AI Technical Summary
Existing small molecule anti-tumor drugs such as perillyl alcohol (PA) and limonene are limited in clinical applications due to their low bioavailability and poor metabolic and excretion properties, resulting in short half-life in the body, harmful to normal cells, and side effects such as nausea, diarrhea and abdominal pain.
A polycyclic terpene precursor (prodrugs) that combine with perelol, lincinol and its derivatives with other drugs such as cholecalciferol, retinoic acid, N-methyl perillyl acid, etc. were developed. These precursors were prepared by chemical synthesis to improve their bioavailability and metabolic and excretion properties, and to reduce damage to normal cells through targeted delivery.
By combining Perelol, Lindol, etc. with other drugs into polycyclic terpene precursors, the bioavailability and metabolic and excretion properties of the drug are significantly improved, the damage to normal cells is reduced, and the targeting effect on tumor cells is enhanced, thereby improving the therapeutic effect and reducing side effects.

Abstract
Description
[Technical field]
[0001] This application claims priority to U.S. Provisional Patent Application No. 63 / 329,272, filed April 8, 2022. The contents of the aforementioned application are incorporated herein by reference. FIELD OF THEINVENTION
[0002] The present invention relates generally to small molecule therapeutics, and more specifically to prodrug compounds with high bioavailability and anti-neoplastic activity. [Background technology]
[0003] Cancer can be defined as a group of diseases involving abnormal cell growth that invades or spreads to other parts of the body. Despite technological advances, cancer continues to be a major cause of death and immense suffering. Cancer is associated with several factors including smoking, obesity, poor diet, lack of physical activity and excessive consumption of alcohol. Other factors include exposure to certain infections, ionizing radiation and environmental pollutants. Certain cancers are associated with infections such as Helicobacter pylori, Hepatitis B, Hepatitis C, Human Papillomavirus infection, Epstein-Barr Virus and Human Immunodeficiency Virus (HIV). Cancer is the second leading cause of death in the United States.
[0004] Cancer patients often have limited treatment options. Traditional cancer treatments are directed at removing cancerous tissue and preventing its spread. Despite advances in research, these treatments have remained relatively unchanged in recent decades. Treatments may include a combination of surgery, radiation therapy, chemotherapy and targeted therapy, hormonal therapy, and palliative care. Treatments are usually pursued based on the type, location, and grade of cancer, as well as the patient's health and preferences. However, these options are limited. They may be ineffective, especially if the cancer has metastasized.
[0005] Cancer cells divide more rapidly than most normal cells, so they may be more sensitive to chemotherapy. However, chemotherapy also attacks other cells in the body, particularly rapidly dividing cells such as blood cells and cells lining the mouth, stomach and intestines. Radiation therapy and chemotherapy usually cause multiple side effects, especially epithelial cell damage. Therefore, there is a need for improved drugs and methods for the treatment of cancer that are more targeted and have fewer harmful side effects.
[0006] Recent efforts have been directed towards small molecule drugs. Perillyl alcohol (PA) is a monoterpene that has been used orally for the treatment of systemic cancer. Perillyl alcohol is a metabolite of limonene, which is itself formed from geranyl pyrophosphate in the mevalonate pathway. Conversion of limonene to perillyl alcohol is achieved via hydroxylation by enzymes belonging to the cytochrome P450 protein superfamily. Perillyl alcohol can be further metabolized to perillaldehyde (perillyl aldehyde) and perillic acid.
[0007] Published in vitro studies have demonstrated that perillyl alcohol (PA) and limonene can both induce apoptosis in cancer cells. Furthermore, under certain conditions, these compounds can restore cancer cell identity without adversely affecting healthy cells in cell culture populations. However, PA and limonene are not used in clinical settings and patient care due to their poor bioavailability and ADME (absorption, distribution, metabolism and excretion) issues. When taken orally, the compounds have negative effects on mucosal tissues and generally have short half-lives. Common side effects include nausea, diarrhea and abdominal pain.
