[0004] Intravenous therapy (IV) or drug delivery delivers fluids directly into a vein. The intravenous route of administration can be used for both injections using a syringe at higher pressure, as well as infusions that typically use only gravity-fed pressure (also called drip). The intravenous route delivers drugs rapidly throughout the body by introducing the therapeutic agent directly into the circulation. This introduction of the drug formulation directly into the bloodstream makes 100% of the formulation bioavailable, but it also means that the formulation must immediately match the subject's physiology, limiting the acceptable ingredients within the formulation. Injectable products can be sterile, pyrogen-free, free of particulate matter when in solution, and isotonic. In addition, the ingredients must be able to withstand terminal sterilization or aseptic manufacturing processes.
[0005] Provided herein are stable, pharma- ceutically acceptable formulations for intravenous injection of tagraxofusp, as well as formulations and methods for making same. Summary of the Invention

[0158] [Figure 1B] 1 provides DSC data from the evaluation of stabilizing excipients detailed in Example 2. DETAILED DESCRIPTION OF THE PREFERRED EMBODIMENTS
[0159] I. Definition As used herein, the terms "administer" and "administered" refer to the delivery of a composition to a subject by a method or route that results in at least partial localization of the composition at a desired site. The composition may be administered by any suitable route that results in effective treatment in the subject, i.e., administration results in delivery to a desired location in the subject, and at least a portion of the composition is delivered to the desired site for a period of time. Modes of administration include injection, infusion, infusion, or ingestion. "Injection" includes, but is not limited to, intravenous, intramuscular, intraarterial, intrathecal, intraventricular, intracapsular, intraorbital, intracardiac, intradermal, intraperitoneal, transtracheal, subcutaneous, subcuticular, intraarticular, subcapsular, subarachnoid, intraspinal, intracerebrospinal, and intrasternal injection and infusion. In some examples, the route is intravenous.
[0160] As used herein, the term "agent" refers to any molecule, compound, and / or substance for use in the prevention, treatment, management and / or diagnosis of disease, including tagraxofusp as disclosed herein.
[0161] As used herein, the term "buffer" or "buffering agent" refers to one or more components that, when added to an aqueous solution, when an acid or alkali is added, or when diluted with a solvent, can protect the solution from pH fluctuations. In addition to phosphate buffers, glycine buffers, carbonate buffers, citrate buffers, etc. can be used, in which sodium, potassium, or ammonium ions can function as counterions.
[0162] As used herein, the term "bulking agent" refers to one or more components that form the majority of the lyophilized product and provide the cake with the appropriate structure. They are generally used for low dose (high potency) drugs that do not have the bulk necessary to support their own structure.
[0163] As used herein, the term "carrier" refers to a diluent, adjuvant, or excipient with which the compositions disclosed herein are administered. In some embodiments, the carrier is sterile. Water can be a carrier. Saline solutions and aqueous dextrose and glycerol solutions can also be used as liquid carriers.
[0164] As used herein, the term "charge variant profile" refers to the relative amounts of various species of a protein that differ in net charge. Proteins are large molecules that contain numerous chemical entities that are subject to various post-translational enzymatic and chemical modifications that can change the overall charge of the molecule. In addition, chemical modifications such as oxidation or deamination can occur during the manufacturing process and storage. The charge state can affect the structure, stability, binding affinity, efficacy, and safety of a biotherapeutic. Analysis of charge variants is usually a regulatory requirement for biotherapeutic proteins and can be accomplished using isoelectric focusing or ion exchange chromatography. Changes in the relative amounts of various charge variants due to process inconsistencies and / or stability-related events can adversely affect the safety and efficacy of a biotherapeutic protein.
[0165] As used herein, the term "excipient" refers to a substance used to formulate a therapeutic agent into a pharmaceutical preparation and can be any solid, liquid, or semi-solid additive such as diluents, solubilizers, stabilizers, adjuvants, thickeners, lubricants, bulking agents, buffers, enhancers, antibacterial agents, wetting agents, and surfactants.
