[0001] The present invention generally relates to a processes for preparing 3- [ (3aS, 6aR) -5- {5-chloro-2- [ (1-methyl-1H-pyrazol-4-yl) amino] pyrimidin-4-yl} -3a-methyl-octahydropyrrolo [3, 4-c] pyrrole-2-yl] -3-oxopropionitrile, a TYK2 inhibitor currently in clinical trials to treat autoimmune diseases. The present invention also relates to novel intermediates used in the process.BACKGROUND OF THE INVENTION

[0002] Janus kinase (JAK) is a non-receptor tyrosine protein kinase consisting of four family members, namely JAK1, JAK2, JAK3 and TYK2. Among them, TYK2 is involved in the signal transduction of inflammatory cytokines such as interferons (IFNs) , IL-12 and IL-23, and plays a key role in immunity.

[0005] The present invention relates to a process for preparing 3- [ (3aS, 6aR) -5- {5-chloro-2- [ (1-methyl-1H-pyrazol-4-yl) amino] pyrimidin-4-yl} -3a-methyl-octahydropyrrolo [3, 4-c] pyrrole-2-yl] -3-oxopropionitrile (Compound I) in high purity and good yield. The process is suitable for large-scale production (over 0.5 kg, preferably over 1 kg, over 2 kg, or over 5 kg) . The process provides purity of Compound I ≥ 90%, or ≥ 95%, or ≥ 98%, or ≥ 99%.

[0006] In a first aspect, the invention provides a process for preparing Compound I. The process comprises the steps of:

[0007] (a) 1 and 2 undergo a cycloaddition reaction in the presence of one or more suitable acids and one or more suitable solvents at -5~50℃ to obtain 3,

[0018] In step (a) , suitable acids include hydrochloric acid, sulfuric acid, perchloric acid, trifluoroacetic acid and trifluoromethanesulfonic acid. Solvents include dichloromethane, 1, 2-dichloroethane, toluene, xylene, tetrahydrofuran, 2-methyltetrahydrofuran, and acetonitrile. The reaction temperature is -5~50℃, or 0~40℃, or 0~30℃, preferably 0~25℃. Reaction time is usually 0.5 to 8 hours.

[0019] In step (b) , the one or more solvents used include water, toluene, xylene, acetic acid, isopropyl alcohol, n-butanol, tetrahydrofuran, 2-methyltetrahydrofuran and mixed solvents thereof. Ammonia or ammonium salts used include ammonium hydroxide, ammonia gas, ammonium acetate, ammonium formate, ammonium carbonate, ammonium bicarbonate and ammonium sulfate. The reaction temperature is 50~140℃, or 80~130℃, or 90~120℃, preferably 95~115℃. Reaction time is usually 8 to 24 hours.

[0020] Step (c) is a chiral separation step, and suitable chiral organic acids include but are not limited to D-tartaric acid, L-tartaric acid, D-camphoric acid, L-camphoric acid, D-malic acid, L-malic acid, D-camphorsulfonic acid, R-mandelic acid, D-dibenzoyltartaric acid, L-dibenzoyltartaric acid, D-di-p-methylbenzoyltartaric acid, L-di-p-methylbenzoyltartaric acid, D-di-paramethoxybenzoyltartaric acid, L-di-paramethoxybenzoyltartaric acid, and ascorbic acid. The solvent or solvents used include water, methanol, ethanol, isopropanol, acetone, acetonitrile and tetrahydrofuran.

[0021] Suitable reducing agents in step (d) include but are not limited to lithium aluminum hydride, diisobutylaluminum hydride (DIBAL) , red aluminum (Red-Al) , lithium borohydride, tri-sec-butyl potassium borohydride, tri-sec-butyl borohydride Sodium borohydride and lithium tri-sec-butylborohydride. Solvents used include tetrahydrofuran, 2-methyltetrahydrofuran, diethyl ether, methyl tert-butyl ether, dichloromethane, and toluene.

