Description

Technical Field

[0001] The present invention relates to a topical skin preparation used as a cosmetic, quasi-drug, or pharmaceutical product.

Background Art

[0002] Hyaluronic acid is a mucopolysaccharide composed of glucuronic acid and N-acetylglucosamine, and is present in large amounts in the dermis as a conjugate substance filling between cells and fibers in vivo, and is also present in the epidermis. Since hyaluronic acid has high water retention and viscoelasticity, it is widely used in topical skin preparations as a moisturizing agent and the like.

[0003] When hyaluronic acid is formulated into a topical skin preparation, a unique feeling of use such as cushioning or thickness can be obtained due to the viscosity and viscoelasticity peculiar to hyaluronic acid. Furthermore, by applying such a preparation to the skin, a moist and cushioned or thick feeling of the skin texture that sticks to the hand can be obtained. This feeling of use is such as cushioning or thickness, and is a unique feeling of use preferred by consumers. On the other hand, since hyaluronic acid is a component with strong drawability, it gives a sticky feeling when formulated. In order to improve the cushioning of the skin during and after application, it is required to formulate hyaluronic acid at a high concentration, but the stickier it becomes as the amount of hyaluronic acid formulated increases. Therefore, it is difficult to achieve both cushioning and a smooth feeling of use.

[0004] Patent Document 1 teaches that acetyl hyaluronic acid has an excellent skin softening effect while improving drawbacks such as the high drawability of hyaluronic acid.

Summary of the Invention

Problems to be Solved by the Invention

[0006] The present invention aims to provide a topical skin preparation that contains hyaluronic acid or its salt, has high cushioning properties, and suppresses stickiness.

Means for Solving the Problems

[0007] In order to solve the above problems, the present inventor has conducted extensive research and found that by combining a high-molecular-weight hyaluronic acid with a weight-average molecular weight of 50,000 or more or its salt and a low-molecular-weight hyaluronic acid with a weight-average molecular weight of less than 50,000 or its salt as hyaluronic acids in a non-emulsified topical skin preparation that contains water, has an oil content of 0 to 10% by mass, and is substantially free of parabens, and further incorporating a hyaluronic acid derivative, it is possible to suppress the amount of hyaluronic acids that cause stickiness while enhancing the thickness or cushioning feeling felt during application and the cushioned texture of the skin after application. As a result, it has been found that high cushioning properties and a smooth usability can be achieved simultaneously. Furthermore, it has been found that by incorporating an alkanediol having 5 to 10 carbon atoms into this topical skin preparation, a decrease in viscosity over time can be suppressed. This suppresses a decrease in the cushioning properties or the thick feeling of the preparation during use, as well as a decrease in the thickness feeling of the preparation itself.

[0008] The present invention has been completed based on the above findings and provides the following topical skin preparation. 〔1〕 A non-emulsified topical skin preparation containing (A) a high-molecular-weight hyaluronic acid with a weight-average molecular weight of 50,000 or more or its salt, (B) a low-molecular-weight hyaluronic acid with a weight-average molecular weight of less than 50,000 or its salt, (C) a hyaluronic acid derivative, and (D) water, having (E) an oil content of 0 to 10% by mass, and (F) being substantially free of parabens. 〔2〕 The topical skin preparation according to 〔1〕, wherein the content of (A) a high-molecular-weight hyaluronic acid with a weight-average molecular weight of 50,000 or more or its salt is 0.001 to 1% by mass based on the total amount of the topical skin preparation. 〔3〕(B) The content of low-molecular-weight hyaluronic acid or its salt with a weight-average molecular weight of less than 50,000 is 0.0001 to 1% by mass based on the total amount of the external preparation for skin, the external preparation for skin according to 〔1〕 or 〔2〕. 〔4〕(C) The content of the hyaluronic acid derivative is 0.00001 to 0.3% by mass based on the total amount of the external preparation for skin, the external preparation for skin according to any one of 〔1〕 to 〔3〕. 〔5〕(C) With respect to 1 part by mass of the hyaluronic acid derivative, the total of (A) high-molecular-weight hyaluronic acid or its salt with a weight-average molecular weight of 50,000 or more and (B) low-molecular-weight hyaluronic acid or its salt with a weight-average molecular weight of less than 50,000 is 0.005 to 200,000 parts by mass, the external preparation for skin according to any one of 〔1〕 to 〔4〕. 〔6〕Furthermore, the external preparation for skin according to any one of 〔1〕 to 〔5〕, which contains an alkanediol having 5 to 10 carbon atoms. 〔7〕The content of the alkanediol having 5 to 10 carbon atoms is 0.0001 to 20% by mass based on the total amount of the external preparation for skin, the external preparation for skin according to 〔6〕. 〔8〕The external preparation for skin according to any one of 〔1〕 to 〔7〕, wherein the L value in the Lab color system is 40 or more. 〔9〕A method for improving the cushioning property or the mochi-mochi feeling in the texture of the preparation (during application) and / or after application while suppressing the stickiness of a non-emulsified external preparation for skin, which contains (A) high-molecular-weight hyaluronic acid or its salt with a weight-average molecular weight of 50,000 or more, (B) low-molecular-weight hyaluronic acid or its salt with a weight-average molecular weight of less than 50,000, and (D) water, has an oil content of 0 to 10% by mass, and is substantially free of parabens, and contains (C) a hyaluronic acid derivative (however, the oil content of 0 to 10% by mass in (E) is the content after adding (C) the hyaluronic acid derivative).

