Abstract

The present utility model provides a novel diagnostic tool for the detection of Salmonella enterica serovar Typhimurium. Specifically, the utility model relates to a developed and optimized simple and fast loop-mediated isothermal amplification (LAMP)-based assay parallel with optimized conventional PCR assay to detect Salmonella Typhimurium targeting the STM4497 gene. A critical feature of the utility model is the utilization of closed-tube protocols calcein-based LAMP detection assay for visualization in post-enrichment steps to prevent cross-contamination events. Another critical feature is the incorporation of optimized betaine concentration to prevent non-specific amplifications. The method is specifically designed for a simple, sensitive, specific and fast surveillance method for S. Typhimurium that lessens the turnaround time and shorter amplification time than conventional methods.
