AI Technical Summary
Existing e-cigarettes face challenges in providing an aerosol that replicates the complex flavor and nicotine delivery of traditional cigarettes, with a balance of sensory factors like flavor, intensity, smoothness, and nicotine reward, while also reducing harmful toxic substances.
An aerosolizable formulation comprising at least 50 wt.% water, nicotine, and one or more acids, which protonates nicotine for controlled release, optimizing sensory properties and reducing toxic substances.
The formulation achieves a controlled nicotine delivery that enhances flavor and sensory experience, with improved throat hit and irritation, while minimizing toxic substances, by using high water content and acid protonation to manage nicotine release.

Abstract
Description
FIELD OF THEINVENTION
[0001] The present disclosure relates to aerosolizable formulations, methods of making aerosolizable formulations, containers containing aerosolizable formulations, devices containing aerosolizable formulations, and methods and uses of aerosolizable formulations.
[0002] Electronic aerosol delivery systems, such as e-cigarettes, generally contain a reservoir of liquid to be vaporized, typically containing nicotine. When a user inhales on the device, a heater is activated to vaporize a small amount of the liquid, which is then inhaled by the user.
[0003] Use of e-cigarettes in the UK is growing rapidly and it is now estimated that there are over one million e-cigarette users in the UK.
[0004] One challenge faced in providing such a system is to deliver an inhaled aerosol from the aerosol delivery device that provides an acceptable experience to the consumer. Some consumers may prefer e-cigarettes that generate an aerosol that "resembles" the smoke inhaled from tobacco products such as cigarettes. The aerosol from e-cigarettes and the smoke from tobacco products such as cigarettes provide the user with a complex chain of flavors in the mouth, nicotine absorption in the mouth and throat, and subsequently nicotine absorption in the lungs. These various aspects are described by users in terms of flavor, strength / quality, impact, spiciness / smoothness, and nicotine reward. Nicotine contributes to several of these factors and is strongly associated with factors such as spiciness, spiciness, and smoothness, which are easily perceived by consumers, and e-cigarettes may give consumers too much or too little of these parameters, depending on personal preferences. Nicotine reward is particularly complex because it is a result of both the amount and rate at which nicotine is absorbed through the oral mucosa (which is typically nicotine in the vapor phase) and the amount and rate at which nicotine is absorbed through the lungs (which is typically nicotine in the particle phase of the inhaled aerosol). Each of these factors, and the balance of factors, can strongly contribute to consumer acceptability of an e-cigarette. Thus, delivery measures that optimize the overall vaping experience are desirable for e-cigarette manufacturers.
[0005] A further challenge facing such systems is the ongoing demand for harm reduction: the harm caused by cigarettes and e-cigarette devices comes primarily from toxins, and therefore it is desirable to reduce or eliminate components that may produce toxins. Summary of the Invention

[0013] The inventors have found that it is possible to provide an advantageous system that contains a high content of water and that is provided with nicotine and an acid. The acid protonates nicotine, and the inventors have found that such protonation in a system that contains at least 50 wt% water provides control of the release of nicotine. In particular, in the system of the present invention, for example, by controlling the degree of protonation, control is provided of the location where nicotine is released or accumulated when inhaled by the user. For example, by controlling the degree of protonation, some nicotine can be delivered to the deep lungs of the user. This is in contrast to "conventional" e-liquids based on glycerol and propylene glycol, where free nicotine is more likely to be trapped in glycerol / propylene glycol than any protonated nicotine.
[0014] As will be appreciated by those of skill in the art, nicotine may exist in unprotonated, monoprotonated or diprotonated forms. The structures of each of these forms are shown below. [ka]

