Description

SOLID FORMS OF AN NLRP3 INHIBITOR AND USES THEREOFRELATED APPLICATION

[0001] This application claims priority to, and the benefit of, International Application PCT / CN2023 / 118568, filed September 13, 2023, the entire contents of which are incorporated herein by reference.BACKGROUND

[0002] Innate immune responses are mediated by different types of receptors termed patternrecognition receptors (PRRs). PRRs recognize the presence of pathogen -associated molecular patterns (PAMPs) and damage-associated molecular patterns (DAMPs). Once engaged these receptors trigger the activation of downstream inflammatory pathways that will help resolve injury. However, in many instances this activation can be uncontrolled and leads to disease.

[0003] The inflammasomes represent a class of PRRs that are crucial components of the innate immune response. Activation of the inflammasomes trigger a cascade of events that releases IL- 1 (3, IL- 18, and promotes an inflammatory form of cell death called pyroptosis induced by the activation of Gasdermin. Pyroptosis is a unique form of inflammatory cell death that leads to the release of not only cytokines but also other intracellular components that promote a broader immune response both of the innate and acquired immune system. Thus, inflammasome activation is a major regulatory of the inflammatory cascade.

[0004] NLRP3 is the most characterized inflammasome and has been shown to be critical in innate immunity and inflammatory responses. While several other NLR complexes, such as NLRC4, are activated under very specific circumstances, NLRP3 can be activated by numerous stimuli and should be seen as a sensor of intracellular homeostatic imbalance. Therefore, its precise functioning is essential. In addition to playing a role in host immune defense, dysregulation of NLRP3 has been linked to the pathogenesis of many inflammatory disorders. These include genetic diseases such as cryopyrin-associated periodic syndromes (CAPS) which is caused by gain-of-fimction mutations in the NLRP3 gene, as well as many prevalent neurologic and systemic diseases. Importantly, NLRP3 hyperactivation has been demonstrated pre-clinically to play a critical role in a plethora of inflammatory and degenerative diseases including, Non-Alcoholic Steatohepatitis (NASH), atherosclerosis and other cardiovascular diseases, Alzheimer’s disease, Parkinson’s disease, diabetes, gout, and numerous other autoinflammatory diseases. See, e.g., Li et al., European Journal of Pharmacology (2022) 928: 175091; Nguyen et al., Journal of Parkinson ’s Disease (2022) 72:2117- 2133; Su et al., Current Medicinal Chemistry (2021) 25:569-582; Zahid et al., Frontiers in Immunology (2019) 10:2538.SUMMARY

The software used for data collection wasDIFFRAC. COMMANDER version 6.5.0.1 and the data was presented using DIFFRAC.EVA version 4.2.1.11. XRPD diffractograms were acquired under ambient conditions (25 °C) via transmission on a 96 well plate. The data collection range was 3.0 - 40.0 °20 (degrees 2-theta) with a step size of 0.020 °20 (degrees 2-theta) and a collection time of 0.20 seconds per step. Samples were prepared by placing the powder or slurry onto a 96 well plate.Single Crystal X-ray Diffraction (SXRD)

[0322] A suitable crystal was selected and mounted on a MITIGEN holder in perfluoroether oil. Data was collected on a Rigaku 007HF diffractometer with HF Varimax confocal mirrors, an UG2 goniometer, and HyPix 6000HE detector at 100(2) K using Cu-Ka radiation (1.54178 A), with the crystal kept at a steady T= 100(2) K during data collection.Polarized Light Microscopy

[0323] Optical microscopy was performed using the Nikon Eclipse LV100ND Optical Microscope equipped with 5, 10, 20, 50 and 100x objective lens. The microscope is coupled with a PROMICRA, PROMICAM 3-5CP digital camera. The camera is controlled by the QuickPHOTO MICRO 3.2 software. The camera software measurement tool is calibrated using a transmitted Illumination slide (76 x 26 mm, crossed scale - 20 mm divided into 0.1 mm divisions, 200 divisions of 0.1 mm (100 pm)). Samples were prepared by distributing a powder over a microscope slide.Differential Scanning Calorimetry (DSC)

[0324] DSC data was collected on a TA Instruments Q2000 DSC. A predefined amount of the sample, 2.0 to 10.0 mg, was placed in an aluminum pan and heated at 10 °C / minute from 40 °C to 300 °C or varied as experimentation dictated. A purge of dry nitrogen at 100 mb / minute was maintained over the sample. The instrument control and data acquisition are acquired using Q Advantage software release version 5.5.23. The data was processed and presented using the TA Universal Analysis 2000 software version 4.5A build 4.5.0.5.Dynamic Vapor Sorption (DVS)

[0325] Dynamic Vapor Sorption (DVS) was carried out using the TA Instruments Q5000 SA. A predefined amount of the sample, 2.0 to 10.0 mg, was placed in a platinum pan. The sample was allowed to equilibrate at 50 °C at 0% RH for a period of 60 minutes. The sample was then equilibrated at 25 °C before ramping the humidity from 0 to 95% RH at 5% increments every hour. A similar ramp profile was used for desorption cycle. XRPD analysis was also performed on post DVS samples. Mass change by DVS is related to hygroscopicity.Thermal Gravimetric Analysis (TGA)

