Description

[Technical field]

[0001] The present invention relates to a proteolysis targeting chimera (PROTAC) protein having an intracellular signaling function and a pharmaceutical composition comprising the same. [Background technology]

[0002] Proteolysis targeting chimeras (PROTACs) are dual-binding molecules that bind to disease-causing proteins and induce the proteasome to degrade the proteins, ultimately inducing selective proteolysis. PROTACs consist of two protein-binding molecules, one for binding to E3 ubiquitin ligase and the other for binding to the target protein. By binding to the two proteins, PROTACs bring the target protein into close proximity with the E3 ligase, inducing the tagging, or ubiquitination, of the target protein for subsequent degradation by the proteasome.

[0003] Ubiquitination involves three steps: activation, conjugation, and ligation, which are performed by ubiquitin-activating enzyme (E1), ubiquitin-conjugating enzyme (E2), and ubiquitin ligase (E3). As a result of this sequential cascade, ubiquitin is covalently attached to the target protein. The ubiquitinated protein is finally degraded by the proteasome.

[0004] Protac technology was first launched in 2001 (Sakamoto et al., Proceedings of the National Academy of Sciences of the United States of America. 98: 8554, 2001). Since then, this technology has been used in various drug designs: pVHL, MDM2, β-TrCP1, cereblon, and c-IAP1. Protac drugs have been developed, mostly consisting of low molecular weight compounds. Various types of low molecular weight compounds derived from the thalidomide series have the structural advantage of being able to simultaneously bind target proteins and E3 ligases. Although these known Protac drugs are very useful, they need to be improved to solve the inherent concerns about reduced protein degradation efficiency and safety due to the low solubility and reduced intracellular permeability of low molecular weight compounds, and there is a need for Protac drugs that improve these problems.

[0005] Therefore, instead of using low molecular weight compounds, the present inventors have made intensive efforts to develop a protac protein that has high target protein degradation efficiency and intracellular delivery function. As a result, they have prepared a target protein binding sequence linked to the Von Hippel-Lindau (VHL) protein sequence by a linker consisting of five glycines as a substrate recognition unit that can transfer E3 ligase, which is an important target in cancer, inflammatory diseases, and infections, to a target protein, and have confirmed that an endogenous target protein for a disease is actually degraded through intracellular delivery, thereby completing the present invention.

[0006] The information provided in this Background section is intended solely to enhance understanding of the background of the invention and may not include information that constitutes prior art already known to a person of ordinary skill in the art to which the invention pertains. Generally, the nomenclature used herein and the laboratory procedures described below are those well known and commonly employed in the art.

[0011] The present invention is based on the discovery that a ubiquitin pathway protein will ubiquitinate any target protein when the ubiquitin pathway protein and the target protein are in close proximity to a chimeric structure that binds the ubiquitin pathway protein and the target protein.

[0012] The main reason for the present invention is to overcome the limitations of PROTAC technology produced by conventional methods, such as low actual solubility or inability to effectively degrade endogenous disease proteins when applied to cells. In one aspect, the present invention relates to a PROTAC protein having the structure of Formula 1 or Formula 2 below: [ka] [ka] In the above Chemical Formula 1 or Chemical Formula 2, (i) PEP is an antibody or a fragment of an antibody, or a cell penetrating peptide; (ii) L1 and L2 are linkers, L1 and L2 may be the same or different, and L1 is linked to TB or L2; (iii) TB is a binder or conjugate that binds to a target protein; (iv) UR is a binding ligand for ubiquitin ligase; (v) n and m each independently represent an integer of 1 to 10.

[0013] As used herein, the term "PROTAC" is an abbreviation for proteolysis targeting chimera, and refers to a bifunctional small molecule composed of two active domains and a linker, which can remove specific unwanted proteins. As used herein, "PROTAC protein" is used interchangeably with "bioPROTAC protein" and "chimeric protein."

[0014] The Protac proteins according to the present invention are capable of inducing ubiquitination of a selected target protein and are designed to link a target protein binding moiety and a linker to contain a ubiquitin ligase binding site.

[0015] In the present invention, the Protac protein comprises a ligand that binds to ubiquitin ligase at one end and a portion that binds to a target protein at the other end, and comprises a cell-permeable domain at the end of the protein that binds to the target protein or at the linker that connects the ubiquitin ligase-binding ligand to the target protein-binding portion, which allows the Protac protein to be located in close proximity to the target protein when introduced into cells and induce protein degradation, and is present at a high concentration in cells depending on the numbers n and m, thereby inducing effective degradation of the target protein. In the present invention, the protac protein can be characterized by being represented by the amino acid sequences of SEQ ID NOs: 1 to 6, but is not limited thereto.

