Description

[Technical field]

[0001] The present invention relates to a salt form of a fibroblast growth factor receptor (FGFR) inhibitor (3-(2,6-difluoro-3,5-dimethoxyphenyl)-1-ethyl-8-(morpholin-4-ylmethyl)-1,3,4,7-tetrahydro-2H-pyrrolo[3',2':5,6]pyrido[4,3-d]pyrimidin-2-one), including a method for its preparation, which is useful in the treatment of FGFR-mediated diseases, such as cancer. [Background technology]

[0002] Fibroblast growth factor receptors (FGFRs) are receptor tyrosine kinases that bind to fibroblast growth factor (FGF) ligands. There are four FGFR proteins with ligand-binding capacity (FGFR1-4), which are involved in the regulation of many physiological processes, including tissue development, angiogenesis, wound healing, and metabolic control. Upon ligand binding, these receptors dimerize and undergo phosphorylation, which stimulates protein kinase activity and recruits many intracellular docking proteins. These interactions promote the activation of an array of intracellular signaling pathways (including Ras-MAPK, AKT-PI3K, and phospholipase C) that are important for cell growth, proliferation, and survival (reviewed in Eswarakumar et al. Cytokine & Growth Factor Reviews, 2005). Aberrant activation of this pathway, by overexpression of FGF ligands or FGFRs or activating mutations in FGFRs, can result in tumorigenesis, tumor progression, and tumor resistance to conventional cancer treatments. Genetic alterations leading to ligand-independent receptor activation have been reported in human cancers, including gene amplification, chromosomal translocations, and somatic mutations. Large-scale DNA sequencing of thousands of tumor samples has revealed that components of the FGFR pathway are among the most frequently mutated in human cancers. Many of these activating mutations are identical to germline mutations that cause skeletal dysplasia syndromes. Mechanisms that lead to aberrant ligand-dependent signaling in human diseases include overexpression of FGFs and altered FGFR splicing that enhances the receptor's promiscuous ligand-binding capacity (reviewed in Knights and Cook Pharmacology & Therapeutics, 2010; Turner and Grose, Nature Reviews Cancer, 2010). Therefore, developing inhibitors that target FGFRs may be useful in the clinical treatment of diseases in which FGF or FGFR activity is elevated.

[0003] Cancer types in which FGF / FGFR is involved include, but are not limited to, carcinomas (e.g., bladder, breast, cervical, colorectal, endometrial, gastric, head and neck, kidney, liver, lung, ovarian, prostate), hematopoietic malignancies (e.g., multiple myeloma, chronic lymphocytic lymphoma, adult T-cell leukemia, acute myeloid leukemia, non-Hodgkin's lymphoma, myeloproliferative neoplasms, and Waldenstrom's macroglobulinemia), and other neoplasms (e.g., glioblastoma, melanoma, and rhabdomyosarcoma). In addition to its role in oncogenic neoplasms, activation of FGFR has also been implicated in skeletal and chondrocyte disorders, including, but not limited to, achondroplasia syndrome and craniosynostosis syndrome.

[0004] Specifically, the FGFR4-FGF19 signaling system is involved in the pathogenesis of several cancers, including hepatocellular carcinoma (Heinzle et al., Cur. Pharm. Des. 2014, 20:2881). Ectopic expression of FGF19 in transgenic mice has been shown to result in liver tumor formation, and neutralizing antibodies against FGF19 have been shown to inhibit tumor growth in mice. Moreover, overexpression of FGFR4 has been observed in multiple tumor types, including hepatocellular carcinoma, colorectal cancer, breast cancer, pancreatic cancer, prostate cancer, lung cancer, and thyroid cancer. Furthermore, activating mutations in FGFR4 have been reported in rhabdomyosarcoma (Taylor et al. JCI 2009, 119:3395).

