[0005] Lisdexamfetamine dimesylate is used to treat attention deficit hyperactivity disorder (ADHD) in adults and children 6 years of age and older. This medicine is also used to treat moderate to severe binge eating disorder (BED). It belongs to the group of medicines called central nervous system (CNS) stimulants.

[0006] Lisdexamfetamine dimesylate increases attention and decreases restlessness in children and adults who are overactive, cannot concentrate for very long, or are easily distracted and impulsive. This medicine is used as part of a total treatment program that also includes social, educational, and psychological treatment.

Because of its hygroscopic nature, Microcrystalline Cellulose is generally not an effective compound especially for those hygroscopic or deliquescent active substances. For the purpose of improving the stability of some active substances, such as lisdexamfetamine dimesylate, the hygroscopicity of microcrystalline cellulose can be problematic.

[0015] Detailed Description of the Invention

[0016] The main object of the present invention is to provide a capsule formulation comprising lisdexamfetamine dimesylate having the high stability and flowability.

[0017] Another object of the present invention is to provide a process for a capsule formulation comprising lisdexamfetamine dimesylate prepared by simple, easy, time-saving and fast manufacturing methods.

[0018] Lisdexamfetamine dimesylate formulation is known in the prior art, but there is still a need for a formulation with high stability and flowability. Calcium phosphate dibasic anhydrous was used as filler in the formulation due to the moisture sensitivity of lisdexamfetamine dimesylate. The calcium phosphate dibasic anhydrous is a non-hygroscopic excipient. The calcium phosphate dibasic anhydrous used as a filler helps to optimize moisture absorption capacity of pharmaceutical product and improve powder flow in manufacturing. We found that the use of anhydrous calcium phosphate dibasic as filler in the formulation increased the stability of the formulation and improved the flowability.

[0019] According to one embodiment of the invention, a capsule formulation comprising lisdexamfetamine dimesylate and at least one filler, wherein filler is calcium phosphate dibasic anhydrous.According to one embodiment of the present invention, the amount of the lisdexamfetamine dimesylate is between 25.0% and 45.0% by weight of the total formulation. Preferably, it is between 33.0% and 42.0% by weight of the total formulation.

CLAIMS1. A capsule formulation comprising lisdexamfetamine dimesylate and at least one filler, wherein filler is calcium phosphate dibasic anhydrous.

2. The capsule formulation according to claim 1, wherein the amount of the lisdexamfetamine dimesylate is between 25.0% and 45.0% by weight of the total formulation.

3. The capsule formulation according to claim 1, wherein the amount of filler is between 45.0% and 65.0% by weight of the total formulation.

4. The capsule formulation according to claim 1, wherein the capsule formulation further comprises at least one pharmaceutically acceptable excipient selected from the group comprising disintegrants, lubricants or mixtures thereof.

The capsule formulation according to claim 4, wherein disintegrants are selected from the group comprising sodium starch glycolate, croscarmellose sodium, crospovidone, low-substituted hydroxypropyl cellulose, starch, calcium carboxymethyl cellulose, pregelatinized starch, docusate sodium, guar gum, sodium alginate, alginic acid, magnesium aluminum silica, poloxamer, sodium glycine carbonate or mixtures thereof.

6. The capsule formulation according to claim 5, wherein the disintegrant is sodium starch glycolate.

7. The capsule formulation according to claim 5 or 6, wherein the amount of disintegrant is between 0.5% and 10.0% by weight of the total formulation.

The capsule formulation according to claim 4, wherein lubricants are selected from the group comprising sodium stearyl fumarate, magnesium stearate, talc, colloidal silicon dioxide, corn starch, calcium stearate, zinc stearate, magnesium lauryl sulfate, sodium oleate, sodium acetate, stearic acid, fumaric acid, glyceryl palmito sulphate or mixtures thereof.

9. The capsule formulation according to claim 8, wherein the lubricant is sodium stearyl fumarate.

10. The capsule formulation according to claim 8 or 9, wherein the amount of lubricant is between 0.1% and 5.0% by weight in the total formulation.

The capsule formulation according to any preceding claims, wherein the capsule formulation comprises;- Lisdexamfetamine dimesylate- Calcium phosphate dibasic anhydrous- Sodium starch glycolate- Sodium stearyl fumarate12. A process for capsule formulation according to any of the preceding claims comprising lisdexamfetamine dimesylate comprising the following steps:a. Sieving and blending lisdexamfetamine dimesylate and filler,b. Sieving disintegrant and blending it with the powder mixture obtained in (a), c. Sieving the lubricant and blending it with the powder mixture obtained in (b), d. Filling the final mixture obtained in (c) into capsules.
