Abstract
Description
[Technical field]
[0001] The present invention relates to arthropod control compositions comprising compounds according to formula (I), methods and uses for controlling arthropods and arthropod control articles comprising the arthropod control compositions.
[0002] 2. Background of the Invention Many mammals, including humans, are plagued by arthropod activity, some of which, such as mosquitoes and ticks, are undesirable to vertebrate, e.g., mammals, particularly human subjects, because their bites can result in itching, disease and / or bacterial transmission, or can be the cause of other diseases and / or conditions.
[0003] The arthropod control composition contains an active material and, when applied to the skin, clothing, or other surface, can prevent arthropods from landing or climbing on the surface. Arthropod control agents are useful for preventing and controlling the occurrence of arthropod-borne diseases, such as malaria.
[0004] The prior art describes structurally diverse arthropod control agents, such as N,N-diethyl-3-methylbenzamide (also known as DEET) or ethyl butylacetylaminopropionate (also known as insect repellent 3535, IR3535).
[0005] However, some known arthropod control agents have certain drawbacks, such as being irritating to the skin, particularly to subjects, children, and / or having negative olfactory properties, e.g., a foul odor.
[0006] There is a need to provide arthropod control compounds and compositions that use limited amounts of irritant arthropod control agents or that do not use irritant arthropod control agents at all.
[0007] EP 1 022 265 describes the olfactory properties of some of the compounds used in the compositions of the present invention and their use as fragrance ingredients providing a lily of the valley type fragrance characteristic, but does not disclose or suggest the compounds as having arthropod control activity.
[0008] None of the prior art discloses or suggests that the compounds according to the present invention have arthropod control activity. [Brief description of the drawings]

[0033] The expression "hydrocarbon group" is understood to mean that said group consists of hydrogen and carbon atoms and may be in the form of an aliphatic hydrocarbon, i.e., a linear or branched saturated hydrocarbon (e.g., an alkyl group), a linear or branched unsaturated hydrocarbon (e.g., an alkenyl or alkynyl group), a saturated cyclic hydrocarbon (e.g., a cycloalkyl) or an unsaturated cyclic hydrocarbon (e.g., a cycloalkenyl), or in the form of an aromatic hydrocarbon, i.e., an aryl group, or in the form of a mixture of the above types of groups, for example, a particular group may contain linear alkyl, branched alkenyl (e.g., having one or more carbon-carbon double bonds), (poly)cycloalkyl and aryl moieties, unless a specific limitation to only one type is mentioned. Similarly, in all embodiments of the present invention, when a group is mentioned as being in one type of saturated or unsaturated form (e.g., an alkyl group), it is meant that said group may be of any type of topology (e.g., linear, cyclic or branched), or may have several moieties with different topologies.
[0034] The expression "optionally substituted" is understood to mean that the optionally substituted group may or may not be substituted with the corresponding substituent.
[0035] The term "arthropod" has its usual meaning to those skilled in the art. Arthropods include invertebrates, such as insects, arachnids, and crustaceans, which have body segments and attached appendages. Arthropods usually have a chitinous exoskeleton that is molted at intervals, and a dorsal forebrain connected to a ventral chain of ganglia.
[0036] Arthropods in the understanding of the present invention relate to undesirable arthropods, which means that their presence in the air, on the surface of objects, on the surface of plants or on vertebrates, such as human subjects or other mammals, preferably on the surface of human subjects, is undesirable. Preferably, undesirable arthropods are pest arthropods that affect plants and animals, such as thrips, aphids, beetles, moths, mealybugs, scale insects, etc., more preferably pest arthropods that affect animals, such as ants, termites, cockroaches, flies, etc., even more preferably blood-sucking arthropods that affect vertebrates, such as stable flies, bedbugs, assassin bugs, fleas, lice, mosquitoes and mites, even more preferably mosquitoes and mites.
[0037] The presence of arthropods may be undesirable because their presence in the air is unpleasant to the subject, contact of arthropods with items may transmit disease and / or bacteria, or arthropods may bite organisms, causing itching and leading to the transmission of disease and / or bacteria, or arthropod ingestion may cause other diseases and / or conditions.
[0038] In certain embodiments, the arthropod is an insect or an arachnid, preferably an insect.
[0039] The term "insect" has its ordinary meaning to one of ordinary skill in the art. Insects are described by a distinct head, thorax and abdomen, no more than three pairs of legs, and typically one or two pairs of wings.
[0040] In certain embodiments, the insect is a mosquito, stable fly, bedbug, assassin bug, flea, lice, ant, termite, cockroach, fly, aphid, beetle, thrips, moth, mealybug or scale insect, more preferably a mosquito.
[0041] The term "arachne" has its ordinary meaning to one of ordinary skill in the art. Arachne is described as having a body segment divided into two regions, the anterior portion of which has four pairs of legs but no antennae.
[0042] In certain embodiments, the arachnid is a tick, mite, chigger or spider, more preferably a mite.
[0043] Expressions such as "control", "arthropod control", "insect control" or "arachnid control" have their ordinary meaning to those skilled in the art.
[0044] In the context of the present invention, "control" is defined as the ability of the compound of formula (I) or arthropod control composition of the present invention to attract, inhibit, kill or repel arthropods, preferably the ability to inhibit or repel arthropods, and even more preferably the ability to repel arthropods.
[0045] According to the present invention, "attract" is defined as the ability of the compound of formula (I) or arthropod attractant composition of the present invention to increase or promote the contact or presence of arthropods at an arthropod attractant source to which the arthropod attractant compound or arthropod attractant composition is applied, for example, in the air, on the surface of an article, or on the surface of a vertebrate such as a human subject or other mammal, preferably on the surface of an article such as a trapping device.
[0046] According to the present invention, "inhibit" is defined as the ability of the compound of formula (I) or arthropod control composition of the present invention to minimize, reduce, deter or prevent the contact or presence of arthropods in the arthropod control source to which the arthropod control compound or arthropod control composition is applied, for example, in the air, on the surface of an article, or on the surface of a vertebrate such as a human subject or other mammal, preferably a human subject. Typically, the inhibitory effect is exhibited when the arthropod control compound or arthropod control composition is used as a feeding deterrent to prevent subsequent feeding or egg-laying of pests after the initial application.
