Description

Method for promoting enzyme reaction

[0001] The present invention relates to a method for promoting an enzyme reaction, an enzyme reaction accelerator, an enzyme reaction accelerator composition, and a dishwasher detergent composition.

[0002] Enzymes such as lipases, amylases, and proteases are used in many industrial sectors, including cleaning, food processing, and papermaking.

[0003] Japanese Patent Laid-Open Publication No. 63-7789 discloses a method for producing fatty acids by treating fats and oils with lipase in a reverse micelle system consisting of an organic solvent, a surfactant, and a buffer solution or water. Japanese Patent Laid-Open Publication No. 2012-75378 discloses a method for enzymatic saccharification of woody biomass, including a powdering step (1) in which at least one type of woody biomass selected from wood chips and waste building materials is pulverized to obtain woody biomass powder, and a saccharification step (2) in which the woody biomass powder is hydrolyzed in the presence of a predetermined anionic surfactant and a hydrolytic enzyme to obtain sugar.

[0004] Enzymes are used in many cleaning applications. Japanese Patent Application Laid-Open Publication No. 2019-182911 discloses a liquid detergent composition for tableware and / or kitchen hard objects, containing (a) an alkyl sulfosuccinate ester or a salt thereof, the alkyl group of which has 5 to 18 carbon atoms; (b) one or more surfactants selected from semi-polar surfactants and amphoteric surfactants; and (c) an enzyme and water under specified conditions. Japanese Patent Application Laid-Open Publication No. 2020-100745 discloses a liquid dishwashing detergent composition containing an anionic surfactant (component (A)) containing one or more (a1) components selected from secondary alkane sulfonates, dialkyl sulfosuccinates, and compounds represented by a specific formula; an amine oxide surfactant (component (B)); a protease (component (C)); and an amylase (component (D)), wherein the mass ratio of component (A) to component (B) is 0.7 to 2.

[0005] Furthermore, with the recent widespread use of automatic dishwashers, many detergent compositions for dishwashers have been proposed. Examples of stains that adhere to tableware include oil stains, starch stains such as rice grains, protein stains such as eggs, and pigment stains such as tea. Among these, oil stains and starch stains are particularly difficult to remove, and washing in a dishwasher poses the problem of the stains reattaching to the dishes and causing glass tableware to become cloudy. Therefore, in order to enhance the detergency and anti-reattachment properties against the stains, various formulations have been proposed that combine, for example, types of active cleaning ingredients such as surfactants, enzymes, and chelating agents.

[0006] Japanese Patent Publication No. 4-72397 discloses a technology that combines a specific nonionic surfactant and a calcium-sequestering chelating agent with alkaline pullulanase and lipase having α-amylase activity to achieve high detergency. Japanese Patent Publication No. 2006-152287 also describes a technology that prevents clouding of glass tableware and improves the finish by combining a high concentration of chelating agent with a low-foaming nonionic surfactant. The aforementioned Japanese Patent Publication No. 2019-182911 discloses a liquid detergent composition containing a sulfosuccinate alkyl ester salt and an enzyme, and a cleaning method in which the composition is brought into direct contact with tableware stained with grease in a foamy state. The aforementioned Japanese Patent Publication No. 2020-100745 discloses that a composition containing a dialkyl sulfosuccinate, a protease, and an amylase has excellent detergency and defoaming properties.

[0007] Summary of the Invention When enzymes are used in various industrial fields, they are often reacted with substrates in aqueous media, necessitating a reaction field where water, substrate, and enzyme come into contact. However, while enzymes are hydrophilic, substrates are often hydrophobic, resulting in low affinity between the substrate and enzyme, making the enzymatic reaction difficult and preventing the expected enzymatic activity from being achieved. To address this issue, for example, methods have been used, such as using large amounts of aqueous solvent or using surfactants in combination. However, the former is uneconomical because it uses aqueous solvent unnecessary for the reaction, and the latter, while improving the affinity between the substrate and enzyme, results in excessive surfactant action on the enzyme, reducing the enzymatic activity itself. Therefore, there is a strong demand for technology that promotes the action of substrates and enzymes while maintaining high enzymatic activity in aqueous media.

[0008] The present invention provides a method for enhancing an enzymatic reaction, for example, an enzymatic reaction in an aqueous medium.

[0009] Furthermore, when it comes to dishwasher detergents, it is desirable to emulsify the oil adhering to dishes with a very small amount of surfactant to prevent foam from overflowing inside the dishwasher and compromising cleaning power. However, because the amount of surfactant is small, no technology has yet been found that combines high cleaning power with improved finish by preventing dirt from re-adhering to dishes.

