[0004] 8p11 myeloproliferative syndrome (EMS) or stem cell leukemia / lymphoma syndrome (SCLL) is an aggressive myeloproliferative neoplasm (MPN) caused by rearrangement of an FGFR1 tyrosine kinase gene on chromosome 8p11-12, resulting in fusion of the FGFR1 gene with other partner genes. It has been recognized as a unique entity in the 2016 World Health Organization (WHO) classification. The most common of the above partner genes are ZNF198 on chromosome 13q11-12 and BCR on 22q11.2, and reported partner genes also include FOP on 6q27 and the like. These gene fusions lead to constitutive activation of the FGFR1 tyrosine kinase, thereby initiating the development and progression of hematological tumors. The clinical manifestations can progress to acute myeloid leukemia (AML) , T-cell lymphoblastic lymphoma (T-LBL) , chronic myelogenous leukemia (CML) , chronic myelomonocytic leukemia (CMML) , or mixed phenotype acute leukemia (MPAL) . In the 2022 WHO classification, EMS belongs to myeloid / lymphoid neoplasms with eosinophilia and tyrosine kinase gene fusions (MLN-TK) . To date, EMS has limited treatment options and a very unfavorable prognosis.
