Anti-ALPP / ALPPL2 Antibodies, Bispecific Binding Proteins and Uses ThereofTechnical Field

[0001] The present application relates to antibodies specifically binding to alkaline phosphatase, placental type (ALPP) and / or alkaline phosphatase placental-like 2 (ALPPL2) , and bispecific binding proteins targeting ALPP / ALPPL2. In addition, the present application further relates to the use of said antibodies and bispecific binding proteins, and methods of using said antibodies and bispecific binding proteins for treating diseases or disorders, such as hematopoietic cancers and solid tumors.Background

[0002] Alkaline phosphatase, placental type (ALPP, also known as placental alkaline phosphatase, PLAP) is a dimeric enzyme anchored by glycosyl phosphatidyl inositol (GPI) to cell membrane. It is thought to play a role in guiding migratory cells and transporting specific molecules across the plasma membrane (Dabare AA et al. Urol Int 1999; 63: 168-174) . Alkaline phosphatase placental-like 2 (ALPPL2, also known as alkaline phosphatase, germ cell, ALPG) is also a GPI-anchored membrane protein, and belongs to the human alkaline phosphatase family. ALPPL2 shares a 98%amino acid (aa) sequence identity with ALPP, 87%homology with the intestinal alkaline phosphatase (ALPI) , and 57%homology with the tissue-nonspecific liver / bone / kidney phosphatase ALPL (Su, Yang, et al. Cancer research 80.20 (2020) : 4552-4564) .

[0003] In normal tissues, ALPP and ALPPL2 are largely expressed in placenta. However, both ALPP and ALPPL2 are expressed at high levels in multiple cancers, including seminoma, ovarian cancer, endometrial cancer, gastric cancer, lung cancer, and pancreatic cancer (Hyrenius-Wittsten, Axel, et al. Science translational medicine 13.591 (2021) : eabd8836) .

[0004] The significant tissue specificity and broad tumor type coverage of ALPP / ALPPL2 render ALPP / ALPPL2 to be excellent target for cancer treatment. International publication NO. WO2017 / 095823 disclosed an anti-ALPP / ALPPL2 antibody, M25, generated by phage display, which was designed in ADC and CAR-T formats. The M25-based ADC showed cell killing activity against pancreatic cancer and lung cancer cell lines, also demonstrated strong tumor growth inhibition in a mesothelioma xenograft model. International publication No. WO2022 / 197890 disclosed an antibody-drug conjugate capable of binding both ALPP and ALPPL2 (referred to as ALPP / ALPPL2-directed ADC) , demonstrating in vitro tumor cytotoxicity and in vivo anti-tumor activity in CAOV3 / NCI-N87 / H1651 mouse models. International publication NO. WO2023 / 104938 disclosed a bispecific antibody comprising an anti-ALPP arm and an anti-CD3 antibody as the other arm, exhibiting T cell activation and tumor cell killing activities.

[0005] Given that ALPP / ALPPL2 is a promising target in cancer treatment and the tremendous therapeutic potential of antibodies, there remains a need in the art to develop diverse anti-ALPP / ALPPL2 molecules with desired properties, for example, specific binding potency, and / or binding sites and / or internalization properties. Additionally, there is a need to explore diversified antibody formats and to expand and / or improve their therapeutic utility and suitability for manufacturing. The present disclosure satisfies these needs to a certain extent.Summary

[0101] Definitions

[0102] Unless otherwise explained, all technical and scientific terms used herein have the same meaning as commonly understood by one of ordinary skill in the art to which this disclosure belongs. Other specifically defined terms are to be construed in a manner consistent with the definitions provided herein.

[0103] Unless otherwise required by context, singular terms shall include pluralities, and plural terms shall include the singular. Also, terms such as "element" or "component" encompass both elements and components comprising one unit and elements and components that comprise more than one subunit unless specifically stated otherwise.

[0104] The term “about” used in conjunction with a numerical value is intended to encompass the numerical values in a range from a lower limit less than the specified numerical value by 5%to an upper limit greater than the specified numerical value by 5%.

[0105] The use of "or" means "and / or" , unless stated otherwise. As used herein, the term “and / or” , when used in conjunction with more than one options, refers to any one of the options, or any two or more or all of the options.

[0106] As used herein, the term “comprise” , “include” or the variants thereof, such as “comprising” , “including” , or “comprised” , “included” is intended to mean that the described elements, integers or steps are included, but not to the exclusion of any other elements, integers or steps. The term “comprise” or “include” used herein, unless otherwise specified, also encompasses the situation where the entirety consists of the described elements, integers or steps. For example, when referring to an antibody variable region “comprising” a particular sequence, it is also intended to encompass an antibody variable region consisting of the particular sequence.

[0107] The term "polypeptide” refers to any polymeric chain of amino acids. The terms "peptide" and “protein” are used interchangeably with the term polypeptide and also refer to a polymeric chain of amino acids. The term "polypeptide" encompasses native or artificial proteins, protein fragments and polypeptide analogs of a protein amino acid sequence. The term "polypeptide” encompasses fragments and variants (including fragments of variants) thereof, unless otherwise contradicted by context. For an antigenic polypeptide, a fragment of polypeptide optionally contains at least one contiguous or nonlinear epitope of polypeptide. The precise boundaries of the at least one epitope fragment can be confirmed using ordinary skill in the art. The fragment comprises at least about 5 contiguous amino acids, such as at least about 10 contiguous amino acids, at least about 15 contiguous amino acids, or at least about 20 contiguous amino acids. A variant of a polypeptide is as described herein.

[0108] The term “isolated protein” or “isolated polypeptide” is a protein or polypeptide that by virtue of its origin or source of derivation is at least partially separated from naturally associated components that accompany it in its native state, is substantially free of other proteins from the same species, is expressed by a cell from a different species, or does not occur in nature. A polypeptide that is chemically synthesized or synthesized in a cellular system different from the cell from which it naturally originates may be “isolated” from its naturally associated components. A protein may also be rendered substantially free of naturally associated components by isolation, using protein purification techniques well known in the art. As measured by Size Exclusion Chromatography SEC-HPLC, an isolated protein may be of more than 80%, 85%, 90%, 95%or 99%purity.

[0109] As used herein, the term "antibody" refers to a binding protein comprising at least a light chain or heavy chain immunoglobulin variable region that specifically recognizes and binds to a particular antigen (e.g., CD3 or ALPP / ALPPL2) . The term encompasses various antibody structures, including, but not limited to, monoclonal antibodies, single-chain antibodies or multi-chain antibodies, single valent or multivalent antibodies, monospecific or multispecific antibodies, chimeric or humanized antibodies, complete antibodies, and antibody fragments, as long as they exhibit the desired antigen-binding activity. Therefore, the term "antibody" includes immunoglobulin molecules comprising four polypeptide chains, two heavy (H) chains and two light (L) chains inter-connected by disulfide bonds, as well as multimers thereof (e.g., IgM) . The term “antibody” also includes any functional fragment, mutant, variant, or derivation of such immunoglobulin molecules, which retains the essential epitope binding features. Such functional fragment, mutant, variant, or derivative antibody formats are known in the art, and non-limiting embodiments (e.g., FIT-Ig and MAT-Fab binding proteins) are discussed below.

[0110] As used herein, the term "immunoglobulin (Ig) " refers to heterodimeric proteins composed of two heavy (H) and two light (L) chains. Each heavy chain is comprised of a heavy chain variable region (abbreviated herein as VH) and a heavy chain constant region. The heavy chain constant region is comprised of three or four domains: CH1, CH2, CH3, and optionally CH4. Each light chain is comprised of a light chain variable region (abbreviated herein as VL, optionally Vκ) and a light chain constant region. The light chain constant region is comprised of one domain, CL. Constant regions are not directly involved in antibody-antigen binding, but they display a variety of effector functions. The light chains of antibodies can be assigned to one of two types, known as kappa (κ) and lambda (λ) , based on the amino acid sequences of light chain constant regions. The heavy chains of antibodies can be divided into five major types / classes, IgG, IgE, IgM, IgD, IgA and IgY depending on the amino acid sequences of heavy chain constant regions, and several of these types can be further divided into subtypes or subclass, such as IgG1, IgG2, IgG3, IgG4, IgA1 and IgA2.

[0111] As used herein, the term "variable region" or "variable domain" refers to the domain of the heavy or light chain of an antibody that is involved in binding the antibody to an antigen. Typically, a heavy chain variable domain (VH) is paired with a light chain variable domain (VL) to confer antigen binding specificity; however, in some cases, a single VH domain (e.g., a single VHH domain from a heavy chain antibody) is also sufficient to confer antigen binding specificity. The VH and VL regions can be further subdivided into regions of hypervariability, termed complementarity determining regions (CDRs) , interspersed with regions that are more conserved, termed framework regions (FRs) . Each VH and VL is comprised of three CDRs and four FRs, arranged from amino-terminus to carboxy-terminus in the following order: FR1-CDR1-FR2-CDR2-FR3-CD3-FR4. First, second and third CDRs of a VH domain are commonly enumerated as CDR-H1, CDR-H2, and CDR-H3, or HCDR1, HCDR2, and HCDR3; likewise, first, second and third CDRs of a VL domain are commonly enumerated as CDR-L1, CDR-L2, and CDR-L3, or LCDR1, LCDR2, and LCDR3.

