[0005] The inventors have surprisingly discovered that administration of a glucokinase activator (e.g., Dorzagliatin) can prevent, alleviate, or treat cognitive impairment or a neurodegenerative disease, delay the progression of cognitive impairment or a neurodegenerative disease, and reduce the risk of developing cognitive impairment or a neurodegenerative disease. Furthermore, the inventors have surprisingly discovered that Dorzagliatin delayed the progression of cognitive impairment or a neurodegenerative disease (e.g., mild cognitive impairment (MCI), Alzheimer's disease) and reduced the risk of developing a cognitive impairment-related disease in an animal model.

[0160] Preparation of the experimental environment: The laboratory environment was kept quiet. A circular maze / pool (135 cm in diameter, 45 cm in height) was set up. Visual references (spatial cues) were placed on the walls of the pool and around the pool for the animals to learn from. The pool area was subdivided into four virtual quadrants (northeast, northwest, southeast, and southwest), with the northeast quadrant defined as the target quadrant, containing a hidden escape platform. Here, we evaluated the effects of Dorzagliatin on the hippocampal energy metabolism and insulin signaling pathway by analyzing the protein expression levels of insulin receptors, key synaptic proteins, and glucose transporters in the hippocampus of GK rats.Method

Long-term treatment of Dorzagliatin can also inhibit the downregulation of two subunits of the key synaptic protein, the ionotropic glutamate receptor, suggesting that this may be a novel mechanism by which Dorzagliatin protects the memory function.
