Description

SALT-INDUCIBLE KINASES (SIK) INHIBITORS AND METHODS OF USES THEREOFCROSS-REFERENCE

[0001] This patent application claims the benefit of International Application No. PCT / CN2024 / 091925, filed May 09, 2024; which is incorporated herein by reference in its entirety.BACKGROUND

[0002] Adenosine Monophosphate-activated Protein Kinases (AMPK) belong to the protein kinase family, which comprises Salt-Inducible Kinases (SIKs) , a family of serine / threonine kinases widely expressed in the body, and involved in particular in cellular energy homeostasis. Three SIK isoforms have been identified, named SIK1 (also referred as SNFI-Like Kinase (SNF1LK) or Myocardial Snfl-like Kinase (MSK) ) , SIK2 (SNF1LK2 or KIAA0781) and SIK3 (KIAA0999) .

[0003] A role for SIK2 in T helper (Th) cell differentiation has recently been described through the regulation of IFNγ and IL-12 signaling, suggesting SIK2 may be an interesting target for inflammatory diseases.

[0004] Despite great advances over the past two decades in the treatments of patients affected by autoimmune disorders, based on antibodies targeting pro-inflammatory cytokines such as anti-TNFα, a significant proportion of patients do not respond to these therapies or experience serious adverse events such as opportunistic infections. Therefore, a large unmet medical need still exists for the treatment of these diseases, and new agents for the treatment of the above-mentioned diseases are needed.SUMMARY

[0005] Disclosed herein are compounds, or a pharmaceutically acceptable salt, or stereoisomer thereof, that are SIK inhibitors, and more particularly SIK1, SIK2 and / or SIK3 inhibitors.

[0006] Disclosed herein is a compound of Formula (I) or (II) , or a pharmaceutically acceptable salt, or stereoisomer thereof:

[0007] Also disclosed herein is a pharmaceutical composition comprising a compound disclosed herein, or a pharmaceutically acceptable salt, or stereoisomer thereof, and a pharmaceutically acceptable excipient.

[0008] Also disclosed herein is a method of inhibiting Salt-Inducible Kinase (SIK) activity in a subject in need thereof, comprising administering to the subject in need thereof a compound disclosed herein, or a pharmaceutically acceptable salt, or stereoisomer thereof, to the subject in need thereof. INCORPORATION BY REFERENCE

[0009] All publications, patents, and patent applications mentioned in this specification are herein incorporated by reference to the same extent as if each individual publication, patent, or patent application was specifically and individually indicated to be incorporated by reference.DETAILED DESCRIPTIONDefinitions

[0144] In certain embodiments, the compositions containing the compound (s) described herein are administered for therapeutic treatments. In certain therapeutic applications, the compositions are administered to a patient already suffering from a disease or condition, in an amount sufficient to cure or at least partially arrest at least one of the symptoms of the disease or condition. Amounts effective for this use depend on the severity and course of the disease or condition, previous therapy, the patient’s health status, weight, and response to the drugs, and the judgment of the treating physician. Therapeutically effective amounts are optionally determined by methods including, but not limited to, a dose escalation and / or dose ranging clinical trial.

[0145] In certain embodiments wherein the patient’s condition does not improve, upon the doctor’s discretion the administration of the compounds is administered chronically, that is, for an extended period of time, including throughout the duration of the patient’s life in order to ameliorate or otherwise control or limit the symptoms of the patient’s disease or condition.

[0146] Once improvement of the patient’s conditions has occurred, a maintenance dose is administered if necessary. Subsequently, in specific embodiments, the dosage, or the frequency of administration, or both, is reduced, as a function of the symptoms.

[0147] The amount of a given agent that corresponds to such an amount varies depending upon factors such as the particular compound, disease condition and its severity, the identity (e.g., weight, sex) of the subject or host in need of treatment, but nevertheless is determined according to the particular circumstances surrounding the case, including, e.g., the specific agent being administered, the route of administration, the condition being treated, and the subject or host being treated.

[0148] In some embodiments, doses employed for adult human treatment are typically in the range of 0.01 mg -5000 mg per day. In some embodiments, the daily dosages appropriate for the compound described herein, or a pharmaceutically acceptable salt thereof, are from about 0.01 to about 50 mg / kg per body weight. In various embodiments, the daily and unit dosages are altered depending on a number of variables including, but not limited to, the activity of the compound used, the disease or condition to be treated, the mode of administration, the requirements of the individual subject, the severity of the disease or condition being treated, and the judgment of the practitioner. [0150] In certain embodiments, a compound as described herein is administered in a local rather than systemic manner, for example, via injection of the compound directly into an organ, often in a depot preparation or sustained release formulation. In specific embodiments, long acting formulations are administered by implantation (for example subcutaneously or intramuscularly) or by intramuscular injection. Furthermore, in other embodiments, the drug is delivered in a targeted drug delivery system, for example, in a liposome coated with organ specific antibody. In such embodiments, the liposomes are targeted to and taken up selectively by the organ. In yet other embodiments, the compound as described herein is provided in the form of a rapid release formulation, in the form of an extended release formulation, or in the form of an intermediate release formulation. [0209] Added 5 μl ADP-Glo reagents. Centrifuged plate at 1000 g for 1 min, and incubated at room temperature for 60 min.

[0210] Added 10 μl kinase detection reagents. Centrifuged plate at 1000 g for 1 min, and incubated at room temperature for 60 min.

[0211] Centrifuged plate at 1000 g for 1 min.

[0212] Read luminescence signal on Envision 2104 plate reader. Data analysis

[0213] %Inhibition is calculated as follow:

[0214] Signal Ave_PC: The average for the positive controls across the plate. 1μM HG-9-91-01 group as positive control.

[0215] Signal Ave_VC: The average for the negative controls across the plate. DMSO group as negative control.

[0216] Calculated IC50 and Plot effect-dose curve of compounds.

[0217] Calculated IC50 by fitting %Inhibition values and log of compound concentrations to nonlinear regression (dose response -variable slope) with Graphpad: Y=Bottom + (Top-Bottom)  /  (1+10^ ( (LogIC50-X) *HillSlope) )  X:log of Inhibitor concentration; Y: %Inhibition.

[0218] The results are found in Table 3a ( [ATP] =1 mM) . Table 3
