmethane and ethylhexyl triazone before exposing said surface to UV radiation, then exposing said surface to UV radiation and optionally appreciating the beneficial effects produced thereby.
[0019] The protection according to the invention against skin bacteria of the genus Lactobacillus, such as in particular Lactobacillus crispatus, is characterized in that after exposure to UV radiation (at 50 SED), the survival of skin bacteria of the genus Lactobacillus, such as in particular Lactobacillus crispatus, is increased by at least 100% compared to a control.
[0020] In an additional and advantageous embodiment, the S. epidermidis are also simultaneously protected from UV radiation. Said protection is characterized in that the survival rate of the S. epidermidis after exposure to UV radiation (at 15 SED) is increased by at least 20% compared to a control.
[0021] Particularly preferred UV filters according to the invention are octocrylene and a mixture of ethylhexyl triazone, butyl methoxydibenzoylmethane and bis-ethylhexyloxyphenol methoxyphenyl triazine, since they lead to particularly good survival of skin bacteria of the genus Lactobacillus, in particular Lactobacillus crispatus.
[0022] The most preferred Lactobacillus species in all embodiments of the present invention is Lactobacillus crispatus CIP 103604.
[0023] The most preferred Staphylococcus epidermidis in all embodiments of the present invention is Staphylococcus epidermidis ATCC 12228.

[0110] In vitro UV model
[0111] The microbial strains used in this study are listed below and their cultures were reconstituted according to the instructions of the supplier's reference strain collection CRBIP (Institut Pasteur Collection, France) or ATCC (American Type Culture Collection):
[0112] -Lactobacillus crispatus CIP 103604
[0113] -Staphylococcus epidermidis ATCC 12228
[0114] Fill culture medium (140 μ L) in 96 well plates, and add the calibrated inoculum (20 μ L) of every kind of bacterial strain to reach the ultimate concentration of 1.106 colony forming units / milliliter (cfu / mL).After contamination, add sample (40 μ L in triplicate; 20% v / v), and fully mix to ensure uniform distribution. Then, according to the UV dosage shown in the following table 1, plate is subjected to UV stress by UV irradiator system (BioSun, Vilber Lourmat, France).Selected specific UV dosage is for making the survival rate of microbial colony between 10% and 49%.
[0115] strain SED(unit) <![CDATA[UVA+UVB(mJ / cm 2 )]]> Survival rate after 4 hours (%) Staphylococcus epidermidis 15 150 ±* Lactobacillus crispatus 50 500 -
[0116] Table 1. Selection of UV irradiation conditions for each microbial strain. The energy ratio between UVA and UVB is 1:19. ±*: the survival rate of the microbial population is between 21% and 49%; -: the survival rate of the microbial population is equal to or lower than 20%.
[0117] For the microbial population counts, aliquots of contaminated samples of Lactobacillus crispatus and Staphylococcus epidermidis were serially diluted (4 ten-fold dilutions followed by 8 two-fold dilutions in triplicate) in a medium on a 96-well plate containing triphenyltetrazol. The plates were then incubated at 37°C ± 2,5°C during 48h, taking into account the respiratory type of each strain. After 48h under optimal growth conditions, the final dilution exhibited a pink color (triphenyltetrazol), or turbidity indicated the contamination rate and allowed the determination of the reduction rate of the microbial population (most probable number MPN method). The control performed during this experiment was a medium containing bacteria incubated without UV stress. The microbial population counts are expressed in cfu / mL and are the average of experiments performed in triplicate.
[0118] The use levels of single UV filters and combinations in the test formulations were calculated to obtain predicted in vitro SPF values of 5.75 + / - 0.15 and 23.45 + / - 0.65, respectively (Table 2). The UV filters were mixed in two oils, Tegosoft XC (55%) and Miglyol 829 Eco (ad 100%), and the emulsifier Tween 80 (5%).
[0119] The effect of selected UV irradiation conditions on the survival of the microbial populations Lactobacillus crispatus and Staphylococcus epidermidis in the presence of 20% v / v of the test samples 1-10 was compared with the corresponding strains in the absence of samples 1-10 and without exposure to UV irradiation (control). The results after 4 hours of incubation were compared with the bacterial population incubated without UV stress (determination of bacterial population survival) and expressed as a percentage of survival. The relative protection percentage (%) was calculated based on the difference between the survival percentages of the sample and the vehicle control (with UV irradiation) divided by the survival percentage of the vehicle control. Therefore, the survival rate of each microbial population and the relative protection achieved by the UV filter were calculated and summarized in Table 2.