[0022] Suitable solvents in step (e) include methanol, ethanol, isopropyl alcohol, tetrahydrofuran, 2-methyltetrahydrofuran, dichloromethane, N, N-dimethylformamide and toluene. Additional organic bases or inorganic bases can also be added in step (e) , including but not limited to triethylamine, N, N-diisopropylethylamine, 1, 8-diazabicyclo [5.4.0] undec-7-ene (DBU) , pyridine, 2, 6-lutidine, imidazole, N-methylimidazole, N-methylmorpholine, sodium hydroxide, trisodium phosphate, disodium hydrogen phosphate, sodium bicarbonate, sodium carbonate, potassium carbonate and cesium carbonate.

[0023] Step (f) is a hydrogenation reaction, and the hydrogen pressure is usually between 0.1 and 4 MPa. Suitable catalysts typically are transition metals supported on different carriers (the metal content is usually between 1%and 20%) , including but not limited to palladium on carbon (Pd / C) , palladium hydroxide on carbon (Pd (OH) 2 / C) , palladium on alumina (Pd / Al2O3) , platinum on carbon (Pt / C) and platinum on alumina (Pt / Al2O3) ; a suitable catalyst can also be a transition metal oxide, such as platinum oxide (PtO2) , etc. Solvents used in step (f) include methanol, ethanol, isopropyl alcohol, tetrahydrofuran, 2-methyltetrahydrofuran, ethyl acetate and toluene. The reaction can be carried out in a conventional reactor or in a flow chemistry reactor. The reaction temperature is 20~100℃ or 30~80℃, preferably 40~70℃.

[0024] A suitable base in step (g) includes triethylamine, N, N-diisopropylethylamine, 1, 8-diazabicyclo [5.4.0] undec-7-ene (DBU) , pyridine, 2, 6-lutidine, imidazole, N-methylimidazole and N-methylmorpholine. Solvents used include methanol, ethanol, isopropyl alcohol, tetrahydrofuran, 2-methyltetrahydrofuran, dichloromethane, methyl tert-butyl ether, N, N-dimethylformamide, and acetonitrile. The reaction temperature is -10~50℃ or -10~30℃, preferably -5~5℃. Reaction time is usually 0.5 to 8 hours.

[0025] Different organic acids can be added to catalyze the reaction in step (h) , including but not limited to trifluoroacetic acid, trifluoromethanesulfonic acid, benzenesulfonic acid, p-toluenesulfonic acid and camphorsulfonic acid. Different inorganic acids can also be added to catalyze the reaction, including but not limited to hydrochloric acid, sulfuric acid, and phosphoric acid. Suitable solvents in step (h) include methanol, ethanol, isopropanol, tetrahydrofuran, 2-methyltetrahydrofuran, N, N-dimethylformamide and acetonitrile. The reaction temperature is 50~100℃ or 60~90℃, preferably 70~80℃. Reaction time is usually 8 to 48 hours.

[0026] Suitable bases in step (i) include, but are not limited to, triethylamine, N, N-diisopropylethylamine, 1, 8-diazabicyclo [5.4.0] undec-7-ene (DBU) , pyridine, 2, 6-lutidine, N-methylimidazole and N-methylmorpholine. Solvents used include methanol, ethanol, isopropanol, tetrahydrofuran, 2-methyltetrahydrofuran, N, N-dimethylformamide, and acetonitrile. The reaction temperature is 10~60℃ or 10~50℃, preferably 20~45℃. Reaction time is usually 4 to 48 hours.

[0035] Scheme 1 summarizes a process of preparing Compound I, which is shown in Examples 1 to 7.

[0036] Example 1. Preparation of 5-benzyl-3a-methyltetrahydropyrrolo [3, 4-c] pyrrole-1, 3 (2H, 3aH) -dione (4)