[0042] The present invention will be described in more detail with reference to the following examples. It will be obvious to those skilled in the art that these examples are merely for the purpose of illustrating the present invention and are not intended to limit the scope of the present invention. EXAMPLES

[0043] Example 1: Expression of cell-permeable protac protein in mammalian cells The present inventors designed an expression vector for a protak protein that degrades a target protein as follows. G12CA cDNA containing a monoclonal antibody targeting VHL protein that binds to E3 ligase and a Kozak sequence that binds to ribosomes and helps transcription was inserted into the pcDNA3.4 vector, cloned, and confirmed by DNA sequencing. The signal peptide, linker, and PEP sequence were added to the constructed plasmid DNA sequence. The signal peptide was located at the N-terminus, followed by a monoclonal antibody targeting mutant human KRAS G12C, VHL, and PEP linked to this by linkers of various lengths in various orders. The plasmid constructed in this way (Figure 1) was transfected into CHO-S cell line, and then enhancer was added and cultured for 7 days.

[0044] The expressed Protac protein was measured by Western blotting using anti-His tag antibody (Abcam, ab18184) and anti-VHL antibody (Invitrogen, MA5-13940), and the Western blotting results are shown in Figure 2. The theoretical protein size is 56 kDa, and Protac protein began to appear from day 3 of culture. Since the His Tag did not appear from day 5 of culture onwards, it is considered to have been cleaved from the protein.

[0045] Example 2: Confirmation of the effect of Protac protein in suppressing cancer cell proliferation A plasmid encoding Protak was transfected into cancer cells, and an MTT assay was performed to examine the anticancer effect of the Protak protein expressed by the plasmid. G12C 5X10 cells of non-small cell lung cancer with the phenotype 3Various amounts of plasmids were transfected into H358 (ATCC, CRL-5807) using Lipofectamine 3000 (Invitrogen, L3000001), and cell viability was evaluated by MTT assay 72 hours later (Figure 3). The higher the concentration of the treated plasmid, the lower the viability of H358 cells. This confirms that the protac protein was expressed and acted on the cells by the plasmid introduced into them.

[0046] Example 3: Confirmation of the effect of Protak protein in suppressing KRAS signal transduction To confirm that the target protein is degraded by the protactic protein expressed in the cells, the plasmid DNA prepared in Example 1 was transfected with human KRAS using Lipofectamine 3000 (Invitrogen, L3000001). G12C The cells were transfected into H358 (ATCC, CRL-5807), a non-small cell lung cancer cell line with the phenotype of . After transfection, the cells were lysed every 16, 24, and 48 hours to obtain cell lysates.

[0047] To isolate active KRAS, 80μg of GST-Raf1-RBD was added using the Active GTPase Kit (Cell signaling, 11860S) and 500μg of protein was dispensed into a spin cup. After 1 hour of reaction at 4℃ and 15 seconds of centrifugation at 6000xg, the column was transferred to a new tube and 400μL of 1x lysis buffer was added. After 15 seconds of centrifugation at 6000xg, the column was transferred to a new tube and 50μL of 2x SDS buffer (5X SDS Sample loading dye 200μl, Water 300μl mixture) was dispensed and reacted for 2 minutes. After 2 minutes of centrifugation at 6000xg, the column was heated at 100℃ for 7 minutes. Proteins (protein loading volume 20μl / lane) were separated by electrophoresis on 11% SDS-PAGE and transferred to a nitrocellulose membrane. To measure ERK1 / 2, a sub-signal of KRAS, 30μg of protein was electrophoresed on 11% SDS-PAGE and transferred to nitrocellulose membrane. After blocking with 5% skim milk dissolved in T-TBS for 1 hour, primary antibody was diluted 1:1000 in T-TBS and reacted overnight at 4℃ with inverting. After washing with T-TBS three times for 10 minutes each, secondary antibody was diluted 1:3000 in T-TBS and reacted at room temperature for 1 hour with inverting. After washing with T-TBS three times for 10 minutes each, chemiluminescence was performed with ECL substrate (Thermo, 34580) and confirmed with Amersham Imager 680 (GE). Information on the antibodies used is shown in Table 2 below.

[0048] [Table 2]

[0049] The protak protein was identified using an anti-His tag antibody by immunoblotting to confirm the expression of all KRAS (pan KRAS) or activated KRAS (active KRAS) in the cells (Figure 4). As the protak protein was expressed, the expression level of activated KRAS decreased, but the expression level of pan KRAS did not change. This confirms that the protak protein selectively degrades only activated KRAS and not normal pan KRAS. Example 4: Intracellular introduction of mRNA encoding a protac protein

[0050] To express the protak protein in cells, it was prepared in the form of mRNA instead of a plasmid. 5' capping and 3' poly A tail were inserted to increase the stability of the mRNA. The cells were transfected using Lipofectamine MessngerMAX (Invitrogen, LMRNA003). 24 hours after transfection, the cells were lysed to obtain cell lysates. Activated KRAS was selected in the same manner as described in Example 3. The protak protein expressed by the mRNA introduced into the cells was confirmed using an anti-His tag antibody (Abcam) by immunoblotting. It was confirmed that the expression level of activated KRAS decreased as the protak protein was expressed (Figure 5). The protak protein introduced in the form of mRNA was expressed, and the target protein, activated KRAS, was degraded. [Industrial Applicability]

[0051] The PROTAC protein according to the present invention has higher solubility than PROTACs produced by conventional methods, and efficiently degrades endogenous disease proteins when applied to cells, making it effective in treating cancer or inflammatory diseases.
