Description

[Technical field]

[0001] Background G protein-coupled receptors (GPCRs) constitute the largest family of membrane receptors in cells. They transduce extracellular signals to intracellular effector systems and are involved in a wide variety of physiological phenomena, making them the most common targets for pharmaceutical drugs, although only a small proportion of GPCRs are currently targeted by therapeutics. [Background technology]

[0002] GPCRs respond to a wide range of ligands. With advances in human genome sequencing, defined, physiologically relevant ligands are still missing for approximately 25% of the more than 400 identified GPCRs (not including olfactory GPCRs). These receptors are known as "orphan GPCRs". "Deorphanization" and identification of their in vivo roles is expected to reveal new regulatory mechanisms and, therefore, disclose new drug targets. Whether GPR17 is such an orphan receptor is still controversial. Phylogenetically, GPR17 is closely related to the nucleotide P2Y receptors and the cysteinyl leukotriene (CysLT1, CysLT2) receptors, with amino acid sequence identity of approximately 30 to approximately 35%, respectively.

[0003] Northern blot and RT-PCR analyses of multiple tissues show predominant expression of GPR17 in the central nervous system (CNS) (Ciana et al., 2006, EMBO J 25(19):4615; Blasius et al., 1998, J Neurochem 70(4):1357) and also in the heart and kidney, organs that typically undergo ischemic injury. Two human GPR17 isoforms have been identified that differ only in the length of their N-terminus. The short GPR17 isoform encodes a 339 amino acid residue protein with seven transmembrane motifs of the typical rhodopsin type. The long isoform encodes a receptor with a 28 amino acid longer N-terminus (Blasius et al., 1998). GPR17 is highly conserved among vertebrate species (approximately 90% identity of amino acid sequence to both mouse and rat orthologues).

[0004] In the original de-orphanization report, GPR17 was: 35Based on SGTPγS binding and cAMP inhibition assays, as well as single-cell calcium imaging, it was identified as a dual receptor for uracil nucleotides and the cysteinyl leukotrienes (cysLTs) LTC4 and LTD4, respectively (Ciana et al., 2006, ibid.). Evidence for GPR17 function was provided in a variety of cell backgrounds, including 1321N1, COS7, CHO, and HEK293 cells (Ciana et al., 2006, ibid.). An independent study subsequently confirmed activation of GPR17 by uracil nucleotides, but failed to reproduce activation by CysLTs (Benned-Jensen and Rosenkilde, 2010, Br J Pharmacol, 159(5):1092). However, recent independent reports (Maekawa et al., 2009, PNAS 106(28), 11685; Qi et al., 2013, J Pharmacol Ther 347,1, 38; Hennen et al., 2013, Sci Signal 6, 298) suggested a lack of GPR17 responsiveness to both uracil nucleotides and CysLT across different cell backgrounds (1321N1, CHO, HEK293 cells) stably expressing GPR17. A novel regulatory role for GPR17 has also been proposed, where GPR17, when co-expressed with the CysLT1 receptor, renders the CysLT1 receptor insensitive to its endogenous lipid mediators LTC4 and LTD4. Additional investigations are required to more deeply explore the pharmacology and function of GPR17.

[0005] Agents that modulate GPR17 activity may have neuroprotective, anti-inflammatory, and anti-ischemic effects and may therefore be useful for the treatment of cerebral, cardiac and renal ischemia, and stroke (WO2006 / 045476) and / or improving recovery from these events (Bonfanti et al, Cell Death and Disease, 2017, 8, e2871).

[0006] GPR17 modulators are also believed to be involved in food intake, insulin and leptin responses and are therefore claimed to have a role in the treatment of obesity (WO2011 / 113032).

[0007] Furthermore, there is strong evidence that GPR17 is involved in the myelination process and that negative GPR17 modulators (antagonists or inverse agonists) may be useful drugs to treat or alleviate myelination disorders such as multiple sclerosis and spinal cord injury (Chen et al, Nature Neuroscience 2009, 12(11):1398-1406; Ceruti et al; Brain: a Journal of Neurology 2009 132(Pt8):2206-18; Hennen et al, Sci Signal, 6, 2013, 298; Simon et al J Biol Chem 291, 2016, 705; Fumagalli et al, Neuropharmacology 104, 2016, 82). More recently, two groups showed that adult GPR17- / - knockout mice remyelination was faster than wild-type littermates after LPC induced demyelination in the spinal cord (Lu et al., Nature Scientific Reports, 2018, 8:4502) or in the corpus callosum (Ou et al., J. Neurosci., 2016, 36(41):10560). In contrast, activation of GPR17 has been shown to inhibit oligodendrocyte precursor cell (OPC) maturation and prevent effective myelination (Simon et al., supra). This again confirmed the potential key role of GPR17 in the remyelination process and as a promising drug target in demyelinating diseases. Thus, the identification of potent and selective GPR17 modulators has important implications for the treatment of myelination disorders.