[0005] Currently, inhibitors of FGFR are being developed to treat cancer. For example, 3-(2,6-difluoro-3,5-dimethoxyphenyl)-1-ethyl-8-(morpholin-4-ylmethyl)-1,3,4,7-tetrahydro-2H-pyrrolo[3',2':5,6]pyrido[4,3-d]pyrimidin-2-one molecules and other small molecule inhibitors of FGFR have been reported, and these reports are described, for example, in U.S. Pat. See, for example, Publication Nos. 2012 / 0165305, 2014 / 0045814, 2013-0338134, 2014 / 0171405, 2014 / 0315902, 2016 / 0115164, 2016 / 0244448, 2016 / 0244449, and 2016 / 0244450. Thus, there is a need for new FGFR inhibitor molecule salts for preparing pharma- ceutical useful formulations and dosage forms having suitable properties, e.g., properties associated with facilitating the manufacture of safe, effective, and high quality pharmaceutical products. [0037] The salts of the present invention may be isolated in one or more solid forms. The phrase "solid form" as used herein refers to the salts of the present invention in either an amorphous state or a crystalline state ("crystalline form" or "crystalline solid"), and the salts of the present invention in a crystalline state may optionally include a solvent or water in the crystal lattice, for example, forming a solvated or hydrated crystal form. In some embodiments, the salts of the present invention are in a crystalline state as described herein. The term "hydrated" as used herein is intended to refer to a crystal form that includes one or more water molecules in the crystal lattice. Examples of "hydrated" crystal forms include hemihydrate, monohydrate, dihydrate, and the like. Other hydrate forms, such as channel hydrates and the like, are also included within the meaning of this term.

[0038] In some embodiments, the salt of the present invention can be prepared by any suitable method for preparing an acid addition salt. For example, the free base of compound 1 can be mixed with the desired acid in a solvent or melt. Alternatively, the acid addition salt of compound 1 can be converted to a different acid addition salt by anion exchange. The salt of the present invention prepared in a solvent system can be isolated by precipitating from the solvent. Precipitation and / or crystallization can be induced, for example, by evaporation, temperature reduction, addition of anti-solvent, or a combination thereof.

[0039] In some embodiments, the salt of the present invention is crystalline, including anhydrous, hydrated, non-solvated, or solvated crystalline forms. Examples of hydrates include hemihydrates, monohydrates, dihydrates, and the like. In some embodiments, the crystalline salt is anhydrous and non-solvated. "Anhydrous" means that the crystalline salt does not contain bound water in the crystal lattice structure, i.e., such compounds do not form crystalline hydrates.

[0040] In some embodiments, the salt of the present invention is substantially isolated. "Substantially isolated" means that the salt is at least partially or substantially separated from the environment in which it is formed or detected. Partial separation can include, for example, enrichment of the salt of the present invention in the composition. Substantial separation can include at least about 50%, at least about 60%, at least about 70%, at least about 80%, at least about 90%, at least about 95%, at least about 97%, or at least about 99% by weight of the salt in the composition.

[0041] The salts of the present invention include all those containing isotopes of atoms in such salts. Isotopes include atoms having the same atomic number but different mass numbers. For example, isotopes of hydrogen include tritium and deuterium.

[0042] The salt forms of the invention have been found to be highly crystalline and have desirable properties that may facilitate purification of the drug, such as by crystallization and, if necessary, recrystallization. Additionally, crystalline forms tend to have improved stability and may be easier to grind or micronize during drug formulation. Crystalline salts also tend to have superior solubility properties and may be more amenable to reproducible manufacture with well-defined acid / base ratios, facilitating the preparation of liquid formulations for oral as well as intravenous use.

[0043] The term "crystal" or "crystalline form" as used herein refers to a crystalline solid form of a compound, including, but not limited to, mono- or poly-crystalline forms (e.g., solvates, hydrates, clathrates, and co-crystals). As used herein, "crystalline form" is intended to refer to a particular lattice arrangement of a crystalline substance. Different crystalline forms of the same substance typically have different crystal lattices (e.g., unit cells), and these crystal lattice differences are attributed to different physical properties unique to each of these crystalline forms. In some cases, the different lattice arrangements differ in water or solvent content. Different crystal lattices can be identified by methods for characterizing the solid state, such as X-ray powder diffraction (XRPD). Other methods for characterization, such as differential scanning calorimetry (DSC), thermogravimetric analysis (TGA), dynamic vapor sorption and desorption (DVS), solid-state NMR, and the like, also further aid in the identification of crystalline forms and the determination of stability and solvent / water content.

