[0002] INTRODUCTION
[0003] The invention relates to a feed additive composition suitable for supplementing the nutrition of livestock, especially poultry, to improve the physical constitution and the health of livestock during growing by strengthening the microbiome of the livestock’s digestive system. The feed additive composition is suitable for the prevention and treatment of dysbacteria infections and lameness resulting from dysbacteria infections in young livestock, can be produced by a simple method, and administered to the livestock during regular feeding procedures in a liquid or in a solid form.
[0004] BACKGROUND OF THE INVENTION
[0005] Successful rearing of livestock on a large scale requires a keen eye on the fed animals’ well-being, which is in a paramount part connected to their digestive tract. It is therefore apparent that diseases associated to an unstable intestinal microbiome and the overall quality of the applied animal feed may go hand in hand. Animals that are capable of utilizing their required nutrients in sufficient amounts and get the chance to develop a strong and balanced microbiome gain the most weight, convert feed better to body mass, stay healthy and are exposed to as little stress as possible.
[0006] Keeping large groups of animals together comes with a limitation on the available space. Such conditions naturally lead to an increased risk for outbursts of infections among the flock or herd. The risk for infections is especially prevalent during the early life stages of a livestock when the immune defense system and the digestive track of the animals is not fully developed yet. This can lead to cases in which microorganisms, that do not cause any infectious outbursts in fully grown animals with a stable immune system, can pose a serious risk for the health of the young livestock and in the further growing period.
[0007] For example, microbes of the genus Enterococcus (E.) are a commonly occurring species of the domain Bacteria known for their ability to produce lactic acid. This feature makes them valuable for the food industry, where they are e. g. utilized in the fermentation of cheeses. They naturally occur in the microbiome of the digestive system in animals as well as humans. Even E. species which are associated with diseases such as E. cecorum can be found under physiological conditions where they do not cause infections in healthy organisms.
[0008] However, there are cases where E. cecorum has been connected to problematic infectious outbursts in livestock. An increasing number of reports concerning young broilers that suffer from sudden paresis or lameness have emerged. Some of these animals may recover on their own, but others may show additional symptoms including brain damage or even death. It is generally assumed that these symptoms are caused by an infection with E. cecorum, transmitted from the digestive system of the parent flock via the eggshell surface. The protective cuticula naturally found on the eggshell can become damaged or even be missing altogether. This in turn can expose the not yet hatched broilers to E. cecorum before their digestive microbiome is stable enough to assimilate the Bacteria properly.
[0009] In the past, infections such as in the stated example have been treated with antibiotics, but there are a number of factors which call this approach into question. For one, the effect of the applied antibiotics is often only temporary, and reinfection occurs shortly after the medication is discontinued. Furthermore, many Bacteria which were predominantly treated in such a way have become resistant to usual antibiotics. Another factor which should not be underestimated is the growing concern in the population regarding animal products which are loaded with antibiotics.
[0010] Therefore, there is an ever increasing need to provide a measure to prevent and / or deal with infectious diseases among young livestock that does not involve treatment with antibiotics.
[0011] STATE OF THE ART
[0012] A number of approaches which involve feed additives dedicated to improving livestock health are known in the general state of the art.
[0013] The document US 2010 / 0233320 A1 concerns an animal feed kibble comprising a probiotic coating. Said kibble is intended for house animals such as dogs and cats and thus the actual kibble is protein based. However, the coating may be composed of compounds which further stimulate the animal’s digestive capability by introducing plant-based nutrients, vitamins and probiotic Bacteria strains.
[0014] A liquid cysteamine hydrochloride composition is described in the application CN 101366452 A. The composition is provided to poultry to increase its weight gain and comprises a cysteamine hydrochloride derivative, p-cyclodextrine, emulsifiers, amino acids as well as a choice of vitamins and nutrients.
[0015] The application CN 102144727 A discloses a liquid functional vitamin premix comprising emulsifiers, fat- and water-soluble vitamins, carbohydrates and nutrients. The invention of CN 102144727 A is intended to be applied to chickens and increases weight gain and the egg to feed ratio.

The method involves feeding said additive to a bred livestock species.

However, the application does not attempt to address multiple issues at once with a combined approach. The compositions may comprise coloring agents, carbohydrates, amino acids, emulsifiers, bacterial, yeast, or viral strains.
[0021] The application CN 116 172 134 A describes a probiotic granule pet feed for dogs and cats. The feed comprises meat and additives comprising starches, vegetables and fruits, nutritional supplements, probiotics and cellulose.