[0112] In some aspects, the variable domain (s) of an antibody may be modified. For example, substitution, deletion and / or addition of one or more amino acid residues, especially conservative residue substitution, may be introduced into one or more CDR regions and / or one or more framework regions to obtain an antibody variant. Alternatively, the variable domain (s) of an antibody may be modified by CDR grafting. As is known in the art, CDRs are primarily responsible for antibody-antigen interactions. Antibody variants can be constructed by grafting CDR sequences from a known antibody onto the framework regions of a different antibody having a desired property. In addition, one to several mutations can be further performed, such as back mutations, to refine the desired properties of the antibody variant. The properties of antibody variants can be assessed in vitro or in vivo assays. Preferably, antibody variants substantially retain at least one desired biological property (e.g., antigen binding ability) of their parent antibodies.

[0113] The term "CDR" refers to the “complementarity determining regions” within antibody variable domain sequences. The growth and analysis of extensive public databases of amino acid sequences of variable heavy and light regions over the past twenty years have led to the understanding of the typical boundaries between framework regions (FRs) and CDR sequences within variable region sequences and have enabled persons skilled in the art to accurately determine the CDRs. The CDR definition schemes available in the art include e.g., Chothia scheme based on the three-dimensional structure of antibodies and the topology of the CDR loops (Chothia et al. (1989) Nature, 342: 877-883; Al-Lazikani et al., Standard conformations for the canonical structures of immunoglobulins, Journal of Molecular Biology, 273: 927-948 (1997) ) , Kabat scheme based on antibody sequence variability (Kabat et al., Sequences of Proteins of Immunological Interest, 4th Ed., U. S. Department of Health and Human Services, National Institutes of Health (1987) ) , AbM scheme (University of Bath) , Contact scheme (University College London) , International ImMunoGeneTics database (IMGT) scheme (imgt. cines. fr / on the World Wide Web) , and North scheme based on the affinity propagation clustering using a large number of crystal structures. The boundaries of each CDRs according to several CDR definition schemes are presented below:

[0114] Unless otherwise indicated, when referring to CDRs contained in a variable domain, the CDRs can be determined using any one or a combination of the antibody CDR definition schemes mentioned above.

[0115] It should be noted that boundaries of CDRs in variable domains of an antibody determined by different definition schemes may differ. Therefore, the CDR sequences of variable regions of the same antibody defined by different schemes may differ. Accordingly, when it comes to defining an antibody with specific CDR sequences according to a specific definition scheme (e.g., Kabat Scheme) in the present disclosure, the scope of antibody encompasses any antibodies whose variable region sequences comprise the specific CDR sequences, but may have different CDR boundaries assigned by application of a different scheme.

[0116] As is known in the art, CDRs vary from antibody to antibody. However, a limited number of the amino acid positions in a set of CDRs are directly involved in antigen binding. Using at least two of Kabat, Chothia, AbM and IMG schemes, the minimum overlapping regions for each CDRs can be determined, thereby providing a "minimum binding unit" for antigen binding. Such a minimum binding unit can be a subset of CDR residues of an antibody, while the remaining CDR residues, as those skilled in the art will appreciate, can be determined by analyzing the structure and folding of the antibody. Therefore, in some instances, CDR amino acid sequences as described herein may comprise a modification (s) . The modification may be amino acid substitution, deletion, and / or addition. Antibodies and antigen-binding fragments obtained in such manner can be easily tested for one or more desired property such as, improved binding specificity, increased binding affinity, improved or enhanced biological properties (as the case may be) , reduced immunogenicity, etc.

[0117] As used herein, unless otherwise indicated, the amino acid positions of CDRs and variable domains of an antibody are numbered according to the Kabat numbering system (Kabat et al., Sequences of Proteins of Immunological Interest, 5th Ed., Public Health Service, National Institutes of Health, Bethesda, MD (1991) ) .

[0118] The term “chimeric” antibody as used herein is intended to an antibody in which a portion of the heavy and / or light chain is derived from a particular source or species, while the remainder is derived from a different source or species; for example, an antibody comprising heavy and light chain variable region sequences from one species and constant region sequences from another species, such as an antibody having murine heavy and light chain variable regions linked to human constant regions.

[0119] The term "CDR-grafted antibody" refers to antibodies that comprise heavy and light chain variable region sequences from one species but in which the sequences of one or more of the CDR regions of VH and / or VL are replaced with CDR sequences of another species, such as antibodies having human heavy and light chain variable regions in which one or more of the human CDRs has been replaced with murine CDR sequences.

[0120] The term "humanized antibody" refers to antibodies that comprise heavy and light chain variable region sequences from a non-human species (e.g., a mouse) but in which at least a portion of the VH and / or VL sequence has been altered to be more "human-like" , i.e., more similar to human germline variable sequences. One type of humanized antibody is a CDR-grafted antibody, in which CDR sequences from a non-human species (e.g., mouse) are introduced into human VH and VL framework sequences. A humanized antibody is an antibody or a variant, derivative, analog or fragment thereof which immunospecifically binds to an antigen of interest and which comprises framework regions and constant regions having substantially the amino acid sequence of a human antibody but complementarity determining regions (CDRs) having substantially the amino acid sequence of a non-human antibody. As used herein, the term "substantially" in the context of a CDR refers to a CDR having an amino acid sequence at least 80%, at least 85%, at least 90%, at least 95%, at least 98%or at least 99%identical to the amino acid sequence of a non-human antibody CDR. A humanized antibody comprises substantially all of at least one, and typically two, variable domains (Fab, Fab', F (ab') 2, Fv) in which all or substantially all of the CDR regions correspond to those of a non-human immunoglobulin (i.e., donor antibody) and all or substantially all of the framework regions are those of a human immunoglobulin consensus sequence. In an embodiment, a humanized antibody also comprises at least a portion of an immunoglobulin constant region (Fc) , typically that of a human immunoglobulin. In some embodiments, a humanized antibody contains both the light chain as well as at least the variable domain of a heavy chain. The antibody also may include the CH1, hinge, CH2, CH3, and CH4 regions of the heavy chain. In some embodiments, a humanized antibody only contains a humanized light chain. In some embodiments, a humanized antibody only contains a humanized heavy chain. In specific embodiments, a humanized antibody only contains a humanized variable domain of a light chain and / or humanized heavy chain.

[0121] The framework and CDR regions of a humanized antibody need not correspond precisely to the parental sequences, e.g., the donor antibody CDR or the acceptor framework may be mutagenized by substitution, insertion and / or deletion of at least one amino acid residue so that the CDR or framework residue at that site does not correspond to either the donor antibody or the consensus framework. In an exemplary embodiment, such mutations, however, will not be extensive. Usually, at least 80%, for instance, at least 85%, at least 90%, or at least 95%of the humanized antibody residues will correspond to those of the parental FR and CDR sequences. Back mutation at a particular framework position to restore the same amino acid that appears at that position in the donor antibody is often utilized to preserve a particular loop structure or to correctly orient the CDR sequences for contact with target antigen.

[0122] A humanized antibody may be selected from any class of immunoglobulins, including IgM, IgG, IgD, IgA and IgE, and any isotype, including without limitation IgG1, IgG2, IgG3, and IgG4. The humanized antibody may comprise sequences from more than one class or isotype, and particular constant domains may be selected to optimize desired effector functions using techniques well known in the art.

[0123] The term “Fc region” is used to define the C-terminal region of an immunoglobulin heavy chain, which may be generated by papain digestion of an intact antibody. The Fc region may be a native sequence Fc region or a variant Fc region. The Fc region of an immunoglobulin generally comprises two constant domains, i.e., a CH2 domain and a CH3 domain, and optionally comprises a CH4 domain, for example, as in the case of the Fc regions of IgM and IgE antibodies. The Fc region of IgG, IgA, and IgD antibodies comprises a hinge region, a CH2 domain, and a CH3 domain. In contrast, the Fc region of IgM and IgE antibodies lacks a hinge region but comprises a CH2 domain, a CH3 domain and a CH4 domain. In one embodiment, a human IgG immunoglobulin Fc region is from Asp221 or from Cys226 or from Asp231 to the C-terminus of heavy chain. However, the lysine (Lys447) or glycine-lysine (Gly446Lys447) at the C-terminal end of the Fc region may be present or absent. Variant Fc regions having substitutions of amino acid residues in the Fc portion to alter antibody effector function are known in the art (see, e.g., Winter et al., US Patent Nos. 5,648,260 and 5,624,821) . The Fc portion of an antibody may mediate one or more effector functions, for example, cytokine induction, ADCC, phagocytosis, complement dependent cytotoxicity (CDC) , and / or half-life / clearance rate of antibody and antigen-antibody complexes. In some cases, these effector functions are desirable for therapeutic antibody but in other cases might be unnecessary or even deleterious, depending on the therapeutic objectives. Certain human IgG isotypes, particularly IgG1 and IgG3, mediate ADCC and CDC via binding to FcγRs and complement C1q, respectively. In still another embodiment at least one amino acid residue is replaced in the constant region of the antibody, for example the Fc region of the antibody, such that effector functions of the antibody are altered. The dimerization of two immunoglobulin Fc regions or two antibody chains may be mediated by the dimerization of CH3 domains, and further stabilized by the disulfide bonds within the hinge region that connects CH1 constant domains to the Fc constant domains (e.g., CH2 and CH3) , and / or artificially introduced into the Fc domains. The anti-inflammatory activity of IgG is dependent on sialylation of the N-linked glycan of the IgG Fc fragment. The precise glycan requirements for anti-inflammatory activity have been determined, such that an appropriate IgG1 Fc fragment can be created, thereby generating a fully recombinant, sialylated IgG1 Fc with greatly enhanced potency (see, Anthony et al., Science, 320: 373-376 (2008) ) .