[0044] Crystalline forms of a substance include both solvated (e.g., hydrated) and non-solvated (e.g., anhydrous) forms. Hydrated forms are crystalline forms that contain water in the crystal lattice. Hydrated forms can be stoichiometric hydrates, in which water is present in the lattice at a specific water / molecule ratio, such as in hemihydrates, monohydrates, dihydrates, etc. Hydrated forms can also be non-stoichiometric, in which the water content is variable and dependent on external conditions (such as humidity).

[0045] The term "substantially crystalline" as used herein means that the majority of the weight of a sample or preparation of the salt of the present invention (or its hydrate or solvate) is crystalline, and the remaining part of the sample is a non-crystalline form (e.g., amorphous form) of the same compound. In some embodiments, a substantially crystalline sample has a crystallinity of at least about 95% (e.g., about 5% is the non-crystalline form of the same compound), preferably a crystallinity of at least about 96% (e.g., about 4% is the non-crystalline form of the same compound), more preferably a crystallinity of at least about 97% (e.g., about 3% is the non-crystalline form of the same compound), even more preferably a crystallinity of at least about 98% (e.g., about 2% is the non-crystalline form of the same compound), even more preferably a crystallinity of at least about 99% (e.g., about 1% is the non-crystalline form of the same compound), and most preferably a crystallinity of about 100% (e.g., about 0% is the non-crystalline form of the same compound). In some embodiments, the term "sufficiently crystalline" means that the crystallinity is at least about 99% or about 100%.

[0046] Crystalline forms are most commonly characterized by XRPD. The reflection (peak) pattern of XRPD is typically considered to be a fingerprint of a particular crystal form. It is widely known that the relative intensity of XRPD peaks can vary widely depending on, among other things, the sample preparation technique, crystal size distribution, filters, sample mounting procedures, and the particular instrument used. In some cases, peaks may be newly observed or existing peaks may disappear depending on the instrument type or settings (e.g., whether a Ni filter is used). As used herein, the term "peak" refers to a reflection having a relative height / intensity of at least about 4% of the maximum peak height / intensity. Furthermore, instrumental differences and other factors may affect the 2θ values. Thus, peak assignments (such as those reported herein) may vary by ± about 0.2° (2θ), and the term "substantially" as used herein in connection with XRPD is intended to encompass the above-mentioned variations.

[0047] Similarly, temperature observations associated with DSC, TGA, or other thermal experiments may vary by about ±3° C. depending on the instrument, the particular settings, sample preparation, etc. For example, in DSC, it is known that the observed temperatures will depend on the rate of temperature change, as well as the sample preparation technique and the particular instrument utilized. Thus, values reported herein for DSC thermograms, as noted above, may vary by ±3° C. Thus, crystalline forms reported herein having DSC thermograms "substantially" as shown in any of the figures should be understood to include such variations.

[0048] The salts and compounds disclosed herein may include all those that contain isotopes of atoms. Isotopes include atoms with the same atomic number but different mass numbers. For example, isotopes of hydrogen include tritium and deuterium. The salts and compounds of the present invention may include all those that contain isotopes of atoms in intermediates or final compounds. Isotopes include atoms with the same atomic number but different mass numbers. For example, isotopes of hydrogen include tritium and deuterium. One or more of the constituent atoms of the compounds of the present invention may be replaced or replaced with an isotope of the atom to achieve natural or non-natural abundance ratios. [0294] Formulation, Dosage Forms, and Administration When the salt of the present invention described herein is utilized as a pharmaceutical, it can be administered in the form of a pharmaceutical composition (referring to a combination of a salt of Compound 1 described herein and at least one pharma- ceutically acceptable carrier). Such compositions can be prepared in a manner well known in the pharmaceutical art, and can be administered by various routes of administration, depending on whether the desired treatment is local or systemic, and on the area to be treated. Administration can be topical (including ocular administration, and administration to mucous membranes (including intranasal, intravaginal, and intrarectal delivery)), pulmonary (e.g., administration by inhalation or insufflation of powder or aerosol (including by nebulizer), intratracheal, intranasal, subcutaneous, and transdermal administration), ophthalmic, oral, or parenteral. Methods for ocular delivery can include topical administration (ocular drops), subconjunctival, periocular, or intravitreal injection, or introduction by a balloon catheter or intraocular insert surgically placed in the conjunctival sac. Parenteral administration includes intravenous, intraarterial, subcutaneous, intraperitoneal, or intramuscular injection or infusion, or intracranial (e.g., intrathecal or intraventricular) administration. Parenteral administration can be in the form of a single bolus dose, or by, for example, a continuous infusion pump. Pharmaceutical compositions and formulations for topical administration can include transdermal patches, ointments, lotions, creams, gels, drops, suppositories, sprays, liquids, and powders. Conventional pharmaceutical carriers, aqueous, powder, or oily bases, thickeners, and the like, may be necessary or desirable.