[0010] Thus, a technology for dishwasher detergents that combines high detergency with prevention of redeposition on dishes using a minimal amount of surfactant is desired. However, conventional technologies have been insufficient in preventing the redeposition of oily and proteinaceous soils on dishes, resulting in the appearance of white spots on the surface of dishes after washing and drying, significantly impairing the finish. The inventors have clarified that the cause of this is that during the drainage process (transition from washing to rinsing), some oil droplets detached from the dishes remain in the circulation line specific to dishwashers. When the oil droplets are carried over into the dishwasher again during the rinsing process, the destabilized oil droplets coalesce and become larger in particle size due to dilution (decrease in surfactant concentration), temperature change (from high temperature to room temperature), and agitation halted during drainage, making them more likely to redeposit on dishes. Therefore, there has been a need for a technology for stabilizing the dispersion of oily soils that takes into account the rinsing process specific to dishwashers.

[0011] The present invention provides a detergent composition for dishwashers that has excellent detergency and excellent finish on tableware. In particular, the present invention provides a detergent composition for dishwashers that has excellent detergency on stubborn stains such as oil and starch, and excellent finish on tableware made of plastic or glass.

[0012] The present invention relates to a method for promoting an enzymatic reaction, which comprises reacting a substrate with an enzyme in the presence of (a) a branched alkyl sulfosuccinate ester having a branched alkyl group having from 9 to 12 carbon atoms or a salt thereof.

[0013] The present invention also relates to an enzyme reaction accelerator containing, as an active ingredient, (a) a sulfosuccinic acid branched alkyl ester having a branched alkyl group having from 9 to 12 carbon atoms or a salt thereof.

[0014] The present invention also relates to an enzyme reaction accelerator composition comprising (a) an enzyme reaction accelerator containing, as an active ingredient, a sulfosuccinic acid branched alkyl ester having a branched alkyl group having from 9 to 12 carbon atoms or a salt thereof [hereinafter referred to as component (a)], and optionally a surfactant other than component (a) [hereinafter referred to as component (b)], wherein, when component (b) is contained, the mass ratio (a) / (b) of the content of component (a) to the content of component (b) is 0.001 or more and 50 or less.

[0015] The present invention also relates to a dishwasher detergent composition containing (a) a sulfosuccinic acid branched alkyl ester having a branched alkyl group having from 9 to 12 carbon atoms or a salt thereof [hereinafter referred to as component (a)], and (d) an enzyme [hereinafter referred to as component (d)].

[0016] According to the present invention, there is provided a method for promoting an enzymatic reaction, for example, an enzymatic reaction in an aqueous medium.

[0017] Furthermore, the present invention can provide a detergent composition for dishwashers that has excellent detergency and excellent finish on tableware. In particular, the present invention can provide a detergent composition for dishwashers that has excellent detergency on stubborn stains such as oil and starch, and excellent finish on tableware made of plastic or glass.

[0018] Modes for Carrying Out the Invention [Method for Promoting an Enzyme Reaction] In the method for promoting an enzymatic reaction of the present invention, a substrate is reacted with an enzyme in the presence of component (a), preferably in the presence of component (a) and water.

[0019] Sulfosuccinate esters or salts thereof are known as components incorporated into detergent compositions for hard articles, as described in Patent Document 3, for example. However, it was not known in the art that compounds having a structure corresponding to component (a) of the present invention can significantly promote enzymatic reactions with various enzymes such as proteases. For example, as shown in the comparative examples described below, a compound having a structure similar to component (a), but with a branched alkyl group having 8 carbon atoms, does not exhibit the excellent effect of improving enzymatic reactions as component (a) of the present invention. In other words, even compounds having a structure similar to component (a) have significant differences in their effects on enzymes, which is an unexpected effect that those skilled in the art could not have predicted. Enzymes are used in various industrial fields, such as the cleaning field, and in such cases, they are often used in combination with other chemicals. The ability to promote enzymatic reactions and improve their effects leads to a reduction in the amounts of surfactants and solvents used, for example, and can reduce CO 2 The inventors believe that this will also contribute to the realization of a sustainable society in terms of reducing CO2 emissions.

[0020] The mechanism by which the enzymatic reaction is promoted by the present invention is presumed to be, but not limited to, the following: In the presence of component (a), the spreading and wetting of the reaction substrate proceeds with high efficiency, making it possible to promote the reactivity of the enzyme contained in water with the substrate. According to the present invention, the use of other surfactants and solvents that cause a decrease in enzyme activity can be minimized, thereby maintaining high enzyme activity and allowing the catalytic reaction by the enzyme to be sustained.