[0124] The terms “antigen-binding portion (ABP) ” , “antigen-binding domain (ABD) ” and “antigen-binding fragment” or “functional fragment” of an antibody are used interchangeably and refer to one or more fragments or portions of an antibody that retain the ability to specifically bind to an antigen, i.e., the same antigen as the full-length antibody from which the portion or fragment is derived. It has been shown that the antigen-binding function of an antibody can be performed by fragments of a full-length antibody. Such antibody embodiments may also be bispecific, dual specific, or multi-specific formats; specifically binding to two or more different antigens. Examples of binding fragments encompassed within the term “antigen-binding portion” of an antibody include (i) a Fab fragment, a monovalent fragment consisting of the VL, VH, CL, and CH1 domains; (ii) a F (ab’ ) 2 fragment, a bivalent fragment comprising two Fab fragments linked by a disulfide bridge at the hinge region; (iii) an Fd fragment consisting of the VH and CH1 domains; (iv) a Fv fragment consisting of the VL and VH domains of a single arm of an antibody, (v) a dAb fragment (Ward et al., Nature, 341: 544-546 (1989) ; PCT Publication No. WO 90 / 05144) , which comprises a single variable domain; and (vi) an isolated complementarity determining region (CDR) . Furthermore, although the two domains of the Fv fragment, VL and VH, are coded by separate genes, they can be joined, using recombinant methods, by a synthetic linker that enables them to be made as a single protein chain in which the VL and VH regions pair to form monovalent molecules (known as single chain Fv (scFv) ; see, for example, Bird et al., Science, 242: 423-426 (1988) ; and Huston et al., Proc. Natl. Acad. Sci. USA, 85: 5879-5883 (1988) ) . Such single chain antibodies are also intended to be encompassed within the term “antigen-binding portion” of an antibody and equivalent terms given above. Other forms of single chain antibodies, such as diabodies are also encompassed. Diabodies are bivalent, bispecific antibodies in which VH and VL domains are expressed on a single polypeptide chain, but using a linker that is too short to allow for pairing between the two domains on the same chain, thereby forcing the domains to pair with complementary domains of another chain and creating two antigen binding sites (see, for example, Holliger et al., Proc. Natl. Acad. Sci. USA, 90: 6444-6448 (1993) . Such antibody binding portions are known in the art (Kontermann and Dübel eds., Antibody Engineering (Springer-Verlag, New York, 2001) , p. 790 (ISBN 3-540-41354-5) ) . In addition, single chain antibodies also include “linear antibodies” comprising a pair of tandem Fv segments (VH-CH1-VH-CH1) which, together with complementary light chain polypeptides, form a pair of antigen binding portions (Zapata et al., Protein Eng., 8 (10) : 1057-1062 (1995) ; and US Patent No. 5,641,870) ) .

[0125] The term “human sequence” or “from human” , in relation to the light chain constant domain CL, heavy chain constant domain CH, and Fc region of the antibody or the binding protein according to the present disclosure, means the sequence is of, or from, human immunoglobulin sequence. The human sequence of the present disclosure may be native human sequence, or a variant thereof including one or more (for example, up to 20, 15, 10) amino acid residue changes.

[0126] The term “monoclonal antibody” or “mAb” refers to an antibody obtained from a population of substantially homogeneous antibodies, i.e., the individual antibodies of the population are identical except for possible naturally occurring mutations that may be present in minor amounts. The modifier “monoclonal” is not to be construed as requiring production of the antibody by any particular method.

[0127] An “antibody that competes with a reference antibody for binding to its antigen” refers to an antibody that blocks 50%, 60%, 70%, 80%, 90%or 95%or more of the binding of the reference antibody to its antigen in a competition assay. Conversely, the reference antibody blocks 50%, 60%, 70%, 80%, 90%or 95%or more of the binding of the antibody to its antigen in a competition assay. Numerous types of competitive binding assays can be used to determine whether an antibody competes with another antibody, such as direct or indirect solid-phase radioimmunoassay (RIA) , direct or indirect solid-phase enzyme immunoassay (EIA) and sandwich competition assay. An antibody that shows the same or similar binding affinity and / or specificity as a reference antibody refers to an antibody that is capable of having at least 50%, 60%, 70%, 80%, 90%or 95%or more of the binding affinity and / or specificity of the reference antibody. This can be determined by any method known in the art for determining binding affinity and / or specificity.

[0128] The term “specific binding” or “specifically binding” means that an antibody selectively or preferentially binds to an antigen. If an antibody binds to antigen with a KD of about 1× 10-7 M or less, about 1× 10-8 M or less, or about 1× 10-9 M or less based on Bio-Layer Interferometry (BLI) or Surface Plasmon Resonance, the antibody is considered as an antibody that “specifically binds to the antigen” .

[0129] The term "KD" , as used herein, is intended to refer to the "equilibrium dissociation constant" , and refers to the value obtained in a titration measurement at equilibrium, or by dividing the dissociation rate constant (koff) by the association rate constant (kon) . The association rate constant (kon) , the dissociation rate constant (koff) , and the equilibrium dissociation constant (KD) are used to represent the binding affinity of an antibody to an antigen. Methods for determining association and dissociation rate constants are well known in the art. Using fluorescence-based techniques offers high sensitivity and the ability to examine samples in physiological buffers at equilibrium. Other experimental approaches and instruments such as a   (biomolecular interaction analysis) assay can be used (e.g., instrument available from BIAcore International AB, a GE Healthcare company, Uppsala, Sweden) . Biolayer interferometry (BLI) using, e.g., the  RED96 system (Pall FortéBio LLC) , is another affinity assay technique. Additionally, a   (Kinetic Exclusion Assay) assay, available from Sapidyne Instruments (Boise, Idaho) can also be used. In some embodiments, KD values of antibodies or binding proteins of the present disclosure are measured by a grating-coupled interferometry (GCI) based assay.

[0130] As used herein, the amino acid positions of all constant regions and domains of the heavy and light chain of an antibody are numbered according to the Kabat numbering system described in Kabat, et al., Sequences of Proteins of Immunological Interest, 5th ed., Public Health Service, National Institutes of Health, Bethesda, MD (1991) and is referred to as “numbering according to Kabat” or “Kabat numbering” herein. Specifically, the Kabat numbering system (see pages 647-660) of Kabat, et al., Sequences of Proteins of Immunological Interest, 5th ed., Public Health Service, National Institutes of Health, Bethesda, MD (1991) is used for the light chain constant domain CL of kappa (Cκ) and lambda (Cλ) isotype, and the Kabat EU index numbering system (see pages 661-723) is used for the constant heavy chain domains (CH1, Hinge, CH2 and CH3, which is herein further clarified by referring to “numbering according to Kabat EU index” or “EU numbering” in this case) .

[0131] General information regarding the sequences of human immunoglobulins light and heavy chains is given in: Kabat, E.A., et al., Sequences of Proteins of Immunological Interest, 5th ed., Public Health Service, National Institutes of Health, Bethesda, MD (1991) . The term "multivalent binding protein" denotes a binding protein comprising two or more antigen binding domains. A multivalent binding protein is, in certain cases, engineered to have three or more antigen binding domains, and is generally not a naturally occurring antibody. The term "multispecific (e.g., bispecific) binding protein" (which can be used interchangeably with the term “multispecific (e.g., bispecific) antibody” , unless stated otherwise) refers to a binding protein capable of binding at least two different target antigens or different epitopes. Hence, a multispecific antigen-binding fragment of an antibody will typically comprise at least two different variable domains, wherein each variable domain is capable of specifically binding to a separate antigen or to a different epitope. FIT-Ig binding proteins of the present disclosure comprise four antigen binding sites and are typically tetravalent binding proteins. MAT-Fab binding proteins of the present disclosure comprise two antigen binding sites and are typically bivalent binding proteins. A FIT-Ig or MAT-Fab according to this disclosure binds both ALPP / ALPPL2 and CD3 and is bispecific.

[0132] As used herein, the term “FIT-Ig binding protein” is used interchangeably with “FIT-Ig antibody” , “FIT-Ig protein” , “FIT-Ig molecules” , “FIT-Ig” and the like, and refers to a multivalent antibody comprising two long (heavy) V-C-V-C-Fc chain polypeptides and four short (light) V-C chain polypeptides, which form a hexamer exhibiting four Fab antigen binding sites (VH-CH1 paired with VL-CL, sometimes notated VH-CH1: : VL-CL) . In the long (heavy) V-C-V-C-Fc chain polypeptides, V-C-V-C represents VL-CL-VH-CH1 structure or VH-CH1-VL-CL structure, in which VL and VH are derived from different antibodies binding different antigens, and complementary pair to corresponding VH and VL derived from the same antibody, Figure 1A exemplified the structure of FIT-Ig. Each half of a FIT-Ig comprises three chains: a long chain polypeptide and two short chain polypeptides, and complementary immunoglobulin pairing of the VH-CH1 and VL-CL elements of the three chains results in two Fab-structured antigen binding sites, arranged in tandem. In the present disclosure, it is preferred that the immunoglobulin domains comprising the Fab elements are directly fused in the long chain polypeptide, without the use of interdomain linkers. That is, the N-terminal V-C element of the long (heavy) polypeptide chains is directly fused at its C-terminus to the N-terminus of another V-C element, which in turn is linked to a C-terminal Fc region. In bispecific FIT-Ig binding proteins, the tandem Fab elements may be reactive with different antigens. Each Fab antigen binding site comprises a heavy chain variable domain and a light chain variable domain with a total of six CDRs per antigen binding site.

