[0154] Compound Dilution Scheme: All compounds supplied were prepared to a working stock of 50x final assay concentration in 100%DMSO. Where appropriate, more concentrated stocks were diluted manually to 50x using 100%DMSO. Compounds supplied as powders were reconstituted to a 10 mM stock in 100%DMSO before further dilution to 50x.

[0155] Assay Procedure: The required volume of the 50x stock of test compound was added to the assay before a reaction mix containing the enzyme and substrate was added. The reaction was initiated by the addition of ATP at the selected concentration. There was no pre-incubation of the compound with the enzyme / substrate mix prior to ATP addition. For further details of each individual assay, please refer to the website or the accompanying protocol document.

[0156] Data Analysis: Data are handled using a custom built in-house analysis software. Results are expressed as kinase activity remaining, as a percentage of the DMSO control. This is calculated using the following formula:

[0157] For IC50 determinations, data are analyzed using XLFit version 5.3 (ID Business Solutions) . Sigmoidal dose-response (variable slope) curves are fit based on the mean result for each test concentration using non-linear regression analysis. Where the top and / or bottom of the curve fall >10%out with 100 and 0, respectively, either or both of these limits may be constrained at 100 and 0, provided that the QC criterion on R2 is met.

[0158] Table 2 include IC50 values for TNIK of selected compounds; compounds having an IC50 value of 1-12 nM as +++, 12 –120 nM as ++, and > 120 nM as +. Table 2 Compounds and Assay Data Example B: Compound 112 in Pain Models Compound 112 were tested in various preclinical pain models to explore its analgesic effects. Unless noted otherwise, two-way (time course*compound treatment) or one-way (compound treatment only) ANOVA with Dunnett’s multiple comparison test is used for statistical analysis. Example B1: Rat and mouse spinal nerve ligation (SNL) model

[0159] SNL model is well established by surgically ligating the lumbar spinal nerves and representative of neuropathic pain, in which the paw withdrawal threshold (PWT) is largely decreased with mechanical stimulation in Von Frey test. Compound 112 was tested in both rat and mouse spinal nerve ligation (SNL) model to explore its analgesic effect. In the rat model, high (500 μg) and low (250 μg) intrathecal (i.t) injection of Compound 112 reduced hypersensitivity to mechanical stimulation (FIG. 1) . The model group underwent the SNL surgery; the control group underwent the same surgical procedure except for the nerve ligation step (sham surgery) .