[0188] Examples <Example 1a and Comparative Example 1a> A triglyceride with a triolein / tristearin ratio of 8.5 / 1.5 (by mass) was dissolved in chloroform to prepare a model stain. A 75 mm (width) x 100 mm (length) x 1 mm (thickness) polypropylene test piece was uniformly coated with the model stain at a coating weight of 0.1 g, and the chloroform was evaporated and dried to prepare a stained piece. The aqueous medium shown in Table 1 was placed in a beaker, and the stained piece was immersed so that the entire area coated with the model stain was in contact with the aqueous medium. The temperature of the aqueous medium was 25°C. After immersion for 10 minutes, hydrochloric acid was added to inactivate the enzyme. The stained piece was removed and rinsed with running distilled water for 15 seconds. All of the water used during rinsing was recovered. After rinsing, the remaining stain on the stained piece was washed away with 50 ml of chloroform, and the resulting mixture was placed in a separatory funnel together with the water recovered during rinsing. After shaking several times, the lower chloroform layer was recovered. Thereafter, 25 ml of chloroform was added again, and after shaking several times again, the chloroform layer was recovered. This operation was repeated three times, after which the recovered chloroform was transferred to a recovery flask, and the chloroform was evaporated using an evaporator. 10 ml of methanol was then added to the recovery flask to dissolve the extract, and the amount of triglyceride (total amount of triolein and tristearin) after immersion was quantified by LC / MS. The triglyceride decomposition rate (I) was calculated from the triglyceride amount of the entire immersion blend using the following formula. The results are shown in Table 1. A higher triglyceride decomposition rate (I) indicates a more accelerated enzymatic reaction. Triglyceride decomposition rate (I) (%) = (total amount of triglycerides before immersion - total amount of triglycerides after immersion) / total amount of triglycerides before immersion × 100

[0189]

[0190] The components in the table are as follows: The mass % of each component in the table is based on the amount of the active ingredient (the same applies hereinafter). Component (a') is a comparative component to component (a). a-1: Sodium bis-(2-propylheptyl) sulfosuccinate a'-1: Sodium bis-(2-ethylhexyl) sulfosuccinate LAS: Sodium dodecylbenzenesulfonate, manufactured by FUJIFILM Wako Pure Chemical Industries, Ltd. AS: Sodium dodecyl sulfate, manufactured by FUJIFILM Wako Pure Chemical Industries, Ltd. FA: Sodium laurate, manufactured by FUJIFILM Wako Pure Chemical Industries, Ltd. AG: Alkyl polyglucoside, product name "Plantacare 2000UP", manufactured by BASF SE, alkyl group having 8 to 16 carbon atoms, glucose condensation degree 1 to 2 Betaine: Lauryl hydroxysulfobetaine, Amphitol 20HD, manufactured by Kao Corporation Amine oxide (1): Lauryl dimethylamine oxide, Amphitol 20N, manufactured by Kao Corporation Amine oxide (2): Octyl dimethylamine oxide, Amphitol 08N, manufactured by Kao Corporation Lipase: product name "Lipex Evity 100L", manufactured by Novozymes, molecular weight approximately 30,000

[0191] Example 2a The aqueous medium shown in Table 2 was filled into a pressure-accumulating foam discharge trigger-type spray container (Cucute Clear Foam Spray, manufactured by Kao Corporation) and sprayed five times onto soiled pieces prepared in the same manner as in Example 1a. The aqueous medium was discharged in foam form. The total amount of aqueous medium discharged was approximately 3 g. The components in Table 2 are the same as those used in Example 1a. After contacting the discharged foam with the soiled pieces for 1 minute, the pieces were rinsed with running distilled water for 15 seconds. The ambient temperature at this time was 25°C. All of the water used during rinsing was recovered. After rinsing, the remaining dirt on the plate was washed away with 50 ml of chloroform, and the mixture was placed in a separatory funnel together with the water recovered during rinsing. After shaking several times, the lower chloroform layer was recovered. 25 ml of chloroform was then added again, and the mixture was shaken several times again, after which the chloroform layer was recovered. This procedure was repeated three times, after which the recovered chloroform was transferred to a recovery flask and evaporated using an evaporator. Then, 10 ml of 2-propanol was added to the recovery flask to dissolve the extract. The amounts of each component, triolein (a), tristearin (b), diolein (c), distearin (d), monoolein (e), monostearin (f), oleic acid (g), stearic acid (h), and glycerin (i), were quantified by LC / MS based on their peak areas. The triglyceride decomposition rate (II) was calculated from the amounts of each component using the following formula. The results are shown in Table 2. A higher triglyceride decomposition rate (II) indicates a more accelerated enzymatic reaction. Triglyceride decomposition rate (II) (%) = {(c) + (d) + (e) + (f) + (g) + (h) + (i)} / {(a) + (b) + (c) + (d) + (e) + (f) + (g) + (h) + (i)} × 100