[0008] Several serious myelination disorders are known to be caused by either impaired myelination, loss of myelin (usually called demyelination), and / or the body's inability to properly form myelin (sometimes called hypomyelination). Myelination disorders can be idiopathic or secondary to a specific triggering event, such as traumatic brain injury or viral infection. Myelination disorders can primarily affect the central nervous system (CNS), but can also involve the peripheral nervous system. Myelination disorders include, among others, multiple sclerosis, neuromyelitis optica (also known as Devic's disease), leukodystrophy, Guillain-Barre syndrome, and many other disorders described in more detail below (see, e.g., Love, J Clin Pathol, 59, 2006, 1151; see also Fumagalli et al., supra). Neurodegenerative diseases such as Alzheimer's disease, Huntington's disease, Parkinson's disease, amyotrophic lateral sclerosis (ALS), and multiple system atrophy (MSA) have also recently been strongly associated with reduced myelination (see, e.g., Ettle et al, Mol Neurobiol 53, 2016, 3046; Jellinger and Welling, Movement Disorders, 31, 2016;1767; Kang et al, Nature Neurosci 6, 2013, 571; Bartzokis, Neurochem Res (2007) 32:1655).

[0009] Multiple sclerosis (MS) is a chronic progressive disease. It is an inflammatory autoimmune disease that causes oligodendrocyte damage, demyelination, and ultimately axonal loss, thus resulting in a wide range of signs and symptoms of severe neurological disease, such as fatigue, dizziness, mobility and walking problems, difficulty speaking and swallowing, and pain. There are several forms of MS, and new symptoms either occur in isolated attacks (relapsing forms) or accumulate over time (progressive forms). While certain symptoms may disappear completely between isolated attacks, severe neurological problems often remain, especially as the disease progresses to more advanced forms. According to the Multiple Sclerosis Association of America, approximately 400,000 people in the United States and 2.5 million worldwide have been diagnosed with MS, with an estimated 10,000 new cases diagnosed in the United States each year. Multiple sclerosis is two to three times more common in women than in men.

[0010] There is no known causal cure or regimen for multiple sclerosis, or for many other myelinating disorders. Treatment is usually symptomatic, addressing the inflammatory component of the disease to improve function after attacks and prevent new attacks. Such immunomodulatory drugs are usually moderately effective, especially as the disease progresses, but they can have side effects and be poorly tolerated. Furthermore, most of the available drugs, such as beta-interferon, glatiramer acetate, or therapeutic antibodies, are only available in injectable form and / or address only the inflammatory component of the disease and not directly the demyelination. Other drugs, such as corticosteroids, have fairly non-specific anti-inflammatory and immunosuppressive effects and therefore can cause chronic side effects, such as those seen in Cushing's syndrome.

[0011] Therefore, there is a strong need for safe and effective drugs for the treatment of myelinating diseases such as MS, preferably drugs suitable for oral administration.Ideally, such drugs would reverse the demyelination process by reducing demyelination and / or promoting remyelination of affected neurons.Compounds that effectively reduce GPR17 receptor activity can meet these requirements.

[0012] However, few compounds that effectively modulate GPR17 activity are known.

[0013] WO2005 / 103291 suggests the endogenous molecules 5-aminolevulinic acid (5-ALA) and porphobilinogen (PBG) as activating ligands for GPR17, discloses the analgesic effects of GPR17 agonists, and proposes the use of GPR17 agonists to treat neuropathic pain and as tools in GPR17 screening assays. However, the reported affinities of 5-ALA and PBG are very low, and the amounts required for the assays are significant; that is, in the three-digit micromolar range for 5-ALA and even in the mM range for PBG, making both compounds poorly suited for routine screening assays or even for therapeutic use. Moreover, PBG is a chemically unstable reactive compound that degrades rapidly on exposure to air and light, making its routine handling impractical. Thus, these compounds do not provide a promising starting point for developing therapeutically effective negative GPR17 modulators.

[0014] Montelukast and pranlukast were originally developed as leukotriene receptor antagonists and were recently found to also act on the GPR17 receptor (Ciana et al., EMBO J. 2006, 25, 4615-4627). However, subsequent results of functional assays were contradictory, with montelukast (Hennen et al., 2013, ibid.), whereas pharmacological inhibition of GPR17 with pranlukast promoted differentiation of primary mouse (Hennen et al., 2013, ibid.) and rat (Ou et al., J. Neurosci. 36, 2016, 10560-10573) oligodendrocytes. Pranlukast phenocopies the effects of GPR17 inhibition in the lysolecithin model of focal demyelination, as both GPR17 knockout and pranlukast-treated wild-type mice show early onset of remyelination (Ou, supra). These results strongly support the hypothesis that GPR17 inhibitors offer a potential treatment for human demyelinating diseases.