[0166] The term "effective amount" or "sufficient amount" as used herein refers to an amount of a therapeutic composition or therapy sufficient to cause prevention of the onset, recurrence, or onset of a disease or disorder and one or more symptoms thereof, reduce the severity or duration of a disease or disorder, alleviate one or more symptoms of a disease or disorder, prevent the progression of a disease or disorder, cause regression of a disease or disorder, and / or enhance or improve the therapeutic effect of another therapy. In embodiments, an "effective amount" or "therapeutically effective amount" refers to an amount of a composition sufficient to cause a beneficial or desired outcome. A therapeutically effective amount may vary depending on one or more of the subject and disease condition being treated, the weight and age of the subject, the severity of the disease condition, the method of administration, and the like, which can be readily determined by one of skill in the art. In certain embodiments, a "sufficient amount" of a therapeutic composition refers to the amount of the composition necessary to prevent, reduce, or alleviate at least one or more signs or symptoms of a disease. Thus, the term "effective amount" refers to an amount of a composition that is sufficient to promote a particular effect when administered to a typical subject (such as a subject having or at risk for cancer or, for example, an autoimmune disease). An effective amount would also include an amount sufficient to delay the onset of, alter the course of, or reverse a disease symptom (such as, but not limited to, slowing the progression of a disease symptom). It will be understood that for any given case, an appropriate "effective amount" may be determined by one of ordinary skill in the art using routine experimentation.
[0167] As used herein, the term "isotonic" refers to a formulation that has essentially the same osmotic pressure as human blood. Isotonic formulations generally have an osmotic pressure of 250-350 mOsmol / KgH2O. Isotonicity can be measured, for example, using a vapor pressure or ice osmometer.
[0168] As used herein, the terms "lyophilization" and "lyophilized" refer to a process in which the material to be dried is first frozen and then the ice or frozen solvent is removed by sublimation in a vacuum environment.
[0169] As used herein, the term "lyophilized solution" refers to a formulation that undergoes the lyophilization process.
[0170] As used herein, the term "lyophile" refers to any solid material obtained by lyophilization of a lyophilization solution (i.e., freeze-drying of an aqueous solution).
[0171] As used herein, the term "lyoprotectant" refers to a molecule that, when combined with a protein of interest, prevents or reduces chemical and / or physical instability of the protein before or during lyophilization or freeze-drying, and during subsequent storage. Lyoprotection is defined as the stabilization of a therapeutic agent and prevention of its degradation both during and after lyophilization.
[0172] As used herein, in the context of administering a therapy to a subject, the terms "manage," "managing," and "management" do not result in a cure of the disease or disorder, but refer to the beneficial effects that a subject derives from a therapy (e.g., a prophylactic or therapeutic composition) or combination of therapies. In certain embodiments, a subject is administered one or more therapies (e.g., one or more prophylactic or therapeutic compositions) to "manage" a disease, to prevent the progression or worsening of the condition.
[0173] As used herein, the term "peroxide" or "peroxides" refers to any of a class of chemical compounds in which two oxygen atoms are linked together by a single covalent bond. Organic peroxides are compounds with the bonds COOC or COOH. An example is tert-butyl hydroperoxide. Peroxides are known to readily decompose into highly reactive free radical-containing moieties (CO· or HO·) that can cause further degradation or destabilization of pharmaceutical formulations.
[0174] As used herein, the term "pharmacologically acceptable" refers to compounds, materials, compositions, formulations, and / or dosage forms that are suitable for use in contact with the tissues of a subject, human, or animal without undue toxicity, irritation, allergic response, or other problem or complication, within the scope of sound medical judgment, commensurate with a reasonable benefit / risk ratio.
[0175] As used herein, in the context of administration of a therapy to a subject, the terms "prevent", "preventing", and "prevention" refer to the prevention or inhibition of recurrence, onset, and / or development of a disease or disorder or a symptom thereof in a subject resulting from administration of a therapy (e.g., a prophylactic or therapeutic agent) or combination of therapies (e.g., a combination of prophylactic or therapeutic agents). In certain embodiments, such terms refer to one, two, three, or more of the following outcomes following administration of one or more therapies: (1) delaying the onset of disease symptoms, (2) altering the course of disease symptoms (e.g., but not limited to, delaying the progression of disease symptoms), (3) reversing disease symptoms, (4) reducing the rate of disease recurrence, (5) extending the time to disease recurrence, (6) increasing the disease-free, recurrence-free, progression-free, and / or overall survival of a patient, and (7) improving disease-related symptoms and / or quality of life. In certain embodiments, such terms refer to a reduction in mortality and / or an increase in survival rate of a patient population. In further embodiments, such terms refer to an increase or improvement in the quality of life of a patient population. In certain embodiments, such terms refer to a reduction in the rate of hospitalization of a patient population and / or a reduction in the length of hospitalization of a patient population.
