[Technical Field]

[0001] The present invention relates to a pyrimido pyrimidinone compound and a pharmaceutical composition comprising the same, and, more specifically, to a pyrimido pyrimidinone compound having an effect of inhibiting PD-L1 expression and a pharmaceutical composition comprising the same as an active ingredient for use in the treatment or prevention of cancer.[Background Art]

[0002] PD-1 (Programmed death-1, CD279) is a receptor protein on T cells, which is known to suppress activation signals from the T cell receptor when binding to its ligands PD-L1 (Programmed death-Ligand 1, CD274) or PD-L2 (Programmed death-Ligand 2, CD273). That is, when the PD-1 receptor protein of the T cells binds to the ligand PD-L1 / 2, the functional activity of the T-cells, including T-cell proliferation, cytokine secretion, and cytolytic activity, is reduced, thereby protecting normal cells. PD-1 and PD-L1 / 2 interaction downregulates the immune response in infections, tumors or autoimmune diseases (Keir Me, Butte MJ, Freeman GJ, et al. PD-1 and its ligands in tolerance and immunity. [0003] Blockade of the PD-1 / PD-L1 interaction using antibodies against PD-1 and PD-L1 has been shown to restore and enhance T cell activation in many systems. In patients with cancer such as melanoma, non-small cell lung cancer, and renal cancer, PD-1 / PD-L1 combination inhibitory therapy using a monoclonal antibody are known.

[0004] In a study on the regulation of PD-L1 expression and immune anticancer response in cancer cells by targeting αvβ3-integrin, it has been found that transplantation of β3-integrin-depleted tumor cells drastically reduces the growth of primary tumors, and the result is attributed to reduced PD-L1 expression. Thus, inhibition of β3-integrin can increase the efficacy of immune anticancer drugs against melanoma (Proc Natl Acad Sci USA., 2019 Oct 1, 116(40), 20141-20150). It is known that retinoblastoma protein RB, known as a tumor suppressor, exhibits anticancer efficacy through inhibition of PD-L1 expression by inhibition of NF-kB activity. Also, in a melanoma model, curcumin and apigenin showed melanoma cell growth inhibitory effects and, in particular, apigenin was found to significantly suppress PD-L1 expression induced by IFN-γ. In addition, it is known that the expression of PD-L1 induced by IFN-γ is reduced by 40 to 80% in the lung cancer cell line A549 pretreated with EGCG and green tea extract (GTE). Meanwhile, in a kinase inhibitor library screening study that can increase immune anticancer effects against melanoma, regorafenib has been identified as the most powerful drug among about 20 drugs that reduced cell surface PD-L1 expression level by more than 50%. The drug strongly promotes anti-tumor efficacy when combined with IFN-γ or immune checkpoint blocker (ICB) in cells and in vivo, strongly suppresses JAK1 / 2-STAT1 and MAPK signaling and then reduces IFN-γ-induced PD-L1 and IDO1 expression (Clin Cancer Res., 2019 Jul 15, 25(14), 4530-4541).

It is also known that 3-phenyl-dihydropyrimido[4,5-d]pyrimidinone derivatives exhibit an inhibitory activity of 1 to 5 nM (IC 50 ) against Src kinase, but their effect on PD-L1 has not been mentioned (International Patent Publication No. WO 2005 / 011597).

WO 2018 / 009544 discloses bicyclic urea kinase inhibitors which are indicated therein as being useful in modulating the activity of a protein kinase such as SIK and hence, potentially useful in treating or preventing various protein-kinase related diseases. Cho et al., J. Med. Chem., 2018, 18, 353-8373 also discloses protein kinase inhibitors which are potentially useful in treating or preventing cancer.

Claims

1. A pyrimido pyrimidinone compound of formula (I) or a pharmaceutically acceptable salt thereof: wherein, R1 is phenyl unsubstituted or substituted with halogen or hydroxyl; R2 is a C1-C4 alkyl; R3 is hydrogen, halogen, a C1-C4 alkyl or a C1-C4 alkoxy; and R4 is phenyl unsubstituted or substituted with a C1-C4 alkyl, a C1-C4 haloalkyl or alkyldihydrooxazolyl, or alkylthiazolyl, wherein the term "alkyldihydrooxazolyl" means a dihydrooxazole group substituted with one or more C1-C4 alkyl groups and the term "alkylthiazolyl" means a thiazolyl group substituted with one or more C1-C4 alkyl groups.

The pyrimido pyrimidinone compound according to claim 1 or pharmaceutically acceptable salt thereof, wherein, R1 is phenyl, fluorophenyl or hydroxyphenyl; R2 is methyl; R3 is hydrogen, chloro, bromo, methyl or methoxy; and R4 is trifluoromethylphenyl, t-butylphenyl, dimethyldihydrooxazolylphenyl or t-butylthiazolyl.

4. A pharmaceutical composition for inhibiting programmed death-ligand 1 (PD-L1) comprising the pyrimido pyrimidinone compound as defined in any one of claims 1 to 3 or a pharmaceutically acceptable salt thereof, together with a pharmaceutically acceptable carrier.

5. A pharmaceutical composition as defined in claim 4 for use in a method of treating or preventing cancer.

6. The pharmaceutical composition for use according to claim 5, wherein the cancer to be treated or prevented is colon cancer, lung cancer, breast cancer, gastric cancer, cervical cancer, bladder cancer, blood cancer or non-Hodgkin's lymphoma.