[0037] Toluene (208.8 kg) , trifluoroacetic acid (2.9 kg) and 1 (31.2 kg) were added to a reaction vessel sequentially. The mixture was stirred at 0~15℃ and then added with 2 (60.0 kg) dropwise over about 3.5 hours. After the addition was completed, the mixture was stirred at 15~25℃ for 0.5 hours and concentrated under reduced pressure at 20~30℃ for about 6 hours until no obvious fraction was collected. Toluene (104.4 kg) , acetic acid (189.0 kg) and ammonium acetate (23.4 kg) were added, and the mixture was heated to 100~110℃ and refluxed for 10 hours to separate water. After cooling to 40~45℃, the mixture was concentrated under reduced pressure until there was no obvious fraction collected and then cooled to 20~25℃. The mixture was added with water (180.0 kg) and ethyl acetate (162.0 kg) , stirred, and slowly added with NaOH solution to adjust the pH of the aqueous phase to 9~11. The organic phase was separated and discarded. The aqueous phase was added with ethyl acetate (162.0 kg) and adjusted the pH to 8~9 with 2 M HCl while stirring. The organic phase was separated, and the aqueous phase was extracted one more time with ethyl acetate (108.0 kg) . Two organic phases were combined, washed with 10%NaCl aqueous solution (120 kg) , dried over anhydrous magnesium sulfate (6 kg) , and filtered. The filtrate was concentrated at 45~50℃ to a volume of about 120 L and then slowly added with n-heptane (205.2 kg) . The mixture was concentrated to a volume of about 120 L again and then added with n-heptane (205.2 kg, 5 L / kg) . The resulting mixture was concentrated to a volume of about 300 L, stirred at 15~20℃ for 2~4 hours and filtered. The filter cake was dried at 40~45℃ for 16~24 h to obtain 4 (30.48 kg) with a yield of 50%.

[0038] Example 2. Preparation of (3aS, 6aR) -5-benzyl-3a-methyltetrahydropyrrolo [3, 4-c] pyrrole-1, 3 (2H, 3aH) -dione (5)

[0039] Acetonitrile (239.9 kg) , water (15.2 kg) and 4 (30.48 kg) were added to a reaction vessel sequentially. After stirring at 20~30℃, L-di-p-methylbenzoyltartaric acid (21.6 kg) was added to the reaction vessel in batches (about 5-6 kg / batch) . After stirring for 0.5 hours, the mixture was heated to 70~80℃ and refluxed for 3 hours. After gradually cooling to 25~35℃ (about 3-4 hours) , the mixture was stirred for 12~16 hours and then filtered. The filter cake was washed with acetonitrile (36.0 kg) and dried to obtain L-di-p-methylbenzoyltartaric acid salt of 5' (the enantiomer of 5) .

[0040] The above filtrate was concentrated at 40~45℃ until no obvious fraction was collected, cooled to 20~30℃ and added with ethyl acetate (82.3 kg) . The mixture was stirred for 0.5~1 hour, slowly added with saturated sodium bicarbonate aqueous solution (335.3 kg) and stirred for 1-2 hours. The organic phase was separated and the aqueous phase was extracted with ethyl acetate (54.9 kg) . The separated organic phases were combined and concentrated at 40~45℃ until no obvious fraction was collected. After cooling to 20~30℃, acetonitrile (239.9 kg) and water (15.2 kg) were added, and the mixture was stirred to dissolve. D-di-p-methylbenzoyltartaric acid (21.6 kg) was added in batches. The mixture was stirred for 0.5 hour, heated to reflux and stirred at 70~80℃ for 3 hours. After about 6-7 hours of gradient cooling to 30℃, the mixture was stirred for 12-16 hours and centrifuged. The filter cake was rinsed with acetonitrile (36.0 kg) , transferred to a reaction vessel, and added with acetonitrile (192.0 kg) and water (9.1 kg) . The mixture was heated to reflux and stirred at 70~80℃ for 1~2 hours. After gradually cooling to 20~30℃ over about 6~7 hours and stirring for 2~3 hours, the mixture was centrifuged, and the filter cake was rinsed with acetonitrile (72.0 kg) . The filter cake was transferred to a reactor, added with ethyl acetate (137.2 kg) and then slowly added with saturated sodium bicarbonate aqueous solution (228.8 kg) while stirring. After stirring for 1~2 hours, the organic phase was separated and the aqueous phase was extracted with ethyl acetate (54.9 kg) . The organic phases were combined, washed with 10%NaCl aqueous solution (63.0 kg) , dried over anhydrous magnesium sulfate (3.0 kg) and filtered. The filtrate was concentrated to about 60 L at 40~45℃, slowly added with n-heptane (104.2 kg) , and concentrated under reduced pressure to about 60 L. n-Heptane (104.2 kg, 5 L / kg) was slowly added, and the mixture was concentrated under reduced pressure to about 150 L. The mixture was cooled to 15~20℃, stirred for 2~3 hours and filtered. The filter cake was dried at 40~45℃ for 10-12 hours to obtain 5 (10.35 kg) with a yield of 34.0%.