[0133] A description of the design, expression, and characterization of FIT-Ig molecules is provided in PCT Publication WO 2015 / 103072, which is incorporated by reference herein in its entirety. An example of such half of FIT-Ig molecules comprises a long (heavy) chain and two different short (light) chains. The long chain comprises the structural formula VLA-CL-VHB-CH1-Fc where CL is directly fused to VHB (namely “Format LH” ) or VHA-CH1-VLB-CL-Fc where CH1 is fused directly to VLB (namely “Format HL” ) , and the two short polypeptide chains of the FIT-Ig correspondingly have the formulas VHA-CH1 and VLB-CL (for “Format LH” ) or VLA-CL and VHB-CH1 (for “Format HL” ) , respectively; wherein VLA is a variable light domain from a parental antibody that binds antigen A, VLB is a variable light domain from a parental antibody that binds antigen B, VHA is a variable heavy domain from a parental antibody that binds antigen A, VHB is a variable heavy domain from a parental antibody that binds antigen B, CL is a light chain constant domain, CH1 is a heavy chain constant domain 1, and Fc is an immunoglobulin Fc region (e.g., the C-terminal hinge-CH2-CH3 portion of a heavy chain of an IgG1 antibody) . In the half of FIT-Ig embodiments, antigen A and antigen B are different antigens, or different epitopes of the same antigen. In the present disclosure, one of antigen A and antigen B is ALPP / ALPPL2 and the other is CD3, for example, antigen A is ALPP / ALPPL2, and antigen B is CD3. Figure 1A provides schematic illustration of the domain structure of a FIT-Ig bispecific antibody.

[0134] As used herein, the term "fused directly" or "directly fused" , when referring to the linear connection of two domains in a polypeptide structure, means that the domains are joined directly by a peptide bond, without the use of an artificial polypeptide linker or connector.

[0135] As used herein, the term “MAT-Fab binding protein” is used interchangeably with “MAT-Fab antibody” , “MAT-Fab protein” , “MAT-Fab molecules” , “MAT-Fab” and the like, and refers to a multivalent binding protein comprising one long (heavy) V-C-V-C-Fc chain polypeptide, two short (light) V-C chain polypeptides, and one immunological Fc chain polypeptide, which form a tetramer exhibiting two Fab antigen binding sites arranged in tandem (VH-CH1 paired with VL-CL, sometimes notated VH-CH1: : VL-CL) , and one Fc: Fc dimer. In the long (heavy) V-C-V-C-Fc chain polypeptides, V-C-V-C represents VL-CL-VH-CH1 structure or VH-CH1-VL-CL structure, in which VL and VH are derived from different antibodies binding different antigens, and complementary pair corresponding VH and VL derived from the same antibody, Figure 1B exemplified the structure of MAT-Fab. Often modifications have been introduced into the CH3 domain of Fc region of the MAT-Fab long chain (abbreviated as CH3m1 domain) and also the CH3 domain of the MAT-Fab Fc polypeptide chain (abbreviated as CH3m2 domain) to favor the heterodimerization of the two CH3 domains. The modifications can be “knob-in-hole” (KIH) mutations, for instance, a mutation is made to form a structural knob in the CH3m1 domain of the long chain for pairing with a CH3m2 domain of the Fc chain that comprises a complementary structural hole or vice versa. However, other modifications such as those introduced into the domains salt bridges or electrostatic interactions are also useful. The constant regions may also include other modifications, for example, Cys residues to stable the MAT-Fab molecule, and / or mutations to prevent or impair the Fc effector functions. Preferably, a feature of the structure of a MAT-Fab bispecific antibody described herein is that all adjacent immunoglobulin heavy and light chain variable and constant domains are linked directly to one another without an intervening synthetic amino acid or peptide linker.

[0136] A description of the design, expression, and characterization of MAT-Fab molecules is provided in PCT Publication WO2018 / 035084, which is incorporated herein in its entirety. One example of such MAT-Fab molecules comprises a long chain with a “knob” in Fc region, two different short chains, and one Fc polypeptide chain with a “hole” . In some embodiments, the long chain comprises the structural formula VLA-CL-VHB-CH1-hinge-CH2-CH3m1 where CL is directly fused to VHB (namely “Format LH” ) , or VHA-CH1-VLB-CL-CH1-hinge-CH2-CH3m1 where CH1 is fused directly to VLB (namely “Format HL” ) , two short polypeptide chains of the MAT-Fab correspondingly have the formulas VHA-CH1 and VLB-CL (for “Format LH” ) or VLA-CL and VHB-CH1 (for “Format HL” ) , and a Fc chain of hinge-CH2-CH3m2 respectively; wherein VLA is a variable light domain from a parental antibody that binds antigen A, VLB is a variable light domain from a parental antibody that binds antigen B, VHA is a variable heavy domain from a parental antibody that binds antigen A, VHB is a variable heavy domain from a parental antibody that binds antigen B, CL is a light chain constant domain, CH1 is a heavy chain constant domain 1, and CH3m1 is a heavy chain constant domain 3 with knob mutation (s) such as T366Y or T366W, or hole mutations such as Y407T, or a combination of T366S, L368A and Y407V. The Fc polypeptide chain may be the C-terminal hinge-CH2-CH3 portion of a heavy chain of an immunoglobulin (such as IgG antibody) , with hole mutations complementary to knob mutation (s) in CH3m2 such as Y407T, or a combination of T366S, L368A and Y407V; or a knob mutation (s) such as T366Y or T366W. In certain embodiments, in bispecific MAT-Fab embodiments, antigen A and antigen B are different antigens, or different epitopes of the same antigen. In the present disclosure, one of antigen A and antigen B is ALPP / ALPPL2 and the other is CD3, for example, antigen A is ALPP / ALPPL2, and antigen B is CD3, or antigen B is ALPP / ALPPL2 and antigen A is CD3. Figure 1B provides schematic illustration of the domain structure of a MAT-Fab bispecific antibody.

[0137] In a particular embodiment, each Fc region of the Fc scaffold further comprises mutations to form non-natural disulfide bond. For example, in one Fc region, the serine residue at position 354 is replaced with a cysteine residue (S354C) or the glutamic acid residue at position 356 is replaced with a cysteine residue (E356C) (in particular, the serine residue at position 354 is replaced with a cysteine residue) ; and in the other Fc region, the tyrosine residue at position 349 is replaced with a cysteine residue (Y349C) (numbering according to the EU index) .

[0138] In yet another embodiment, in the CH3 region of an Fc region, the threonine residue at position 366 is replaced with a tryptophan residue (T366W) , and the serine residue at position 354 is replaced with a cysteine residue (S354C) or the glutamic acid residue at position 356 is replaced with a cysteine residue (E356C) (in particular, the serine residue at position 354 is replaced with a cysteine residue) ; whereas in the CH3 region of the other Fc region, the threonine residue at position 366 is replaced with a serine residue (T366S) , the leucine residue at position 368 is replaced with an alanine residue (L368A) , and the tyrosine residue at position 407 is replaced with a valine residue (Y407V) (numbering according to the EU index) , optionally the tyrosine residue at position 349 is replaced with a cysteine residue (Y349C) (numbering according to the EU index) . In a particular embodiment, one Fc region comprises the amino acid substitutions S354C and T366W and the other Fc region comprises the amino acid substitutions Y349C, T366S, L368A and Y407V (numbering according to the EU index) .

[0139] In another embodiment, one or more salt bridges between the CH3m1 and CH3m2 domains of a MAT-Fab antibody described herein can be introduced by mutating either or both domains such that a residue in one of the domains is able to hydrogen bond and electrostatically interact (bond) with a residue in the other domain. For example, a salt bridge may be introduced by changing (mutating) an amino acid residue in the CH3m1 domain to a glutamate or aspartate residue and changing (mutating) a residue in the CH3m2 domain to a lysine or arginine residue such that the glutamate or aspartate residue in the CH3m1 domain can hydrogen bond and electrostatically interact with the lysine or arginine residue in the CH3m2 domain.

[0140] The term "CD3" , as used herein, refers to an antigen which is expressed on T cells as part of the multimolecular T cell receptor (TCR) and which consists of a homodimer or heterodimer formed from the association of two of four receptor chains: CD3-epsilon, CD3-delta, CD3-zeta, and CD3-gamma. An amino acid sequence of a human CD3-epsilon is set forth under UniProtKB accession number P07766; an amino acid sequence of a human CD3-delta under UniProtKB accession number P04234; an amino acid sequence of a human CD3-zeta under UniProtKB accession number P20963; and an amino acid sequence of a CD3-gamma under UniProtKB accession number P09693. All references to CD3 proteins, polypeptides and protein fragments herein are intended to refer to the human version of the respective protein, polypeptide or protein fragment unless explicitly specified as being from a non-human species.

[0141] The term “ALPP” is used interchangeably herein with “alkaline phosphatase, placental” or “PLAP” and refers to placental alkaline phosphatase (Enzyme Commission (EC) number 3.1.3.1) . Human PLAP is a 535 amino-acid glycosylated protein. All references to ALPP proteins, polypeptides and protein fragments herein are intended to refer to the human version of the respective protein, polypeptide or protein fragment unless explicitly specified as being from a non-human species. The term also encompasses naturally occurring variants of ALPP, e.g., splice variants or allelic variants, isoforms, and vertebrate species homologs of human ALPP.

[0383] In one embodiment, the anti-ALPP / ALPPL2×CD3 FIT-Ig or MAT-Fab binding protein as described herein has high stability in temperature and freezing-thawing stress assay.

[0384] Nucleic acid, vector, and host cells

[0385] The present disclosure provides isolated nucleic acids, recombinant expression vectors and host cells, which is useful for making the antibodies, functional antibody fragments, and binding proteins described herein.