[0008] Due to the shortcomings of the use of PA as described above, there is a need for improved formulations and methods of administering compounds. Such formulations and methods must improve their bioavailability and ADME. The embodiments of the present invention meet these needs and provide related additional advantages as outlined below. [0047] The term "tumor" refers to a disease caused or resulting from inappropriately high levels of cell division, inappropriately low levels of apoptosis, or both. For example, cancer is an example of a tumor. Examples of cancer include, but are not limited to, leukemia (e.g., acute leukemia, acute lymphocytic leukemia, acute myeloid leukemia, acute myeloblastic leukemia, acute promyelocytic leukemia, acute myelomonocytic leukemia, acute monocytic leukemia, acute erythroleukemia, chronic leukemia, chronic myelogenous leukemia, chronic lymphocytic leukemia), polycythemia vera, lymphoma (e.g., Hodgkin's disease and non-Hodgkin's disease), Waldenstrom's macroglobulinemia, heavy chain disease, and solid tumors such as sarcomas and carcinomas (e.g., fibrosarcoma, myxosarcoma, liposarcoma, chondrosarcoma, osteogenic sarcoma, chordoma, angiosarcoma, endothelial sarcoma, lymphangiosarcoma, lymphoma ... These include intraductal sarcoma, synovium, mesothelioma, Ewing's tumor, leiomyosarcoma, rhabdomyosarcoma, colon cancer, pancreatic cancer, breast cancer, ovarian cancer, prostate cancer, squamous cell carcinoma, basal cell carcinoma, adenocarcinoma, sweat gland carcinoma, sebaceous gland carcinoma, papillary carcinoma, papillary adenocarcinoma, cystadenocarcinoma, medullary carcinoma, bronchogenic carcinoma, renal cell carcinoma, hepatoma, cholangiocarcinoma, choriocarcinoma, seminoma, embryonal carcinoma, Wilms' tumor, cervical cancer, uterine cancer, testicular cancer, lung cancer, small cell lung carcinoma, bladder carcinoma, epithelial carcinoma, glioma, astrocytoma, medulloblastoma, craniopharyngioma, ependymoma, pinealoma, hemangioblastoma, acoustic neuroma, oligodendroglioma, schwannoma, meningioma, melanoma, neuroblastoma, and retinoblastoma. Lymphoproliferative disorders are also considered to be proliferative disorders.
[0048] The term "glioma" refers to a type of tumor that arises beginning in the glial cells of the brain or spinal cord. Gliomas comprise approximately 30% of all brain and central nervous system tumors and 80% of all malignant brain tumors.
[0049] The term "glioblastoma" refers to an aggressive type of cancer that can occur in the brain or spinal cord. Glioblastomas form from cells called astrocytes, which support nerve cells. Glioblastomas can occur at any age, but tend to occur more often in older people. Initially, signs and symptoms of glioblastoma are nonspecific. They may include headaches, personality changes, nausea, and symptoms similar to those of a stroke. Symptoms often worsen rapidly and may progress to loss of consciousness. The cause of most cases of glioblastoma is unknown. Uncommon risk factors include genetic disorders such as neurofibromatosis and Li-Fraumeni syndrome, as well as previous radiation therapy. Glioblastomas represent 15% of all brain tumors. They can begin in normal brain cells or develop from pre-existing low-grade astrocytomas. Diagnosis is typically made by a combination of CT scans, MRI scans, and tissue biopsies. Treatment usually involves surgery, followed by chemotherapy and radiation therapy. The drug temozolomide is often used as part of chemotherapy. High-dose steroids may also be used to help reduce swelling and decrease symptoms. Surgical removal of the tumor is associated with increased survival, but only for a few months.