The feed additive is intended to promote pig health and lead to lean meat products. The contributions to the state of the art up until this point are mostly concerned with either introducing a beneficial bacterial species or providing a mixture of high energy nutrients to the livestock. However, so far no composition is available which strengthens the microbiome and also the feed conversion especially suited to livestock in its early life stages, particularly in poultry. Furthermore, none of the mentioned documents are concerned with the treatment of enterococcus-based infections.
[0024] OBJECT OF THE INVENTION
[0025] The objective of the present invention was to provide an improved feed additive composition for life stock. The novel feed additive composition should be suitable for easily supplementing the nutrition of livestock. The novel feed additive composition should be able to improve the physical constitution and the health of livestock during growing or to improve the intestinal health and feed utilization of livestock in its early life stages. In a particular aspect, a novel feed additive composition should be made available, which may serve as a substitute for antibiotic treatment regimens. The novel feed additive composition should in particular be suitable for supplementing the nutrition of poultry, especially chicks, in poultry growing.
[0026] The inventors of the present invention surprisingly found that a feed additive composition comprising a combination of oligosaccharides, small organic acids or their esters as well as emulsifiers improves the cultivation conditions for obligate anaerobic Bacteria in the digestive system of livestock while simultaneously increasing the nutrient utilization in said livestock. As short-chained saccharides undergo fast resorption in the upper gastrointestinal tract of livestock, usually animal feed contain longer-chain saccharides to achieve sufficiently slow resorption. For example, in pig breeding short-chain saccharides are not suitable due to the fast resorption. The inventors of the present invention found that surprisingly oligosaccharides can provide a nutritional benefit when administered in the novel animal feed without the disadvantage of fast resorption, in particular when administered to early-life livestock, such as particularly to poultry, like young chicks, in the first days or 1 or 2 weeks after hatching. With the surprising suitability of oligosaccharides, the novel feed additives can benefit from better solubility, which allows to more easily formulate the components when manufacturing the novel feed additive. This further allows more freedom in the choice of the form of the formulation and providing the feed additive in liquid form, e.g. for administration via drinking water, becomes possible.
[0027] The thusly achieved combined beneficial effect on the livestock’s health in its early life stages improves its immune response to infectious disease and leads to increased intestinal and common health in a similar manner as an antibiotic treatment regime without the disadvantages, side-effects and risks arising from antibiotic treatment. Furthermore, the feed conversion rate of the treated animals is in combination with better / dryer litter and consequently better footpad quality improved as well. BRIEF DESCRIPTION OF THE INVENTION

[0139] The present invention concerns a feed additive composition for improving the growing of livestock, preferably poultry. In the sense of the invention, growing of livestock means any form of breeding, keeping and / or farming a given livestock which involves an aspect of the livestock’s life wherein they increase in size, weight or both.
[0140] The feed additive composition of the invention is especially suitable for facilitating the food utilization of livestock in its very early live stages. For achieving the best results, the feed additive composition of the invention shall be administered to the livestock early after birth, most preferably starting on the first day after birth. Therewith, surprisingly, remarkable improvements in the livestock’s microbiome can be achieved and therewith the best possible feed can be provided to be optimally digested by the young livestock. The inventors of the present invention surprisingly found that with the feed additive compositions of the present invention the objects of the invention as described above can be solved. As previously mentioned, the choice of components in the additive composition of the invention provides excellently digestible nutrient sources to livestock, especially young livestock in its early live stages, and furthermore promotes the development of a healthy microbiome in such animals. The presence of a healthy microbiome increases the digestive capability of the livestock and thus naturally enhances the nutrient uptake. With the specific selection and combination of nutrients, the feed additive composition of the invention surprisingly allows to achieve the beneficial effects described herein.

The addition of choice probiotics can help to provide the microbiome with important microbes which increase the livestock’s resistance to the spread of disease.

Once said high pH-value diarrheas accumulate in the ground litter, the footpads of the livestock can be damaged due to the caustic environment. Therefore, the feed additive composition according to the invention does not only increase the livestocks health through a strong and infection resistant gut microbiome but also by improving the living conditions of the livestock by the described approach.

[0388] I. Feed conversion efficiency and weight gain: effects of different ingredients of the feed additive composition
[0389] Broiler chickens were supplied as day-old chicks from the same hatchery and reared on one fattening farm in Germany with enterococcal spondylitis history in four chicken houses (-40,000 birds each) for about five weeks (37 to 38 days). One identical conventional complete diet was offered ad libitum in all chicken houses. Chickens in each chicken house were treated either with Lincomycin / Spectinomycin (LincospectinOPulver, Zoetis Deutschland GmbH, Berlin, Germany, 250 g / 1000 L) or one of three different additives administered via drinking water during the first days of life. Chickens in the first chicken house were treated with lincomycin at the first day of life up to the age of two days (three days in total). At the first day of life up to the age of three days (four days in total), water soluble esterified butyrins were administered to the chickens in the second chicken house (0.12-0.24 mL / kg body weight). Over the same period, the chickens in the third chicken house were offered drinking water containing Bacillus licheniformis (70 g / 1000 L), while palm oil (2 L / 1000 L) was given to the chickens in the fourth chicken house during the first seven days of life. Table 3 provides an overview on the average body weights (g) measured at slaughterhouses at the respective days of life.