[0015] However, the affinity of Monteclast and Planclast for GPR17 is only in the high micromolar range (Koese et al, ACS Med. Chem. Lett. 2014, 5, 326-330). Given the high protein binding and poor brain penetration of both compounds, it is unlikely that they will reach high enough free concentrations to bind to the GPR17 receptor in amounts suitable for human therapy. Furthermore, results obtained in vivo with these compounds are difficult to interpret due to their confounding high affinity for the CYSLT1 receptor. Cross-reactivity to other receptors further complicates their use to target GPR17.

[0016] US Patent No. 8,623,593 discloses certain indole-2-oxolinic acids as GPR17 agonists and their use in screening assays. However, all of these derivatives are strong agonists and are not suitable for downregulating GPR17 activity, which is necessary for the treatment of myelination disorders such as MS. Moreover, this class of GPR17 activators does not cross the blood-brain barrier well due to the easily ionized oxol group, and therefore was not a suitable lead compound for developing negative GPR17 modulators. See also Baqi et al., Med.Chem.Commun., 2014, 5, 86 and Koese et al., 2014, ibid.

[0017] WO2013 / 167177 suggests certain phenyltriazole and benzodiazepine compounds as GPR17 antagonists. However, the disclosed compounds were selected solely based on the results of in silico screening, and no biological data was provided. The inventors of the present application have not been able to confirm the GPR17 antagonistic modulation activity of any of the ligands purportedly proposed by the authors of this previous patent application.

[0018] WO2018 / 122232 discloses (Aaza) indole-benzothiophene- and benzofuran-3-sulfonamides as potent GPR17 antagonists. However, all of the disclosed compounds have an indole moiety with an unsubstituted nitrogen, and therefore differ from the compounds of the present application, which always have N-substitution at the indole group. Thus, the present application provides potent GPR17 modulators that surprisingly offer additional structural options by allowing the nitrogen of the indole ring to be substituted with various residues with or without polar groups. [0067] Therapeutic and diagnostic applications In one aspect, the invention relates to any of the compounds described herein for use in therapy or diagnosis, particularly in the treatment of animals, especially humans.

[0068] Due to their GPR17 modulating properties, the compounds of the invention can be used as medicines and can be used for the treatment and / or prevention of various diseases of the CNS system.

[0069] Thus, one embodiment of the present disclosure is a compound as described herein for use as a medicament, in particular for use as a medicament for the treatment and / or prevention of a GPR17 related disease.

[0070] GPR17-related diseases or disorders are diseases associated with dysfunction of the GPR17 signaling system, such as overexpression and / or overactivity of the GPR17 receptor. Without wishing to be bound by theory, the activity of GPR17 may increase, expand, or otherwise change in certain tissues, such as oligodendrocyte precursor cells (OPCs), or during oligodendrocyte maturation, potentially to activate endogenous stimuli, such as inflammatory factors. High activity of GPR17 may impede oligodendrocyte differentiation and efficient myelination, promoting the emergence or further development of myelination disorders (see Chen et al., supra). Thus, negative GPR17 modulators may promote myelination by reducing or turning off GPR17 activity and by supporting OPC maturation into myelin-producing oligodendrocytes (see, e.g., Simon et al., supra).

[0071] In a preferred aspect, the present invention relates to any of the compounds described herein for use in the diagnosis, prevention or treatment of disorders or syndromes selected from and / or related to myelination disorders, particularly demyelinating disorders such as disorders of the central nervous system. In one embodiment, the compounds of the present invention are for use in promoting, stimulating and / or accelerating remyelination in an animal in need thereof. In one embodiment, remyelination associated with administration of the compounds of the present invention will prevent or treat a demyelinating disease, such as, but not limited to, multiple sclerosis.

[0072] The compounds of the invention may also be useful in the treatment or prevention of disorders or syndromes associated with brain tissue injury, cerebrovascular disorders, and certain neurodegenerative diseases.

[0073] Neurodegenerative disorders have recently been strongly associated with loss of myelination. Thus, preserved oligodendroglial and myelin function is considered to be an important prerequisite for preventing axonal and neuronal degeneration (see, for example, Ettle et al., supra). Thus, negative GPR17 modulators may represent a good therapeutic option for any neurodegenerative disease associated with demyelination and / or affected myelination, such as ALS, MSA, Alzheimer's disease, Huntington's disease, or Parkinson's disease.