[0176] As used herein, the term "reconstituted" or "reconstituted formulation" refers to a formulation prepared by dissolving a lyophilisate in an aqueous carrier. In some embodiments, the reconstituted formulation is suitable for intravenous injection (IV) in a patient in need thereof.
[0177] As used herein, the term "stable" refers to a formulation or composition in which the therapeutic agent contained therein essentially retains its physical and chemical stability and integrity during processing and storage. The stability of a formulation can be measured at a selected temperature after a selected period of time. For example, the increase in particles, the formation of aggregates, or the presence of other impurities after lyophilization and storage are indicators of instability of a lyophilized formulation. In addition to the formation of aggregates, the retention of original clarity, color, and odor throughout the shelf life are indicators that can be used to monitor a stable solution.
[0178] As used herein, the terms "subject" and "patient" are used interchangeably and refer to a human subject, although it is to be understood that the methods described herein are effective with respect to all vertebrate species intended to be encompassed by the term "subject." [0220] III. Impurities Although several trace impurities are present in tagraxofusp or tagraxofusp formulations, one impurity is associated with a single oxidation of the drug. By mass spectrometry, the intact mass of the oxidized impurity is presented as a single peak at +16 Da from tagraxofusp. Although the impurity is a single peak, peptide mapping shows that it represents several species, each with a single oxidation at one of several sites on tagraxofusp. [0254] Tagraxofusp is a fusion protein produced by fermentation in E. coli constructs genetically engineered to produce the target protein in inclusion bodies. The tagraxofusp protein is a CD123-directed cytotoxin composed of a fusion protein of human IL-3 and a truncated diphtheria toxin (DT) that targets CD123-expressing cells. It has an N-terminal methionine, and the DT portion contains the first 388 amino acids of diphtheria toxin (DT), including the catalytic and translocation domains. The amino acid sequence of tagraxofusp is as follows: (SEQ ID NO: 1).
[0255] Example 1: Baseline Aqueous Formulation The baseline aqueous formulation of tagraxofusp for injection was not a lyophilized formulation, and this formulation did not exhibit the desired shelf life at the preferred storage temperatures, as illustrated in this example.
[0256] Tagraxofusp for Injection, 1 mg / mL (1 mL / vial), was provided as a liquid drug product in a clear, colorless 2 cc glass vial. The composition of the drug product solution was 1.0 mg / mL tagraxofusp in 75 mM NaCl (USP, EP), 5% (w / w) sorbitol (USP / NF, EP), and 20 mM Tris buffer (USP, EP), pH 7.5. The formulation was intended to be stable under refrigerated storage conditions (5±3° C.), however, during generation of long-term stability data, it was found that storage at −20° C. was beneficial in achieving the desired shelf life. Stability studies, including evaluation by various analytical techniques such as reversed-phase ultra-high performance chromatography (RP-UPLC), size-exclusion high performance chromatography (SEC-HPLC), sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE), AEX-UPLC, and potency by cytotoxicity bioassay, showed that the product began to fall outside the desired acceptance criteria within 12-18 months at 5±3°C, but met all acceptance criteria up to 36 months of testing without any significant decrease in purity or potency when stored at -20°C.
[0257] A lyophilized formulation of Tagraxofusp was developed to obtain the desired shelf life at the preferred storage temperature. Examples 2-5 below provide some of the experimental processes and procedures used to arrive at the final improved formulation for lyophilization, an example of which is shown in Example 6.
[0258] Example 2: Evaluation of Surfactants for Use in Improved Formulations for Lyophilization To address the tendency of proteins to form particulates, especially at the lower protein concentrations used for dose delivery, a suitable surfactant is added to the formulation as a precaution.