[0027] The relevant amount of nicotine present in the formulation in protonated form is specified herein. These amounts can be easily calculated by those skilled in the art. Nicotine (3-(1-methylpyrrolidin-2-yl)pyridine) is a dibasic acid with a pKa of 3.12 for the pyridine ring and 8.02 for the pyrrolidine ring. Nicotine can exist in protonated (mono- and di-) and non-protonated (free base) forms with different bioavailability depending on pH. [ka]
[0028] The distribution of protonated and unprotonated nicotine will change at various pH increments. [ka]
[0029] The proportion of unprotonated nicotine dominates at high pH levels, while lowering the pH will see an increase in the proportion of protonated nicotine (mono- or di-, depending on the pH). If the relative proportion of protonated nicotine and the total amount of nicotine in a sample are known, the absolute amount of protonated nicotine can be calculated.
[0030] The relative proportion of protonated nicotine in a formulation can be calculated by using the Henderson-Hasselbalch equation, which expresses pH as a derivation of the acid dissociation constant equation, and which is widely used in chemical and biological systems. Consider the following equilibrium: [ka]
[0031] The Henderson-Hasselbalch equation for this equilibrium is:
number
[0032] where [B] is the amount of unprotonated nicotine (i.e., the free base), [BH+] is the amount of protonated nicotine (i.e., the conjugate acid), and pKa is the reference pKa value of the pyrrolidine ring nitrogen of nicotine (pKa=8.02). The relative proportion of protonated nicotine can be derived from the alpha value of unprotonated nicotine calculated from the Henderson-Hasselbalch equation as follows:

[0033] Determination of the pKa values of nicotine formulations was performed using the basic method described in "Spectroscopic investigations into the acid-base properties of nicotine at different temperatures", Peter M. Clayton, Carl A. Vas, Tam TTBui, Alex F. Drake and Kevin McAdam, Anal. Methods, 2013, 5, 81-88.

[0063] The inventors have identified an aqueous system that delivers water droplets containing an active agent to desired areas of the lungs, such as the deep lung, by carefully selecting the particle size distribution of the aerosol. The inventors have determined that in aqueous aerosols, the droplets tend to evaporate and reduce in size when inhaled by the user. Thus, the specific selection of particle size described herein addresses the size reduction due to evaporation and still delivers the active agent to desired areas of the lungs. These issues and the selections made in the present invention are in contrast to conventional liquids based on glycerol and propylene glycol, where the droplets tend to adhere to moisture inside the lungs when inhaled, thereby increasing in size.
[0064] The formulation may be contained or delivered by any means. In one embodiment, the present invention provides a contained aerosolizable formulation comprising (a) one or more containers and (b) an aerosolizable formulation as defined herein. The container may be any suitable container, for example to allow for storage or delivery of the formulation. In one embodiment, the container is configured to engage with an electronic aerosol delivery system. The container may be configured to be in fluid communication with an electronic aerosol delivery system such that the formulation can be delivered to the electronic aerosol system. As mentioned above, the present disclosure relates to a container that can be used in an electronic aerosol delivery system, such as an e-cigarette. Throughout the following description, the term "e-cigarette" is used, which term can be used interchangeably with electronic aerosol delivery system.
[0065] As discussed herein, the container of the present invention is typically provided for delivery of an aerosolizable formulation to or within an e-cigarette. The aerosolizable formulation may be retained within the e-cigarette or sold as a separate container for subsequent use with or within the e-cigarette. As will be understood by those skilled in the art, the e-cigarette may include a unit known as a removable cartomizer, which typically includes a reservoir of the aerosolizable formulation, a wick material, and a heating element for vaporizing the aerosolizable formulation. In some e-cigarettes, the cartomizer is part of an integrally molded device and is not removable. In one embodiment, the container is the cartomizer or is part of the cartomizer. In one embodiment, the container is not the cartomizer or is not part of the cartomizer, but is a container such as a tank, which can be used to deliver a nicotine formulation to or within the e-cigarette.
[0066] In one embodiment, the container is part of an e-cigarette.Accordingly, in a further embodiment, the present invention provides an electronic aerosol delivery system comprising an aerosolizable formulation as defined herein, an aerosolization device for aerosolizing the formulation for inhalation by a user of the electronic aerosol delivery system, and a power source comprising a cell or battery for powering the aerosolization device.
[0067] In addition to the aerosolizable formulations of the invention and systems, such as containers and electronic aerosol delivery systems, containing the same, the invention provides methods for improving the sensory properties of aerosolized nicotine. In a further aspect, the invention provides methods for improving the storage stability of aerosolized nicotine formulations.
[0068] Reference to improving the sensory properties of the vaporized nicotine solution may include improving the smoothness of the vaporized nicotine solution as perceived by the user.
[0069] Methods of the present invention may include additional steps either before the recited steps, after the recited steps, or between one or more of the recited steps.