[0327] Column: Poroshell 120 Phenyl-Hexyl, 150 x 4.6 mm, 2.7 pm; Flow Rate: 1.0 mL / min;Column Temperature: 40 °C; Mobile Phase A: Water + 0.1% (v / v) trifluoroacetic acid (TFA); Mobile Phase B: Acetonitrile + 0.05% (v / v) TFA; Run Time: 22 minutes; Injection Volume: 5 pL; Detection: UV at 262 nm. Gradient: see Table A.Karl Fisher Titration

[0328] KF titration for water determination was performed using a Mitsubishi Moisture meter Model CA-200. Aquamicron AX or CXU was used as the reagent. Samples of approximately 100 mg were weighed in a glass weighing boat (variable W). After transferring the sample into the KR titration cell, the empty glass weighing boat was weighed (variable w). Direct titration was performed in duplicate, and mean water content was reported in % w / w.

[0329] The water content in % w / w in the sample was calculated according to the following equation: Water content of sample (% w / w) = (M / W-w) x (100 / 1,000,000), where M = amount of water measured in the sample titration (ug); W = weight of the weighing boat and sample (g); w = weight of empty weighing boat after transferring sample to the KF titration cell (g); (W -w) = weight of material transferred (g); 1,000,000 = factor to convert ug to g; and 100 = factor to convert result to 0 / / o.Example 1. Crystalline Form I and II Freebases(i) Form I / II mixture

[0330] 40 g of Compound 1 free base was dissolved in 330 mb ethanol (EtOH), and the solution was separated by chiral HPLC with the following conditions: CHIRAL ART Amylose-SA, 7*25cm,10um; mobile phase A: hexanes, mobile phase B: ethanol (EtOH) with 0.1% 7M NH3in methanol(MeOH); isocratic 50% B in 15 min; detector: UV 220 nm. The purified Compound 1 was obtained as light-yellow solid after concentration, which was then suspended in 200 mL n-heptanes and treated by wet milling at 20-25 °C. The solid was filtered, washed with 40 mL n-heptanes, and dried at approximately 50 °C under vacuum.

[0331] The solid was determined to be a mixture of two crystalline Compound 1 freebase forms: designated Form I and Form II freebases by XRPD. Competitive slurry experiments were performedby stirring a saturated suspension of the Form I / II freebase mixture in either methanol (MeOH) or ethanol (EtOH) at 500 RPM at 25 °C for a period of time. For example, in one experiment, approximately 200 mg of the Form I / II freebase mixture was added to a 7 mb vial. An appropriate amount of MeOH was added to form a slurry and the vial was capped and stirred at 25 °C at 500 RPM for 24 hours. The solids were then filtered and analyzed by XRPD, confirming Form II freebase reverted to Form I freebase, thereby identifying Form I freebase as the more thermodynamically stable form of the two forms. See FIG. 1A, comparing the XRPD of the Form I / II freebase mixture to the XRPD after the slurry experiment; see FIG. 2A, providing the XRPD of pure-phase Form I freebase; and see FIG. IB, identifying the signals attributed to Form II freebase. See also Table B(it) Form I

[0332] Single crystals suitable for single crystal X-ray diffraction (SXRD) analysis were obtained from dissolving 40 mg of Compound 1 freebase in 20% v / v methanol / isopropyl acetate. The sample was temperature cycled between 0-50 °C at 5 °C / min heating rate and 0. 1 °C / min cooling rate and 20 minutes isotherm at 0 °C after each cycle. A total of 15 cycles were applied. Thin block like crystals approximately 50-190 pm in length were obtained and analyzed by polarized light microscopy (PLM). See FIG 2D. A comparison of a simulated XRPD of the single crystal to the experimental Form I freebase XRPD, confirming the single crystals are consistent with, and thus correspond to, Form I freebase. See FIG. 2E.

[0333] Table D provides the crystal structure data and structural refinement parameters from the SXRD analysis. The crystal structure was refined into the tetragonal space group P4;,2|2 (no. 96). The asymmetric unit comprises one molecule of Compound 1 freebase, with eight asymmetric units in the unit cell and with no solvent accessible voids. See FIGS. 2F-2H, providing the crystalline packing viewed down the a axis (FIG. 2F), b axis (FIG. 2G), and c axis (FIG. 2H).

[0334] Large-scale preparation of Form I from ethanol / water, followed by ethanol / n-heptanes, provided bulk crystalline material having a particle size distribution diameter DIO to D90 range of about 9 microns to about 142 microns, and having a particle size distribution mean D50 value of about 22 microns. See Table E. The large-scale preparation is as follows: (1) Charge Compound 1 freebase (5 kg) and water (15 V). (2) Slurry for at least 2 hours. (3) Filter, wash w / 2 V water. (4) Recharge water-wet cake to reactor. (5) Charge water (2.5 V minus water on cake), and ethanol (42.5 V). (6) Polishing fdtration. (7) Concentrate to 5.5 V at <50 °C. (8) Add ethanol (10 V) and concentrate to 5.5 V. (9) Add ethanol (5 V), concentrate to 5.5 V. (10) Target <2.5% wt / v water. (11) Charge n-heptanes (11 V). (12) Filter, wash with 2 V of 1:2 ethanol-n-heptanes twice. (13) Dry at 45 °C. and (14) Store at 15-25 °C.