[0386] The term "isolated nucleic acid" , as used herein, means a polynucleotide (e.g., of genomic, cDNA, or synthetic origin, or some combination thereof) that, by human intervention, is not associated with all or a portion of the polynucleotides with which it is found in nature; is operably linked to a polynucleotide that it is not linked to in nature; or does not occur in nature as part of a larger sequence.

[0387] The term "vector" , as used herein, is intended to refer to a nucleic acid molecule capable of transporting another nucleic acid to which it has been linked. One type of vector is a "plasmid" , which refers to a circular double stranded DNA loop into which additional DNA segments may be ligated. Another type of vector is a viral vector, wherein additional DNA segments may be ligated into the viral genome. Certain vectors are capable of autonomous replication in a host cell into which they are introduced (e.g., bacterial vectors having a bacterial origin of replication and episomal mammalian vectors) . Other vectors (e.g., non-episomal mammalian vectors) can be integrated into the genome of a host cell upon introduction into the host cell, and thereby are replicated along with the host genome. Moreover, certain vectors are capable of directing the expression of genes to which they are operatively linked. Such vectors are referred to herein as "recombinant expression vectors" (or simply, "expression vectors" ) . In general, expression vectors of utility in recombinant DNA techniques are often in the form of plasmids. In the present specification, "plasmid" and "vector" may be used interchangeably as the plasmid is the most commonly used form of vector. However, the disclosure is intended to include such other forms of expression vectors, such as viral vectors (e.g., replication defective retroviruses, adenoviruses and adeno-associated viruses) , which serve equivalent functions.

[0388] The term "operably linked" refers to a juxtaposition wherein the components described are in a relationship permitting them to function in their intended manner. A control sequence "operably linked" to a coding sequence is ligated in such a way that expression of the coding sequence is achieved under conditions compatible with the control sequence. "Operably linked" sequences include both expression control sequences that are contiguous with the gene of interest and expression control sequences that act in trans or at a distance to control the gene of interest. The term "expression control sequence" as used herein refers to polynucleotide sequences that are necessary to affect the expression and processing of coding sequences to which they are ligated. Expression control sequences include appropriate transcription initiation, termination, promoter and enhancer sequences; efficient RNA processing signals such as splicing and polyadenylation signals; sequences that stabilize cytoplasmic mRNA; sequences that enhance translation efficiency (i.e., Kozak consensus sequence) ; sequences that enhance protein stability; and when desired, sequences that enhance protein secretion. The nature of such control sequences differs depending upon the host organism; in prokaryotes, such control sequences generally include promoter, ribosomal binding site, and transcription termination sequence; in eukaryotes, generally, such control sequences include promoters and transcription termination sequence. The term "control sequences" is intended to include components whose presence is essential for expression and processing, and can also include additional components whose presence is advantageous, for example, leader or signal sequences and fusion partner sequences.

[0389] The vector (s) comprising nucleic acid (s) encoding the antibodies, functional antibody fragments, and binding proteins described herein can be introduced into a host cell (s) by transformation. The term "transformation" , as defined herein, refers to any process by which exogenous DNA enters a host cell. Transformation may occur under natural or artificial conditions using various methods well known in the art. Transformation may rely on any known method for the insertion of foreign nucleic acid sequences into a prokaryotic or eukaryotic host cell. The method is selected based on the host cell being transformed and may include, but is not limited to, transfection, viral infection, electroporation, lipofection, and particle bombardment. Such "transformed" cells include stably transformed cells in which the inserted DNA is capable of replication either as an autonomously replicating plasmid or as part of the host chromosome. They also include cells which transiently express the inserted DNA or RNA for limited periods of time.

[0390] The term "recombinant host cell" (or simply "host cell" ) , is intended to refer to a cell into which exogenous DNA has been introduced. In an embodiment, the host cell comprises two or more (e.g., multiple) nucleic acids encoding antibodies, such as the host cells described in US Patent No. 7, 262, 028, for example. Such terms are intended to refer not only to the particular subject cell, but also to the progeny of such a cell. Because certain modifications may occur in succeeding generations due to either mutation or environmental influences, such progeny may not, in fact, be identical to the parent cell, but are still included within the scope of the term "host cell" as used herein. In an embodiment, host cells include prokaryotic and eukaryotic cells selected from any of the Kingdoms of life. In another embodiment, eukaryotic cells include protist, fungal, plant and animal cells. In another embodiment, host cells include but are not limited to the prokaryotic cell line Escherichia coli; mammalian cell lines CHO, HEK293, COS, NS0, SP2 and PER. C6; the insect cell line Sf9; and the fungal cell Saccharomyces cerevisiae.

[0391] Standard techniques may be used for recombinant DNA, oligonucleotide synthesis, and tissue culture and transformation (e.g., electroporation, lipofection) . Enzymatic reactions and purification techniques may be performed according to manufacturer's specifications or as commonly accomplished in the art or as described herein. The foregoing techniques and procedures may be generally performed according to conventional methods well known in the art and as described in various general and more specific references that are cited and discussed throughout the present specification. See e.g., Sambrook et al., Molecular Cloning: A Laboratory Manual, 2nd ed. (Cold Spring Harbor Laboratory Press, Cold Spring Harbor, N.Y., 1989) .

[0392] Antibodies, functional fragments thereof, and binding proteins according to the present disclosure may be purified (for an intended use) by using one or more of a variety of methods and materials available in the art for purifying antibodies and binding proteins. Such methods and materials include, but are not limited to, affinity chromatography (e.g., using resins, particles, or membranes conjugated to Protein A, Protein G, Protein L, or a specific ligand of the antibody, functional fragment thereof, or binding protein) , ion exchange chromatography (for example, using ion exchange particles or membranes) , hydrophobic interaction chromatography ( "HIC" ; for example, using hydrophobic particles or membranes) , ultrafiltration, nanofiltration, diafiltration, size exclusion chromatography ( "SEC" ) , low pH treatment (to inactivate contaminating viruses) , and combinations thereof, to obtain an acceptable purity for an intended use. A non-limiting example of a low pH treatment to inactivate contaminating viruses comprises reducing the pH of a solution or suspension comprising an antibody, functional fragment thereof, or binding protein of the present disclosure to pH 3.5 with 0.5 M phosphoric acid, at 18℃ -25℃, for 60 to 70 minutes.

[0421] EXAMPLES

[0422] The following examples further illustrate the present disclosure, however, it should be understood that the examples are described in an illustrative rather than a limiting manner, and that various modifications may be made by those skilled in the art.

[0423] Unless expressly stated to the contrary, conventional methods from chemistry, biochemistry, organic chemistry, molecular biology, microbiology, recombinant DNA technology, genetics, immunology, and cell biology will be used. Experimental methods, unless otherwise specified, are conventional methods in the art using default parameters, steps, etc. ; the experimental materials used, unless otherwise specified, are all commercially available products. If no specific techniques or conditions are noted, they shall be executed according to the techniques or conditions described in the literature in the art, or according to corresponding product instructions. The reagents or instruments used without indicating the manufacturer are all standard products available through usual channels.

[0424] Generally, in the present disclosure, conventional hybridoma screening protocol was used to generate ALPP / ALPPL2-targeting monoclonal antibodies with improved properties. The anti-ALPP / ALPPL2×CD3 chimeric bispecific antibodies were further constructed using selected anti-ALPP / ALPPL2 antibodies and anti-CD3 antibodies, and screened via multiple in vitro assays. The selected anti-ALPP / ALPPL2 antibodies were humanized by conventional CDR-grafting method.

[0425] Example 1: Generation of anti-ALPPL2 antibodies

[0426] Anti-ALPPL2 antibodies were obtained by immunizing Balb / c mice with a human ALPPL2 (NP_112603.2) encoded mRNA-LNP complex. A sequence of HUMAN_ALPPL2_CDS is shown as follows:

[0427] >HUMAN_ALPPL2_CDS

[0428] Mice were immunized at 2-week intervals and monitored for serum titer once a week after the second injection. After 2 immunizations, splenocytes were harvested and fused with mouse myeloma cells to form hybridoma cell lines, when serum titer achieved desired level. Hybridoma cells were plated in 384-well plates containing selection media with hypoxanthine-aminopterin-thymidine (HAT) at a density of 2.5×104 cells per well. Seven to ten days later, macroscopic hybridoma colonies were observed. Supernatants of hybridoma cells were then screened and selected to identify cell lines producing anti-ALPPL2 mouse antibodies. Upon preliminary characterization, several anti-ALPPL2 antibodies were selected and sequenced.

[0429] Example 1.1: Heavy and light chain variable region sequences

[0430] To amplify heavy and light chain variable regions of these selected antibodies, total RNA of each hybridoma clone was isolated from more than 5×106 cells with TRIzolTM RNA extraction reagent (Invitrogen, Cat. #15596018) . cDNA was synthesized using an InvitrogenTM SuperScriptTM III First-Strand Synthesis SuperMix kit (ThermoFisher Scientific Cat. #18080) following manufacturer's instructions, and the cDNAs encoding the variable regions of light and heavy mouse immunoglobulin chains were amplified using a MilliporeSigmaTM NovagenTM Mouse Ig-Primer Set (Fisher Scientific Cat. #698313) . PCR products were analyzed by electrophoresis on a 1.2%agarose gel with SYBRTM Safe DNA gel stain (ThermoFisher Cat. #S33102) . DNA fragments with correct size were purified using a  Gel and PCR Clean-up kit (Macherey-Nagel, Cat. #740609) according to manufacturer's instructions and were subcloned into pMD18-T vector individually. Fifteen colonies from each transformation were selected and sequences of insert fragments were analyzed by DNA sequencing. The protein sequences of murine mAb variable regions were analyzed by sequence homology alignment.