[0120] Table 2. Test formulations of UV filters and ingredients in % by weight (* mixed with 55% TegosoftXC, ad100% Miglyol 829Eco, 5% Tween 80). The survival results for each microbial population are calculated in % compared to the untreated and non-irradiated control. In brackets: the relative protection achieved by the UV filters is given in % relative to the vehicle control.

[0124] In vivo study setup
[0125] This clinical single-blind placebo-controlled trial specifically assessed the effects of UV irradiation on the composition of the skin microbiome of human skin and the protection of the skin microbiome by a representative SFP20 sunscreen versus a placebo formulation.
[0126] The investigational product consisted of two test formulations, one containing four UV filters (i.e. Shield, 340, EHS and 1789) for broad spectrum UV protection, and a vehicle formulation without UV filters (“placebo”) to assess the potential to protect the skin microbiome from UV exposure:
[0127] 1. Placebo formulation, which contained vehicle only; the UV filter had been replaced by a higher oil content.
[0128] 2.SPF20 sunscreen formula
[0129] Table 3: List of ingredients for placebo formulation and SPF20 sunscreen formulation
[0130]
[0131]
[0132] After obtaining informed consent from the volunteers, the test formulation was applied to the backs of 10 healthy female subjects (22-43 years old, Fitzpatrick skin type II-III), followed by two minimal erythema doses (2 MED) of UV irradiation. 2 Approximately 2 mg of both test formulations were applied to approximately 25 cm of the body surface. 2 Two test areas of skin. The individual MED of the subjects was determined one day before treatment. Prior to UV irradiation, skin surface swabs were taken from untreated, non-irradiated areas for analysis. Three defined areas on the back (approximately 25 cm 2 ) were exposed to 2 MED of UV radiation using a single-port solar simulator. Two hours after UV exposure, skin swab samples were collected on the three irradiated areas for analysis.
[0133] DNA was extracted from swab samples of nine completers using the ZymoBIOMICS DNA Miniprep kit according to the manufacturer's protocol and sequenced on an Illumina MiSeq in a 2×300 paired configuration. Primers targeted the V3-V4 region of the bacterial 16S rRNA gene. Sequences were then processed into amplicon sequence variants (ASVs).
[0134] A software tool called Songbird, as listed in Morton et al. (Nat. Commun., 2019.10(1): p. 2719), was applied to the categorical variables of radiation, treatment, and verum to study the differential ranking of taxonomy from the perspective of 16S rDNA. Songbird models the differential ranking to provide the relationship between each feature and the classification or quantitative measurement in question. These different features then contain coefficients to describe their relationship with the modeled variables.
[0135] In vivo results:
[0136] Ten Caucasian female subjects aged between 22 and 43 years (mean age 32±2 years, Fitzpatrick skin types II (4 subjects) and III (6 subjects)) were enrolled and completed the study. No adverse reactions were observed during the study.
[0137] Based on 16S rRNA sequencing analysis of samples from nine completers, Lactobacillus crispatus was identified as positively associated only with the formulation containing UV filters, whereas it showed negative associations with irradiation and with the placebo formulation.
[0138] Table 4: Coefficients for two Lactobacillus crispatus species from Songbird. T:YES is the association of these traits with verum, compared to T:NO which is the control for this variable.
[0139]
[0140] Specific microorganisms will benefit from additional UV protection, as observed with Lactobacillus crispatus. Interestingly, Lactobacillus crispatus is rarely described as a skin commensal. These major vaginal bacteria play a key role by forming beneficial biofilms to protect host tissues and act as components of the innate immune response. They metabolize glucose to lactic acid, thereby creating an acidic environment (~ pH 4-4.5), competitive exclusion, and release antimicrobial compounds (e.g., H2O2, bacteriocins, and other byproducts), thereby maintaining the skin's self-defense and the symbiosis of the skin microbial flora.