[0074] Thus, in certain preferred aspects, the compounds of the invention can be used for the prevention and / or treatment of peripheral or central myelination disorders, particularly myelination disorders of the central nervous system. In one aspect, the compounds of the invention are used in the treatment and / or prevention and / or diagnosis of myelination disorders by oral administration. In a preferred embodiment, the myelination disorder treated with the compounds of the invention is a demyelinating disorder.

[0075] Examples of such myelination disorders to be treated and / or prevented by the presently disclosed compounds are, inter alia: - Multiple sclerosis (MS), including its various subforms; - Neuromyelitis optica (also known as Devic's disease), - chronic relapsing inflammatory optic neuritis, acute disseminated encephalomyelitis, - Acute hemorrhagic leukoencephalitis (AHL), - Periventricular leukomalacia - demyelination due to viral infections, e.g. HIV or progressive multifocal leukoencephalopathy, - central pontine myelinolysis and extrapontine myelinolysis, - demyelination due to traumatic brain tissue injury, including compression demyelination, e.g. due to a tumor; - demyelination in response to hypoxia, stroke, ischemia, or other cardiovascular diseases, -Demyelination due to exposure to carbon dioxide, cyanide, or other central nervous system toxins - Schilder's disease, - Baroconcentric sclerosis, - perinatal encephalopathy, and - Neurodegenerative diseases, especially: o Amyotrophic lateral sclerosis (ALS). o Alzheimer's disease (AD). oMultiple system atrophy Parkinson's disease Spinocerebellar ataxia (SCA), also known as spinocerebellar atrophy Huntington's disease - Psychiatric disorders such as schizophrenia and bipolar disorder (see, for example, Fields, Trends Neurosci 31, 2008, 361; Tkachev et al, Lancet 362, 2003, 798). - peripheral myelinating disorders such as leukodystrophies, peripheral demyelinating neuropathies, Dejerine-Sottas syndrome, or Charcot-Marie-Tooth disease

[0076] Treatment or prevention of a central nervous system disease, such as a demyelinating disease, also includes treatment of the signs and symptoms associated with such disease.

[0077] For example, the use of the compounds of the invention for the treatment and / or prevention of MS also includes the treatment and / or prevention of signs and symptoms associated with MS, such as adverse effects on the optic nerve (loss of vision, double vision), spinal column (loss of sensation), corticospinal tract (spastic weakness), cerebellar pathways (loss of coordination, dysarthria, dizziness, cognitive impairment), medial longitudinal fasciculus (double vision in lateral gaze), spinal trigeminal tract (facial numbness or pain), muscle weakness (dysphagia, bladder or bowel control, spasms), or psychological effects associated with the underlying disease, such as depression, anxiety or other mood disorders, general weakness or insomnia.

[0078] Thus, the compounds of the present invention are for use in treating the signs and symptoms of myelinating diseases, particularly demyelinating diseases such as multiple sclerosis. Such signs and symptoms of MS include, but are not limited to, the following groups: vision loss, blurred vision, double vision, loss or impairment of sensation, weakness such as spastic weakness, impaired motor coordination, dizziness, cognitive impairment, facial numbness, facial pain, impaired swallowing, impaired speech, impaired bladder and / or bowel control, convulsions, depression, anxiety, mood disorders, insomnia, and fatigue.

[0079] In one preferred embodiment, the compound of the present invention is for use in treating multiple sclerosis. MS is a heterogeneous myelinating disease and can present in a variety of different forms and stages, including but not limited to relapsing-remitting MS, secondary progressive MS, primary progressive MS, and progressive relapsing MS, each of which depends on the activity and progression of the disease. Thus, in one embodiment, the compound of the present invention is for use in treating multiple sclerosis in its various stages and forms, as described herein.

[0080] In one aspect, the compounds of the invention are for use in the treatment and / or prevention of neuromyelitis optica (also known as Devic's disease or Devic's syndrome). Neuromyelitis optica is a complex disorder characterized by inflammation and demyelination of the optic nerve and spinal cord. Many of the associated symptoms are similar to MS, including muscle weakness, especially in the hands and feet, loss of sensation, and loss of bladder control.

[0081] In one aspect, the compounds of the invention are for use in the prevention and / or treatment of ALS, which has recently been associated with increased oligodendrocyte degeneration and demyelination, suggesting that ALS is a target disease for negative GPR17 modulators (Kang et al., supra; Fumagalli et al., Neuropharmacology 104, 2016, 82).

[0082] In one aspect, the compounds of the invention are for use in the prevention and / or treatment of Huntington's disease, which is well described as being associated with affected myelination (Bartzokis et al., supra; Huang et al., Neuron 85, 2015, 1212).

[0083] In one aspect, the compounds of the invention are for use in the prevention and / or treatment of multiple system atrophy. MSA has recently been strongly linked to demyelination (Ettle supra; Jellinger supra), suggesting a remyelination strategy to treat or prevent MSA.