[0192]

[0193] In Table 2, the triglyceride decomposition rate (II) was high in the Examples using an aqueous medium containing component (b). This is thought to be because the quality of the foam formed from the aqueous medium differs depending on whether or not component (b) is present. That is, the foam produced from an aqueous medium containing component (b) has a finer texture and a longer lasting foam state than the foam produced from an aqueous medium not containing component (b). This is presumably because the opportunity for contact between the substrate and the enzyme, whose activity has been increased by component (a), is increased, resulting in a higher triglyceride decomposition rate (II).

Electrophoresis was performed at 40 mA for 80 minutes (WSE-1100PageRun-R (manufactured by ATTO Corporation)). One-step Ruby (manufactured by Aprosciences) was used to stain the gel. Fluorescent images of the gel were taken, and the band intensity in the 50-80 kDa region was measured from the acquired images using image analysis software (manufactured by Bio-Rad). The BSA degradation rate was calculated as an index of enzyme activity using the following formula, and the value of the BSA degradation rate was evaluated as follows. The results are shown in Table 3. In this evaluation, a BSA degradation rate of 30% or more is preferable, and a larger value is more preferable. BSA degradation rate (%) = 100 - (band intensity after degradation / undegraded band intensity) x 100

[0197] In the table, the hardness of the aqueous medium is German hardness (° dH) measured by the following method. The hardness in the table was adjusted by dilution to the required hardness of the aqueous medium using a mother liquor of a 4000° DH hardness solution prepared by mixing 1 L of water, 83.9 g of calcium chloride dihydrate, and 29.0 g of magnesium chloride hexahydrate. In this specification, German hardness (° dH) refers to the concentration of calcium and magnesium in water expressed as CaCO 3German hardness refers to a concentration expressed as 1 mg / L (ppm) = approximately 0.056°dH (1°dH = 17.8 ppm). The calcium and magnesium concentrations for this German hardness are determined by chelate titration using disodium ethylenediaminetetraacetic acid. The specific method for measuring German hardness of aqueous media in this specification is described below. <Method for Measuring German Hardness of Aqueous Media> [Reagents] 0.01 mol / L EDTA·2Na solution: 0.01 mol / L aqueous solution of disodium ethylenediaminetetraacetate (titration solution, 0.01 M EDTA-Na2, manufactured by Sigma-Aldrich) Universal BT indicator (product name: Universal BT, manufactured by Dojindo Laboratories, Inc.) Ammonia buffer solution for hardness measurement (67.5 g of ammonium chloride was dissolved in 570 ml of 28 w / v% ammonia water, and the total volume was adjusted to 1000 ml with ion-exchanged water) [Hardness Measurement] (1) 20 ml of the sample aqueous medium was placed in a conical beaker using a volumetric pipette. (2) 2 ml of ammonia buffer solution for hardness measurement was added. (3) 0.5 ml of Universal BT indicator was added. It was confirmed that the solution after addition was reddish purple. (4) While shaking the conical beaker well, 0.01 mol / L EDTA·2Na solution was added dropwise from the burette, and the end point of the titration was the point when the aqueous medium sample turned blue. (5) The total hardness was calculated using the following formula: Hardness (°dH) = T × 0.01 × F × 56.0774 × 100 / A T: Titration volume (mL) of 0.01 mol / L EDTA·2Na solution A: Sample volume (20 mL, volume of aqueous medium sample) F: Factor of 0.01 mol / L EDTA·2Na solution

[0200] <Method for Evaluating Cleaning Performance> (1) Method for Evaluating Cleaning Performance for Rice Staining Contaminated bowls were prepared by applying 0.5 g of rice stain to a ceramic bowl and letting it dry overnight. Two contaminated bowls were placed inside an automatic dishwasher (Panasonic, model number: NP-45MS8S), with an identical, uncontaminated bowl placed inside to prevent direct contact with water. Six grams of the dishwasher detergent composition shown in the table were added, and the bowls were washed using the standard cycle. After washing, the remaining stains were visualized by staining with iodine, and the bowls were then rated according to the following criteria. The concentration of component (a) in the washing solution (formed by diluting the dishwasher detergent composition with water) used for the dishes was 10 ppm when 6 g of the composition was added. *Criteria for evaluating cleaning performance for rice staining: 3: The stain was completely removed. 2: Most of the stain was removed, but some remained. 1: Some stain remained, and the stain was mostly visible.