[0050] The term "astrocytoma" refers to a type of brain tumor. They begin in a specific type of glial cell in the brain, a star-shaped brain cell called an astrocyte. This type of tumor does not usually spread outside the brain and spinal cord, and does not usually affect other organs. Astrocytomas are the most common gliomas and can occur in most parts of the brain, and occasionally the spinal cord. Within astrocytomas, two broad classes are generally recognized, which share various characteristics, including (a) narrow areas of infiltration that are often clearly delineated on diagnostic images (predominantly non-invasive tumors; e.g., pilocytic astrocytoma, subependymal giant cell astrocytoma, pleomorphic xanthoastrocytoma), but (b) diffuse areas of infiltration that share various characteristics, including being able to arise anywhere in the central nervous system, but are more prevalent in the brain hemispheres (e.g., high-grade astrocytoma, anaplastic astrocytoma, glioblastoma); they usually arise in adults and have an inherent tendency to progress to a more aggressive form. People can develop astrocytomas at any age. Low-grade types are more often found in children or young adults, while high-grade types are more common in adults. Basilar astrocytomas are more common in young people and account for about 75% of neuroepithelial tumors.
[0051] The term "cell identity" can be defined by the expression or activity of certain genes in its DNA and the resulting production of certain proteins. The increase in biological complexity is made possible by the ability of cells to differentiate and achieve a distinct "identity" in systems that reflect morphological or functional differences from precursor cells. This identity is defined primarily in terms of cell type and cell state. The precise definition of these terms remains elusive: cell types have historically been described by observing reproducible functional differences in vivo or in vitro (often together with the expression of a set of marker genes), whereas cell state refers to dynamic responsive changes that modify the phenotype and function of the cell, but not significant changes that result in the acquisition of a new cell type.
[0052] The term "blood-brain barrier" or "BBB" refers to a highly selective semi-permeable boundary of endothelial cells that prevents solutes in circulating blood from crossing nonselectively into the extracellular fluid of the central nervous system where neurons reside. The blood-brain barrier is formed by endothelial cells of the capillary walls, the foot processes of astrocytes that encase the capillaries, and pericytes embedded in the capillary basement membrane. This system allows the passage of some small molecules by passive diffusion, as well as the selective active transport of various nutrients, ions, organic anions, and macromolecules such as glucose and amino acids that are crucial for neuronal function.
[0053] The term "perillic alcohol", "PA" or "POH" refers to a naturally occurring monoterpene. It is also identified as [4-(prop-1-en-2-yl)cyclohex-1-en-1-yl]methanol) (i.e., IUPA name). PA and its precursor limonene are monocyclic terpenes of natural origin that can be derived from the mevalonate pathway in plants. PA has also shown some antitumor activity in laboratory and animal studies. Human clinical trials were inconclusive due to adverse side effects. PA can be found in the essential oils of various plants such as lavender, lemongrass, sage, and peppermint. It has many manufacturing, household, and medical applications.
[0054] The term "perillic acid" refers to the metabolite / conjugate acid of perillic alcohol. Perillic acid is an alpha, beta unsaturated monocarboxylic acid and a cyclohexene carboxylic acid. It has a role as an antitumor drug, human metabolite and mouse metabolite.
[0055] The term "limonene" refers to a colorless liquid aliphatic hydrocarbon classified as a cyclic monoterpene. It is the major component in citrus peel oil. It is also identified as 1-methyl-4-(prop-1-en-2-yl)cyclohex-1-ene (i.e., IUPA name).
[0056] The term "cholecalciferol" or "vitamin D3" refers to an analog of vitamin D. It is naturally synthesized in the skin, where it functions as a prohormone and is converted to calcitriol. In the liver, it is converted to calcifediol (25-hydroxyvitamin D), which is then converted to calcitriol (1,25-dihydroxyvitamin D) in the kidneys. One of its actions is to increase calcium uptake by the intestine.
[0057] The term "prodrug" refers to a derivative of a known direct acting drug, which has enhanced delivery properties and therapeutic value compared to the drug, and which is converted to the active drug by enzymatic or chemical processes.
[0058] The term "terpene" refers to a compound of the formula (C5H8) n Terpenes refers to a class of compounds having the formula: 1. Terpenes include over 30,000 compounds and are primarily produced by plants, especially conifers. Terpenes can be further classified by the number of carbon atoms (e.g., monoterpenes (C 10 ), sesquiterpenes (C 15 ), diterpenes (C 20 ), etc. Terpenes comprise a diverse group of naturally occurring compounds. They are mostly found in plants and form the major constituents of plant-derived essential oils. Terpenes have a wide range of medical uses, among which antiplasmodial activity is notable as its mechanism of action is similar to commonly used antimalarial drugs (i.e., chloroquine). Monoterpenes have been particularly widely studied for their antiviral properties. With the modern era seeing an increase in the incidence of cancer and diabetes, terpenes also have the potential to serve as anticancer and antidiabetic agents.