[0047] According to the present invention, "killing" is defined as the ability of the compound of formula (I) or arthropodicidal composition of the present invention to kill arthropods at the arthropod killing source to which the arthropodicidal compound or arthropodicidal composition is applied, for example, in the air, on the surface of an article, or on the surface of a vertebrate such as a human subject or other mammal, preferably a human subject. When the arthropodicidal composition is applied to a plant, animal, or human subject, the arthropodicidal composition is applied in an amount that kills arthropods but does not kill the subject.
[0048] In accordance with the present invention, "repellency" is defined as the ability of the compound of formula (I) or arthropod repellent composition of the present invention to minimize, reduce, deter or prevent the approach or presence of arthropods at an arthropod-inhibiting source to which the arthropod repellent compound or arthropod repellent composition is applied, for example, in the air, on the surface of an article, or on the surface of a vertebrate such as a human subject or other mammal, preferably a human subject.
[0049] In certain embodiments, the arthropod control source is a surface and / or air in the vicinity of an article, preferably a candle, coil, electric diffuser, wristband, patch, collar, ear tag, clothing, fabric, paper, biochar, cardboard, cellulose pad, bed net, screen, curtain, furniture, wall, ground or paint, or a surface of a subject, preferably a vertebrate such as a human subject or other mammal, preferably a human subject, i.e., the skin of a human subject that has been treated with a product such as a spray, aerosol, cream, roll-on, wristband, lotion, soap, shampoo, sunscreen or patch, or a fabric surface that has been treated with a product such as a laundry powder, liquid detergent, spray, lotion, powder, etc.
[0050] Arthropod control efficacy according to the present invention is determined for mosquitoes using an adapted warm body assay as defined in Kroeber T, Kessler S, Frei J, Bourquin M, Guerin PM. An in vitro assay for testing mosquito controlling compounds employing a warm body and carbon dioxide as a behavioral activator. J Am Mosq Control Assoc. 2010; 26:381-386.
[0051] The control efficacy of the present invention is determined by testing the warm-body assay against the yellow fever mosquito, Aedes aegypti Rockefeller strain. A. aegypti is a model organism for control testing and is one of the model organisms recommended by the World Health Organization (WHO) since it is a highly aggressive and human-parasitic mosquito species that generally shows low sensitivity to arthropod control compounds. Observations of control efficacy were performed on host-seeking females of uniform age, 5-10 days old, selected as mentioned in the above-mentioned publication. The starved females tested had access to 10% sugar solution but were not allowed to feed.
[0052] The published protocol was adapted to count landing mosquitoes automatically using automated counting software instead of manually, and switching from Anopheles gambiae to A. aegypti reduced the number of mosquitoes placed in the tested cages due to size differences (i.e., 30 mosquitoes instead of 50), and also increased the lighting (i.e., 150 lux instead of 4 lux) as A. aegypti is a diurnal mosquito.
[0053] In addition, by using the in vitro system mentioned above, it is possible to evaluate at what concentration the compounds according to formula (I), either alone or in a composition, control all the arthropods tested.
[0054] Furthermore, the control effect of the compounds according to formula (I) can be tested at various concentrations, either alone or in a composition, so that the efficacy at equivalent doses can be compared with a reference compound, such as DEET or IR3535.
[0055] In certain embodiments, the arthropod control composition of the present invention comprises an arthropod-controlling effective amount of a compound of formula (I).The compound can be used alone or in combination with other components of formula (I) and / or with arthropod-controlling co-components.Therefore, it is understood that the concentration of the compound according to formula (I), either alone or in the composition, provides a higher repellency than pure (i.e., 100%) ethanol.
[0056] In a particular embodiment, the composition comprises the compound according to formula (I) in an amount of 0.02-80% by weight, more preferably 0.05-70% by weight, and even more preferably 0.1-60% by weight, based on the total weight of the composition, whereby the composition is understood to preferably comprise the compound according to formula (I) in a minimum amount of at least 0.2% by weight, at least 0.05% by weight, or at least 0.1% by weight, and in a maximum amount of 80% by weight or less, 70% by weight or less, or 60% by weight or less, based on the total weight of the composition.
[0057] In a particular embodiment, the composition comprises a compound according to formula (I) in an amount of 0.1-5% by weight, preferably 0.15-4% by weight, more preferably 0.2-3% by weight or 11-35% by weight, preferably 15-30% by weight, more preferably 18-25% by weight, based on the total weight of the composition. Thus, in one embodiment, the composition comprises a compound according to formula (I) in a minimum amount of at least 0.1% by weight, at least 0.15% by weight or at least 0.2% by weight, and a maximum amount of 5% by weight or less, 4% by weight or less, or 3% by weight or less, based on the total weight of the composition, and in another embodiment, a compound according to formula (I) in a minimum amount of at least 11% by weight, at least 15% by weight or at least 18% by weight, and a maximum amount of 35% by weight or less, 30% by weight or less, or 25% by weight or less.
[0058] According to certain embodiments, the arthropod control compositions of the present invention comprise one or more arthropod control co-ingredients.
[0059] An "arthropod control co-ingredient" is understood to be an ingredient capable of imparting an arthropod control benefit in addition to the arthropod control effect of the compounds of formula (I) described above.
[0060] In one embodiment, the compounds of formula (I) can modify, enhance or improve the arthropod control efficacy of an arthropod control co-ingredient, for example, by reducing the amount of the arthropod control co-ingredient in the composition. This can be particularly beneficial when the arthropod control co-ingredient is harmful to human subjects at a particular dose or when the arthropod control co-ingredient has negative olfactory properties at a particular dose.
[0061] According to certain embodiments, the combination of a compound of formula (I) with an arthropod control co-ingredient provides a synergistic arthropod control effect.
[0062] The arthropod control co-ingredients are N,N-diethyl-3-methylbenzamide (DEET), ethyl butylacetylaminopropionate (IR3535), (13Z)-hexadec-13-en-11-yn-1-yl acetate, [0089] Non-limiting examples of liquid carriers can include emulsification systems, i.e. systems based on solvents and surfactants, or solvents commonly used in perfumery.Detailed descriptions of the nature and type of solvents may generally not be exhaustive. On the other hand, non-limiting examples can include solvents such as butylene glycol or propylene glycol, glycerol, dipropylene glycol and its monoethers, 1,2,3-propanetriyl triacetate, dimethyl glutarate, dimethyl adipate, 1,3-diacetyloxypropan-2-yl acetate, diethyl phthalate, isopropyl myristate, benzyl benzoate, benzyl alcohol, 2-(2-ethoxyethoxy)-1-ethanol, triethyl citrate, 2-methylprop-1-ene and 2-(2-ethoxyethoxy)ethanol or mixtures thereof, with dipropylene glycol, 2-methylprop-1-ene and 2-(2-ethoxyethoxy)ethanol and mixtures thereof being particularly suitable.