Advantages of the Invention

[0009] The external preparation for skin of the present invention contains water, has an oil content of 0 to 10% by mass, is a non-emulsified external preparation for skin that is substantially free of parabens, and is formulated by combining three specific types of hyaluronic acids. As a result, the cushioning property or the feeling of thickness possessed by hyaluronic acid or its salt is effectively enhanced. Therefore, since the blending amount of hyaluronic acids that causes stickiness can be suppressed, it has both a high cushioning property and a dry and comfortable feeling of use. Thus, the external preparation for skin of the present invention, in addition to the feeling of use of the preparation such as the cushioning property or the feeling of thickness during application, gives a unique firm and cushioned or thick feeling to the skin after application, like being sucked by the hand, and since stickiness is suppressed, it has a high commercial value.

In the present invention, hyaluronic acid having a weight-average molecular weight of 50,000 or more or a salt thereof is referred to as "high-molecular-weight hyaluronic acid or a salt thereof", and hyaluronic acid having a weight-average molecular weight of less than 50,000 or a salt thereof is referred to as "low-molecular-weight hyaluronic acid or a salt thereof".

[0011] (A) High molecular weight hyaluronic acid or its salt Hyaluronic acid is a kind of acidic mucopolysaccharide and is a polysaccharide containing a disaccharide of glucuronic acid and N-acetylglucosamine as a structural unit. Hyaluronic acid can be extracted and recovered from animal tissues such as chicken combs, shark skins, umbilical cords, eyeballs, skin, and cartilage, hyaluronic acid-producing microorganisms such as Streptococcus microorganisms, animal cells, and plant cell cultures. Also, commercial products can be purchased. As the high molecular weight hyaluronic acid having a weight average molecular weight of 50,000 or more, such natural hyaluronic acid can be used. The weight average molecular weight of natural hyaluronic acid is about 500,000 to 7,000,000.

[0012] In addition, degraded hyaluronic acid obtained by decomposing natural hyaluronic acid can also be used as long as its weight average molecular weight is 50,000 or more. Degraded hyaluronic acid can be prepared, for example, by hydrolysis in the presence of an acid or an alkali, enzymatic treatment such as hyaluronidase, physical cleavage such as ultrasonic waves or shearing, fermentation by microorganisms, etc. Also, commercially available products can be used.

The L value in the Lab color system of cosmetics represents the transparency value when the transparency of purified water is set to 100 and the case where there is no light transmission at all is set to 0. That is, the closer the L value is to 100, the higher the transparency. In the present invention, the L value is a value measured using a spectrophotometer. Specifically, it is a value measured by putting the test solution in a glass cell (CM-A99, thickness 20 mm) and using a spectrophotometer CM-5 (manufactured by Konica Minolta Inc.). Examples

[0093] Hereinafter, the present invention will be described in more detail with reference to examples, but the present invention is not limited to these examples. (1) Effect of acetyl hyaluronic acid Skin external preparations having the compositions shown in Tables 1 and 2 were prepared according to a conventional method. These skin external preparations are lotion agents that do not contain emulsified particles. Also, the visual appearance of these skin external preparations was transparent. The viscosities of the external preparations were measured, and the viscosity increase rate was calculated according to formula (1). In formula (1), the comparative examples corresponding to Examples 1 to 4 are Comparative Examples 1 to 4, respectively. Viscosity increase rate = [(viscosity of the example) / (viscosity of the corresponding comparative example)] × 100 ····· (1)

[0094] (Viscosity measurement method) Measurement was carried out using VISCOMETER TV-10M (manufactured by Toyo Sangyo Co., Ltd.) under the conditions of a temperature of 25 °C, rotor M-2, a speed of 60 rpm, and for 20 seconds.

[0095] The results are shown in Tables 1 and 2. The unit of the component content in Tables 1 and 2 is mass%. As shown in Table 1, the viscosities of Reference Examples 1 to 3, which contain acetylated hyaluronic acid but do not contain hyaluronic acid or its salt, were zero, similar to Reference Example 4, which does not contain both acetylated hyaluronic acid and hyaluronic acid or its salt. It can be seen that acetylated hyaluronic acid itself has no viscosity increasing effect.