[0084] In one embodiment, the compounds of the present invention are for use in the prevention and / or treatment of Alzheimer's disease. AD has recently been observed to be associated with increased oligodendronecytic cell death and focal demyelination, which represent the pathological process of AD (Mitew et al., Acta Neuropathol 119, 2010, 567).

[0085] One aspect of the invention relates to a method for treating any of the diseases or disorders described herein, particularly myelination diseases such as MS, neuromyelitis optica, ALS, Huntington's chorea, Alzheimer's disease, etc., by administering to a subject, including a human patient, in need of treatment a therapeutically effective amount of a compound of the invention.

[0086] In another aspect, the compounds of the present invention can be used for the prevention and treatment of spinal cord injury, perinatal encephalopathy, stroke, ischemia, or cerebrovascular accidents.

[0087] In one aspect, the present invention relates to a method for the prevention and / or treatment of disorders associated with impaired myelination, or disorders or syndromes associated with brain tissue damage, comprising administering to a patient in need thereof a therapeutically effective amount of a compound as described herein. A patient in need of such treatment may be any patient who has suffered brain tissue damage, such as by mechanical, chemical, viral, or other trauma.

[0088] In one aspect, the present invention relates to a method for the prevention and / or treatment of disorders of myelination or syndromes or disorders associated with stroke or other disorders or syndromes associated with cerebral ischemia, comprising administering a therapeutically effective amount of a compound as described herein to a patient in need thereof. The patient in need thereof may be any patient who has recently experienced cerebral ischemia / stroke, which may have been caused, for example, by occlusion of a cerebral artery, either by embolism or local thrombosis.

[0089] GPR17 has also recently been associated with food intake, insulin regulation, and obesity. According to various reports, negative modulators of GPR17 may be useful in controlling food intake and treating obesity (see, for example, Ren et al, Diabetes 64, 2015;3670). Thus, one embodiment of the present invention relates to the use of the compounds herein for the prevention and / or treatment of obesity, as well as methods of treating obesity.

[0090] Furthermore, the compounds of the present invention can be used for the prophylactic treatment of tissues in which GPR17 is expressed, such as the heart, lungs, or kidneys. In one embodiment, the compounds of the present invention can be used to treat or prevent ischemic damage to the kidney and / or heart.

[0091] GPR17 has also been implicated in lung inflammation and asthma, such as that induced by dust mites (Maekawa, J Immunol 2010, 185(3), 1846-1854). Thus, the compounds of the present invention can be used to treat asthma or other lung inflammation.

[0123] The present invention also includes all suitable isotopic variants of the compounds of the present invention. An "isotopic variant" of a compound of the present invention, or simply "isotope", is defined as one in which at least one atom is replaced by an atom having the same atomic number but a different atomic mass than the atomic mass usually found in nature, with the most abundant isotope being preferred. Examples of isotopes that can be incorporated into the compounds of the present invention include: 2 H, 3 H, 11 C. 13 C. 14 C. 15 N, 17 O. 18 O. 35 S, 18 F, and 36 Specific isotopic variations of the invention include isotopes of hydrogen, carbon, nitrogen, oxygen, sulfur, fluorine, and chlorine, such as Cl. 3 H or 14 Those incorporating radioactive isotopes such as C are useful in drug and / or substrate tissue distribution studies. [0183] A-III. Analysis method Commercially available solvents and reagents were generally used without further purification, including anhydrous solvents where necessary (usually Sure-Seal™ products from Aldrich Chemical Company or AcroSeal™ from ACROS Organics). Reactions were generally followed by thin layer chromatography or liquid chromatography mass spectrometry.

[0184] Mass spectrometry measurements in LCMS mode are performed using different methods and instruments:

[0185] - Basic LCMS method 1: Mass spectrometry (MS) spectra were recorded on a LCMS-2010EV mass spectrometer (Shimadzu) with electrospray ionization (ESI) connected to a HPLC Modular Prominence (Shimadzu) using an Xbridge C18-2.1x30mm, 2.5μm (Waters) column. A volume of 3μL of sample solution with a concentration of approximately 1mg / mL was injected. The mobile phase for the basic conditions was a mixture of A) 5mM ammonium formate + 0.1% aqueous ammonia B) 5% mobile phase A + 0.1% ammonia in acetonitrile. The gradient used was as follows: 5:95 (B / A) to 95:5 (B / A) in 4 min, then hold at 95:5 (B / A) for the next 1 min.