[0090] For compositions comprising a carrier, other suitable carriers than those specified above may be ethanol, water / ethanol mixtures, limonene or other terpenes, isoparaffins, for example known under the trademark Isopar® (supplier: Exxon Chemical), or glycol ethers and glycol ether esters, for example known under the trademark Dowanol® (supplier: Dow Chemical Company), or hydrogenated castor oil, for example known under the trademark Cremophor® RH40 (supplier: BASF).
[0091] Solid carrier is meant to refer to a material that arthropod control composition or some elements of arthropod control composition can be chemically or physically bound. Generally, such solid carrier is utilized either to stabilize the composition or to control the evaporation rate of the composition or some components. The use of solid carrier is currently used in the art, and those skilled in the art know how to achieve the desired effect. Meanwhile, non-limiting examples of solid carrier can include absorbent gum or polymer or inorganic material, such as porous polymer, cyclodextrin, wood-based material, organic or inorganic gel, clay, gypsum talc or zeolite.
[0092] Other non-limiting examples of solid carriers can include encapsulation materials.Examples of such materials can include wall-forming and plasticizing materials, such as monosaccharides, disaccharides or trisaccharides, natural or modified starches, hydrocolloids, cellulose derivatives, polyvinyl acetate, polyvinyl alcohol, proteins or pectins, or further materials cited in references, such as H. Scherz, Hydrokolloide: Stabilisatoren, Dickungs- und Geliermittel in Lebensmitteln, Band 2 der Schriftenreihe Lebensmittelchemie, Lebensmittelqualitaet, Behr's Verlag GmbH & Co., Hamburg, 1996.Encapsulation is a process well known to those skilled in the art, and can be carried out by using techniques such as spray drying, agglomeration or even extrusion, or can consist of coating encapsulation, including coacervation and complex coacervation techniques.
[0093] Non-limiting examples of encapsulation include core-shell capsules comprising aminoplast, polyamide, polyester, polyurea or polyurethane type resins or mixtures thereof (all of the above resins are well known to those skilled in the art) using techniques such as phase separation processes induced by polymerization, interfacial polymerization, coacervation or all of these (all of the above techniques are described in the prior art), optionally in the presence of polymeric stabilizers or cationic copolymers.
[0094] The resins can be prepared by polycondensation of aldehydes (e.g., formaldehyde, 2,2-dimethoxyethanal, glyoxal, glyoxylic acid or glycolaldehyde and mixtures thereof) with amines, such as urea, benzoguanamine, glycoluril, melamine, methylolmelamine, methylated methylolmelamine, guanazole, and the like and mixtures thereof. Alternatively, preformed resin alkylated polyamines can be used, such as those commercially available under the trademarks Urac® (supplied by Cytec Technology Corp.), Cymel® (supplied by Cytec Technology Corp.), Urecoll® or Luracoll® (supplied by BASF).
[0095] Other resins are those prepared by polycondensation of polyols, such as glycerol, with polyisocyanates, such as the trimer of hexamethylene diisocyanate, the trimer of isophorone diisocyanate or xylylene diisocyanate or the biuret of hexamethylene diisocyanate or the trimer of xylylene diisocyanate with trimethylolpropane (known under the trade name Takenate®, supplier: Mitsui Chemicals), among others the trimer of xylylene diisocyanate with trimethylolpropane and the biuret of hexamethylene diisocyanate.
[0096] Some of the prominent literature related to encapsulation by polycondensation of amino resins, i.e. melamine-based resins with aldehydes, includes those represented by articles such as Acta Polymerica, 1989, vol. 40, pages 243, 325 and 683 and 1990, vol. 41, page 91, published by K. Dietrich et al., which already describe the various parameters influencing the preparation of such core-shell microcapsules, according to the methods of the prior art, which are further detailed and exemplified in the patent literature. U.S. Patent No. 4,396,670 to Wiggins Teape Group Limited is a good early example of the latter. Since then, many other authors have enriched the literature in this field, and it is not possible to cover all the developments published here, but a general knowledge in encapsulation technology will be very important. Relatively recent relevant publications disclosing suitable uses of such microcapsules are represented, for example, by the article by H.Y. Lee et al. in Journal of Microencapsulation, 2002, vol. 19, pages 559-569, WO 01 / 41915 or also by the article by S. Bone et al. in Chimia, 2011, vol. 65, pages 177-181.

[0124] 2. Model Organisms Control efficacy was tested against the yellow fever mosquito, A. aegypti Rockefeller strain.
[0125] A. aegypti is a model organism for control testing and is one of the model organisms recommended by the World Health Organization (WHO) as it is a highly aggressive, human-parasitic mosquito species that generally shows low sensitivity to arthropod control compounds.
[0126] Control efficacy was observed for females seeking hosts of uniform age (5–10 days after emergence from pupae). Starved females tested had access to 10% sugar solution but were not allowed to feed on blood.
[0127] The repellent efficacy of different compounds was evaluated against the sheep mite, Ixodes ricinus, one of the recommended model organisms mentioned in the Guidance on the European Biological Products Regulation [Vol II, Efficacy - Assessment & Evaluation (Parts B+C), v. 3.0, April 2018]. Repellent efficacy observations were performed on terminal nymph stages.
[0128] 3. Description of Arthropod Control Test Arthropod repellency is tested using the in vitro and in vivo test methods described below.
[0129] 3.1. In vitro K&D assay In vitro control efficacy was measured using the Klun & Debboun (K&D) assay test module [Klun JA, Kramer M, Debboun M. A new in vitro bioassay system for discovery of novel human-use mosquito controlling compounds. J Am Mosq Control Assoc. 2005; 21:64-70.], which is an adaptation of the K&D module originally developed and used to quantitatively measure the efficacy of mosquito control compounds in human subjects [Klun JA, Debboun M. A new module for quantitative evaluation of controlling efficacy using human subjects. J Med Entomol. 2000; 37:177-81.].
