[0002] This application is based on the PCT application with Application No. PCT / CN2023 / 143381, filed on December 29, 2023, and PCT / CN2024 / 140003 filed on December 17, 2024, and claims priority to the aforementioned applications, the entire contents of which are hereby incorporated by reference into this application.Technical Field

[0003] The present disclosure relates to engineered T cell receptors (TCRs) , specifically to TCR-based bispecific / multispecific fusion proteins comprising one or more additional antigen-binding domains, the compositions thereof, and methods of using the same.Background

[0004] T cell receptors (TCRs) are the adaptive immune system’s tool for recognizing and eliminating “non-self” intracellular antigens. One TCR is composed of two different protein chains. In humans, the majority T cells express TCRs consisting of alpha (α) chain and beta (β) chain (encoded by TRA and TRB, respectively) , whereas a small population of T cells express TCRs consisting of gamma (γ) and delta (δ) chains (encoded by TRG and TRD, respectively) . α / β TCRs are expressed on both CD8+ and CD4+ T cells and recognize peptide antigens displayed on cell surfaces when complexed with human leukocyte antigens (HLA)  / major histocompatibility complex (MHC) and this interaction induces T cell activation and elimination of tumor cells displaying the specific MHC / peptide complexes.

[0005] It is widely established that tumor immune-rejection is T cell mediated and the antitumor T cell response is antigen-specific1. Approaches of TCR-engineered adoptive cell transfer (ACT) treatment showed objective tumor regressions in the clinic2 3 4. Alternatively, the use of the soluble extracellular region of the TCR fused to an antibody fragment that recognizes an activating TCR subunit, commonly CD3ε, has been used to redirect endogenous T cells to target tumor cells5 6 7. The first TCR / anti-CD3 bispecific fusion protein Tebentafusp showed clinical benefits in uveal melanoma patients8 and was approved by FDA recently.

[0006] However, TCR assembly and expression remains challenging. α / β TCRs express poorly with less reliable assembly than antibodies5. Tebentafusp molecule is constructed with “ImmTac” technology which fuses an anti-CD3 scFv to the c-terminus of an optimized heterodimeric α / β TCR. But the molecule is expressed in a complex Escherichia coli (E. coli) expression system. Instead of heterodimeric α / β TCRs, some researchers have tried using only the variable domain regions of TCRs in a single chain format or scTv, like an antibody single chain Fv (scFv) , for targeting specific HLA / peptide complexes. While scFvs have shown a propensity for instability, aggregation, and low solubility, scTvs have generally shown even worse expression and stability issues. Great efforts have been made to engineer scTvs for therapeutic and diagnostic use. Unfortunately, the high diversity of Vα / Vβ germlines (higher than antibody VH / VL germlines) , renders each set of stabilizing designs within scTvs to be unique to the individual Vα / Vβ subunit and unlikely to find general use.

[0007] Considering the expression challenges of TCR-based fusion proteins and its significance in treatment of cancers, there is still need in the art to develop alternative TCR-based fusion protein modalities which are not only potent in targeting antigen specific tumor cells but also suitable for industrial scale production in mammalian expression system.Summary of Invention

[0008] Provided herein is a TCR-Fab in tandem (T-FIT) fusion protein.

[0009] In some embodiments, the fusion protein comprises a T cell receptor (TCR) moiety and an immunoglobulin (Ig) moiety, wherein the TCR moiety comprises an antigen binding variable (V) region of a TCR, and the immunoglobulin moiety comprises an antigen binding portion (ABP) comprising a heavy chain variable region and a light chain variable region of an immunoglobulin.

[0298] The T2 cell line (Shanghai Jiya Biotechnology, XY-XB-3269) is a negative mutant for “Transporter associated with antigen processing” (TAP protein) , expressing an empty HLA-A*02: 01 allele of the MHC class I molecule on the cell surface. To confirm that MHC peptides indeed bind to HLA-A*02, T2 cell line is often used.

[0299] Briefly, T2 cells were washed once with growth medium (RPMI1640+10%FBS) , NY-ESO-1 peptide (SLLMWITQC) was added to cells at 6 ug / ml. After 3 hours of incubation in tissue culture incubator (37℃, 5%CO2) , cells were washed three times with PBS, then seeded at 300,000 cells per well into 96 well plate. After 10min of incubation with Fc blocker (Biolegend Cat#422302) at room temperature, 100ul test articles (10X serial dilution, from 30ug / ml to 0.03ug / ml) were added and gently mixed with pipette tips. After 45 minutes of incubation at 4℃, plates were washed to remove excess test articles. Fluorochrome-conjugated goat anti-human IgG antibody (Jackson Immunoresearch, Cat#109-606-098) was then added and incubated with cells at 4℃ for 20 minutes. After another round of wash, cells were resuspended in assay buffer (1X PBS containing 2%FBS) for reading on a flow-cytometer. The results shown in FIG. 12 illustrate the binding activity of FIT2022-29a to NY-ESO-1 peptide pulsed T2 cells.

[0314] Anti-tumor efficacy was evaluated in A375 xenograft model in PBMC engraft mice. Female NCG mice aged 8-10 weeks (Gempharmatech Co., Ltd) were used in this study. Briefly, 5x106 A375 cells suspended in 0.2ml DPBS containing 50%ABW Matrigel were subcutaneously injected into the right flank of mice. On the same day, these mice received a single intraperitoneal injection of 8x106 human PBMC. On 14 days post tumor cell inoculation, huCD45+ cells in mouse peripheral blood were detected by using FACS, indicating successful PBMC engraftment. Tumor bearing mice with tumor size of 130-200 mm3 were selected and randomly grouped (5 mice per group) . Treatment was initiated on the same day (Day 0) . Mice were administered intraperitoneally (i.p. ) with vehicle control (DPBS, Basal Media, Cat#B210KJ) or 16 mg / kg of FIT2022-30a on Day 0, Day 4 and Day 9. Tumor volumes were measured 2-3 times per week.

[0318] Binding kinetics and affinities of CD3 binding arm and pMHC binding arm were measured by grating coupled interferometry (GCI) on a WAVEsystem (Malvern Panalytical) according to the manufacturer’s instructions.

[0319] Briefly goat anti-human IgG Fc antibody was immobilized onto a PCP WAVEchip (Malvern Panalytical) to a density of ~6000 pg / mm2 by amine coupling. Antibodies were then captured onto the 4PCP WAVEchip to a density of 150-350 pg / mm2. For FIT2022-130a, recombinant human CD3ε / γ (KACTUS, Cat#CD3-HM157) was injected at concentrations of 246.9 pM, 740.7 pM, 2.2 nM, 6.7 nM and 20 nM respectively in kinetics buffer (GE, cat#BR100669) . For FIT-2022-132a and ABBV-184, human CD3ε / γ was injected at concentrations of 31.25 nM, 62.5 nM, 125 nM, 250 nM, 500 nM and 1000 nM respectively. Recombinant pMHC complex (KACTUS, cat#MHC-HM474T) was injected at concentrations of 274.3 pM, 823.0 pM, 2.5 nM, 7.4 nM, 22.2 nM, 66.7 nM and 200 nM respectively. After each round of association and dissociation, WAVEchip surface was regenerated by injection of Glycine-HCl buffer at pH 1.5 for 120s. All sensorgrams were recorded at 25 ℃ and data were analyzed with the WAVEcontrol software (Malvern Panalytical) . Data were double referenced by substracting the signals from blank injections and from the reference channel. A Langmuir 1: 1 model or a conformational change model were used for data fitting.