[0186] -Neutral LCMS Method 2: Mass spectrometry (MS) spectra were recorded on an LCMS instrument (Applied Biosystems API 2000 LC / MS / MS, HPLC Agilent 1100) using the following procedure: 1.0 mg mL in ACN (solvent A) or water (containing 2 mM ammonium acetate):MeOH 90:10 (solvent B). ‐1Compounds were dissolved at a concentration of 0.01% and sonicated if necessary until completely dissolved. 10 μL of the solution was then injected onto a Phenomenex Luna C18 HPLC column (50 × 2.00 mm, 3 μm particle size) and eluted with a gradient of water:ACN (gradient A) or water:MeOH (gradient B) from 90:10 to 0:100 within 10 min. The gradient was started after 1 min, followed by 300 μL min of pure organic solvent. ‐1 The elution was performed for 10 min at a flow rate of 100 rpm. UV absorption was detected at 220–400 nm using a diode array detector (DAD).

[0187] The crude material could be purified by normal phase chromatography or recrystallization.

[0188] Normal phase chromatography was performed using a silica gel column (100:200 mesh silica gel or cartridges for flash chromatography systems such as the Isolera™ Four from Biotage® or Teledyne Isco CombiFlash®).

[0189] NMR spectra were recorded on a variety of instruments. - Varian 400 MHz NMR spectrometer. Acquisition time (at) = 2.0 s, relaxation delay (d1) = 2.0 s, line broadening (lb) = 0.5 Hz. -Bruker Avance III 600 MHz NMR spectrometer

[0190] Chemical shifts are referenced to signals due to residual protons of the deuterated solvent (DMSO-d6 or CDCl3). Chemical shifts are in parts per million (ppm) and coupling constants (J) are expressed in Hertz (Hz). [0371] E.5. Synthesis of 1-(2-azidoethyl)-N-(4-bromo-2,5-difluorophenyl)-6-chloroindole-3-sulfonamide I-36 [ka]

[0372] A solution of I-13a (200 mg, 0.38 mmol) in 5 mL of dry dimethyl sulfoxide was treated with potassium iodide (75 mg, 0.45 mmol) and sodium azide (30 mg, 0.45 mmol) and heated at 80° C. for 16 h. The mixture was poured into ice water and the precipitate was filtered under reduced pressure to give 1-(2-azidoethyl)-N-(4-bromo-2,5-difluorophenyl)-6-chloro-N-(methoxymethyl)-1H-indole-3-sulfonamide, which was characterized only by TLC (one clear spot of product) and used directly in the next step. The intermediate thus obtained was dissolved in methanol (15 mL), treated with concentrated HCl (5 equiv.) and stirred for 16 h. The solvent was removed under reduced pressure and the residue was treated with ethyl acetate (50 mL) and a saturated solution of sodium bicarbonate (50 mL). The organic layer was separated and the aqueous phase was extracted with ethyl acetate (2x50 mL). The combined organic layers were washed with brine, dried over MgSO4, filtered and concentrated in vacuo to give 140 mg of 1-(2-azidoethyl)-N-(4-bromo-2,5-difluorophenyl)-6-chloroindole-3-sulfonamide I-36 as a colorless solid.

[0379] B. Biology / Pharmacology: B-I.Cell culture GPR17 recombinant cell line: Flp-In T-REx CHO cells stably expressing the human GPR17 receptor (CHO hGPR17) from the laboratory of Evi Kostenis (University of Bonn, Germany) were cultured at 37 °C in a humidified atmosphere of 5% CO2. Cells were cultured in DMEM containing nutrient mixture F-12 supplemented with hygromycin B (500 μg / ml) and blasticidin (30 μg / ml). Expression from the Flp-In locus was induced by treatment with doxycycline (1 μg / ml) for 16–20 h before assay.

[0380] Primary oligodendrocytes: Primary oligodendrocyte precursor cells (OPCs) were isolated from the forebrains of Wistar rat pups on postnatal days 0–2. The cerebrum was mechanically dissociated with a syringe and two different hollow needles (first 1.2x40 and then 0.60x30). The clump-free cell suspension was filtered through a 70 μm cell strainer and plated on poly-D-lysine-coated 75 cm plates with medium changes every 2 days with DMEM supplemented with 10% (v / v) heat-inactivated fetal bovine serum, penicillin (100 units / ml), and streptomycin (0.1 mg / ml). 2OPCs were plated in culture flasks. After 8–11 days at 37°C in a humidified atmosphere of 5% CO2, the mixed culture was shaken at 240 rpm for 14–24 h to separate the OPCs from the astrocytes and microglia. To further enrich for OPCs, the suspension was plated on uncoated Petri dishes for 45 min. OPCs were then seeded on poly-L-ornithine-coated plates and maintained at 37°C in a humidified atmosphere of 5% CO2 with medium changes every 2 days in growth Neurobasal medium supplemented with 2% (v / v) B27, 2 mM GlutaMAX, 100 units / ml penicillin, 0.1 mg / ml streptomycin, 10 ng / ml PDGF-AA, and 10 ng / ml basic FGF.