[0041] Example 3. Preparation of tert-butyl (3aS, 6aR) -3a-methylhexahydropyrrolo [3, 4-c] pyrrole-2 (1H) -carboxylate (8)

[0042] Toluene (69.6 kg) and 5 (8.0 kg) were added to a reaction vessel. After stirring to dissolve, the solution was cooled to -5~5℃ and added with red aluminum (Red-Al, 70%toluene solution, 47.2 kg) dropwise over 3-4 hours. After the temperature was adjusted to 20~30℃, the solution was stirred for 0.5 hours, heated to 50~60℃ and stirred for 12~16 hours. After cooling to -10~0℃, the solution was added with 40%potassium sodium tartrate aqueous solution (66.6 kg) dropwise over about 2 hours and stirred for 0.5~1 hour. The toluene phase was separated, and the remaining was extracted with toluene (13.6 kg) . Two toluene phases were combined, washed with 10%NaCl aqueous solution (16.0 kg) , dried over anhydrous magnesium sulfate (1.2 kg) and filtered. The filtrate was concentrated to dryness to obtain 6.

[0048] Ethanol (48.4 kg) , maleate of 8 (16.52 kg) , N, N-diisopropylethylamine (21.4 kg) , and ethanol (18.7 kg) were added to a reaction vessel. The resulting mixture was stirred until dissolved. After cooling to -10~10℃, 9 (8.6 kg) was slowly dripped into the reaction vessel. After the addition was complete, the vessel was rinsed with ethanol (1.7 kg) and the reaction was continued at -10~10℃ for 0.5~72 hours. The temperature was raised to 15~35℃ and water (86 kg) was dripped into the vessel over 0.5~12 hours. After the temperature was lowered to -8~8℃ over 0.5~12 hours, the mixture was stirred for another hour and then filtered. The wet cake was washed with ethanol aqueous solution (30.6 kg, 1: 1 v / v) precooled to 0~5℃, and then vacuum dried at 40~60℃ and ≤-0.08 MPa for 18~120 hours to obtain 10 (15.26 kg) with a yield of 87.2%.

[0049] Example 6. Preparation of 5-chloro-N- (1-methyl-1H-pyrazol-4-yl) -4- ( (3aR, 6aS) -3a-methylhexahydropyrrolo [3, 4-c] pyrrol-2 (1H) -yl) pyrimidin-2-amine hydrochloride salt (12)

[0050] 10 (15 kg) , 11 (7.5 kg) and ethanol (118.5 kg) were added to a reaction vessel. The mixture was heated to 65~78℃ for 6-10 hours and then added with a solution of hydrochloric acid in ethanol (2 M, 1.80 kg) . After the reaction was continued for 10-60 hours, to the mixture was added ethyl acetate (112.5 kg) . The mixture was cooled to 15~35℃ over 0.5-24 hours and then stirred for 0.5-24 hours. The obtained mixture was centrifuged, and the wet product was washed with a mixed solvent of ethanol and ethyl acetate (51 kg, 1: 1 v / v) .

[0051] The obtained wet product was placed in a reactor and added with ethanol (94.8 kg) . After heating to 55~78℃, the mixture was stirred for 4-48 hours and then cooled to 15~35℃over 6 hours. After further stirring for 1-48 hours, the mixture was centrifuged and the wet cake was washed with a mixture of ethanol and ethyl acetate (51 kg, 1: 1 v / v) and vacuum dried at 40~70℃ and ≤-0.08 MPa for 24-144 hours to obtain 12 (14.755 kg) with a yield of 89.7%.

[0052] Example 7. Preparation of 3- [ (3aS, 6aR) -5- {5-chloro-2- [ (1-methyl-1H-pyrazol-4-yl) amino] pyrimidin-4-yl} -3a-methyl-octahydropyrrolo [3, 4-c] pyrrole-2-yl] -3-oxopropionitrile (Compound I)

[0053] Acetonitrile (17.1 kg) and DBU (14.6 kg) were added to a reaction vessel under a nitrogen atmosphere. 12 (14.5 kg) was slowly added at 35~55℃, and then 13 (8.12 kg) was used to rinse the reactor wall. After the addition, the temperature was controlled at 35~55℃and the mixture was stirred for 4~24 hours. Isopropanol (171.1 kg) was added to the vessel, the temperature was raised to 30~70℃, and the mixture was stirred for 1-24 hours. After the temperature was slowly lowered to -15~5℃ over 1-12 hours, the mixture was stirred for 2 hours and centrifuged. The wet cake was rinsed with isopropanol (58.0 kg) and vacuum dried at 40-60℃ and ≤-0.08 MPa for 6-192 hours to obtain I (12.6 kg) with a yield of 87.8%.