All-trans retinoic acid is required in chordates, which includes all higher animals from fish to humans. All-trans retinoic acid (ATRA) is the predominant retinoic acid present, while isomers such as 13-cis- and 9-cis-retinoic acid are also present at much lower levels in mammals.
[0061] The term "mevalonate pathway" refers to a metabolic pathway that plays an important role in multiple cellular processes by synthesizing sterol isoprenoids, such as cholesterol, and non-sterol isoprenoids, such as dolichol, heme-A, isopentenyl tRNA, and ubiquinone.
[0062] The term "linker" refers to a functional group that covalently bonds two or more moieties in a compound or substance. For example, a linker can serve to covalently bond a prodrug moiety to an active agent.
[0063] The term "alkyl" refers to a straight or branched chain saturated aliphatic radical having the indicated number of carbon atoms. Alkyl can be any number of carbons, e.g., C 1-2 , C 1-3 , C 1-4 , C 1-5 , C 1-6 , C 1-7 , C 1-8 , C 1-9 , C 1-10 , C2-3 , C 2-4 , C 2-5 , C 2-6 , C 3-4 , C 3-5 , C 3-6 , C 4-5 , C 4-6 and C 5-6 For example, C 1-6 Alkyl includes, but is not limited to, methyl, ethyl, propyl, isopropyl, butyl, isobutyl, sec-butyl, t-butyl, pentyl, isopentyl, hexyl. "Substituted alkyl" groups can be substituted with one or more groups selected from halo, hydroxy, amino, oxo (=O), alkylamino, amido, acyl, nitro, cyano, and alkoxy. The term "alkylene" refers to a divalent alkyl radical.
[0064] The term "heteroalkyl" refers to an alkyl group, as described herein, in which one or more carbon atoms are optionally and independently replaced with a heteroatom selected from N, O, and S. The term "heteroalkylene" refers to a divalent heteroalkyl radical.

[0072] The term "passive transport" refers to a type of membrane transport that does not require energy to move substances across the cell membrane. The four main types of passive transport are simple diffusion, facilitated diffusion, filtration, and / or osmosis. Passive transport follows Fick's first and second laws of thermodynamics.
[0073] The term "active transport" refers to the movement of molecules across a cell membrane from an area of low concentration to an area of high concentration (i.e., movement against a concentration gradient). Unlike passive transport, which uses the kinetic energy and natural entropy of the molecule moving along the gradient, active transport uses cellular energy to move molecules against a gradient, polar repulsion, or other resistance. Active transport is usually associated with the accumulation of high concentrations of molecules needed by cells, such as ions, glucose, and amino acids. Specialized transmembrane proteins recognize and move substances across the membrane that would not otherwise occur because the phospholipid bilayer of the membrane is impermeable to the substance being moved, or because the substance moves against the direction of its concentration gradient. There are two types of active transport: primary active transport and secondary active transport. In primary active transport, the proteins involved are pumps that use chemical energy, usually in the form of ATP. Secondary active transport uses potential energy, usually obtained by exploiting an electrochemical gradient.
[0074] The term "administration" refers to the introduction of a predetermined amount of a substance into a patient by a specific suitable method. The compositions disclosed herein can be administered via any of the common routes, such as, but not limited to, intraperitoneal, intravenous, intramuscular, subcutaneous, intradermal, oral, topical, intranasal, pulmonary, or rectal administration, as long as they can reach the target tissue. However, since peptides are digested when administered orally, the active ingredients of compositions for oral administration should be coated or formulated to protect against degradation in the stomach.
[0075] Where applicable, the term "about" or "generally" as used herein and in the appended claims means a tolerance of ±20% unless otherwise indicated. Where applicable, the term "substantially" as used herein and in the appended claims means a tolerance of ±10% unless otherwise indicated. It should be understood that not all uses of the above terms are necessarily quantifiable such that a reference range can be applied.
