[0003] [Patent Document 1] JP 2015-051947 A Summary of the Invention [Problem to be solved by the invention]
[0004] The present inventors have studied a patch containing bisoprolol, and found that the content of bisoprolol in the patch decreases with time. When the content of bisoprolol decreases, it may affect the performance of bisoprolol, such as the skin permeability. Therefore, the object of the present invention is to provide a patch in which the content of bisoprolol is unlikely to decrease even with time.In addition, the object of the present invention is to provide a method for improving the storage stability of bisoprolol or its pharma-ceutically acceptable salt in a patch that has an adhesive layer on a support. [Means for solving the problem]
[0005] As a result of intensive research, the inventors discovered that a patch containing 2-mercaptobenzimidazole and a silicic acid compound in the adhesive layer suppresses the decrease in bisoprolol content over time, and thus completed the present invention.

[0042] The patch can be produced, for example, by the following method, but is not limited thereto, and any known method can be used. First, the components constituting the adhesive layer are mixed in a solvent (toluene, ethyl acetate, etc.) in a predetermined ratio to obtain a uniform solution of the adhesive composition. Next, the solution of the adhesive composition is spread on a peelable film (release liner), and the solvent is then dried and removed to form an adhesive layer of a predetermined thickness. Furthermore, a support is pressed onto the adhesive layer so that the adhesive layer is sandwiched between the release liner and the support. Finally, the patch can be obtained by cutting into the desired shape and dimensions. In this case, the release liner is removed when the patch is applied. The area of the patch is 5 cm 2 ~100cm 2 5 cm 2 ~50cm 2 The shape and dimensions of the patch may be, for example, a rectangle with short sides of 2 cm to 10 cm and long sides of 3 cm to 15 cm, or a circle with a diameter of 1 cm to 8 cm. EXAMPLES
[0043] Test Example 1: Evaluation of the Decrease in Bisoprolol Content 1. Preparation of the Patch As per the compounding ratios in Tables 1 to 4 below, each component was thoroughly mixed in toluene to obtain a solution of the pressure-sensitive adhesive composition. After spreading the obtained solution of the pressure-sensitive adhesive composition on a release liner (a polyethylene terephthalate film subjected to a release treatment), toluene was removed by drying to form a pressure-sensitive adhesive layer with a mass per unit area of 37 g / m 2 A support layer (a polyethylene terephthalate film) was laminated on the opposite surface of the obtained pressure-sensitive adhesive layer to obtain a patch laminated in the order of support layer / pressure-sensitive adhesive layer / release liner.
[0044] 2. Measurement of the Content of Bisoprolol The content of bisoprolol was measured using the following procedure with the patch before storage and the patch enclosed in a package and stored in a constant temperature machine at 60°C for 1 month, both of which were cut to an area of 2.5 cm 2 . After removing the release liner, the patch was put into a 50 mL test tube, and 10 mL of a tetrahydrofuran solution containing propyl paraoxybenzoate as an internal standard substance was added thereto. After dissolving by shaking for 30 minutes and ultrasonic treatment for 30 minutes, it was diluted 5-fold with a 0.2% aqueous phosphoric acid solution. After mixing for 10 minutes, it was filtered through a 0.45 μm membrane filter to obtain a sample solution. Thereafter, bisoprolol contained in the sample solution and the standard sample solution was eluted for 15 minutes in high performance liquid chromatography (HPLC) using the following mobile phase, detected, and then the concentration of bisoprolol in the sample solution was quantified using the calibration curve obtained from the standard sample solution. By calculating the content of bisoprolol in the patch and correcting it using the measured value of the mass of the pressure-sensitive adhesive layer of each patch, the content of bisoprolol in the patch before storage and after storage at 60°C for 1 month was calculated. <HPLC Analysis Conditions> Column: Inertsil ODS-3 (ODS particle size: 3 μm, inner diameter: 4.6 mm, length: 15 cm) Mobile phase: Aqueous mobile phase (aqueous solution of 20 mM sodium heptanesulfonate and 0.2% phosphoric acid):acetonitrile = 60:40 Measurement wavelength: 225nm Flow rate: 0.8mL / min Sample injection volume: 10 μL Column temperature: 40℃ The theoretical value of the bisoprolol content at the time of preparation of the patch was set to 100, and the theoretical values of the bisoprolol content in the patch before storage and after one month of storage at 60°C were calculated respectively. The reduction amount was calculated by subtracting the theoretical value of the bisoprolol content in the patch after one month of storage at 60°C from the theoretical value of the bisoprolol content in the patch before storage.
[0045] [Table 1]
[0046] [Table 2]
[0047] [Table 3]

[0049] As is clear from the results shown in Tables 1 to 4, the patch containing 2-mercaptobenzimidazole and light anhydrous silicic acid was able to suppress the decrease in the content of bisoprolol during storage. Also, as is clear from the results shown in Tables 1 and 2, the patch not containing either 2-mercaptobenzimidazole or light anhydrous silicic acid was unable to suppress the decrease in the content of bisoprolol during storage.