[0130] For the in vitro assay, this module is connected to a membrane-blood reservoir consisting of a unit with 6 wells filled with 7 mL of defibrinated porcine blood maintained at a constant temperature of 38 °C and covered with a bovine collagen film (Devro plc, Edicol-MX400 CLR 100M REEL) as a membrane. On top of this membrane, a piece of cotton fabric (Baptist Roh, 100% cotton, mass: 69.0 g / m 2 , yarn density: warp and weft 26.5×26.5 yarns / cm, Modessa Basel) were placed after being treated with 80 μL of the test stimuli.
[0131] Finally, the K&D units, containing 5 mosquitoes in each cell, were placed on the treated fabric. The number of mosquitoes that bit into each cell within a 3-minute exposure was then recorded. This procedure was repeated until 6 replicate exposures and observations (i.e., 30 mosquitoes) had been performed.
[0132] 3.2. In vivo K&D assay For the K&D in vivo study [Klun JA, Debboun M. A new module for quantitative evaluation of controlling efficacy using human subjects. J Med Entomol. 2000; 37:177-81.], three study participants (two women and one man) with low or no skin reaction to mosquito bites were recruited at the Swiss Tropical and Public Health Institute (Swiss TPH). The study participants' ages were 28 and 32 years for the women and 28 years for the male participant. The study was approved by the Ethics Commission of Northwest and Central Switzerland (study number.PB_2017-00436-220 / 11), and all study participants signed an informed consent form. To avoid undesirable bias, participants were asked to avoid alcohol and products such as fragrances, colognes and lotions for at least 12 hours before and during the experiment. During the experiment, participants were asked to avoid rubbing, touching or wetting the arthropod control compound treated areas and any activity that may lead to increased sweating.
[0133] For the in vivo assay, a template was used to delineate 4 cm x 5 cm areas on the skin corresponding to each of the six cell openings (3 cm x 4 cm) on the bottom of the K&D module. The five areas were treated with various test stimuli. Modules containing five host-seeking 5- to 10-day-old mosquitoes in each cell were then placed on top of the treated skin areas. The number of mosquito bites within a 2-minute exposure was recorded. This procedure was repeated for three study participants until six replicate exposures and observations (i.e., 30 mosquitoes) had been performed.
[0134] 3.3. In vitro warm body assay 3.3.1. In Vitro Warm Body Assays for Insects, e.g., Mosquitoes The control efficacy of the present invention is determined by testing the warm-body assay against the yellow fever mosquito A. aegypti Rockefeller strain. A. aegypti is a model organism for control testing and is one of the model organisms recommended by the World Health Organization (WHO) since it is a highly aggressive and human-parasitic mosquito species that generally shows low sensitivity to arthropod control compounds. Observations of control efficacy were performed on host-seeking females of uniform age 5-10 days selected as mentioned in the above-mentioned publication. The starved females tested had access to 10% sugar solution but were not allowed to feed.
[0135] The control efficacy of the present invention was evaluated using an adapted warm body assay as defined in Kroeber T, Kessler S, Frei J, Bourquin M, Guerin PM. An in vitro assay for testing mosquito controlling compounds employing a warm body and carbon dioxide as a behavioral activator. J Am Mosq Control Assoc. 2010; 26:381-386.
[0136] The published protocol was adapted to count landing mosquitoes automatically using automated counting software instead of manually, and due to the switch from Anopheles gambiae to Aedes aegypti, the number of mosquitoes placed in the tested cages was reduced due to size differences (i.e., 30 mosquitoes instead of 50), and lighting was increased (i.e., 150 lux instead of 4 lux) as Aedes aegypti is a diurnal mosquito.
[0137] In this in vitro assay, the number of A. aegypti landings on warm bodies treated with the tested irritants was measured to evaluate their repellency. Three different doses: 0.0016%, 0.04% and 1% in ethanol were used in three replicates, with 25 A. aegypti (females, 10–12 days old) per cage.
[0138] 3.3.2. In vitro hot plate assay for arachnids, e.g. mites Repellency efficacy was evaluated using the in vitro hot plate assay protocol as defined in Kroeber T, Bourquin M, Guerin PM. A standardized in vivo and in vitro test method for evaluating tick repellents. Pesticide Biochemistry and Physiology. 2013; 107(2):160-168.
[0139] 3.4. In vivo arm in cage test The arm in the box method was adapted from the WHO guidelines for efficacy testing of mosquito repellents on human skin (WHO / CDS / NTD / WHOPES / 2009.4). The probe readiness of 100 starved female mosquitoes A. aegypti was assessed by inserting their untreated arm into a cage (40 x 40 x 40 cm) for 30 seconds (negative control) three times (once at the beginning, once at the 4th hour and once at the 8th hour) to determine the probe activity. The products were then applied to the skin of the forearm of human volunteers (600 cm 2 After 5 min, the arm is inserted into the cage and exposed for 3 min. The assay is performed in a temperature (27±2° C.) and humidity (80±10% RH) controlled room on 3 different volunteers.
[0140] 4. Evaluation of the arthropod control effect of the present invention The arthropod repellency of the present invention is tested using in vitro and in vivo test methods, as generally described below.
[0141] 4.1. In vitro K&D assay In vitro repellency against the mosquito Aedes aegypti was measured in the K&D test against various dilutions of (2,5-dimethyl-2,3-dihydro-1H-inden-2-yl)methanol and is shown in Table 1 below.
[0142] [Table 1]
[0143] The in vitro repellency of the mosquito Aedes aegypti was measured in a K&D test against various dilutions of (2,5-dimethyl-2,3-dihydro-1H-inden-2-yl)methanol according to Table 1 and is shown in FIG.
[0144] The percent repellency of Aedes aegypti increases with increasing test doses of (2,5-dimethyl-2,3-dihydro-1H-inden-2-yl)methanol, demonstrating the biological effectiveness of (2,5-dimethyl-2,3-dihydro-1H-inden-2-yl)methanol (see Table 1 and Figure 1).
[0145] At low concentrations (0.0008%, 0.004% and 0.02%), the repellency was comparable to that induced by the solvent ethanol, indicating that (2,5-dimethyl-2,3-dihydro-1H-inden-2-yl)methanol itself was not effective at these concentrations (see Table 1 and Figure 1). At concentrations of (2,5-dimethyl-2,3-dihydro-1H-inden-2-yl)methanol above 0.02%, the % repellency was consistently higher than that induced by the solvent ethanol (see Table 1 and Figure 1).