[0076] The terms "medicament", "active agent" or "active ingredient" refer to a substance, compound, or molecule that is biologically active or that induces a biological or physiological effect in a subject to which it is administered. In other words, an "active agent" or "active ingredient" refers to the component or components of a composition to which all or part of the effect of the composition is due. The active agent may be the primary active agent, or in other words the component of the composition to which all or part of the effect of the composition is due. The active agent may be a secondary agent, or in other words the component of the composition to which additional parts of the composition and / or other effects are due.
[0077] The term "bioavailability" refers to the proportion of the administered dose of unchanged drug that reaches the systemic circulation. For example, when a drug is administered intravenously, its bioavailability is 100%. However, when a drug is administered via other routes (e.g., orally), its bioavailability is usually reduced due to incomplete absorption and the first-pass effect. Bioavailability is one of the essential tools in pharmacokinetics, since it must be taken into account when calculating dosage for non-intravenous administration routes. In one embodiment, the bioavailability of an agent is increased by converting it into a prodrug.
[0078] The term "pharmaceutical composition" is intended to include the combination of an active agent (i.e., a prodrug therapeutic agent) with an inert or active carrier in a sterile composition suitable for in vitro, in vivo or ex vivo diagnostic or therapeutic use. In one aspect, the pharmaceutical composition is substantially free of endotoxin or is non-toxic to a recipient at the dosage or concentration employed.
[0079] The term "pharmaceutically acceptable salt" refers to an acid or base salt generally regarded in the art as suitable for use in contact with the tissues of humans or animals without undue toxicity, irritation, allergic response, or other problems or complications. Such salts include inorganic or organic acid salts of basic residues such as amines, as well as alkali or organic salts of acidic residues such as carboxylic acids. Specific pharmaceutical salts include, but are not limited to, alkanoic acids such as hydrochloric acid, phosphoric acid, hydrobromic acid, malic acid, glycolic acid, fumaric acid, sulfuric acid, sulfamic acid, sulfanilic acid, formic acid, toluenesulfonic acid, methanesulfonic acid, benzenesulfonic acid, ethanedisulfonic acid, 2-hydroxyethylsulfonic acid, nitric acid, benzoic acid, 2-acetoxybenzoic acid, citric acid, tartaric acid, lactic acid, stearic acid, salicylic acid, glutamic acid, ascorbic acid, pamoic acid, succinic acid, fumaric acid, maleic acid, propionic acid, hydroxymaleic acid, hydroiodic acid, phenylacetic acid, acetic acid, HOOC-(CH2) n-COOH (n=0-4) and the like. Similarly, pharma-ceutically acceptable cations include, but are not limited to, sodium, potassium, calcium, aluminum, and ammonium. In general, pharma-ceutically acceptable acid or base salts can be synthesized from a parent compound that contains a basic or acidic moiety by any conventional chemical method. For example, such salts can be prepared by reacting the free acid or base form of these compounds with a stoichiometric amount of the appropriate base or acid in a suitable solvent.
[0080] The term "effective amount" refers to the amount of a desired chemical composition or a specified component sufficient to achieve a desired biological and / or therapeutic result. The desired result may be the alleviation or amelioration of the signs, symptoms or causes of a disease, or any other desired change in a biological system. If the desired result is a therapeutic response, the effective amount will vary depending on, but is not limited to, the specific disease or condition being treated or alleviated, the age, sex and weight of the subject being treated, the dosage schedule of the formulation, the severity of the condition, the method of administration, etc. These factors can be easily determined by those skilled in the art.

DETAILED DESCRIPTION OF THE PREFERRED EMBODIMENTS
[0087] Perillyl alcohol (PA) and other cyclic monoterpenes have demonstrated antineoplastic activity in cell and animal models. The mode of action of PA in programmed cell death of cancer cells is under investigation. However, studies have shown that PA can increase mannose-6-phosphate and insulin-like growth factor II receptors. In addition, treatment of cell lines with PA has been demonstrated to increase tissue growth factor beta receptor and BAK, a proapoptotic Bcl-2 family member. PA exposure has also been shown to decrease RAS protein prenylation, decrease ubiquinone synthesis, and induce phase I and phase II detoxification systems.
