Description

Cell freezing medium for high-throughput cryopreservation of Chinese hamster ovary cells Technical Field

[0001] The present invention relates to the field of cell preservation, and in particular to a cryopreservation solution for high-throughput deep-low temperature preservation of Chinese hamster ovary cells and a cryopreservation method using the cryopreservation solution. Background Art

[0002] Cryopreservation is one of the primary methods for cell preservation. Using cryopreservation technology to store cells at deep temperatures in liquid nitrogen allows for long-term storage. Furthermore, high-quality cryopreservation ensures that cells retain the same pre-cryopreservation characteristics upon thawing, such as highly stable and consistent cell doubling times and the quantity and quality of expressed products. This provides the necessary support for the long-term stability of production and experimental work.

[0003] In the field of biopharmaceuticals, engineered cell lines are important raw materials for production and experiments. Chinese Hamster Ovary (CHO) cells are currently an important host cell line for protein production and have many advantages over other expression systems. An increasing number of pharmaceutical proteins have been efficiently expressed in CHO cells and commercialized. The effective preservation and culture of CHO cells play a vital role in the stability, continuity, reproducibility and controllability of production. Large-scale, standardized CHO cell banks with long-term storage stability can continuously provide companies with CHO cells that stably express target proteins. The storage stability of such cell banks mainly depends on the quality of cell freezing, among which the cell freezing medium is an important factor affecting the quality of cell freezing.

[0004] Currently, cell cryopreservation often uses cryopreservation fluid containing serum components for cell cryopreservation, and the number of cells frozen is generally around 20-50. For biopharmaceutical companies, the scale of a cell bank of 20-50 cells cannot meet their long-term stable and continuous production needs. At the same time, the cost of cell bank preparation and testing is high, especially the time period for external testing of cell banks is long. Therefore, there is still a demand in this field for high-throughput (for example, more than 200 cells) and high-quality cell cryopreservation technology.

[0005] The inventors of the present invention have conducted research on establishing a high-throughput standardized cell bank for Chinese hamster ovary cells. The core of the research is to provide a cell freezing solution for high-throughput cell freezing, and a method for freezing cells using this cell freezing solution.

[0051] In some embodiments, the serum-free cryopreservation solution in the present application is particularly suitable for high-throughput deep-low temperature preservation of CHO cells. In the process of laboratory and industrial cell freezing, the sedimentation of cells in the cryopreservation solution limits the number of cryopreservation tubes that can be frozen at one time, making it difficult to obtain a large-scale, highly consistent and long-term storage stable CHO cell bank. The serum-free cryopreservation solution in the present application, based on the combination and content of its added components, unexpectedly enables cells to be stably suspended in the cryopreservation solution for a long time (e.g., 1, 1.5, 2, 2.5, 3 hours), so as to allow large-scale cell freezing at one time. Specifically, cryopreservation solution can be provided for a CHO cell bank containing 200, 300, 400, 450, or 500 or even more cryopreservation tubes (2 mL / tube) at one time, thereby achieving high-throughput freezing. At the same time, the cryopreservation solution of the present application allows the initial freezing concentration of the frozen cell suspension (i.e., the cell suspension after the Chinese hamster ovary cells and the cryopreservation solution are mixed) to be greater than 1×10 7 cells / mL, for example, 5×10 7 cells / mL, 6×10 7 cells / mL, 7×10 7 cells / mL, 8×10 7 cells / mL, 9×10 7 cells / mL, 1×10 8 cells / mL, 2×10 8 cells / mL, 3×10 8 cells / mL, 4×10 8 cells / mL or 5×10 8 The initial freezing concentration of the cell suspension containing Chinese hamster ovary cells and the freezing solution of the present application is much higher than the initial concentration of the cell suspension frozen with the general freezing solution in the art.

[0052] Another aspect of the present application provides a method for cryopreserving Chinese hamster ovary cells, comprising cryopreserving the Chinese hamster ovary cells using the serum-free cryopreservation solution disclosed herein, mixing the Chinese hamster ovary cells with the cryopreservation solution to form a cell suspension, cooling the cell suspension to below -80°C within 90 minutes, and then transferring the cell suspension to liquid nitrogen for storage. The cell cryopreservation method provided herein precisely controls the changes in the cell suspension temperature and the ambient temperature by setting multiple temperature change steps (including the starting and ending temperatures of the temperature change, as well as the cooling rate), achieving strong controllability and fully protecting the cell performance from damage during the cryopreservation process.

[0053] In certain embodiments, the cell suspension is cooled from 2°C, or 3°C, or 5°C, or 6°C, or 7°C, or 8°C to below -80°C within 90 minutes. In certain embodiments, the cell suspension is cooled from 4°C to -80°C within 90 minutes. In certain embodiments, the cell suspension is cooled from 4°C to -90°C within 90 minutes. In certain embodiments, the cell suspension is cooled from 4°C to -100°C within 90 minutes. In certain embodiments, the cooling to below -80°C is completed within 90 minutes. [0110] (4) Results and analysis

[0111] The cell activities after recovery of each control example and experimental example are shown in Table 3 below.