[0402] In a further study, three flocks comprising 40000 hatchlings which were 35 days old were kept in separate stables. The hatchlings were provided with a fat-enriched nutrition ad libitum day and had water at their disposable.

The conducted study highlights how advantageous effects occur through feeding of the composition according to the invention even in comparison of two flocks which have both been treated with antibiotics. While it is a goal of the invention to provide feed compositions which make the use of antibiotics become obsolete, some breeders may choose to utilize antibiotics for specific reasons. The herein described study shows that these breeders may benefit from the composition according to the invention as well.
[0409] In this study, two houses containing broiler flocks were investigated. One of the houses contained 24800 Cobb broiler on 1080 m2, the second house contained 11700 Cobb broiler on 510 m2. The broilers were provided from the same hatchery and both houses received the same feeds in the same amount each.

The microbes 4b1823i Bacillus velezensis ATCC PTA6737 (1011 CFUkg-1) and 4b1830 Clostridium butyricum FERM BP- 2789 (3 * 1010 CFU kg-1) were added to this mixture. Furthermore, Lysine and Methionine were added.

[0421] VI. Process for Preparing a Liquid Feed Additive Composition
[0422] This example describes the general process to produce a liquid feed additive composition according to the invention without specification of the utilized components.
[0423] In a first step, the necessary tools (such as mixing tanks, stirrers etc.) are thoroughly cleaned so as to prevent any contaminations. Thereafter, the chosen components are measured, i. e. with a scale or suitable volumetric device. Water and the emulsifier are provided to the mixing tank and the remaining components are slowly added to the solution while it is constantly stirred.

Claims
CLAIMS1 . The use of a composition comprising a combination of the following components: a) one or more food-safe oligosaccharides; and b) one or more food-safe organic acids and / or food-safe organic acid esters; and c) optionally one or more emulsifiers as a feed additive for poultry.
2. The use according to claim 1 , wherein a) the food-safe oligosaccharides are composed of linear or branched chains of 20 to 50 monosaccharides, preferably of 30 to 40 monosaccharides and are selected from mannan or fructans, preferably from fructans, more preferably from inulins; and b) wherein one or more food-safe organic acids and / or food-safe organic acid esters are selected from compounds according to formula (I),wherein R1is hydrogen, or a linear C1-C4 alkyl; and wherein R2is hydrogen, a linear or branched Ci-Ce alkyl or any saturated or unsaturated fatty acid; and c) wherein optional emulsifiers are selected from lecithin, ascorbyl palmitate, glycerides, and mixtures thereof, preferably at least one emulsifier is lecithin.
3. The use according to any one of claims 1 to 2, wherein the feed additive composition further comprises one or more amino acids, including arginine, glutamine, glutamic acid, lysine, methionine and mixtures thereof, preferred are arginine, glutamic acid and mixtures thereof.
4. The use according to any one of claims 1 to 3, wherein the feed additive composition further comprises one or more probiotics, wherein probiotics comprise one or more strains of the genus Bacillus, including: Bacillus subtilis, Bacillus lichiniformis, Bacillus amyloliquefaciens, Bacillus coagulans, and mixtures thereof, preferably one or more members of the Bacillus subtilis group, further preferably Bacillus velezensis; and / orprobiotics comprise one or more strains of the genus Bacteria, selected from one or more strains of Clostridium, including: Clostridium butyricum, Clostridium estertheticum, Clostridium pasteurianum, Clostridium putrefaciens, Clostridium sporogenes, Clostridium tyrobutyricum, and mixtures thereof, preferred is Clostridium butyricum; and / or preferably probiotics comprise combinations of probiotics selected from the genus Bacillus and selected from the genus Clostridium, further preferred are combinations of one or more members of the Bacillus subtilis group, preferably Bacillus velezensis with Clostridium butyricum.
5. The use according to any one of claims 1 to 4, wherein the feed additive composition comprises(a) one or more oligosaccharides selected from fructans, preferably inulin; and(b) one or more food-safe organic acids and / or food-safe organic acid esters selected from butyric acid ester, formic acid and mixtures thereof; and(c) one or more optional emulsifiers; and(d) one or more amino acids selected from arginine, glutamic acid and mixtures thereof; and(e) one or more probiotics selected from any Bacillus genus, Clostridium butyricum, and mixtures thereof, preferable is a mixture of one or more members of the Bacillus subtilis group, preferably Bacillus velezensis, with Clostridium butyricum.
6. The use according to any one of claims 1 to 3, wherein the feed additive composition further comprises a bile salt hydrolase inhibitor (BSH), including: KCI, riboflavin (vitamin B2), caffeic acid, and mixtures thereof.