[0295] The present disclosure also includes pharmaceutical compositions that include a salt of Compound 1 as an active ingredient in combination with one or more pharma- ceutically acceptable carriers. In preparing the compositions described herein, the active ingredient is typically mixed with or diluted by an excipient, or enclosed within such a carrier, for example, in the form of a capsule, sachet, paper, or other container. When an excipient serves as a diluent, it can be a solid, semi-solid, or liquid material, which serves as a vehicle, carrier, or medium for the active ingredient. Thus, the composition can take the form of a tablet, pill, powder, lozenge, sachet, cachet, elixir, suspension, emulsion, solution, syrup, aerosol (as a solid or in a liquid medium), ointment (e.g., containing up to 10% by weight of the active compound), soft and hard gelatin capsules, suppositories, sterile injectable solutions, and sterile packaged powders.

[0296] In preparing the formulation, active compound can be milled to suitable particle size and then mixed with other components. If active compound is substantially insoluble, such active compound can be milled to particle size of less than 200 mesh.If active compound is substantially water-soluble, such active compound can be substantially uniformly distributed in the formulation by adjusting particle size by milling (for example, about 40 mesh).

[0297] Some examples of suitable pharmaceutical excipients include lactose, dextrose, sucrose, sorbitol, mannitol, starch, gum acacia, calcium phosphate, alginic acid, tragacanth, gelatin, calcium silicate, crystalline cellulose, polyvinylpyrrolidone, cellulose, water, syrup, and methylcellulose. The formulation may additionally include lubricants (such as talc, magnesium stearate, and mineral oil), wetting agents, emulsifying and suspending agents, preservatives (such as methyl and propyl hydroxybenzoates), sweeteners, and flavoring agents. The compositions described herein may be formulated to provide quick, sustained, or delayed release of active ingredient after administration to a patient by using procedures known in the art.

[0298] The compositions can be formulated in unit dosage form, each dosage containing from about 5 to about 100 mg, more usually from about 10 to about 30 mg, of the active ingredient. The term "unit dosage form" refers to physically discrete units suitable as unitary dosages for human subjects and other mammals, each unit containing a predetermined quantity of active material calculated to produce a desired therapeutic effect in association with suitable pharmaceutical excipients.

[0299] Active compound can be effective over a wide range of doses and is generally administered in a medicamentously effective amount. However, it will be understood that the amount of compound actually administered will usually be determined by a physician according to the relevant circumstances, including the condition to be treated, the route of administration selected, the compound actually administered, the age, weight and response of individual patient, the severity of the patient's symptoms, and the like.

[0300] For the preparation of solid compositions (such as tablets), the primary active ingredient is mixed with excipients to form a solid preformulation composition containing a homogeneous mixture of a salt of Compound 1. When such preformulation compositions are referred to as homogeneous, the active ingredient is typically evenly dispersed throughout the composition such that the composition can be readily subdivided into equally effective unit dosage forms (such as tablets, pills, and capsules). This preformulation solid is then subdivided into unit dosage forms of the type described above (e.g., containing from 0.1 to about 500 mg of the active ingredient of the present disclosure).

[0301] The tablets or pills of the present disclosure may be coated or otherwise treated to provide the dosage form with the advantage of extended action. For example, the tablets or pills may include an inner dosage component and an outer dosage component, the outer dosage component being in the form of an outer coating that encases the inner dosage component. These two components may be separated by an enteric layer that serves to resist disintegration in the stomach and allows the inner component to reach the duodenum intact or to delay the release of the inner component. Such enteric layers or coatings may be made of a variety of materials, including some polymeric acids and mixtures of polymeric acids with materials such as shellac, cetyl alcohol, and cellulose acetate.