[0112] Table 3 Cell activity after thawing of frozen cells in control and experimental examples

[0113] Compared with Control Example 1 and Control Example 2, the addition of 0.05M trehalose and 0.1M trehalose, 2g / L methylcellulose and 5g / L methylcellulose in the experimental examples with single components (i.e., Experimental Examples 1-1 to 1-6) had a certain degree of effect on improving cell recovery activity, but the effect was limited.

[0114] In the experimental groups to which trehalose and HSA were added in combination (i.e., Experimental Examples 1-7 to 1-9), the cell recovery activity was improved to a certain extent. In the experimental groups to which trehalose, HSA and methylcellulose were added in combination (i.e., Experimental Examples 1-10 to 1-14), the cell recovery activity was significantly improved. Specifically, in the experimental groups to which trehalose, HSA and methylcellulose were added in combination (i.e., Experimental Examples 1-10 to 1-14), the cell recovery activity was higher than 95%, and even reached 97.8%. In addition to having a better freezing effect, the freezing solution of the present invention is very suitable for large-scale cell freezing in production due to its low cost and ease of operation.

[0115] Example 2

[0116] The preparation of the freezing solution, cell freezing, cell recovery, and activity testing procedures were consistent with those of Example 1. A recombinant CHO-2 cell line (the CHO cells expressing the CD147 antibody comprising a heavy chain variable region and a light chain variable region, wherein the heavy chain variable region comprises the amino acid sequence shown in SEQ ID NO: 11 and the light chain variable region comprises the amino acid sequence shown in SEQ ID NO: 12) was used to study the applicability of the freezing solution formulation. The volume ratio of serum-free medium and DMSO in the freezing solution, as well as the final concentrations of trehalose, human albumin, and methylcellulose, were shown in Table 4 below.

[0117] Table 4 Components and concentrations of cell freezing solution

[0118] Note: "S3CD" in Table 4 is a serum-free medium prepared using S3CD medium according to step (4) in Example 1 above, wherein the CD CHO medium of ThermoFisher Scientific in the serum-free medium in step (4) of "Preparation of Cryopreservative Solution" in Example 1 is replaced with S3CD medium of Shanghai Duoning Biotechnology Co., Ltd., product number MNP014.

[0119] Table 5 shows the cell viability of cells cryopreserved using the freezing solutions of Experimental Examples 2-1 to 2-5 after cell thawing. Table 5 shows that the cell thawing activities of cells cryopreserved using the five freezing solution formulations were all high. This indicates that these freezing solution formulations can achieve similarly good cryopreservation results for different CHO cell types.

[0120] Table 5 Cell activity after thawing of frozen cells in experimental example

[0121] Example 3

[0122] The freezing solutions of Experimental Examples 1-11 and 1-14 were used to freeze recombinant CHO-1 and recombinant CHO-2 cells, respectively, according to the steps and methods in Example 1. The frozen cells were revived and evaluated after 24 hours, 1 month, 3 months, 6 months, 1 year, and 3 years of freezing, respectively, to investigate the effect of the freezing formula on the long-term storage stability of the cells. Specifically, at 24 hours, 1 month, 3 months, 6 months, 1 year, and 3 years of freezing, 3 cryopreservation tubes were taken out and the frozen cells were revived, and indicators such as cell activity, cell morphology, doubling period, and the binding activity of the target gene (nucleic acid sequence encoding anti-CD-147 antibody) and the antibody produced were investigated. Tables 6 and 7 below list the stability investigation results of freezing recombinant CHO-1 cells using the freezing solution of Experimental Examples 1-11 and freezing recombinant CHO-2 cells using the freezing solution of Experimental Examples 1-14, respectively.

[0123] Table 6 Stability study results of recombinant CHO-1 cells frozen using the freezing solution of Experimental Examples 1-11

[0124] Note: The expression level of anti-CD147 antibody in recombinant CHO-1 cells before freezing is 2.5±0.2g / L.

[0125] Table 7 Stability study results of recombinant CHO-2 cells frozen in cryopreservation solution of Experimental Examples 1-14

[0126] Note: The expression level of anti-CD147 antibody in recombinant CHO-2 cells before freezing is 3.5±0.2g / L.

[0127] As shown in Tables 6 and 7, cells frozen using the freezing solution formulations of Experimental Examples 1-11 and 1-14 recovered 24 hours, 1 month, 3 months, 6 months, 1 year, and 3 years after freezing. The cells showed good morphology, stable doubling time, and the target gene sequence was consistent with the theoretical sequence. The antibody expression level and antibody binding activity were minimally affected by freezing. It can be seen that the cell freezing solution containing 10% (v / v) DMSO + 0.05M trehalose + 10g / L HSA + 2g / L MC and the cell freezing solution containing 7.5% (v / v) DMSO + 0.1M trehalose + 5g / L HSA + 2g / L MC played a better role in the long-term deep cryopreservation of CHO cells.

[0128] At the same time, the cell activity of cells frozen using the freezing solution formulas of Experimental Examples 1-11 and Experimental Examples 1-14 after thawing is shown in Figure 1. As can be seen from Figure 1, the cells frozen using the freezing solution formulas of Experimental Examples 1-11 and Experimental Examples 1-14 were revived after 24 hours, 1 month, 3 months, 6 months, 1 year, and 3 years of freezing, and the cell activity was greater than 90%, which is much higher than the requirement of cell thawing activity > 80% in the "Chinese Pharmacopoeia" (2020 Edition, Part III).
