[0094] Step i: 20 g of 4-ethoxyphenol (1) and 12.9 mL 100% sulfuric acid were mixed at room temperature and placed in a 30 °C water bath and stirred. The reaction was swirled occasionally as heating continued. After about 1.5 hr., the entire mass set up to a semi-solid. The reaction was cooled, semi-hard solids were broken up, and crushed ice was added with stirring; more crushed ice was added as the hydrolysis continued. The flask was washed out with some of the cold aqueous material from the hydrolysis. When the hydrolysis was complete, the solids of 5-ethoxy-2-methoxybenzenesulfonyl chloride (2) were filtered and washed with ice water. The yield was about 24 g.

The melting point was 69-71 °C.

[0107] Step viii: The (5-ethoxy-2-methoxy-4-(2-nitrovinyl)phenyl)(propyl)sulfane from the previous step was reduced using a solution of aluminum hydride (AIH3; alane) in tetrahydrofuran (THF). Alane is generated in situ from lithium aluminum hydride (LAH) as follows: a 3 L round bottom reaction flask was equipped with a temperature probe, ice bath, Friedrich condenser, pressure-equalizeddropping funnel, and overhead stirrer. A I M solution of LAH in THF (400 mL, 400 mmol) was charged to the reaction flask and stirred until the solution reached a temperature of 2 °C. One hundred percent sulfuric acid (10.6 mL, 200 mmol), produced by treating 96% H2SO4 with oleum, was added to the cold LAH solution, dropwise with vigorous stirring to avoid charring, over a period of one hour. Hydrogen was evolved and lithium sulfate precipitated to give a clear solution of alane in THF. The (5-ethoxy-2- methoxy-4-(2-nitrovinyl)phenyl)(propyl)sulfane was dissolved in 50 m L THF, then added rapidly to the round bottom flask with stirring while the temperature was maintained in the range of 0-4 °C. After the addition was complete, the reaction was stirred for 30 min. on ice, then the ice bath was removed and replaced with a heating mantle. The reaction was heated to reflux for 3 hr. then returned to the ice bath. Glauber's salt (Na2SO4[H2O]i0; 45 g, 140 mmol) was added in portions with vigorous stirring to quench the reaction while the temperature was maintained below 10 °C. The formed greyish-white solids were removed by suction filtration through Celite 545 and the filter cake was washed with fresh THF. The THF was removed by distillation, then the crude product was distilled using a Kugelrohr to obtain a nearly colorless oil of the amine free base.

Diethyl ether or t-butyl methyl ether was added to initiate crystal formation.