[0434] Example 1.2: Generation of anti-ALPP / ALPPL2×CD3 FIT-Ig chimeric bispecific antibodies

[0435] A group of chimeric FIT-Ig proteins recognizing both human ALPP / ALPPL2 and human CD3 were constructed utilizing VH / VL sequences in Table 1 as anti-ALPP / ALPPL2 moiety, and VH / VL sequences published in WO2022 / 042488 as anti-CD3 moiety (Table 2) , as well as human constant region sequences, as shown in Table 3. The obtained antibodies are named with prefix “FIT” .

[0436] Table 2: Amino acid sequences of anti-CD3 variable domains

[0437] *CDRs defined according to Kabat scheme are underlined.

[0438] Table 3: Amino acid sequences of human IgG constant regions

[0439] *Fc mutations are shown in bold italic.

[0440] FIT-Ig molecules were constructed following the general procedures described in PCT Publication No. WO2015 / 103072. Each FIT-Ig consists of two long chains, two first short chains and two second short chains. These polypeptide chains have the following structures:

[0441] Chain #1 (long chain) : VLA-CL-VHB-CH1-hinge-CH2-CH3;

[0442] Chain #2 (first short chain) : VHA-CH1;

[0443] Chain #3 (second short chain) : VLB-CL;

[0444] wherein A stands for antigen ALPP / ALPPL2 and B stands for antigen CD3, therefore, VLA and VLALPP / ALPPL2 are used interchangeably and means the light chain variable domain of a mouse derived antibody recognizing ALPP / ALPPL2 (for example, those screened in table 1 shown above) , VHB and VHCD3 are used interchangeably and means the heavy chain variable domain of a humanized monoclonal antibody recognizing CD3 (for example, those recorded in table 2 shown above) , VHA and VHALPP / ALPPL2 are used interchangeably and means the heavy chain variable domain of a mouse derived antibody recognizing ALPP / ALPPL2 (for example, those screened in table 1 shown above) , VLB and VLCD3 are used interchangeably and means the light chain variable domain of a humanized monoclonal antibody recognizing CD3 (for example, those recorded in table 2 shown above) ; each CL is a light chain constant domain (for example, as shown in SEQ ID NO: 46 ) , each CH1 is a first heavy chain constant domain (for example, as shown in SEQ ID NO: 47) , and CH1-hinge-CH2-CH3 is the C-terminal heavy chain constant region from CH1 through the terminus of the Fc region (for example, as shown in SEQ ID NO: 45) .

[0445] To construct the long chain, cDNA encoding the VLALPP / ALPPL2-CL-VHCD3 segment was synthesized de novo and inserted into the multiple cloning site (MCS) of a vector including coding sequences for human CH1-hinge-CH2-CH3. In the resulting vector, the MCS sequence was eliminated during homologous recombination to ensure that all the domain fragments were in the correct reading frame. Similarly, to construct the first and second short chains, cDNA encoding the VHALPP / ALPPL2 or the VLCD3 segment was de novo synthesized and inserted into the MCS of the appropriate vectors including coding segments for human CH1 and CL domains, respectively.

[0446] The pairing of the VH and the VL creating chimeric ALPP / ALPPL2×CD3 FIT-Ig binding proteins are listed in Table 4.

[0451] Example 1.4: Cell surface binding of anti-ALPP / ALPPL2×CD3 chimeric FIT-Ig bispecific antibodies

[0452] Cell binding activity of anti-ALPP / ALPPL2×CD3 antibodies were measured with human ALPP transfected HEK293 cell line (HEK293-huALPP, Genomeditech, GM-C26401) and human ALPPL2 transfected CHO-K1 cell line (CHOK1-huALPPL2, constructed in house) . Briefly, 2×105 cells were seeded into each well of a 96-well plate. Cells were centrifuged at 400g for 5 minutes and supernatants were discarded. For each well, 100 μl of serially diluted antibodies were then added and mixed with the cells. After 40 minutes incubation at 4℃, plates were washed several times to remove excess antibodies. Secondary fluorochrome-conjugated goat anti-human IgG antibody was then added and incubated with cells at 4℃ for 30 minutes. After another round of centrifugation and a washing step, cells were resuspended in FACS buffer for reading on an iQue3 (Sartorius) . Median Fluorescence Intensity (MFI) readouts were plotted against antibody concentration and analyzed with GraphPad Prism 6.0 software. The results shown in Figure 2 illustrate the binding activities of anti-ALPP / ALPPL2×CD3 chimeric bispecific antibodies to ALPP or ALPPL2-expressing cells. An irrelevant hIgG1 was used as negative control.

[0454] Example 1.5: Epitope binning of anti-ALPP / ALPPL2×CD3 chimeric FIT-Ig bispecific antibodies

[0455] The binding epitope of anti-ALPP / ALPPL2×CD3 antibodies were identified with a competition ELISA. Briefly, 96 well plates were coated with 1ug / mL purified antibodies and incubated overnight at 4℃. After washing with PBS containing 0.05%Tween 20, plates were blocked with blocking buffer (PBS containing 0.05%Tween 20 and 2%BSA) at 37℃ for 2 hours. Biotinylated human ALPPL2 protein pre-mixed with anti-ALPP / ALPPL2×CD3 antibody (sample) or irrelevant human IgG (baseline) was added into plate wells and incubated at 37℃ for 1 hour, then wells were washed 3 times. Streptavidin-HRP (1: 5000 dilution) was then added into each well and incubated at 37℃ for 1 hour, then wells were washed 3 times. Tetramethylbenzidine (TMB) chromogenic solution was added for color development for 5 minutes then the reaction was stopped with 1M HCl. Absorbance at 450 nm (OD450) was measured on a microplate reader. The OD450 baseline represents the level of human ALPPL2 binding to ALPP / ALPPL2 antibodies at absence of competition, while the difference between OD450baseline and OD450sample reflects the competition between the anti-ALPP / ALPPL2 antibody coated on plate and the antibody in solution. The inhibition percentage was calculated by following equation:

[0456] Inhibition %= (1-OD450sample  / OD450baseline) × 100%

[0457] Table 7 shows results of the competition ELISA; wherein FIT2024-7ac and FIT2024-10ac might recognize similar epitope or lapping epitope in / on antigen ALPP / ALPPL2, while other antibodies recognize different epitope in / on anti-ALPP / ALPPL2.

[0459] Example 1.6: Redirected CD3 activation of chimeric FIT-Ig bispecific antibodies

[0460] To measure redirected CD3 activation by anti-ALPP / ALPPL2×CD3 bispecific FIT-Ig antibodies, a co-cultured reporter gene assay was used. In this assay, Jurkat-NFAT-luc cells trigger downstream luciferase signal when cell surface CD3 is activated. ALPP / ALPPL2 over-expressing cells (HEK293-huALPP, CHOK1-huALPPL2) were used as the target cells, which can crosslink CD3 / TCR complex on T cells via bispecific anti-ALPP / ALPPL2×CD3 antibodies upon ALPP / ALPPL2 binding.

[0461] Jurkat-NFAT-luc cells (ChemPartner) and ALPP / ALPPL2 over-expressing cells were washed and resuspended in assay medium (RPMI1640 with 2%FBS) separately. Both cell types were seeded into 96-well plates (Costar #3903) in a ratio of 1: 1, each of cell types were seeded at 4×104 cells per well. FIT-Ig antibodies were added and mixed with the cells and incubated for 4 hours at 37℃. At the end of incubation, ONE-GloTM luminescence assay kit (Promega, Cat. #E6130) reagents were prepared and added into wells according to the manufacturer's instructions. Plates were read for luminescence signals with VarioskanTM LUX microplate reader (ThermoFisher Scientific) . This assay was conducted in the absence of cells expressing a co-target (ALPP / ALPPL2) for the bispecific binding proteins. Non-target redirected CD3 activation was tested using a Jurkat-NFAT-luc based reporter gene assay in the absence of target cells. The results are shown in Figure 3. Bispecific anti-ALPP / ALPPL2×CD3 antibodies show directly activation of CD3 in the presence of ALPP / ALPPL2-expressing target cells (Figure 3A-B) , and less non-target redirected activation than anti-CD3 antibody (huEM0006-01-24) , in the absence of ALPP / ALPPL2-expressing target cells (Figure 3C) .

[0462] Example 1.7: In vitro killing of anti-ALPP / ALPPL2 chimeric FIT-Ig bispecific antibodies

[0463] To evaluate in-vitro killing efficacy of anti-ALPP / ALPPL2×CD3 chimeric bispecific antibodies on tumor cell lines, we chose N87 (ATCC, CRL-5822) and PANC-1 (ATCC, CRL-1469) tumor cell lines with differentiate ALPPL / ALPPL2 expression levels, mixed them with human primary PBMC. Test articles can mediate the killing of target cells by PBMC.

[0464] Briefly, 2×104 cells / well tumor cells and 1×105 cells / well human primary PBMC were seeded into 96-well plates (Costar #3903) at 5: 1 effector-to-target [E / T] ratio. Then serial dilutions of test articles, positive and negative control were added into cells. Following incubation for 48h at 37℃, 5%CO2, supernatant and cells therein were discarded. After wells were gently washed twice. CTG was added to quantitate the number of viable cells present in wells. The results are shown in Figure 4.

[0465] As shown in Figure 4, the higher ALPP / ALPPL2 expression level of tumor cell line, the more potent killing efficacy by human PBMC in the presence of the anti-ALPP / ALPPL2×CD3 chimeric bispecific antibodies. Furthermore, the killing efficacy of the anti-ALPP / ALPPL2×CD3 chimeric bispecific antibodies is in a concentration-dependent manner.