[0259] Table 1 shows the results of a study in which lyophilized tagraxofusp was added to a 50 cc saline infusion bag and stirred with various amounts of surfactant for 6 hours.
[0260] The freeze-drying step was carried out as follows: the product-filled vials were semi-stoppered with elastomeric stoppers and loaded onto the shelves of the freeze-drying chamber, which were maintained at 10° C. for pre-cooling. In the step designated as heat treatment or annealing, the shelf temperature was ramped down from 10° C. to −40° C. in 3 hours, held at −40° C. for 1 hour, ramped up to −10° C. in 1 hour, held at −10° C. for 1 hour, ramped down to −40° C. in 1 hour, and held at −40° C. for an additional hour. Following the procedure above, the condenser was cooled to below −60° C. and the vacuum pump was primed. The chamber pressure was then reduced to 0.133 mBar to initiate sublimation of the ice (also known as the primary drying step). In the primary drying stage, the shelf temperature was ramped up from −40° C. to −25° C. in 1 hour and held at this temperature for 40 hours. The shelf temperature was then ramped up to +25° C. in 14 hours. After this, the shelf temperature was maintained for an additional 23.3 hours in the secondary drying step to complete the drying cycle. Finally, the chamber was evacuated with nitrogen to an atmosphere of 900 mBar, and the vials were then fully stoppered under nitrogen and removed from the chamber.
[0261] The results showed that a small amount of surfactant was sufficient to prevent loss of product due to aggregation (as seen from recovery studies using SEC-HPLC) as well as the formation of visible particles. It was found that while both Polysorbate 80 and Poloxamer 188 were able to reduce particle formation, Polysorbate 80 was able to achieve this at a lower concentration than that of Poloxamer 188. Based on these results, Polysorbate 80 was selected as the surfactant and was included in all subsequent development studies. [Table 1]
[0262] Studies have shown a correlation between the amount of peroxide in the surfactant and the amount of impurity identified as an oxidized species of Tagraxofusp measured in the drug product upon release and during stability studies. Higher levels of peroxide in the surfactant (e.g., peroxide value >2 meqO2 / kg) result in higher levels of this impurity in the final drug product (e.g., >1%). The peroxide content of the surfactant was assessed using potentiometric titration according to the peroxide values of the European Pharmacopoeia (EP) 2.5.5.
[0263] Quantification of oxidized species of tagraxofusp by RP-UPLC The purity of Tagraxofusp liquid drug product (DP) was determined using reversed phase ultra-performance liquid chromatography (RP-UPLC). The method was performed on a reversed phase ultra-performance liquid chromatography system (RP-UPLC) using an Agilent Zorbax 300SB-C3, 2.1×100 mm, 1.8 μm column. The column was eluted using a discontinuous gradient of water and acetonitrile with 0.1% TFA. Detection was performed at A280. The peak areas (%) of the main peak and major impurities relative to the total peak area are reported.
[0264] Equipment: A liquid chromatography system capable of handling 1.8 μm particle size reversed-phase columns equipped with an ultraviolet (UV) detector, a degassing module, a thermostatically controlled column compartment, and a refrigerated autosampler (e.g., a Waters Acquity H class UPLC with a 250 μL mixer and a 50 μL injection loop).
[0265] Solutions: Mobile phase A: purified water containing 0.1% trifluoroacetic acid (TFA), Mobile phase B: acetonitrile (ACN) containing 0.1% TFA.
[0266] Reference Standards and Sample Preparation: Reference substances were tested undiluted. The injection volume was calculated to deliver a target amount of 4.0 μg. For reference substances, the target load was 4.0 μg (2.7 μL of 1.5 mg / mL reference substance injected). The target load for drug substance was 4.0 μg (4.0 ul of 1 mg / mL DP injected).
[0267] Equipment setup: analysis-specific conditions (injection loop size: 50 μL) [Table 2]
[0268] Sample Setup: Run sequence for sample analysis [Table 3] *Sample volume determined by calculating a sample injection load of 4.0 μg (4 μL for DP of 1.0 mg / mL).