[0381] B-II: Functional in vitro GPR17 assay B-II-A: Calcium mobilization functional assay GPR17 is a G protein-coupled receptor. Activation of GPR17 triggers Gq-type G protein signaling, leading to upregulation of cytosolic endoplasmic reticulum calcium (Ca 2+ ) stores, which releases cytosolic Ca 2+ The levels of calcium in the blood can be measured using calcium 5 dye, a fluorescent indicator dye for calcium levels, as described further below. 2+ The antibodies were assessed in either the GPR17 cAMP assay or the GPR17 cAMP assay. As shown in Table 3 below, several representative examples were measured in both activity tests.

[0382] Ca 2+ Assay Description: CHO hGPR17 were thawed and seeded into black, clear-bottom 384-well plates at a density of 20,000 cells per well. Cells were incubated overnight at 37 °C in a humidified atmosphere of 5% CO2. 16–20 h after seeding, CHO hGPR17 were enriched in cytosolic Ca 2+ The indicator fluorescent dye, calcium 5 dye, was loaded for 60 min according to the manufacturer's instructions. 2+The fluorescence signal versus concentration was recorded over time on a FLIPR Tetra reader at room temperature. Cells were first treated with increasing concentrations of test compounds (usually 10 -11 M to 10 -6 The cells were incubated in HBSS Hepes buffer pH 7.4 containing 100 mM M for 30 min at room temperature. Then, 50 nM MDL29,951, a GPR17 agonist, was added to the cells. The inhibitory effect of various concentrations of the test compound was measured and the resulting pIC 50 was determined. All incubations were performed in duplicate and results were compared to concentration-response curves of GPR17 agonist and antagonist reference compounds. Analysis and curve fitting was performed in ActivityBase XE using the XLfit4 parametric logistic equation y=A+((BA) / (1+((C / x)^D))), where A, B, C, and D are the minimum y, maximum y, and IC, respectively. 50 , which represents the gradient.

[0383] Ca 2+ Assay Results: Ca 2+ When tested in the recruitment assay, the compounds of the Examples typically have a pIC of 6.5 or greater, more preferably 7.5 or greater, and even more preferably 8.5 or greater. 50 The activity of the tested Example compounds is shown in Table 3 in Section B-IIB below. Activity ranges A, B, and C are 2+ Assay pIC 50 The values are referenced as follows: "A":pIC 50 6.5≦x<7.5, "B": pIC 50 7.5≦x<8.5, “C”:8.5≦pIC 50

[0384] B-IIB. cAMP accumulation functional assay Activation of GPR17 can also recruit Gi-type G-protein signaling, resulting in a decrease in intracellular cyclic adenosine monophosphate (cAMP). Changes in intracellular cAMP can be measured using the HTRF cAMP dynamic assay kit from CisBio (Codolet, France). Using homogeneous time-resolved fluorescence technology (HTRF), the assay is based on the competition between native cAMP produced by the cells and cAMP labeled with the dye d2. Tracer binding was determined by an anti-cAMP antibody labeled with cryptate.

[0385] cAMP Assay Description CHO hGPR17 were dissociated in PBS containing EDTA and dispatched to black 384-well plates containing 5,000 cells per well. Cells were first incubated for 30 min at room temperature in HBSS Hepes (pH 7.4) containing vehicle or various concentrations of test GPR17 antagonist / inverse agonist compounds. Dose-response curves of MDL29,951 GPR17 agonists (usually 10 -5 M to 10 -10 M) was added to vehicle and the concentration of GPR17 antagonist / inverse agonist compound in each test was brought to a final volume of 20 μL of HBSS Hepes buffer (pH 7.4) containing 1% DMSO, 5 μM forskolin and 0.1 mM IBMX. After 60 min incubation at room temperature, the reaction is stopped and cells are lysed by adding d2 detection reagent and cryptate reagent in 10 μL each of lysis buffer according to the manufacturer's instructions. After 60 min incubation, changes in cAMP concentration are measured according to the manufacturer's instructions using an Envision plate reader equipped with laser excitation. All incubations were performed in duplicate. Data are presented as MDL29,951 pEC in the absence and presence of GPR17 antagonist / inverse agonist test compounds. 50The affinity of GPR17 antagonists / inverse agonists was determined by plotting the dose ratio (DR) versus antagonist concentration and analyzing the effect of schizophrenia on the affinity of GPR17 antagonists / inverse agonists.

[0386] cAMP Assay Results: When tested in the cAMP assay, the compounds of the examples typically exhibit pA2 values of 6.5 or more, preferably 7.5 or more, more preferably 8.5 or more. The activities of the compounds of the examples that were tested are shown in the table below. Activity ranges A, B, and C refer to the pA2 values of the cAMP assay as follows: "A": pA26.5≦x<7.5, "B": pA27.5≦x<8.5, "C": 8.5≦pA2.