[0072] The invention will now be described with reference to the following non-limiting examples.
[0073] Example 1 A series of tests were conducted using a commercially available vibrating mesh nebulizer device. A formulation containing 90.0% (w / w) water, 9.0% 2-hydroxypropyl-β-cyclodextrin, 0.9% l-menthol, and 0.1% nicotine was loaded into a "free base nicotine control" device.
[0074] Similar devices were prepared in which 0.05% w / w citric acid was added to the formulation and the nicotine content was correspondingly reduced to 0.05% (w / w). -1 ) and citric acid (192.123g / mol -1 ), a 1:1 molar equivalent ratio would be expected, suggesting a level of nicotine protonation close to 100%.
[0075] Each of these devices was given to two subjects, including e-cigarette users, who were asked to puff on each device five times in a sequential monadic fashion and were asked to identify a preference for free base or protonated nicotine.
[0076] Both monitors demonstrated a strong preference for the formulation containing protonated nicotine, and both cited improved flavor delivery over the free base nicotine sample, as well as improved sensory attributes of nicotine, such as improved throat stickiness and irritation.
[0077] Example 2 A series of tests were conducted using a commercially available vibrating mesh nebulizer device. The device was filled with a formulation containing 99.6% water (w / w) and 0.4% nicotine.
[0078] Similar devices were prepared in which 0.48% (w / w) citric acid was added to the formulation and the water content was correspondingly reduced to 99.12% (w / w). -1 ) and citric acid (192.123g / mol -1 ), a 1:1 molar equivalent ratio would be expected, suggesting a level of nicotine protonation close to 100%.
[0079] Similar devices were prepared in which 0.4% (w / w) malic acid was added to the formulation and the water content was correspondingly reduced to 99.2% (w / w). -1) and malic acid (134.09 g / mol -1 ), a 1:1 molar equivalent ratio would be expected, suggesting a level of nicotine protonation close to 100%.
[0080] Similar devices were prepared with a formulation containing 77.6% (w / w) water, 15.0% (w / w) 2-hydroxypropyl-β-cyclodextrin, 7.0% (w / w) apple flavor, and 0.4% (w / w) nicotine.
[0081] Similar devices were prepared in which 0.4% (w / w) benzoic acid was added to the formulation and the water content was correspondingly reduced to 77.2% (w / w). -1 ) and benzoic acid (122.12 g / mol -1 ), a 1:1 molar equivalent ratio would be expected, suggesting a level of nicotine protonation close to 100%.
[0082] Similar devices were prepared in which 0.4% (w / w) malic acid was added to the formulation and the water content was correspondingly reduced to 77.2% (w / w). -1 ) and malic acid (134.09 g / mol -1 ), a 1:1 molar equivalent ratio would be expected, suggesting a level of nicotine protonation close to 100%.
[0083] [Table 1]
[0084] The droplet size distribution (DSD) generated upon device actuation was measured by laser diffraction using a Malvern Instruments (Worcestershire, UK) Spraytec system. The position of the aerosol source (e.g., device outlet) was kept constant at a fixed distance (2.5 cm) from the laser beam during measurements to ensure proper actuation and aimed in the path of the laser beam. Measurements were performed in triplicate and the average D50 values are reported in Table 1. Typical particle size distributions for each formulation are shown in Figures 2A-2F.
[0085] Seven panelists participated in the sensory evaluation of the unflavored formulations. Only one of the seven panelists (a heavy nicotine user) preferred the non-protonated formulation over the protonated form. Six of the seven panelists preferred the protonated form (one preferred the citrate form and five preferred the malate protonated form).
[0086] Five panelists participated in a sensory evaluation of the apple flavored formulations. Only one of the five panelists preferred the non-protonated formulation over the protonated form. Four of the five panelists preferred the protonated form (one preferred the citric acid form and three preferred the benzoic acid protonated form). Overall nicotine hit, chest impact, and flavor intensity were rated higher in the protonated formulation. The results of this evaluation are shown in Figures 3A and 3B.