[0269] Before sample analysis, a blank injection was performed followed by five injections of a reference standard for system suitability. After nine sample injections, a bracket injection of a reference material (system suitability) was performed after the final sample injection. Each sample was injected in triplicate.
[0270] Peak Integration: The identity of the peaks in the samples was confirmed by comparison with the most recently injected reference material. The main peak retention time, impurity 1 RRT, and impurity 2 RRT of the tagraxofusp reference material were within the ranges specified under system suitability.
[0271] System Suitability. The chromatograms of the reference material were visually comparable to the exemplary chromatograms. The bracketed reference material injections had main peak purity within ±2.0% of the average purity (%) of the first five system suitability injections of the reference material. The main peak purity CV of the system suitability injections of the reference material was 2% or less. The USP theoretical plate criteria of the main peak of each system suitability injection was 11000 or greater. The USP tailing of the system suitability injections of the reference material was 2.2 or less. The main peak of the reference material varied between 13.9 and 16.0 minutes. The RRT of impurity 1 in the reference material was 0.91-0.92. The RRT of impurity 2 in the reference material was 0.93-0.95.
[0272] Assay acceptance criteria: All samples were run before and after bracketing standards. For triplicate injections of samples, the purity column volume (CV) (%) was ≤ 2% (n=3) for the purity of the main peak. For triplicate injections of samples, the CV was ≤ 2% (n=3) for the main peak retention time.
[0273] Example 3: Evaluation of stabilizers for use in improved formulations for lyophilization The thermal stability of Tagraxofusp bulk drug substance (BDS) was evaluated in the presence of various generally recognized as safe (GRAS) stabilizers by differential scanning calorimetry (DSC) studies and among the excipients evaluated, sucrose was found to provide the greatest increase in melting temperature (Tm). Table 2 and Figure 1 present these results. A high Tm value indicates that the excipient provides protection to the Tagraxofusp molecule by increasing the denaturation temperature, which is an indication that the excipient better stabilizes the molecule. As seen in Table 2, sucrose and mannitol rank highest among the excipients evaluated. Sucrose can also act as a cryoprotectant that protects against denaturation during the lyophilization process, both in the freezing and drying stages of the lyophilization cycle. Furthermore, mannitol can also act as a caking agent, providing structure to the cake and aiding in the removal of moisture during the sublimation and secondary drying processes. These tests were performed on non-lyophilized samples. [Table 4]
[0274] Example 4: Evaluation of buffers for improved formulations for lyophilization Four different buffers suitable for adjustment to the desired pH of 7.5 were evaluated in short-term stability studies of lyophilized Tagraxofusp samples. All formulations were prepared by diafiltration of bulk drug substance (including Tris buffer) into 10 mM buffer (either phosphate, arginine, histidine, or Tris buffer at pH 7.5). Sucrose (10% w / v) and polysorbate 80 (0.25% w / v) were added to these solutions. The solutions were filtered through a 0.22 μM low protein binding filter. 1.1 mL of the filtered solution was added to a clean 3 cc glass vial. The solution-filled vials were semi-stoppered with coated stoppers and placed in the lyophilizer chamber (model: Epsilon 2-6D, Ima Life, Italy). The lyophilization cycle used was as follows: shelf cooling to -40°C for 3 hours, followed by heat treatment at -15°C for 1 hour. The shelves were then cooled back to -45°C and the vacuum was started. Once the pressure was below 0.133 mBar, primary drying was carried out for approximately 16 hours at a shelf temperature of -30°C. The shelf temperature was then increased to 25°C over 14 hours and maintained at 25°C for an additional 12 hours during secondary drying. At the end of the cycle, the chamber was vented with dry air and the vials were stoppered in the chamber. The stoppered vials were removed and crimped with aluminum crimps.