[0146] Using the K&D in vitro assay, (2,5-dimethyl-2,3-dihydro-1H-inden-2-yl)methanol by itself exhibited significant repellent effects against Aedes aegypti at concentrations of 0.1% or greater in ethanol.
[0147] 4.2. In vivo K&D assay The in vivo repellency of the mosquito Aedes aegypti measured against various dilutions of (2,5-dimethyl-2,3-dihydro-1H-inden-2-yl)methanol in the K&D test is shown in Table 2 below.
[0148] [Table 2]
[0149] The in vivo repellency of the mosquito Aedes aegypti measured against various dilutions of (2,5-dimethyl-2,3-dihydro-1H-inden-2-yl)methanol according to Table 2 in the K&D test is shown in FIG.
[0150] The percent repellency of Aedes aegypti increases with increasing test doses of (2,5-dimethyl-2,3-dihydro-1H-inden-2-yl)methanol, demonstrating the biological effectiveness of (2,5-dimethyl-2,3-dihydro-1H-inden-2-yl)methanol (see Table 2 and Figure 2).
[0151] At low concentrations (0.02% and 0.1%), the repellency exceeded that induced by the vehicle, but there was a large variability such that a clear repellent effect of (2,5-dimethyl-2,3-dihydro-1H-inden-2-yl)methanol per se could not be claimed at these concentrations (see Table 2 and Figure 2). On the other hand, at concentrations of (2,5-dimethyl-2,3-dihydro-1H-inden-2-yl)methanol above 0.1%, the % repellency was consistently higher than that induced by the vehicle ethanol (see Table 2 and Figure 2).
[0152] Using the K&D in vivo assay, (2,5-dimethyl-2,3-dihydro-1H-inden-2-yl)methanol by itself exhibited significant repellent effects against Aedes aegypti at concentrations of 0.5% or greater in ethanol.
[0153] 4.3. In vitro warm body assay The average in vitro numbers of Aedes aegypti mosquitoes landing on hosts challenged with various dilutions of DEET, IR3535, (2,5-dimethyl-2,3-dihydro-1H-inden-2-yl)methanol or the solvent ethanol are shown below in Table 3. Due to variability between studies, the number of mosquitoes landing during the 100% ethanol control was used to normalize the number of mosquitoes landing using the irritants.
[0154] [Table 3]
[0155] The number of Aedes aegypti mosquitoes landing on the host decreased with increasing tested doses of stimuli, demonstrating the biological effectiveness of all stimuli (see Table 3 and Figure 3).
[0156] At concentrations below 0.04%, IR3535 and (2,5-dimethyl-2,3-dihydro-1H-inden-2-yl)methanol showed only low repellency (see Table 3 and Figure 3). At concentrations of irritants of 0.20% or higher, repellency was consistently high, with fewer mosquitoes landing on host bodies loaded with the tested compounds (see Table 3 and Figure 3).
[0157] Using a warm body in vivo assay, fragrances containing (2,5-dimethyl-2,3-dihydro-1H-inden-2-yl)methanol demonstrated significant repellent effects against Aedes aegypti, similar to that of IR3535.
[0158] 4.4. In vitro warm body assay The mean ± SD in vitro values of Aedes aegypti mosquitoes landing on hosts loaded with different dilutions of two fragrances containing 33% and 10% (2,5-dimethyl-2,3-dihydro-1H-inden-2-yl)methanol (Fragrances 1a and 1b, respectively), two reference compounds: DEET and IR3535, or the solvent ethanol are shown in Table 4 below.
[0159] [Table 4]
[0160] The in vitro repellency of the mosquito Aedes aegypti, measured in a warm body assay according to Table 4, is shown in Figure 4 below.
[0161] The number of Aedes aegypti mosquitoes landing on the host increases with increasing tested doses of irritant, demonstrating the biological effect of the irritant (see Table 4 and Figure 4).
[0162] At low concentrations (0.0016%), the tested stimuli did not induce repellency; the number of mosquitoes that landed was similar between all stimuli and the solvent ethanol (see Table 4 and Figure 4). On the other hand, at stimuli concentrations of 0.04% and above, repellency was consistently higher, with fewer mosquitoes landing on the host loaded with the tested stimuli, compared to ethanol (see Table 4 and Figure 4). Two fragrances containing (2,5-dimethyl-2,3-dihydro-1H-inden-2-yl)methanol showed similar control efficacy as IR3535 at equivalent doses (see Table 4 and Figure 4).
[0163] Using a warm body in vivo assay, fragrances containing (2,5-dimethyl-2,3-dihydro-1H-inden-2-yl)methanol demonstrated significant control against Aedes aegypti, similar to the control effect of IR3535.
[0164] 4.5. In vitro warm body assay, combined with known repellents A mosquito Aedes aegypti landing on a host loaded with 5 μL of a 1% blend of one fragrance containing 33% or 6.5% (2,5-dimethyl-2,3-dihydro-1H-inden-2-yl)methanol (fragrance 1a and 1e, respectively) and 7.5% or 12.5% IR3535 is shown in FIG. 5.
[0165] The number of Aedes aegypti mosquitoes landing on the warm bodies increases over time. Less than 60% of the mosquitoes landed on the warm bodies in the attraction assay within 6 h, demonstrating the repellent effect of the three blends (see Figure 5).
[0166] Figure 6 shows mosquitoes Aedes aegypti landing on a host 4 hours after application of 5 μL of a blend containing 0.25% or 1% fragrance 1a, which contains 33% (2,5-dimethyl-2,3-dihydro-1H-inden-2-yl)methanol and 12.5% IR3535.
[0167] The number of mosquitoes Aedes aegypti that landed on the host decreased with increasing amounts of fragrance 1a in the formulation (see Figure 6).
[0168] 4.6. In Vitro Incubation Assay, Testing in Combination with Co-ingredients that Contribute to Both the Arthropod Control Activity and Olfactory Properties of the Composition % mosquitoes Aedes aegypti landing on host bodies loaded with blends prepared from 50% (2,5-dimethyl-2,3-dihydro-1H-inden-2-yl)methanol + 50% (1R)-cis-3-oxo-2-pentyl-1-cyclopentane methyl acetate or pure (1R)-cis-3-oxo-2-pentyl-1-cyclopentane methyl acetate demonstrated similar repellency at the three concentrations tested, as shown in Figure 7.