[0302] Liquid forms into which the salts of Compound 1, or compositions described herein, may be incorporated for oral or injectable administration include aqueous solutions, suitably flavored syrups, aqueous or oily suspensions, and emulsions flavored with edible oils, such as cottonseed oil, sesame oil, coconut oil, or peanut oil, as well as elixirs and similar pharmaceutical vehicles.

[0303] Compositions for inhalation or insufflation include solutions and suspensions in pharma- ceutically acceptable aqueous or organic solvents or mixtures thereof, as well as powders. Liquid or solid compositions may contain suitable pharma- ceutically acceptable excipients as described above. In some embodiments, the compositions are administered by oral or nasal respiratory routes to obtain local or systemic effects. The compositions may be nebulized by using inert gases. Nebulized solutions may be directly inhaled from the nebulizing device, or the nebulizing device may be attached to a face mask tent or intermittent positive pressure respirator. Solution, suspension, or powder compositions may be administered orally or nasally from a device that delivers the formulation in an appropriate manner.

[0304] The amount of compound or composition administered to a patient will vary depending on what is being administered, the purpose of the administration (such as prophylaxis or treatment), the condition of the patient, the mode of administration, and the like. In therapeutic applications, the compositions may be administered to a patient already suffering from a disease in an amount sufficient to cure or at least partially arrest the symptoms of such disease and its complications. The effective dose will depend on the condition being treated and the judgment of the attending physician depending on several factors, such as the severity of the disease, the age, weight, and general condition of the patient, and the like.

[0305] The compositions administered to a patient may take the form of pharmaceutical compositions described above. Such compositions may be sterilized by conventional sterilization techniques or may be sterile filtered. Aqueous solutions may be packaged for use as is or lyophilized, and lyophilized preparations are mixed with a sterile aqueous carrier prior to administration. The pH of the compound preparations is typically 3-11, more preferably 5-9, and most preferably 7-8. It will be appreciated that the use of the aforementioned pharmaceutical additives, carriers, or stabilizers will result in the formation of pharmaceutical salts.

[0306] The therapeutic dose of a salt of Compound 1 may vary depending, for example, on the particular application for which the treatment is made, the manner in which the compound is administered, the health and condition of the patient, and the judgment of the prescribing physician. The proportion or concentration of a salt of Compound 1 in a pharmaceutical composition may vary depending on several factors, including dosage, chemical characteristics (e.g., hydrophobicity), and route of administration. For example, a salt of Compound 1 may be provided in an aqueous physiological buffer containing about 0.1 to about 10% w / v of the compound for parenteral administration. Some typical dose ranges are about 1 μg / kg body weight / day to about 1 g / kg body weight / day. In some embodiments, the dose range is about 0.01 mg / kg body weight / day to about 100 mg / kg body weight / day. The dose may depend on variables such as the type and progression of the disease or disorder, the general health of the particular patient, the relative biological potency of the compound selected, the formulation of excipients, and its route of administration. Effective doses may be extrapolated from dose-response curves derived from in vitro or animal model test systems.

[0307] The salts of Compound 1 may also be formulated in combination with one or more additional active ingredients, which may include any pharmaceutical agent, such as an antiviral agent, a vaccine, an antibody, an immunostimulant, an immunosuppressant, an anti-inflammatory agent, and the like. [0332] Example A FGFR enzyme assay Inhibitor potency of compound 1 was measured in an enzyme assay that detects product formation by measuring peptide phosphorylation using FRET measurements. Compound 1 was serially diluted in DMSO and 0.5 μL volumes were transferred to wells of a 384-well plate. For FGFR3, FGFR3 enzyme (Millipore) diluted in assay buffer (50 mM HEPES, 10 mM MgCl2, 1 mM EGTA, 0.01% Tween-20, 5 mM DTT, pH 7.5) was added in 10 μL volumes to the plate and pre-incubated for 5-10 min. Appropriate controls (enzyme blank and enzyme without inhibitor) were included in the plate. The assay was initiated by adding 10 μL of a solution of biotinylated EQEDEPEGDYFEWLE peptide substrate (SEQ ID NO:1) and ATP (final concentrations of 500 nM and 140 μM, respectively) in assay buffer to the wells. The plate was incubated at 25°C for 1 h. The reaction was stopped by adding 10 μL / well of stop solution (50 mM Tris, 150 mM NaCl, 0.5 mg / mL BSA, pH 7.8; 30 mM EDTA containing Perkin Elmer Lance Reagent (Eu-antibody PY20 (3.75 nM) and APC-streptavidin (180 nM)). Plates were allowed to equilibrate for approximately 1 hour before scanning the wells on a PheraStar plate reader (BMG Labtech).