[0275] Cake quality was assessed by appearance and reconstituted solutions were tested by SEC-HPLC for clarity and area (%) of tagraxofusp monomer. Tests were performed immediately after lyophilization and after 4 weeks of storage at 50°C. Table 3 summarizes the results. It shows that each buffer performed similarly. Use of Tris buffer can simplify the manufacturing process since it is also the buffer for the bulk drug substance. Therefore, it was selected as the buffer for subsequent testing. [Table 5] PT = phosphate buffer containing polysorbate 80, AT = arginine buffer containing polysorbate 80 HT = histidine buffer with polysorbate 80, TT = Tris buffer with polysorbate 80
[0276] Example 5: Evaluation of bulking agents for improved formulations for lyophilization Initial freeze-drying trials conducted in a laboratory-scale freeze-dryer utilizing sucrose (10% w / v) as a bulking agent / stabilizer yielded good cakes and demonstrated acceptable stability of freeze-dried tagraxofusp. However, in scale-up trials in larger freeze-dryer units, it was observed that the cake appearance was not consistently acceptable, with failures due to issues such as shrinkage. To address this, additional freeze-drying experiments were conducted in the laboratory where mannitol, another excipient commonly used in injectable products, was used as a bulking agent along with sucrose. The purpose of the mannitol was for the sucrose to act as an amorphous freeze / lyoprotectant during the freeze-drying process to protect the protein from the stresses of freezing and / or dehydration while providing a stiffer cake structure that ensured the cake would not shrink.
[0277] After modifying the formulation to include mannitol as a bulking agent, additional lyophilization experiments were performed to identify the amount of surfactant required to ensure control of fine particles in the new mannitol / sucrose matrix. Dried samples were evaluated for cake appearance, reconstitution time, pH, appearance after reconstitution, percent monomer by RP-HPLC, and residual water by Karl Fischer analysis. The results of these studies are summarized in Table 4 and concluded that polysorbate 80 surfactant at a level of 0.25% w / v remained adequate for acceptable stability of tagraxofusp lyophilized drug product for up to 11 months at room temperature storage. [Table 6] * After storage at room temperature for approximately 11 months.
[0278] Example 6: Final composition of the solution for lyophilization: Based on the above results and confirmation results from the formulation development run, a formulation of Tagraxofusp solution (1 mg / mL) for lyophilization was recommended for the proposed clinical trial as shown in Table 5. The formulation was consistently scaled for all materials when presenting at 1.5 mg. The table lists the composition of the solution for lyophilization as well as the rationale for the addition of each component. [Table 7] *This amount does not include 4% overfill by weight **Removed during freeze-drying
[0279] Physicochemical and biological properties: The pH of the drug product was set at 7.5, close to the physiological pH of 7.4, and consistent with the pH of both the drug product (DS) and conventional liquid drug formulations. This pH provides security against the known physical instability / loss of activity (potency) of DS at pH below 6.5. When the lyophilisate is reconstituted with Water for Injection (WFI), a clear, colourless liquid containing 1 mg / mL of Tagraxofusp is delivered, essentially free of particulates. The presence of low levels of surfactant prevents protein aggregation, a known attribute of DS, at lower dilutions such as those used for infusion.
[0280] The following is an example of a procedure for making one of the formulations disclosed herein.
[0281] Example 7: Preparation of injectable tagraxofusp The manufacturing process for Tagraxofusp for injection (1 mg / vial) consists of the following standard unit operations: compounding the solution for lyophilization, filtration through a grade 0.22 μm sterile filter, filling of the filtered sterile solution into glass vials, and lyophilization of the filled vials to obtain the final drug product cake.
[0282] Formula: The formulation of the lyophilized product was established as outlined herein above. In-process specifications were established to verify the pH, density, and Tagraxofusp concentration of the formulated drug product solution prior to proceeding to the sterilization procedure.
[0283] Sterilized through a 0.22 μm membrane filter: The bulk solution of Tagraxofusp for injection, like all protein therapeutics, cannot be terminally sterilized due to the heat sensitivity of the protein molecule. Thus, a standard sterile filtration operation was designed to perform sterilization of the formulated bulk solution by membrane filtration through one 0.45 μm pre-filter sterile filter and then through two 0.22 μm hydrophilic polyvinylidene fluoride (PVDF) membranes contained in a polycarbonate housing. The formulated bulk is passed through two sterile membranes in series, as is typical in sterile filtration operations, to provide redundant sterilization capacity.