[0169] Thus, replacement of half of the (1R)-cis-3-oxo-2-pentyl-1-cyclopentaneacetic acid with (2,5-dimethyl-2,3-dihydro-1H-inden-2-yl)methanol did not affect the repellent efficacy of the irritant but favorably improved its pleasantness.
[0170] 4.7. In-vitro anaerobic assays using two additional sets of arthropod control compounds In this in vitro assay, the number of Aedes aegypti that landed on a host treated with the tested irritants was measured to assess repellency, with three replicates at three different doses: 0.0016%, 0.04% and 1% in ethanol.
[0171] Additional compounds demonstrated variable repellent effects against Aedes aegypti using an in vitro repellent assay. All compounds tested demonstrated significant repellent effects.
[0172] The number of Aedes aegypti mosquitoes landing on the host decreases with increasing doses of the stimuli tested, demonstrating the biological effect of the stimuli (Figures 8, 9 and 10).
[0173] 4.7.1. First set of arthropod control compounds At low concentrations (0.0016%), two compounds, 4a,8-dimethyl-indano[1,2-d]-1,3-dioxan-2-one and (2,4,6-trimethyl-2,3-dihydro-1H-inden-2-yl)methanol, provided greater than 25% reduction in landing, similar to DEET (Figure 8).
[0174] At a concentration of 0.04%, (2,4,6-trimethyl-2,3-dihydro-1H-inden-2-yl)methanol remained as efficient as DEET in preventing mosquito landings, reducing landings by 88% and 92%, respectively (Figure 5). Four additional compounds tested at 0.4%, 4a,8-dimethyl-indano[1,2-d]-1,3-dioxan-2-one, 5-ethyl-2-methyl-2-indanethanol, 1-(2,5-dimethyl-2-indanyl)-1-ethanol and (2,5-dimethyl-2,3-dihydro-1H-inden-2-yl)methanol, induced a reduction in landings of approximately 70%, similar to the effect of IR3535 (Figure 8).
[0175] All compounds tested at 1% and the reference demonstrated significant avoidance by mosquitoes with less than 15% of mosquitoes landing on the warm body (Figure 8).
[0176] It is also worth noting that when (2,4,6-trimethyl-2,3-dihydro-1H-inden-2-yl)methanol was applied at 1%, no mosquitoes landed (Figure 8).
[0177] 4.7.2. Second set of arthropod control compounds At low concentrations (0.0016%), five tested compounds, 2,5-dimethyl-2-indanyl)methyl formate, (2,5-dimethyl-2,3-dihydro-1H-inden-2-yl)methyl methyl ether, 2,5-dimethyl-2,3-dihydro-1H-indene-2-carboxylic acid, (2,5-dimethyl-2,3-dihydro-1H-inden-2-yl)methanol, and 5-isopropyl-2-methyl-2-indanethanol, provided similar or greater reductions in landing than IR3535. Landings were reduced by 4% to 29% (Figure 9).
[0178] At a concentration of 0.04%, these compounds reduced landing numbers by 29% to 52%, which was lower than the reductions obtained with IR3535 or DEET (Figure 9).
[0179] At a concentration of 1%, all compounds reduced the number of landings by more than 50%. (2,5-Dimethyl-2,3-dihydro-1H-inden-2-yl) methyl methyl ether induced a similar effect to IR3535: a landing reduction of 86% (Figure 9).
[0180] 4.7.3. Third set of arthropod control compounds 2-(hydroxymethyl)-2,6-dimethyl-2,3-dihydro-1H-inden-1-ol (orange) and 5-tert-butyl-2-methyl-2-indanethanol (pink) showed low and decreasing landings with increasing amounts of stimuli tested (Figure 10).
[0181] Other irritants, such as blends of (1RS,2RS)-2-((2-methoxypropan-2-yloxy)methyl)-2,6-dimethyl-2,3-dihydro-1H-inden-1-ol + (1RS,2SR)-2-((2-methoxypropan-2-yloxy)methyl)-2,6-dimethyl-2,3-dihydro-1H-inden-1-ol (brown) and blends of (1RS,2RS)-2-(hydroxymethyl)-2,6-dimethyl-2,3-dihydro-1H-inden-1-ol) + (1RS,2SR)-2-(hydroxymethyl)-2,6-dimethyl-2,3-dihydro-1H-inden-1-ol) (green), demonstrated reduced landings with increasing amounts of irritant, but were clearly less repellent than DEET or IR3535 (Figure 10).
[0182] The compounds of the third set of arthropod control compounds showed a significant repellent effect, whereas these compounds induced a lower repellent effect compared to the compounds of the first two sets mentioned above (Figures 8, 9 and 10).
[0183] 4.7.4. Fourth set of arthropod control compounds [Table 5-1] [Table 5-2]
[0184] All compounds tested reduced A. aegypti landing numbers by more than 50% at the intermediate dose of 0.04%, except for (2,6-dimethyl-1,2,3,4-tetrahydro-2-naphthalenyl)methanol, (4aRS,9bRS)-2,2,4a,7-tetramethyl-4,4a,5,9b-tetrahydroindeno[1,2-d][1,3]dioxin and (4aRS,9bRS)-2,2,4a-trimethyl-4,4a,5,9b-tetrahydroindeno[1,2-d][1,3]dioxin. This decrease was observed for seven compounds; 2,3-dihydro-1H-inden-2-ylmethanol, a blend of (2RS,4aRS,9bSR)-2-methyl-4,4a,5,9b-tetrahydroindeno[1,2-d][1,3]dioxine + (2RS,4aSR,9bRS)-2-methyl-4,4a,5,9b-tetrahydroindeno[1,2-d][1,3]dioxine, (4aRS,9bS It was even greater than 90% for R)-2,2-dimethyl-4,4a,5,9b-tetrahydroindeno[1,2-d][1,3]dioxin, (2RS,4RS,4ASR,9BRS)-2,4-dimethyl-4,4a,5,9b-tetrahydroindeno[1,2-d][1,3]dioxin and 4,4A,5,9B-tetrahydro-indeno[1,2-d]-1,3-dioxin. Moreover, this reduction was greatest for two compounds; (4aRS,9bRS)-4a,7-dimethyl-4,4a,5,9b-tetrahydroindeno[1,2-d][1,3]dioxin and the blend of (2RS,4aRS,9bRS)-2,4a,7-trimethyl-4,4a,5,9b-tetrahydroindeno[1,2-d][1,3]dioxin + (2RS,4aSR,9bSR)-2,4a,7-trimethyl-4,4a,5,9b-tetrahydroindeno[1,2-d][1,3]dioxin, such that no mosquitoes landed at all (Table 6).