[0088] Another proposed mechanism of action of PA in cancer cell apoptosis involves the modulation of CDC42 gene expression, including enzymatic interaction with rho GTPases and pausing of spindle fiber formation. In particular, the dynamics between CDC42 and the response proteins γ-H2AX and p-Chk1 have been proposed to be altered in the presence of these compounds. There is also evidence suggesting that increased arrest through the G2 / M checkpoint may affect the relative expression levels of the response proteins γ-H2AX and p-Chk1. Although additional studies are required to confirm the exact mode of action of PA in cancer cell apoptosis, the conversion of PA, limonene and related compounds into prodrugs represents a novel strategy for the treatment of cancer. However, the compounds have been ineffective in human models due to poor bioavailability and ADME (absorption, distribution, metabolism and excretion) issues.
[0089] The structures of (R)-(+)PA and (S)-(-)PA are shown in Figure 1A. In nature, limonene is formed from geranyl pyrophosphate via the neryl carbocation or its equivalent. The final step involves the loss of a proton from the cation to form an alkene. The stepwise oxidation of limonene to perillic acid is shown in Figure 1B.
[0090] A prodrug is a drug or compound that is metabolized (i.e., converted in the body) into a pharmacologically active drug after administration. Instead of administering a drug directly, the corresponding prodrug can be used to improve the way the drug is absorbed, distributed, metabolized, and excreted (ADME). It can be considered that about 10% of all drugs sold worldwide are prodrugs. For example, aspirin was originally synthesized in the 19th century with the aim of producing a prodrug of salicylic acid that was acceptable when taken orally.
[0091] At least 30 prodrugs have been approved by the FDA since 2008. Prodrugs can be classified into two major types based on the manner in which the body converts the prodrug into the final active drug form. Type I prodrugs are bioactivated inside the cell (i.e., intracellularly). Examples include antiviral nucleoside analogs, which must be phosphorylated, and lipid-lowering statins. Type II prodrugs are bioactivated outside the cell (i.e., extracellularly), such as in the digestive fluids or in the body's circulatory system. Examples of Type II prodrugs include salicin, and certain antibody-targeted, gene-targeted, or virus-targeted enzyme prodrugs used in chemotherapy or immunotherapy. Either type can be further classified into subtypes based on factors such as whether the site of bioactivation within the cell is also the site of therapeutic action (Type 1), or whether bioactivation occurs in the digestive fluids or in the circulatory system (Type 2).
[0092] The present invention overcomes the potential dangers inherent in the prior art by providing novel compositions and methods for use in the treatment of diseases such as cancer. Specifically, the present invention provides cyclic monoterpenes as prodrug compositions. As prodrugs, bioavailability and ADME issues are improved. Conjugation of cyclic monoterpenes can also increase the lipophilicity of the compound. This can increase the penetration of the conjugate through the blood-brain barrier. Furthermore, smaller drug doses can be administered to achieve the desired effect while minimizing toxicity.
[0093] In some embodiments, the prodrugs described herein can target specific tissues, such as tumors. The compounds can be used to treat diseases, such as cancer. In other embodiments, the disease is an infection, Parkinson's disease (PD), gallstones, cholecystitis, or cholangitis.
[0094] In some embodiments, one or more monoterpenes are conjugated to a prodrug moiety such as cholecalciferol / vitamin D, retinoic acid, N-methylperillic acid, tyrosine and / or valproic acid. The molecules can be conjugated via an ester bond. In other embodiments, a linker molecule is used to form a covalent bond. Table 1 lists some drugs, their respective prodrug moieties and transporters. [Table 1]
[0095] Cholecalciferol is one of the five forms of vitamin D (i.e., vitamin D3). It is naturally synthesized in the skin, where it functions as a prohormone and is converted to calcitriol. Cholecalciferol is synthesized by the body and functions as a prehormone. Cholecalciferol is converted to its active form, calcitriol, by two reactions: first in the liver and second in the kidney, the action of which is mediated by the vitamin D receptor. This receptor regulates the synthesis of hundreds of enzymes and is present in virtually every cell of the body.