[0333] Measurements of FGFR1 and FGFR2 were performed under equivalent conditions with varying enzyme and ATP concentrations (0.02 nM and 210 μM for FGFR1, and 0.01 nM and 100 μM for FGFR2, respectively). These enzymes were purchased from Millipore or Invitrogen.

[0334] Data were analyzed using GraphPad prism3. 50Values were derived by fitting the data to an equation for a sigmoidal dose-response with variable slope: Y=Min+(Max-Min) / (1+10^((LogIC 50 -X) * Hill slope) (where X is the logarithm of the concentration and Y is the response). IC 50 Compounds with a concentration of 1 μM or less are considered to be active.

[0335] Compound 1 of the present invention was found to be an inhibitor of one or more of FGFR1, FGFR2, and FGFR3 by the above assay. IC 50 are shown in Table 12 below. The "+" symbol indicates the IC 50 The "++" symbol indicates that the IC is less than 100 nM. 50 is 100 to 500 nM. [Table 12]

[0336] Example B FGFR cell proliferation / survival assay The ability of exemplary compounds to inhibit the proliferation of cells that depend on FGFR signaling for survival was measured using a viability assay. A recombinant cell line overexpressing human FGFR3 was created by stably transfecting a plasmid encoding full-length human FGFR3 into mouse pro-B Ba / F3 cells (obtained from Deutsche Sammlung von Mikroorganismen und Zellkulturen). Cells that were puromycin resistant and proliferated in the presence of heparin and FGF1 were continuously selected. A single cell clone was isolated and characterized for functional expression of FGFR3. The Ba / F3-FGFR3 clone was used in a cell proliferation assay and compounds were screened for their ability to inhibit cell proliferation / survival. The Ba / F3-FGFR3 cells were seeded at 3500 cells / well in 96-well black cell culture plates containing RPMI1640 medium with 2% FBS, 20 μg / mL heparin, and 5 ng / mL FGF1. Cells were treated with 10 μL of serially diluted 10× compound (diluted in serum-free medium from 5 mM DSMO) in a final volume of 100 μL / well. After 72 hours of incubation, 100 μL of Cell Titer Glo® reagent (Promega Corporation), which measures cellular ATP levels, is added to each well. Luminescence readings are taken on a plate reader after 20 minutes of incubation with shaking. Luminescence readings are converted to percent inhibition relative to DMSO-treated control wells and IC values are calculated using GraphPad Prism software by fitting the data to an equation for a sigmoidal dose-response with variable slope. 50 The value is calculated. 50Compounds with IC values of 10 μM or less are considered to have activity. Cell lines representing a variety of tumor types are used in similar proliferation assays, including KMS-11 (multiple myeloma, FGFR3 translocation), RT112 (bladder cancer, FGFR3 overexpression), KatoIII (gastric cancer, FGFR2 gene amplification), and H-1581 (lung, FGFR1 gene amplification). In some experiments, MTS reagent (Cell Titer 96® AQueous One Solution Reagent (Promega Corporation)) is added in place of Cell Titer Glo to a final concentration of 333 μg / mL and read at 490 / 650 nm on a plate reader. IC 50 Compounds with a concentration of 5 μM or less are considered to have activity.

[0337] Experiment C Cell-based FGFR phosphorylation assay The inhibitory effect of compounds on FGFR phosphorylation in relevant cell lines (Ba / F3-FGFR3, KMS-11, RT112, KatoIII, H-1581, and HUVEC) can be assessed using an immunoassay specific for FGFR phosphorylation. After starvation in low serum (0.5%) medium without FGF1 for 4-18 hours depending on the cell line, cells are treated with various concentrations of individual inhibitors for 1-4 hours. For some cell lines (e.g. Ba / F3-FGFR3 and KMS-11), cells are stimulated with heparin (20 μg / mL) and FGF1 (10 ng / mL) for 10 minutes. Whole cell protein extracts are prepared by incubation at 4° C. in lysis buffer containing protease and phosphatase inhibitors (50 mM HEPES (pH 7.5), 150 mM NaCl, 1.5 mM MgCl2, 10% glycerol, 1% Triton X-100, 1 mM sodium orthovanadate, 1 mM sodium fluoride, aprotinin (2 μg / mL), leupeptin (2 μg / mL), pepstatin A (2 μg / mL), and phenylmethylsulfonyl fluoride (1 mM). Protein extracts are clarified to remove cell debris by centrifugation at 14,000×g for 10 min and quantified using BCA (bicinchoninic acid) microplate assay reagent (Thermo Scientific).