Claims

1.A method for preparing 3- [ (3aS, 6aR) -5- {5-chloro-2- [ (1-methyl-1H-pyrazol-4-yl) amino] pyrimidin-4-yl} -3a-methyl-octahydropyrrolo [3, 4-c] pyrrole-2-yl] -3-oxopropionitrile (Compound I) comprising the steps of:(a) reacting 1 and 2 to undergo a cycloaddition reaction in the presence of one or more acids at -5~50℃ to obtain 3,where PG is benzyl or substituted benzyl, preferably benzyl;(b) reacting 3 with ammonia or an ammonium salt at 50~140℃ to obtain 4,(c) chirally separating 4 using a chiral organic acid to obtain two enantiomer salts 5 and 5',(d) selecting and reducing 5 with a reducing agent to obtain 6,(e) reacting 6 with Boc anhydride to give 7,(f) deprotecting 7 in a hydrogen atmosphere in the presence of a catalyst to obtain 8 or its salt thereof,(g) reacting 8 or its salt with 9 in the presence of a base at -10~50℃ to obtain 10,(h) reacting 10 with 11 at 50~90℃ to obtain 12 or its salt,(i) reacting 12 or its salt with 13 in the presence of a base at 10~60℃ to obtain Compound I,wherein R1 is alkyl or cycloalkyl, wherein steps (a) - (i) occur in one or more suitable solvents that may be the same or different.2.The method according to claim 1, wherein the chiral organic acid used in step (c) is selected from the group consisting of: D-tartaric acid, L-tartaric acid, D-camphoric acid, L-camphoric acid, D-malic acid, L-malic acid, D-camphorsulfonic acid, R-mandelic acid, D-dibenzoyltartaric acid, L-dibenzoyltartaric acid, D-di-p-methylbenzoyltartaric acid, L-di-p-methylbenzoyltartaric acid, D-di-p-methoxybenzoyltartaric acid, L-di-p-methoxybenzoyltartaric acid, and ascorbic acid.3.The method according to claim 1, wherein the reducing agent used in step (d) is selected from the group consisting of: lithium aluminum hydride, diisobutylaluminum hydride (DIBAL) , red aluminum (Red-Al) , lithium borohydride, potassium tri-sec-butylborohydride, sodium tri-sec-butylborohydride, and lithium tri-sec-butylborohydride.4.The method according to claim 1, wherein the catalyst used in step (f) is palladium on carbon (Pd / C) , palladium hydroxide on carbon (Pd (OH) 2 / C) , palladium on alumina (Pd / Al2O3) , platinum on carbon (Pt / C) , platinum on alumina (Pt / Al2O3) , or platinum oxide (PtO2) .5.The method according to claim 1, wherein the base used in step (g) is selected from the group consisting of: triethylamine, N, N-diisopropylethylamine, 1, 8-diazabicyclo [5.4.0] undec-7-ene (DBU) , pyridine, 2, 6-lutidine, imidazole, N-methylimidazole, and N-methylmorpholine.6.The method according to claim 1, wherein the base used in step (i) is selected from the group consisting of: triethylamine, N, N-diisopropylethylamine, 1, 8-diazabicyclo [5.4.0] undec-7-ene (DBU) , pyridine, 2, 6-lutidine, N-methylimidazole, and N-methylmorpholine.7.A compound selected from the group consisting of where,x is from 0 to 1, y is from 0 to 2, and z is from 1 to 3;A1 is D-dibenzoyltartaric acid, D-di-p-methylbenzoyltartaric acid, or D-di-p-methoxybenzoyltartaric acid;A2 is L-dibenzoyltartaric acid, L-di-p-methylbenzoyltartaric acid, or L-di-p-methoxybenzoyltartaric acid, andA3 is maleic acid.
