A conjugate of a dual agonist and a saccharide antigen, and methods of making and using the same

By conjugating the iNKT cell agonist KRN7000 with the mincle receptor agonist vizantin to a glycoantigen, a dual agonist-glycoantigen conjugate was constructed, which solved the problem of insufficient immunogenicity of fully synthetic tumor glycoantigen vaccines and achieved a stronger immune response and tumor cell killing effect.

Abstract

This invention relates to a conjugate of a dual agonist and a glycoantigen, wherein the conjugate contains a dual agonist and a glycoantigen, the dual agonist being an iNKT cell agonist and a mincle receptor agonist, the iNKT cell agonist being an α-galactosylceramide analog, and the mincle receptor agonist being a trehalose derivative; the general structural formula of the conjugate is shown in Formula (I): X is the glycoantigen, and Y is the linker. The vaccine prepared from the conjugate of this invention can simultaneously activate iNKT cells and mincle receptors, inducing the production of high-titer IgG antibodies, effectively improving the immunogenicity of the glycoantigen, and showing good prospects for anti-tumor applications.