[0096] Retinoids are a class of compounds that contain four isoprenoid units in their molecular structure. Examples of retinoids include retinol (vitamin A), retinal, retiferol, tretinoin (all-trans retinoic acid, e.g., retinoic acid, retin A), isotretinoin, alitretinoin (9-cis-retinoic acid), etretinate, acitretin, tazarotene, bexarotene, and adapalene. Retinoids and retinoid-related compounds have important and diverse functions. Retinoic acid is a metabolite of vitamin A1 (all-trans retinol) and mediates the functions of vitamin A1, which are necessary for growth and development. All-trans retinoic acid (ATRA) is the predominant retinoic acid present, but isomers such as 13-cis- and 9-cis-retinoic acid also exist at fairly low levels in mammals.
[0097] Tyrosine is a non-essential amino acid that the body makes from another amino acid called phenylalanine. It is an essential component in the production of several important brain chemicals called neurotransmitters, including epinephrine, norepinephrine, and dopamine. LAT1 or SLC7A5 is a major nutrient transport protein involved in the transport of large neutral aromatic or branched amino acids from the extracellular fluid into cells. These include many essential amino acids such as phenylalanine, leucine, isoleucine, valine, tryptophan, histidine, and methionine. Tyrosine is a LAT1 substrate that has a phenolic hydroxyl group that can undergo conjugation with a variety of structurally different drugs, providing a biodegradable linkage.
[0098] Valproic acid is a branched short-chain fatty acid and the 2-n-propyl derivative of valeric acid. Prodrugs of valproic acid include propyl valproic acid (P-VPA), butyl valproic acid (B-VPA, isobutyl valproic acid (IB-VPA), isoamyl valproic acid (IA-VPA), and hexyl valproic acid (H-VPA). Due to the rapid conversion of the prodrug to the parent drug, the concentration of VPA in plasma after administration of the prodrug peaks soon after administration (i.e., at 6 to 26 minutes). Valproic acid (or sodium valproate) is commonly used for the treatment of epilepsy and bipolar disorder, as well as to prevent migraines. Active Transport Prodrugs
[0099] There are two major modes of transport of molecules across biological membranes: passive and active transport. Passive transport (e.g., diffusion) occurs along a concentration gradient from a higher concentration to a lower concentration. This mode of transport does not require energy. In contrast, active transport is an energy-driven process in which membrane proteins transport molecules through cells. Active transport can be classified as primary or secondary based on how energy is coupled to fuel these mechanisms.
[0100] In some embodiments, the prodrugs described herein are actively transported into target cells: a transmembrane protein recognizes the prodrug (or a portion of the prodrug) and can translocate it across the membrane by utilizing an energy source.
[0101] The chemical structures of some active transport prodrugs, their transport proteins and metabolites are shown in FIG. 4. N-methylperillic acid can be transported by FAH. The metabolites produced are perillic acid and methylamine. Similarly, tyrosine can be conjugated to perillic acid to generate a prodrug. Compounds can be transported into target cells by LAT1. Valproic acid can be conjugated to perillic acid to generate a prodrug. Compounds can be transported into target cells by MCT1. Retinoic acid can be conjugated to perillic acid to generate a prodrug. Compounds can be transported into target cells by protein carriers. Additionally, vitamin D can be conjugated to perillic acid to generate a prodrug that can be transported into target cells by protein carriers. Passive Transport Prodrugs
[0102] In some embodiments, the prodrugs described herein are passively transported into target cells.Prodrugs that can enter target cells via passive transport are shown in Figure 5, and include, for example, perillyl alcohol, perillyl acid, perillyl alcohol acetate, perillyl acid methyl ester, perillyl alcohol isobutyrate, perillyl acid ethyl ester, perillyl alcohol pivaloate, perillyl acid isopropyl ester, perillyl perillate, and perillyl carbonate.
