Aspects of the disclosure encompass improvements of immune cell therapies. To improve the cells, reduced activity or expression of a gene of interest, e.g., but not limited to, MED12, CCNC, ARIH2, and / or TGFBR2, may be induced. Engineered cells of the disclosure may comprise increased activity of expression of heterologous genes and / or genetic constructs to improve immune cell activity. Engineered cells of the disclosure may be more effective in immunosuppressive environments and / or against cancer.