[0185] At 1%, 16 compounds reduced A. aegypti landing numbers by >90% (Table 6). At this 1% dose, five compounds; 2,3-dihydro-1H-inden-2-ylmethanol, 2,3-dihydro-1H-indene-2-carbonitrile, (4aRS,9bRS)-4a,7-dimethyl-4,4a,5,9b-tetrahydroindeno[1,2-d][1,3]dioxin, (2RS,4RS,4ARS,9BSR)-2,4-dimethyl-2,4a,5,9b-tetrahydroindeno[1,2 -d][1,3]dioxin and a blend of (2RS,4aRS,9bRS)-2,4a,7-trimethyl-4,4a,5,9b-tetrahydroindeno[1,2-d][1,3]dioxin + (2RS,4aSR,9bSR)-2,4a,7-trimethyl-4,4a,5,9b-tetrahydroindeno[1,2-d][1,3]dioxin even managed to keep mosquito landings to zero (Table 6).
[0186] 4.8. In vitro thermophilic assay with blends In this in vitro assay, the number of Aedes aegypti that landed on a warm body treated with the tested irritants was measured to evaluate the repellency. Various doses ranging from 0.0016% to 1% in ethanol were administered in one replicate.
[0187] Using an in vitro repellent assay, various stimuli containing one component related to formula I demonstrated variable repellent effects against A. aegypti. The number of A. aegypti mosquitoes landing on the repellent tended to decrease with increasing amounts of the stimuli tested, demonstrating the biological effectiveness of the stimuli (Table 7).
[0188] [Table 6]
[0189] All blends and fragrances at concentrations above 0.2% induced a significant repellent effect, since they were able to reduce the number of landings by more than 80% (Table 7). This threshold was achieved even at lower concentrations (80% reduction at a concentration of 0.04%) for blends containing known insect repellent compounds, i.e., Blends #2 and #3.
[0190] Moreover, at low concentrations (0.0016%), Blends #2 and #3 showed significantly higher reductions in landing compared to the single compounds that made up the blends, e.g., a 77% reduction in landing in Blend #3 compared to 5% and 11% reductions for 4,4A,5,9B-tetrahydro-indeno[1,2-D]-1,3-dioxin and 2-(2-hydroxypropan-2-yl)-5-methylcyclohexan-1-ol, respectively, at this same dose.
[0191] It is also worth noting that when Blend #3 was applied at 0.04% and 1%, no mosquitoes landed (Table 7).
[0192] Using the same in vitro setup, Blend #3, prepared from 75% 4,4A,5,9B-tetrahydro-indeno[1,2-D]-1,3-dioxin and 25% 2-(2-hydroxypropan-2-yl)-5-methylcyclohexan-1-ol, was evaluated over time.
[0193] [Table 7]
[0194] As shown in Table 8, 2 and 4 hours after Blend #3 was applied, no Aedes aegypti mosquitoes landed on the host. Six hours after application, the blend was no longer effective, with the same number of landings as the control.
[0195] 4.9. In vivo arm in cage testing A 20% solution of (2,5-dimethyl-2,3-dihydro-1H-inden-2-yl)methanol prepared by diluting with ethanol was applied to the forearms of three volunteers. As shown in Table 15, (2,5-dimethyl-2,3-dihydro-1H-inden-2-yl)methanol was able to reduce the number of mosquitoes landing on the volunteers' arms by more than 90% for up to 7 hours.
[0196] [Table 8]
[0197] 5. In vitro hot plate assay with two arthropod control compounds 5.1 In vitro hot plate assay with single compounds The mean ± SD numbers of Ixodes ricinus mites walking down (affected), falling over (affected) or walking up (unaffected) from hot plates treated with ethanol, N,N-diethyl-3-methylbenzamide diluted to 0.4 and 10 mg / mL in ethanol (component 2) or (2,4,6-trimethyl-2,3-dihydro-1H-inden-2-yl)methanol (component 1) were assessed for n groups of 12 mites (n = 2–5).
[0198] [Table 9]
[0199] When the hot plate was treated with the solvent ethanol, only 6.6% ± 3.5% of the mites were affected. An average of 0.8 mites across 12 mites tested per group did not show the natural behavior of exploring the feeding spot (Table 5 and Figure 11).
[0200] On the other hand, the two tested ingredients and DEET induced a repellent effect at the two concentrations tested (0.04% and 1%; Table 5 and Figure 11).
[0201] (2,5-Dimethyl-2,3-dihydro-1H-inden-2-yl)methanol was at least effective at 0.04%, but only one tick out of 12 was unaffected at the 1% concentration in both replicates, reaching results similar to those obtained with DEET applied at a similar concentration (Table 5 and Figure 11).
[0202] (2,4,6-trimethyl-2,3-dihydro-1H-inden-2-yl)methanol was the most effective, even more so than DEET, at both concentrations tested, with all ticks affected at the high concentration (1%) and only 3 of 12 and 5 ticks unaffected at the lowest concentration tested (0.04%) (Table 5 and Figure 11).
[0203] Further compounds were evaluated using the hot plate assay for the tick I. ricinus applying the protocol mentioned above (Tables 9, 10 and 11).
[0204] [Table 10]
[0205] In all tests, a clear tendency can be observed for the mite I. ricinus to be affected in relation to increasing amounts of irritant applied to the hot plate (Table 9). This demonstrates a clear biological dose-response effect of the irritant. At the highest concentrations, all compounds were effective in repelling the mites, with threshold values greater than 0.447% for 4,4A,5,9B-tetrahydro-indeno[1,2-D]-1,3-dioxin and 2,4-dimethyl-4,4a,5,9b-tetrahydroindeno[1,2-d][1,3]dioxin and greater than 0.2% for (2,5-dimethyl-2,3-dihydro-1H-inden-2-yl)methanol (Table 9).
[0206] [Table 11]
[0207] It can be observed that in all tests there is a clear tendency for more mites I. ricinus to be affected in relation to increasing amounts of irritant applied to the hot plate (Table 10). This demonstrates a clear biological dose-response effect of the irritant. At the highest concentration tested (1%), all eleven compounds affected more than 85% of the mites (Table 10). (5-Methyl-2,3-dihydro-1H-inden-2-yl)methanol, 2,3-dihydro-1H-inden-2-ylmethanol and 5-isopropyl-2-methyl-2-indanethanol reached this level of efficacy at an even lower dose of 0.2% (Table 10).