The use according to claim 6, wherein the feed additive composition comprises(a) one or more oligosaccharides selected from fructans, preferably inulin; and(b) one or more food-safe organic acids and / or food-safe organic acid esters selected from butyric acid ester, formic acid and mixtures thereof; and(c) one or more emulsifiers; and(d) one or more amino acids selected from arginine, glutamic acid and mixtures thereof; and(e) bile salt hydrolase inhibitors selected from caffeic acid, KCI, riboflavin (vitamin B2) an mixtures thereof.
8. The use according to any one of claims 1 to 7, wherein the feed additive composition further comprises one or more higher order carboxylic acids or higher order carboxylic esters according to the following formula (II),wherein R3is hydrogen or a linear C5-C12 alkyl, preferably wherein R3is a C5, C7, C9 or C12 alkyl, and wherein R4is hydrogen, a linear or branched Ci-Ce alkyl or any saturated or unsaturated fatty acid.
9. The use according to any one of claims 1 to 8, wherein the feed additive composition is in a liquid form, preferably in the form of an emulsion.
10. The use according to any one of claims 1 to 4, wherein the feed additive composition is in a solid form, preferably in the form of a powder.11 . The use according to claim 10, wherein the feed additive composition comprises(a) one or more oligosaccharides selected from fructans, preferably inulin; and(b) one or more food-safe organic acids and / or food-safe organic acid esters selected from butyric acid calcium salts, encapsulated butyric acid and mixtures thereof; and(c) one or more amino acids selected from arginine, glutamic acid, and mixtures thereof; and(d) one or more probiotics selected from any Bacillus genus, Clostridium butyricum, and mixtures thereof, preferable is a mixture of one or more members of the Bacillus subtilis group, preferably Bacillus velezensis, and Clostridium butyricum.
12. The use according to claim 10 or 11 , wherein the feed additive further comprises one or more higher order carboxylic acids or higher order carboxylic esters as defined in claim 8.
13. The use according to claim 12, wherein the feed additive composition comprises(a) one or more oligosaccharides selected from fructans, preferably inulin; and(b) one or more food-safe organic acids and / or food-safe organic acid esters selected from butyric acid calcium salts, encapsulated butyric acid and mixtures thereof; and(c) one or more amino acids selected from arginine, glutamic acid, and mixtures thereof; and(d) one or more probiotics selected from any Bacillus genus, Clostridium butyricum, and mixtures thereof, preferable is a mixture one or more members of of the Bacillus subtilis group, preferably Bacillus velezensis, and Clostridium butyricum; and(e) one or more higher order carboxylic acids or higher order carboxylic esters selected from formula (II) embodiments wherein R3is a Cs, C7, C9 or C12 alkyl.
14. The use of the feed additive composition according to any one of claims 1 to 13 for supplementing the nutrition of poultry by admixing the feed additive composition to the nutrition of poultry for promoting the health and growth of poultry.
15. The use of the feed additive composition according to any one of claims 1 to 14 for supplementing the nutrition of poultry by admixing the feed additive composition to the nutrition of poultry for strengthening the microbiome of the digestive system of poultry.
16. The use according to claim 15 for improving the feed utilization of poultry, for improving the weight gain of poultry, for improving litter quality and foot pad health of the poultry and / or for reducing the late mortality rate of poultry.
17. The use according to claim 15 or 16 wherein the feed additive composition as defined in any one of claims 1 to 5, and 8 to 13 is first administered within 24 h after birth.
18. The use according to claim 15 or 16 wherein the feed additive composition as defined in claim 6, 7 or 8 is first administered not earlier than the 10thday following birth.
19. The feed additive composition according to any one of claims 1 to 13 for the use in the treatment and prevention of dysbacteriosis, lameness and / or paresis in poultry in its early life stages.
20. The feed additive composition for the use according to claim 19 in the treatment and prevention of dysbacteriosis, lameness and paresis caused by infections with E. cecorum in poultry in its early life stages and for the prevention of damaged footpads from high pH-value poultry excrements.