[0284] Aseptic filling of sterile solutions: The vial and stopper combination selected for Tagraxofusp for injection was previously qualified on the contract manufacturer's production line. For each vial, 1.04 mL (1.06 grams) or 1.56 mL (1.59 grams) of membrane filtered sterile solution of Tagraxofusp is filled into a Type I pyrogen-free glass vial and the vial is semi-stoppered with a sterile rubber stopper. The filled vial is then transferred to a shelf in the lyophilizer chamber for lyophilization.
[0285] Overfill: The clinical drug product has a labeling of either 1 mg / vial and a target fill of 1.06 gm (1.04 mL) per vial or 1.5 mg / vial and a target fill of 1.59 gm (1.56 mL) per vial. Each vial contains a 4% overfill of tagraxofusp to allow for cake displacement during reconstitution and to ensure that the product has a concentration of exactly 1.0 mg / mL after reconstitution with 1.0 mL / 1.5 mL WFI during dose preparation. This overfill allows for accurate dose preparation on a dose / kg basis.
[0286] Freeze-drying cycle development: The main excipients of the injectable Tagraxofusp solution, sucrose and mannitol, are present at 25 mg / mL each and therefore in considerably larger amounts compared to the active ingredient, which is present at 1 mg / mL. The thermal properties of the frozen solution are therefore mainly determined by the properties of the frozen state of these two components. The freezing behavior of the Tagraxofusp solution for lyophilization by differential scanning calorimetry (DSC) showed minor endothermic events even at about 32°C. The lyophilization parameters during primary drying were selected such that the temperature of the product remained below -32°C during the sublimation stage of drying. A heat treatment step during freezing at -15°C for about 1 hour was included to ensure complete crystallization of mannitol and other metastable phases. Once the primary (sublimation) drying is complete, the shelf temperature is increased to 25°C and maintained at 25°C for the remainder of the drying cycle to ensure that the secondary (desorption) drying stage is completed and the dried product is obtained with low residual water levels.
[0287] After several experimental trials using various combinations of shelf temperature, chamber pressure, and duration of each stage, a freeze-drying cycle was finally determined that consistently yielded a well-formed cake with low residual water.
[0288] Table 6 summarizes the finalized process parameters used for the lyophilization of Tagraxofusp for injection. In addition to the cycle parameters listed above, additional specified loading times and extended freezing times were included in the final cycle as required for standard operation of the production scale equipment. Upon completion of the lyophilization process, the dried vials are fully stoppered in the lyophilization chamber, unloaded, sealed with aluminum crimps, and then rinsed. The crimped vials are then subjected to 100% visual inspection, quality testing, labeling, and packaging. [Table 8]
[0289] Density of tagraxofusp bulk solution for freeze-drying: The density of the tagraxofusp bulk solution for lyophilization was determined using an Anton-Paar densitometer at room temperature and found to be 1.02 g / mL.
[0290] Determination of the displacement volume of lyophilisates The displacement volume of the lyophilisate is the volume of reconstituted solution in milliliters displaced by 1.0 g or 1.5 g of lyophilized dry material. The displacement volume is required to determine the overfill required to achieve a 1 mg / mL solution when reconstituted with exactly 1.0 mL or 1.5 mL of WFI. The displacement volume of lyophilized tagraxofusp for injection was determined by placing 1000 mg of lyophilized material in a 10 mL volumetric flask and then adding 10.0 mL of water. The volume of solution over 10.0 mL caused by the displacement due to the solid content was then measured. It was found that when 1 gm of lyophilized material was dissolved, approximately 0.73 mL of water was displaced.
[0291] Using this relationship, the displacement value of the lyophilized cake was calculated as follows: total weight of active pharmaceutical ingredient (API) + excipients added per mL of drug product (DP) solution is 55.94 mg. Thus, for a fill volume of 1.0 mL at 1.0 mg / mL tagraxofusp, the displacement value of the dry material is 0.04 mL, resulting in a total volume of 1.04 mL or 0.96 mg / mL active substance. To account for this discrepancy caused by the displacement volume, approximately 1.04 mL or 1.56 mL of solution must be filled and lyophilized for the 1.0 mg / vial fill and 1.5 mg / vial fill, respectively. The resulting dry product then contains sufficient tagraxofusp active substance to deliver the target concentration in the reconstituted solution.