[0208] [Table 12]
[0209] It can be observed that in all tests there is a clear tendency for more I. ricinus mites to be affected in relation to increasing amounts of irritant applied to the hot plate (Table 11). This demonstrates a clear biological dose-response effect of the irritant. All 11 compounds affected more than 75% of the mites at a concentration of 1% (Table 11).
[0210] 5.2. In vitro hot plate assay for blends 5.2.1. Testing at various concentrations In this in vitro assay, the % of mites Ixodes ricinus affected by irritants loaded onto a hot plate was measured to assess repellency. Twelve mites were evaluated per concentration ranging from 0.0016% to 1% in ethanol.
[0211] It can be observed that in all tests there is a clear tendency for more I. ricinus mites to be affected in relation to increasing amounts of irritant applied to the hot plate (Tables 12 and 13), demonstrating a clear biological dose-response effect of the irritant.
[0212] [Table 13]
[0213] All blends and fragrances at concentrations above 0.45% induced a significant repellent effect, as they were able to increase the number of affected mites above the 85% threshold (Table 12), which was reached even at lower concentrations for Blend #1 (0.06%) and Blend #2 (0.2%; Table 12).
[0214] Moreover, at low concentrations, Blends #2 and #3 showed a significantly higher reduction in landings compared to the single compounds that constituted the blends, for example at a dose of 0.89%, with 83% of the mites affected by Blend #2, compared to 29% and 46% for (2,5-dimethyl-2,3-dihydro-1H-inden-2-yl)methanol and 3-[Nn-butyl-N-acetyl]aminopropionic acid ethyl ester, respectively, at the same dose; similarly, at a dose of 0.598%, 43% of the mites were affected by Blend #3, compared to 6% and 19% for 4,4A,5,9B-tetrahydro-indeno[1,2-D]-1,3-dioxin and 2-(2-hydroxypropan-2-yl)-5-methylcyclohexan-1-ol, respectively, at the same dose (Table 12).
[0215] [Table 14]
[0216] All three fragrances significantly affected the mites when applied to the hot plate at a dose of 1% (Table 13).Fragrances 1a and 1e affected all 12 mites tested at this dose of 1% (Table 13).
[0217] 5.2.2 Time-course testing Using the same in-vitro setup, the effects of different irritants diluted to 20% (w / w) in ethanol on tick behavior (n≧12) were assessed over time.
[0218] [Table 15]
[0219] As shown in Table 14, the relative effectiveness of Blend #2 over time can be attributed primarily to (2,5-dimethyl-2,3-dihydro-1H-inden-2-yl)methanol, which in this assay exhibited better mite repellency than the known repellent 3-[Nn-butyl-N-acetyl]aminopropionic acid ethyl ester. However, a synergistic effect appears to have occurred at 3 and 4 hours, as the blend was more effective than the two single compounds during these time points (Table 14).
[0220] Similarly, the good activity over time in Blend #3 can be attributed to the dual and synergistic action of 4,4A,5,9B-tetrahydro-indeno[1,2-D]-1,3-dioxin and 2-(2-hydroxypropan-2-yl)-5-methylcyclohexan-1-ol (Table 14). Indeed, 4,4A,5,9B-tetrahydro-indeno[1,2-D]-1,3-dioxin was very effective at the first time points (0.25 hours and 0.5 hours). Meanwhile, 2-(2-hydroxypropan-2-yl)-5-methylcyclohexan-1-ol was not effective at these first time points, but became more effective thereafter, thus extending the effectiveness of Blend #3 (Table 14).
[0221] 1. Novel Compounds of the Present Invention 1.1. 2-(hydroxymethyl)-2,6-dimethyl-2,3-dihydro-1H-inden-1-ol / (1RS,2SR)-2-(hydroxymethyl)-2,6-dimethyl-2,3-dihydro-1H-inden-1-ol MS(EI):192(M + ) 1.2. 4a,8-Dimethyl-indan[1,2-d]-1,3-dioxan-2-one MS(EI):218(M + ) 1.3. (2,5-Dimethyl-2-indanyl)methyl formate MS(EI):204(M + ) 1.4. 8-Methyl-4,4a,5-9b-tetrahydroindeno[1,2-d][1,3]dioxine / (4aRS,9bSR)-8-Methyl-4,4a,5,9b-tetrahydroindeno[1,2-d][1,3]dioxine MS(EI):190(M + ) 1.5. 5-(tert-butyl)-1,3-dihydrospiro[indene-2,3'-oxetane] MS(EI):216(M + ) Preparation of the compounds disclosed herein A three-neck flask was charged with LiAlH4 (6.19 g, 163 mmol, 1.0 equiv.) under N2, followed by 200 mL of anhydrous Et2O. A solution of 2-(hydroxymethyl)-2,6-dimethyl-2,3-dihydro-1H-inden-1-one (31 g, 163 mmol, 1.0 equiv.) in 150 mL of anhydrous Et2O was added to the mixture over 2 h. The resulting grey suspension was left stirring at room temperature for 3 h.
[0222] The reaction mixture was cooled to 0° C. and 5 mL of H2O, 5 mL of NaOH, and 15 mL of H2O were added in that order.
[0223] The suspension was stirred at room temperature and then treated with MgSO4. After stirring for 30 min, the white solid was filtered off through Celite and washed with Et2O. The volatiles were removed under reduced pressure to give the crude desired alcohol as a viscous pale yellow oil.
[0224] The residue was purified by Kugelrohr distillation to give the pure alcohol 2-(hydroxymethyl)-2,5-dimethyl-2,3-dihydro-1H-inden-1-one as a colorless oil (31.3 g, 98% yield). [Table 16]
[0225] [(4aR,9bR)-4a,7-dimethyl-4,4a,5,9b-tetrahydroindeno[1,2-d][1,3]dioxin-2-one] 1,1'-Carbonyldiimidazole (4.5 g, 28 mmol, 1.5 equiv.) and N,N-dimethylpyridin-4-amine (0.57 g, 5 mmol, 0.25 equiv.) were dissolved in 88 mL of dichloromethane. The resulting solution was added dropwise over 30 min to a solution of 2-(hydroxymethyl)-2,5-dimethyl-2,3-dihydro-1H-inden-1-one in 88 mL of dichloromethane.