[0338] Phosphorylation of FGFR receptors in protein extracts was determined using immunoassays, including Western blot, enzyme-linked immunosorbent assay (ELISA), or bead-based immunoassay (Luminex). For detection of phosphorylated FGFR2, a commercially available ELISA kit (DuoSet IC Human Phospho-FGF R2α ELISA Assay (R&D Systems, Minneapolis, MN)) can be used. For this assay, KatoIII cells in Iscove's medium supplemented with 0.2% FBS are plated (50,000 cells / well / 100 μL) in 96-well flat-bottom tissue culture treated plates (Corning, Corning, NY) and incubated for 4 hours at 37°C in a 5% CO2 atmosphere in the presence or absence of a range of test compounds. The assay is stopped by adding 200 μL of chilled PBS and centrifugation. Washed cells were lysed in cell lysis buffer (Cell Signaling, #9803) containing protease inhibitors (Calbiochem, #535140) and PMSF (Sigma, #P7626) for 30 min on wet ice. Cell lysates were frozen at -80°C until aliquots were tested using the DuoSet IC Human Phospho-FGF R2α ELISA Assay Kit. Data were analyzed using GraphPad prism3. IC 50 Values were derived by fitting the data to an equation for a sigmoidal dose-response with variable slope.

[0339] For the detection of phosphorylated FGFR3, a bead-based immunoassay was developed. Anti-human FGFR3 mouse mAb (R&D Systems, Cat. No. MAB7661) was conjugated to Luminex MAGplex microspheres (bead region 20) and used as the capture antibody. RT-112 cells were seeded in multi-well tissue culture plates and cultured to 70% confluence. Cells were washed with PBS and starved for 18 hours in RPMI containing 0.5% FBS. Cells were treated with 10 μL of serially diluted 10× compounds for 1 hour at 37°C in a 5% CO2 atmosphere, and then stimulated with 10 ng / mL human FGF1 and 20 μg / mL heparin for 10 minutes. Cells were washed with cold PBS, lysed with Cell Extraction Buffer (Invitrogen), and centrifuged. Clarified supernatants were frozen at -80°C until analysis.

[0340] For this assay, cell lysates are diluted 1:10 in assay diluent and incubated with capture antibody-conjugated beads in a 96-well filter plate for 2 hours at room temperature on a plate shaker. The plate is washed 3 times using a vacuum manifold and incubated with anti-phospho-FGF R1-4 (Y653 / Y654) rabbit polyclonal antibody (R&D Systems Cat. No. AF3285) for 1 hour at room temperature with shaking. The plate is washed 3 times. Diluted reporter antibody (goat anti-rabbit-RPE conjugated antibody (Invitrogen Cat. No. LHB0002)) is added and incubated for 30 minutes with shaking. The plate is washed 3 times. The beads are suspended in wash buffer for 5 minutes at room temperature with shaking before being read on a Luminex 200 instrument (gate settings 7500-13500) set to count 50 events per sample. Data are presented as mean fluorescence intensity (MFI). Percent inhibition was determined by dividing the MFI from compound-treated samples by the MFI value from the DMSO control, and IC was calculated using GraphPad Prism software. 50 The value is calculated. 50Compounds with a concentration of 1 μM or less are considered to be active.

The medium is aspirated and cells are treated with 30 μL of 1× concentration of compounds serially diluted (diluted from 5 mM DSMO solution in serum-free medium) to a final volume of 30 μL / well and incubated at room temperature for 45 min. Cells are stimulated by adding 10 μL of a solution containing heparin (100 μg / mL) and FGF1 (50 ng / mL) to each well and incubated at room temperature for 10 min.
