[0007] In recent years, formyl peptide receptors (FPR1, FPR2, FPR3) as therapeutic targets are gaining importance due to their useful biological properties. The FPRs are G protein-coupled receptors that are activated via formylpeptides recognition and then transduce chemotactic signals in phagocytes and mediate host-defense as well as inflammatory responses including cell adhesion, directed migration, granule release and superoxide production. The restoration of body homeostasis after injuries or pathogen infections is critical to ensure the survival of an organism. The physiological wound healing and innate immune responses are initiated by the release of soluble mediators from the invading pathogen or injured lesions. The temporally regulated interactive repairing processes involve, for example, many chemokines, cytokines, acute phase proteins, infiltrating and residential cells, fibroblasts, nerve cells, and vasculature. If the injury persists or is of an extensive magnitude, the physiological wound repairing or anti-infection responses can become pathological, leading to excessive inflammation, edema, unwarranted fibrogenic repair, organ dysfunction, acute respiratory distress syndrome (ARDS) , sepsis, ultimately organ failure and / or death. Therefore, effective regulations of the magnitude and the duration of the inflammation and resolution responses can be critical in the injury repair. After tissue injury or pathogen infection (by bacteria, virus, fungus, and / or microbes) , a set of formyl-peptides, damage-associated molecular pattern molecules (DMAPs) , inflammatory lipid mediators (such as leukotrienes and lipoxins) , and acute phase proteins (such as annexins) are released from the invading pathogens, the injured cells, and the lesion tissues. And the three FPR receptors serve as the key sensors for these chemotactic and activating molecules in humans and they are highly expressed on neutrophils, macrophages, T lymphocytes, dendritic cells, epithelial cells, fibroblasts, microglia, and astrocytes. The binding of these chemo-active molecules and acute proteins to the FPR receptors recruit leukocytes, stimulate superoxide and cytokine production, activate microglia, astrocytes, and other inflammatory and resolution responses for injury repair and host defense.

[0478] DPPH reagent (Solarbio, D9370-250mg) stock solution (20 mM) and compounds stock solution were prepared with DMSO. Then, 10 μL / well of compounds (final concentration 100 μM) and 190 μL / well DPPH reagent (1: 100 dilution from DPPH stock solution with 70%ethanol, pH 4.0) were added into the 96-well plate (Costar, 3590) , respectively, and further incubated at room temperature for 5h. OD value at 515 nm was recorded and the antioxidant effect of testing compounds (compared to vehicle control) was calculated by the following equations:

[0485] IMR-32 cells were plated in 96-well plate (Corning, 3603) and test compounds are prepared from stock solution with corresponding culture medium. Cells were incubated with test compounds (final concentration: 10 μM) for 1h under 37℃ and 0.5 μM fMLP was added into 96-well plate, except for the control wells, for another 1h under 37℃. Then, 10 uM DCFH-DA was added to the plate for another 30min under 37℃ and fluorescence intensity was measured with an emission at 538 and excitation at 485 nm by a fluorescence plate reader. The antioxidant effect of testing compounds (compared to vehicle control) was calculated by the following equations: Antioxidant effect (%) = { (F0-F1)  / F0} x100, Where F0 is the Fluorescence intensity of the vehicle control, and F1 is the Fluorescence intensity of the testing compounds. The results of cellular antioxidant assays are shown in the following Table 3.
