AI Technical Summary

Technical Problem

Existing immunoglobulin-cleaving enzymes like IdeS are pathogenic and induce anti-drug antibodies, limiting their therapeutic use, while non-human counterparts like IdeZ have reduced activity against human IgG.

Method used

Identification and characterization of a novel polypeptide, IdeSORK2.0, from Streptococcus kroesus, with enhanced expression and immunoglobulin cleavage activity, minimizing pre-existing immunity in humans.

🎯Benefits of technology

IdeSORK2.0 effectively cleaves human IgG into Fc and F(ab')2 fragments, offering therapeutic potential without inducing anti-drug antibodies, thus treating autoimmune diseases and IgG-mediated conditions.

Abstract

The present invention relates to novel polypeptides that exhibit protease activity against immunoglobulins, particularly human IgG, and their uses in vivo, in vitro and ex vivo. Uses of said polypeptides include methods for the analysis of IgG and the generation of antibody fragments, as well as methods for the prevention or treatment of IgG-mediated diseases and conditions. The inventors have identified, purified and characterized a novel polypeptide. The polypeptide was identified in a previously unknown Streptococcus species that was isolated as part of studies in multiple rodents.