[0466] Example 2: Humanization of anti-ALPP / ALPPL2 antibodies

[0467] The anti-ALPP / ALPPL2 variable region-encoding genes of the anti-ALPP / ALPPL2 antibodies were employed for humanization design. In the first step of this process, the amino acid sequences of the VH and VL domains of the anti-ALPP / ALPPL2 antibodies were compared against the available database of human Ig V-gene sequences to find the overall best-matching human germline Ig V-gene sequences. Additionally, J-region sequences of the VH or VL was compared against the J-region database to find the human framework having the highest homology to the murine VH and VL regions, respectively. Three-dimensional Fv models of anti-ALPP / ALPPL2 antibodies were then generated to determine if there were any framework positions where mouse amino acids were involved in supporting loop structures or the VH / VL interface. These residues in humanized sequences could be back mutated to mouse residues at the same positions to retain affinity / activity. The obtained humanized antibodies, humanized VH or VL are all named based on corresponding parent antibodies with prefix “hu” .

[0468] Several desirable back-mutations were introduced to EM1032-A49-VH / Vκ, EM1032-A67-VH / Vκ, EM1032-A86-VH / Vκ, and EM1032-A51-VH / Vκ; thereby several modified VH or Vκ with mutants were constructed, as shown in Table 8 below. As the "NG" (Asn-Gly) pattern found in LCDR1 or “NS” (Asn-Ser) in HCDR1 is prone to deamination, "DG" (Asp-Gly) in HCDR2 / 3 and LCDR1, or “DS” (Asp-Ser) in LCDR1 / 2 is prone to isomerization, which may result in heterogeneity during manufacturing, VH and VL domains containing NG (Asn-Gly) to NA (Asn-Ala) , NS (Asn-Ser) to NA (Asn-Ala) , DG (Asp-Gly) to DA (Asp-Ala) and / or DS (Asp-Ser) to DA (Asp-Ala) modifications (highlighted in bold italic in Table 8) were also designed and evaluated. The obtained corresponding VH / VL is named by specific mutations in parenthesis, for example, "EM1032-A49-VH (G55A) " means EM1032-A49-VH with an extra mutation G55A, and "EM1032-A86-VH (G100aA) " represents EM1032-A86-VH plus mutation G100aA.

[0470] *CDR sequences according to the Kabat scheme are single underlined; framework back-mutations are double underlined; NG (Asn-Gly) to NA (Asn-Ala) , NS (Asn-Ser) to NA (Asn-Ala) , DG (Asp-Gly) to DA (Asp-Ala) and DS (Asp-Ser) to DA (Asp-Ala) mutations are in bold italic.

[0471] Example 3: Generation and characterization of humanized anti-ALPP / ALPPL2×CD3 FIT-Ig bispecific antibodies

[0472] A group of FIT-Ig proteins recognizing both human ALPP / ALPPL2 and human CD3 were constructed utilizing VH / VL sequences in Table 8 as anti-ALPP / ALPPL2 moiety, VH / VL sequences in Table 2 as anti-CD3 moiety, and human constant region sequences in Table 3.

[0473] The specific methods referred to those recorded in Example 1.2 above, except that anti-ALPP / ALPPL2 VH and VL used are humanized sequences as shown in Table 8.

[0474] Information of constructed humanized ALPP / ALPPL2×CD3 FIT-Ig proteins are listed in Table 9.

[0475] Recombinant FIT-Ig listed in Table 9 were transiently expressed and purified. Specifically, for each FIT-Ig construct, 3 plasmids encoding each of the 3 polypeptide chains were co-transfected into Expi293 cells. After approximately six days of post-transfection cell culture, supernatants of each of cell cultures were separately harvested and subjected to Protein A affinity chromatography, followed by size exclusion chromatography (SEC) using a TSKgel SuperSW3000, 300 x 4.6 mm, SEC column (TOSOH) . A DIONEXTM UltiMate 3000 HPLC instrument (Thermo Scientific) was used for SEC using UV detection at 280 nm and 214 nm. The expression and SEC-HPLC results were shown in Table 10 below.

[0476] The FIT-Ig proteins were assayed for and ranked by dissociation rate constant (koff, "off-rate" ) using an  biolayer interferometry (Pall FortéBio LLC) . Specifically, anti-hIgG Fc Capture (AHC) Biosensors (Pall) were first exposed to antibody at a concentration of 100 nM for 120 seconds to capture antibodies, then dipped into running buffer (1X PBS pH 7.2, 0.05%Tween 20, 0.1%BSA) for 30 seconds to check baseline. Sensors with captured antibodies were dipped into 6.05 ug / ml recombinant human ALPPL2 protein solutions for 200 seconds to measure association, followed by dipped into running buffer for 500 seconds to measure dissociation. The association and dissociation curves were fitted to a 1: 1 Langmuir binding model using FortéBio Data Analysis software (Pall) . Results are shown in Table 10 below. The off-rate ratios were calculated by the off-rate of antibody to that of chimeric bispecific FIT-Igs, respectively. Lower ratio indicates slower dissociation of the antibody in comparison with the parental chimeric antibodies.

[0479] To eliminate heterogeneity during manufacturing, point-mutations evaluated in Example 2 were further introduced into humanized VH / VL sequences of anti-ALPP / ALPPL2 antibodies, as shown in Table 11 below.

[0480] Table 11: VH / VL point-mutation design for anti-ALPP / ALPPL2 antibodies

[0481] *CDR sequences according to the Kabat scheme are single underlined; framework back-mutations are  NG (Asn-Gly) to NA (Asn-Ala) , NS (Asn-Ser) to NA (Asn-Ala) , DG (Asp-Gly) to DA (Asp-Ala) and DS (Asp-Ser) to DA (Asp-Ala) mutations are in bold italic.

[0482] Example 4.1 Construction and expression of anti-ALPP / ALPPL2×CD3 humanized FIT-Igs

[0483] The FIT-Ig humanized bispecific antibodies with high or medium-affinity anti-CD3 module were further constructed employing the anti-CD3 VH / VL sequences in Table 2 as well as humanized anti-ALPP / ALPPL2 modules in Table 11, following the same method described in Example 3. FIT-Ig identifier with suffix (h) indicates a FIT-Ig protein carrying a high affinity anti-CD3 module, while the one with suffix (m) represents a FIT-Ig having a medium affinity anti-CD3 module, as shown in Table 12. No linkers between the immunoglobulin domains were used. The pairing of VH / VL sequences in those FIT-Ig binding proteins is listed in Table 12, and the amino acid sequences are provided in Table 13.

[0484] Table 12: Production of humanized FIT-Ig proteins

[0485] Recombinant FIT-Ig proteins in Table 13 were transiently expressed and purified following the procedure in Example 3. Specifically, for each FIT-Ig construct, 3 plasmids encoding each of the 3 polypeptide chains were co-transfected into Expi293 cells. After approximately six days of post-transfection cell culture, the supernatants of each of cell cultures were harvested and subjected to Protein A affinity chromatography, followed by size exclusion chromatography (SEC) using a TSKgel SuperSW3000, 300 x 4.6 mm, SEC column (TOSOH) . A DIONEXTM UltiMate 3000 HPLC instrument (Thermo Scientific) was used for SEC using UV detection at 280 nm and 214 nm. The expression and SEC-HPLC results were shown in Table 14 below.

[0487] Example 4.2 Construction of anti-ALPP / ALPPL2×CD3 humanized MAT-Fabs

[0488] A group of anti-ALPP / ALPPL2×CD3 humanized MAT-Fab proteins were also generated according to the method as described in International publication No. WO2018035084, incorporated by reference herein in its entirety. The obtained proteins are named with prefix “MAT” as shown in Table 15 and Figure 1B, and the pairing of variable domains for each MAT-Fab protein is also provided herein.

[0491] Example 5.1: Binding kinetics

[0492] Binding affinities and kinetics constants of humanized ALPP / ALPPL2 FIT-Igs were determined using a grating-coupled interferometry (GCI) based WAVEsystem (Creoptix AG, Switzerland) , a label-free biosensor. Briefly, goat anti-human IgG Fc antibody was immobilized onto a PCP WAVEchip (Creoptix AG) at a density of 2248.7 pg / mm2 on Channel 1, 2215.1 pg / mm2 on Channel 2, 2224.6 pg / mm2 on Channel 3 and 2442.5 pg / mm2 on Channel 4 by amine coupling. FIT-Ig was then captured onto the PCP WAVEchip at a density of 348-161.1 pg / mm2. Recombinant human / cyno ALPP or ALPPL2 proteins were injected at 3-fold serial diluted concentrations (1.23 nM, 3.7 nM, 11.11 nM, 33.33 nM, 100 nM) in kinetics buffer (BR100669Cat No 22064428-AD) . Recombinant proteins were injected for 250s at a flow rate of 60 μL / min and dissociation was set to 1000s. After each round of association and dissociation, WAVEchip surface was regenerated by injection of Glycine-HCl buffer at pH 1.5 for 120s at a flow rate of 80 μL / min. All sensorgrams were recorded at 25℃ and the data analyzed on the WAVEcontrol (Creoptix AG) . Data was double normalized by subtracting the signals from blank injections and from the reference channel. A 1: 1 Kinetic model was used for data fitting. Kinetics constants are shown below in Table 16.