[0335] Thermogravimetric Analysis (TGA) of Form I shows a decomposition temperature of270.5 °C. No significant weight loss was observed between 20-150 °C, corroborating Form I freebase is anhydrous and not solvated. See FIG. 2B.

[0336] Differential Scanning Calorimetry (DSC) of Form I shows no endothermic events observed in the DSC scan between 30-170 °C, further corroborating Form I is anhydrous and not solvated. The endothermic melt was observed at 205.8 °C (onset), with a peak at 207.2 °C, and a heat of fusion of 123.8 J / g. See FIG. 2C.

[0337] Dynamic Vapor Sorption (DVS) indicated Form I had water uptake of 2.5% weight gain between 0-80% relative humidity (RH), with <1% weight gain between 30-60% RH, but that all the adsorbed water was lost during the drying cycle, indicating Form I is reversibly hygroscopic. XRPD of the material after the DVS analysis showed that the Form I crystalline phase was unchanged, supporting the position that Form I is a stable crystalline form.

[0338] Long term stability studies (at 25 °C ± 2 °C / 60% ± 5% relative humidity (RH)) and accelerated stability studies (at 40 °C ± 2 °C / 75% ± 5% RH) were also conducted on Form I freebase. See Tables F and G, respectively. Assay (% w / w) and purity levels (% area) were determined following HPLC Assay Method 1, and water content determined by Karl Fisher titration. As shown, the Form I freebase form is maintained over a period of 3 months under both low and high stress conditions, confirming Form I freebase is stable and exhibits low water absorption.(iii) Form II

[0339] 'H NMR analysis confirmed that the crystalline Form I / II freebase mixture contained approximately 0.3 molar equivalents of ethanol (EtOH). Data not shown. Taken together with the above analysis of Form I freebase, the NMR result suggests Form II freebase is an EtOH solvate, containing EtOH trapped in the Form II freebase crystalline lattice.

[0340] Thermogravimetric Analysis (TGA) and Differential Scanning Calorimetry (DSC) of the Form I / II freebase mixture further supports the suggestion that Form II freebase is an EtOH solvate.

[0341] For example, TGA of the Form I / II mixture demonstrated approximately 1.7% weight loss attributed to residual EtOH solvent loss and another 3.5% step weight loss attributed to loss of approximately 0.3 equivalents of bound EtOH (trapped solvent), followed by onset of thermal decomposition at 275 °C. See FIG. 1C, lower scan.

[0342] DSC of the Form I / II freebase mixture demonstrated a broad endothermic event with onset temperature at 115 °C and peak at 117 °C, and the high temperature at which this event occurred suggested solvent loss, and that the solvent was strongly bound within the crystalline lattice. This endothermic event was followed by a second endothermic melt with onset at 201 °C and peak at 204 °C, with a heat of fusion of 96.08 J / g. See FIG. 1C, upper scan. This second endothermic melt is alsoconsistent with conversion of Form II freebase into Form I freebase, as Form I freebase exhibited a similar endothermic melt. Compare the upper scan, second endothermic event of FIG. 1C to the single endothermic event observed in the DSC of pure Form I provided in FIG. 2C.

[0343] Form I / II freebase mixture experienced approximately 4.3% water uptake between 0-80% relative humidity (RH) by Dynamic Vapor Sorption (DVS), demonstrating the mixture is hygroscopic, a result which is also consistent with the DVS analysis of pure Form I freebase, finding Form I freebase to be reversibly hygroscopic. The XRPD post-DVS analysis also confirmed Form II freebase had fully desolvated to Form I freebase. See Form I / II freebase post-DVS XRPD in FIG. 1A.Example 2. Crystalline Form III

[0344] Approximately 150 mg of a mixture of Form I / II freebase was added to a 14 mL vial. 11 mL of 27.3% v / v methanol (MeOH) / 2 -butanol were added to fully dissolve solids. The vial was then uncapped and left to fast evaporate at 50 °C under N2 flow until all solvents were evaporated. The crystalline solid was collected and analyzed by XRPD to confirm a new freebase form of Compound 1, referred to herein as Form III freebase. See FIG. 3A for the XRPD of Form III freebase. See also Table H, providing the XRPD signals for Form III freebase.

[0345] Differential scanning calorimetry (DSC) of Form III freebase showed a single endothermic event at 200 °C (onset), suggesting Form III freebase is anhydrous and non-solvated, with a peak at 204 °C, and heat of fusion of 123.7 J / g, See FIG. 3B forthe DSC scan of Form III freebase.

[0346] Competitive slurry experiments were performed with a mixture of Form I / III freebase at 5 °C, 25 °C and 50 °C in methanol (MeOH). All solids isolated after 24 hours had fully converted to Form I freebase, as determined by XRPD, thereby identifying Form I freebase as the more thermodynamically stable form of the two forms.OTHER EMBODIMENTS