[0096] Also, as shown in Table 2, by using high molecular weight sodium hyaluronate with a weight average molecular weight of 50,000 or more and low molecular weight sodium hyaluronate with a weight average molecular weight of less than 50,000 as hyaluronic acid or its salt, and adding a trace amount of acetylated hyaluronic acid thereto, the viscosity increased significantly. It can be seen that the viscosity increased by combining high molecular weight sodium hyaluronate with a weight average molecular weight of 50,000 or more, low molecular weight sodium hyaluronate with a weight average molecular weight of less than 50,000, and acetylated hyaluronic acid. Since viscosity is one of the indicators showing the cushioning property of the composition, it was shown that by including hyaluronic acid or its salt with a weight average molecular weight of 50,000 or more and those with a weight average molecular weight of less than 50,000, the cushioning property becomes very good by blending acetylated hyaluronic acid.

[0099] (2) Effect of alkanediol External skin preparations shown in Tables 3 to 5 were prepared according to a conventional method and contained 50 mL in a glass container with a capacity of 50 mL. These external skin preparations are lotion agents that do not contain emulsified particles. Also, the visual appearance of these external skin preparations was transparent. Each container was stored at 50 °C for 1 week, the viscosity was measured before and after storage, and the viscosity reduction rate was calculated according to formula (2). Viscosity reduction rate = [(viscosity after storage - viscosity before storage) / viscosity before storage] × 100 ····· (2) Furthermore, the improvement rate of viscosity reduction by blending alkanediol was calculated according to formula (3). In formula (3), the corresponding examples are: for Example 6, it is Example 5; for Example 8, it is Example 7; for Example 10, it is Example 9; for Example 12, it is Example 11; for Example 14, it is Example 13; for Example 16, it is Example 15; for Example 18, it is Example 17; for Example 20, it is Example 19; for Example 22, it is Example 21; for Example 24, it is Example 23; for Example 26, it is Example 25; for Example 28, it is Example 27. Viscosity reduction improvement rate = [(viscosity reduction rate of the example - viscosity reduction rate of the corresponding example) / viscosity reduction rate of the corresponding example] × 100 ····· (3)

[0102]

Table 5

[0103] The results are shown in Tables 3 to 5. The unit of the component content in Tables 3 to 5 is mass%. The viscosity reduction rate under severe conditions at 50°C for one week was significantly improved by the addition of alkanediol. On the other hand, even when dipropylene glycol, polyethylene glycol 400, which are the same diol compounds, or concentrated glycerin, which is the same polyol compound, were added, the viscosity reduction rate was not improved at all.

[0104] (3) Evaluation of usability Each topical skin preparation was applied to the arms of 5 subjects who understood the evaluation criteria, and the touch of the applied area was evaluated on a scale of 1 to 5 (integer values). That is, regarding the stickiness during application, those who felt it strongly were given 1 point, and those who did not feel it were given 5 points, and the value corresponding to the touch felt by the subject was shown. Similarly, regarding the stickiness of the back of the skin 1 minute after application, those who felt it strongly were given 1 point, and those who did not feel it were given 5 points. Also, regarding the cushioning property of the back of the skin 1 minute after application, those who did not feel it were given 1 point, and those who felt it strongly were given 5 points. Next, the average value of the scores for each example was calculated and used as the evaluation value of the topical skin preparation.

[0105]

Table 6

[0106] The results are shown in Table 6. From the comparison between Example 1 and Comparative Example 1, Example 2 and Comparative Example 2, and Example 4 and Comparative Example 4, it can be seen that for a topical skin preparation containing high molecular weight hyaluronic acid or its salt with a weight average molecular weight of 50,000 or more and low molecular weight hyaluronic acid or its salt with a weight average molecular weight of less than 50,000, by blending acetylated hyaluronic acid, the stickiness does not change or rather improves, but the cushioning property of the skin after application increases significantly. In addition, since Reference Example 2 does not contain hyaluronic acid or its salt, the stickiness was low but the cushioning property was very low. It was confirmed that acetylated hyaluronic acid itself has no cushioning property. Also, as shown in Comparative Example 5, when the blending amount of the high molecular weight hyaluronate was increased instead of blending acetylated hyaluronic acid, good cushioning properties were obtained, but the stickiness became extremely strong. It was confirmed that the cushioning properties can be enhanced by blending acetylated hyaluronic acid, and accordingly, the blending amount of hyaluronic acid or its salt can be reduced to suppress stickiness.

[0107] The weight average molecular weight of the sodium acetylated hyaluronate used in the examples is 100,000.

[0108] Formulation examples of the external preparation for skin of the present invention are shown below. The unit of the component contents in Tables 7 and 8 is mass%, and the numerical values in parentheses are the weight average molecular weights.