[0493] Table 16: Binding kinetics of anti-ALPP / ALPPL2 humanized FIT-Ig and MAT-Fab bispecific antibodies

[0494] Example 5.2: Cell surface binding activity

[0495] Cell binding activity of anti-ALPP / ALPPL2×CD3 antibodies were measured with a human Jurkat cell line and ALPP / ALPPL2 over-expressing cell lines (CHOK1-huALPPL2, HEK293-huALPP) . After another round of centrifugation and a washing step, cells were resuspended in FACS buffer for reading on a CytoFLEX Flow Cytometer (Beckman Coulter) . Median Fluorescence Intensity (MFI) readouts were plotted against antibody concentration and analyzed with GraphPad Prism 6.0 software. As shown in Figure 5, FIT-Igs have more cells binding potency than MAT-Fabs, due to different valence against an antigen. Figure 5 shows constructed humanized anti-ALPP / ALPPL2×CD3 FIT-Ig and MAT-Fab bispecific antibodies all can bind to Jurkat cells, CHOK1 cells over-expressing ALPPL2, and / or HEK-293 over-expressing ALPP cells.

[0496] Example 6: Redirected CD3 activation of humanized FIT-Ig and MAT-Fab bispecific antibodies

[0497] To measure redirected CD3 activation by anti-ALPP / ALPPL2×CD3 bispecific FIT-Ig and MAT-Fab antibodies, a co-cultured reporter gene assay was conducted, in which Jurkat-NFAT-luc cells were used. The specific method referred to Example 1.6.

[0498] The results are shown in Figure 6. Bispecific anti-ALPP / ALPPL2×CD3 antibodies in two formats all can activate CD3 in the presence of ALPP-expressing target cells (Figure 6A-B) or in the presence of ALPPL2-expressing target cells (Figure 6C-D) , and have higher activity potency than anti-CD3 antibody (huEM0006-01-24) . Bispecific anti-ALPP / ALPPL2×CD3 antibodies all showed less non-target redirected activation than anti-CD3 antibody (huEM0006-01-24) , in the absence of ALPP / ALPPL2-expressing target cells (Figure 6E-F) .

[0499] Example 7: Redirected T cell cytotoxicity of humanized FIT-Ig and MAT-Fab bispecific antibodies

[0500] The tumor cell killing potency of anti-ALPP / ALPPL2×CD3 bispecific antibodies was measured in a redirected T cell cytotoxicity assay using the human pancreatic cancer cell line Capan-1 (ATCC, HTB-79) and NCI-H1650 (Cobioer, CBP60114) as target cells and human T cells as effector cells. Briefly, cells were harvested, washed, and suspended with assay medium (RPMI1640 with 10%FBS) . Target cells were seeded into flat-bottom 96-well plates (Corning, Cat. #3599) at 5×104 cells per well. T cells were purified from human PBMC with a commercial PBMC isolation kit (EasySepTM, Stemcell Technologies, Cat. #17951) and were added to the wells at 2×105 cells per well. Test articles were added and incubated with the mixture of the cells for 48 hours at 37℃. Lactate dehydrogenase (LDH) release was measured with a CytoTox cytotoxicity assay kit (Promega, Cat. #G1780) . OD490 readouts were obtained following the manufacturer's instructions. The max and min lysis were also generated according the CytoTox kit (Promega, #G1780) instruction. The max lysis was generated by adding lysis buffer to samples which only have tumor cells. The min lysis was generated from the culture medium background. The min lysis was subtracted from the readouts of all samples. Target cells max lysis (100%) minus minimal lysis (0%) was presented as the normalization denominator. The percentage of LDH release was plotted against the concentrations of bispecific antibodies.

[0501] As shown in Figure 7, anti-ALPP / ALPPL2×CD3 bispecific antibodies in two formats all redirect T cell cytotoxicity to Capan-1 and NCI-1650 tumor cells.

[0502] Example 8: Stability of anti-ALPP / ALPPL2×CD3 humanized FIT-Igs

[0503] To measure the stability of the humanized FIT-Igs, an accelerated stability assay and a freezing and thawing assay were used.

[0504] In the accelerated stability assay, FIT2024-10ah-4 (m) and FIT2024-10ah-4 (h) were reconstituted to 1 mg / ml, 50μl of the samples were incubated at 40℃ for 1 / 3 / 7 / 10 / 14 days respectively. Samples were detected by SEC-HPLC and BioDrop for purity identification.

[0505] In the freezing and thawing assay, 50μl of the samples were freezing and thawing for 5 cycles and detected by SEC-HPLC and BioDrop.

[0506] Results are shown in Figure 8 and indicate that all tested antibodies are stable in high temperature and freezing-thawing stress.

[0507] Example 9: Cytokine release induced by anti-ALPP / ALPPL2×CD3 bispecific antibodies

[0508] The ability of test articles to induce in vitro cytokine release from PBMC was assessed. Cryopreserved PBMCs from normal healthy human volunteers were thawed and cultured in RPMI 1640 medium (Gibco RPMI-1640 Medium supplemented with 10%FCS, 100 IU / mL penicillin, 100 pg / mL streptomycin) with viability >85%as determined automated cell counter prior to experiments. PBMC were seeded at a density of 2×107 cells per well into 96-well-plate, followed by incubation at for 48 hours at 37℃, 5%CO2. After incubation, the cells were stimulated with the test articles, positive controls, or isotype controls for 24 hours at 37℃, 5%CO2. Then 96-well plates were centrifuged, supernatants were harvested, respectively. Detection of the IFNγ and TNFα in the supernatants was performed using an antibody capture bead-based kit (LEGENDplexTM HU Th1 Panel (5-plex) w / VbP V02, Biolegend) and flow cytometric analysis.

[0509] Figure 9 shows that FIT2024-10ah-4 (m) and FIT2024-10ah-4 (h) induced much lower cytokines release compared to control molecules.

[0510] Example 10: Redirected T cell cytotoxicity of anti-ALPP / ALPPL2×CD3 FIT-Igs

[0511] The tumor cell killing potency of anti-ALPP / ALPPL2×CD3 bispecific antibodies was further measured in a redirected T cell cytotoxicity assay using the human pancreatic cancer cell line NCI-H1650, CaoV-3 (Cobioer, CBP60284) and RMUG-S (JCRB Cell Bank, IFO50320) as target cells and human T cells as effector cells.

[0512] Briefly, cells were harvested, washed, and suspended with assay medium (RPMI1640 with 10%FBS) . Target cells were seeded into flat-bottom 96-well plates (Corning, Cat. #3599) at 5×104 cells per well. T cells were purified from human PBMC with a commercial PBMC isolation -kit (EasySepTM, Stemcell Technologies, Cat. The max and min lysis were also generated according to the CytoTox kit (Promega, #G1780) instruction.

[0513] As shown in Figure 10, FIT2024-10ah-4 (h) -YTE stimulates T cell cytotoxicity to NCI-H1650, CaoV-3 and RMUG-Stumor cells with greatly higher potency than reference molecules including parent anti-CD3 mab (HuEM0006-01-24) , h12F3 (derived from WO2022197890A1) conjugates such as h12F3-MMAE and h12F3-DXD, irrelevant Ab, and hIgG controls.

[0533] Anti-tumor efficacy was further evaluated in Capan-1 human pancreatic cancer xenograft model in PBMC engraft NCG mice, which is an immunodeficient strain lacking T cells, B cells and natural killer cells. 8-10 weeks female NCG mice (GemPharmatech) were used for human PBMC reconstitution and Capan-1 tumor model establishment. Briefly, the mice received a single intraperitoneal injection of 8×106 human PBMC. After 5 days, 5×106 Capan-1 cells suspended in 0.2ml DPBS (Dulbecco's Phosphate-Buffered Saline) containing 50%ABW Matrigel per animal were subcutaneously injected into the right flank of female NCG mice. Seven days post tumor cell inoculation, huCD45 cells in mouse peripheral blood were detected using FACS, and tumor bearing mice with tumor size of 82.55-167.92 mm3 were selected. Based on the tumor size and huCD45 cells counts, the selected mice were randomized (Stratified blocked randomization) and divided into 6 groups of 8 mice each, and treatment was initiated on the same day (Day 0) . Mice were treated with FIT2024-10ah-4 (h) -YTE (5 mg / kg, i.p., weekly) , RMC-6236 (MCE, HY-148439, a pan-KRAS mutant inhibitor, 10 mg / kg or 25 mg / kg, p.o., daily) , a combination of FIT2024-10ah-4 (h) -YTE (5 mg / kg, i.p., weekly) and RMC-6236 (10 mg / kg or 25 mg / kg, p.o., daily) , or DPBS (i.p., weekly) as control.

[0534] Tumor volumes and the percentage of tumor growth inhibition (TGI) compared to vehicle control (DPBS) are measured and defined following the method mentioned in Example 11.1.

[0535] All treatment regimens were well tolerated by the animals (data not shown) . Figure 15 illustrates the average tumor volumes measured at multiple time points, demonstrating that various treatments led to inhibition of tumor growth. On Day 18, tumor growth inhibition (TGI) ratios calculated based on tumor volume were as follows: 96.57%for FIT2024-10ah-4 (h) -YTE alone, 68.82%for RMC-6236 at 10 mg / kg, 94.91%for RMC-6236 at 25 mg / kg, 114.34%for the combination of FIT2024-10ah-4 (h) -YTE and RMC-6236 at 10 mg / kg, and 121.98%for the combination of FIT2024-10ah-4 (h) -YTE and RMC-6236 at 25 mg / kg.

[0536] Statistical analysis showed significant differences between the combination group and the respective monotherapy groups. Administration of either FIT2024-10ah-4 (h) -YTE antibody or compound RMC-6236 alone led to inhibition of tumor growth while co-administration of the antibody and RMC-6236 induced almost complete regression of established tumors, indicating that the combination achieved a synergistic anti-tumor growth inhibition.