[0387] The following Table 3 shows the Ca 2+ and pIC of the Example Compounds Tested in the cAMP Assay 50 and pA2 values are shown. pA2 or Ca 2+ A blank in an assay column indicates that the respective compound has not yet been tested in the respective assay or that results are not yet available.

[0389] B-IIC: Oligodendrocyte maturation / myelination assay The effect of negative modulators of GPR17 on primary oligodendrocyte maturation / myelination can be assessed in vitro by immunoassay using an antibody against myelin basic protein (MBP) as a marker for mature oligodendrocytes.

[0390] Description of the MBP Western Blot / Oligodendrocyte / Myelination Assay After 3–4 days in growth medium, rat primary OPCs were plated at 1 cm in 12-well tissue culture plates.2 Cells were seeded at 25,000 cells per well and switched to growth factor-free Neurobasal medium to induce spontaneous in vitro differentiation and GPR17 protein expression. After 24–48 h, growth factor-free medium was supplemented with 0.20 ng / mL triiodothyronine (T3) and 10 ng / mL ciliary neurotrophic factor along with 1 μM GPR17 antagonist / inverse agonist test compound or vehicle for an additional 3 days for cell culture. After compound treatment, cells were washed twice with ice-cold PBS and lysed in ice-cold lysis buffer (25 mM Tris, pH 7.4, 150 mM NaCl, 1 mM EDTA, 1% Triton X-100, 1% IGEPAL) supplemented with a protease inhibitor mixture. Lysates were spun at 4 °C for 20 min and centrifuged at 15,000 x g for 10 min at 4 °C. Protein concentrations were determined using the Pierce BCA Protein Assay according to the manufacturer's instructions. 7.5–15 μg of protein was separated by 10% SDS-polyacrylamide gel electrophoresis and transferred to nitrocellulose membranes by electroblotting. After washing, the membranes were blocked with Roti-Block for 1 h at room temperature and incubated overnight at 4°C in Roti-Block containing MBP antibody (1:5000, LifeSpan BioSciences). The membranes were washed three times with PBS containing 0.1% Tween and then incubated with horseradish peroxidase-conjugated goat anti-mouse IgG antibody in Roti-Block for 1 h at room temperature. Immunoreactive proteins were visualized by chemiluminescence using Amersham Biosciences ECL Prime Western blotting detection reagent and quantified by densitometry using Gelscan software. To normalize for equal loading and protein transfer, membranes were reprobed with an antibody against β-actin (1:2500, BioLegend; secondary antibody goat anti-rabbit IgG HRP (ABIN)). Changes in MBP expression levels in the presence of test compounds were compared to MBP expression in control conditions.

[0391] Description of the MBP Fiber Plate / Oligodendrocyte Maturation / Myelination Assay OPCs were placed in a Mimetix Aligned 96-well fiber plate (Electrospining company) at 1 cm 2 Cells were seeded at 16,000–22,000 cells per well. After 2 days in proliferation medium and 2 days in growth factor-free Neurobasal medium to induce spontaneous in vitro differentiation and GPR17 protein expression, vehicle or 1 μM antagonist / inverse agonist test compounds were added in terminal differentiation medium supplemented with 0.20 ng / mL triiodothyronine and 10 ng / mL ciliary neurotrophic factor for 6 days, and the medium was replaced after 3 days. Cells were then fixed with 4% paraformaldehyde, followed by washing with PBS, permeabilization with 0.1% Triton X-100 in PBS, and blocking with 10% goat serum and 1% bovine serum albumin in phosphate-buffered saline. MBP antibody was diluted in blocking buffer (1:2000) and incubated for 1 h at 37 °C. Cells were washed again with PBS and incubated with Cy2-labeled secondary antibody against mouse IgG (Millipore, 1:500) for 1 h. After washing with PBS, cells were stained with 0.2 μg / mL DAPI, washed again and mounted with Mowiol. Fluorescent images were taken using a Zeiss AxioObserver.Z1 microscope equipped with an ApoTome Imaging System and a Plan-Apochromat 20x / 0.8 objective, eGFP filter (excitation 470 / 40 nm, emission 525 / 50 nm) and DAPI filter (excitation 365 nm; emission 445 / 50 nm). At least 15 random fields of control (terminal differentiation medium containing 0.1% DMSO) and test compounds were imaged using the same settings processed with Zeiss ZEN2.3 software. Changes in the number of myelinated fibers were reported by fiber length groups (0–40 μm, 41–60 μm, 61–80, 81–100, 101–120, and >120 μm) in the absence or presence of GPR17 negative modulators.