[0201] (2) Evaluation Method for Protein Cleaning Performance (Finish) (2-1) Method for Creating Protein Stain After preparing a soft-boiled egg (with the yolk soft-boiled and solidified) in a frying pan, only the egg liquid was collected using a strainer. (2-2) Evaluation Method Two clean glass cups were placed in an automatic dishwasher, and 10 g of the soft-boiled egg collected in (2-1) and 6 g of the dishwasher detergent composition shown in the table were added, followed by washing on the standard cycle. After washing, the dishes were scored based on the following criteria. The concentration of component (a) in the washing liquid applied to the dishes (the washing liquid formed by diluting the dishwasher detergent composition with water) was 10 ppm when 6 g of the composition was added. *Evaluation Criteria for Protein Cleaning Performance 3: Approximately 10 water spots and almost no dullness. 2: Approximately 20 to 50 water spots, or approximately 25% of the total surface is white and dull. 1: More than half of the surface is dull.

[0202] (3) Evaluation Method for Oily Stain Cleaning Performance Four polypropylene dishes were placed in an automatic dishwasher. Two of the dishes were contaminated with a composite soiling mixture of lard and rapeseed oil (1:1 by mass) at 3 g per dish, while the remaining two were uncontaminated. Six grams of the dishwasher detergent composition shown in the table was added, and the dishes were washed using the standard cycle. A total of three panelists touched the washed dishes and rated them according to the following criteria. Tables 4 and 5 show the average scores, rounded to the nearest whole number. The concentration of component (a) in the washing liquid (a washing liquid formed by diluting the dishwasher detergent composition with water) applied to the dishes was 10 ppm when 6 g of the composition was added. *Evaluation Criteria for Oily Stain Cleaning Performance 4: The stains were removed, and the dish felt a pleasant, squeaky feeling when touched. 3: The stains were removed, but the dish felt no squeaky feeling when touched. 2: Most of the dirt appears to have been removed, but rubbing with a finger leaves streaks. 1: Oil remains.

[0203] <Method for measuring pH> The dishwasher detergent composition of each Example and Comparative Example was diluted with water to a concentration of 0.2 mass %, and the pH of the cleaning liquid at 25° C. was measured by the glass electrode method. The results are shown in Tables 4 and 5.

[0204] <Method of measuring viscosity> The viscosity at 25°C of the dishwasher detergent compositions of the Examples and Comparative Examples in Table 5 was measured using a Brookfield viscometer (rotor No. 2, 6 rpm, measurement time 1 minute, liquid temperature 25°C). The results are shown in Table 5.

[0205]

[0206] Comparing Example 1b-1 with Comparative Examples 1b-1 and 1b-3 in Table 4, it is presumed that component (a) of the present invention further improves the decomposition reaction of the substrate (dirt) by component (d). Although Table 4 does not include an enzyme (lipase) that acts on oil, the high oil cleaning performance of Example 1b-1 and Comparative Example 1b-2 is thought to be due to the high washing temperature of the automatic dishwasher and the oil stain removal and dispersion effects based on the incorporation of component (a).

[0207]

[0208] In Table 5, comparing Example 2b-1 with Comparative Examples 2b-2 and 2b-5, it is inferred that the (a) component of the present invention further improves the decomposition reaction of the substrate (dirt) by the (d) component. In this evaluation, there was no difference in the cleaning performance of rice stains between Examples 2b-1 to 2b-3 and Comparative Example 2b-5. This is because the amylase used here was relatively easy to perform in a liquid composition, so no apparent difference occurred in this evaluation. For example, when evaluated under more severe cleaning conditions (such as when the cleaning solution is difficult to contact with the substrate during packed washing, or when targeting complex soiling containing rice and lipids), it is thought that differences may occur between these Examples and Comparative Examples. Furthermore, in Table 5, no enzyme (lipase) that acts on oil is blended. However, the high oil stain cleaning performance of Examples 2b-1 to 2b-3 and Comparative Example 2b-1 is thought to be due to the high washing temperature of the automatic dishwasher and the oil stain removal and dispersion effect based on the blending of component (a).

[0209] In Tables 4 and 5, the blending amount of component (d) is expressed as mass % in terms of enzyme protein.
