Description

Technical Field

[0001] This application claims the priority of Korean Patent Application No. 10-2022-0035142, filed on Mar. 22, 2023, the entire text of which is incorporated by reference into this application.

[0002] The present invention relates to novel compounds that induce the expression of the anti-aging gene klotho. Background Art

[0003] The senescence-accelerated mice (SAM) reported in 1981 revealed the possibility of genes that can regulate animal aging. During the mating of AKR / J rats, the rats produced by chance aged more rapidly than rats of the same series, and it was confirmed that the rats had multiple gene mutations. Later, a group of genes related to aging were reported in the 1990s. The reported genes belong to the RecQ family, which is a gene that expresses a DNA helicase. It is reported that if a mutation occurs in this gene, it will cause premature aging or cancer, and it is known that it affects DNA repair. The single gene related to aging is the klotho gene reported in 1997. The Klotho gene was accidentally discovered when creating a transformed animal model of hypertensive rats. In rats that cannot express this gene, premature aging occurs and the life span is shortened. More interestingly, the life span of male rats that later increased the expression of this gene was extended by 20.0% to 30.8%, and the life span of female rats was extended by 18.8% to 19.0%. This is the first time that the world has been informed of the fact that the life span of rats can be extended or shortened by the expression of a single gene. In addition, the base sequence of the Klotho gene is very similar between animals, and it is reported that rats and humans have 98% of the same sequence. This means that humans can also regulate their life span by adjusting the expression of the Klotho gene.

[0004] In humans, the klotho gene is located in chromosome 13 and produces a membrane protein with a base sequence similar to that of β-glucosidase. It is reported that the Klotho protein is mainly expressed in renal tubular epithelial cells and the choroid plexus of the brain, and in some parathyroid glands. The Klotho gene is a gene associated with various aging phenotypes. In rats lacking the klotho gene, syndromes similar to the aging process such as shortened lifespan, decreased activity, growth retardation, atherosclerosis, arterial calcification, osteoporosis, immature genitals, infertility, skin atrophy, and emphysema occur. In Klotho mutant rats, arteriosclerosis similar to Monckeberg-type arteriosclerosis caused by human aging is observed in all arteries from the aorta to the arterioles, and disorders occur in angiogenesis and vasculogenesis.

[0005] The expression of Klotho mRNA in kidney tissue is significantly higher than that in other tissues, but the expression of klotho mRNA is reduced in the kidneys of rats with hypertension, type 2 diabetes, diabetic nephropathy, and chronic renal failure models. In rats with reduced Klotho expression, the production of nitric oxide (NO), a relaxing factor derived from the endothelium, is reduced. If the klotho gene is injected into Otsuka Long-Evans Tokushima fatty rats (OLETF) that have many risk factors for cardiovascular disease, the vascular intimal dysfunction is improved, the production of NO is increased, and vascular thickening and fibrosis are inhibited, thereby lowering blood pressure. In addition, the klotho gene affects glucose and insulin metabolism in rats, and statins, which are representative hypercholesterolemia treatment agents, increase the expression of klotho mRNA in renal proximal tubular cells. In rats with reduced Klotho expression, osteopenia with low bone turnover caused by impaired differentiation of osteoblasts and osteoclasts is induced, which is similar to the characteristics of human age-related bone loss and senile osteoporosis. In addition, in Klotho mutant rats, abnormal elongation of epiphyseal trabeculae and abnormal trabecular organization in micro-computed tomography imaging were observed, which is caused by obstacles in the bone resorption process. The clinical phenotypic changes caused by Klotho gene mutations observed in humans are diverse. Functional variants of klotho (KL-VS) with mutations in three sites of Klotho gene exon2 are associated with lipid metabolism, blood pressure, life span, cognitive function, coronary artery disease and cerebrovascular disease, and microsatellite polymorphisms and single nucleotide polymorphisms of Klotho gene are associated with bone density. It has been reported that single nucleotide polymorphisms of Klotho gene are also associated with risk factors for cardiovascular disease and bone density in healthy adult women. Recently, the association between the Klotho gene and Alzheimer's disease has been reported by multiple papers. It is reported that in Alzheimer's disease and dementia mouse models, if klotho is overexpressed, the life span of the mice is extended by 30% and cognitive decline is suppressed. It was also observed that the production of beta-amyloid protein in the brain was reduced by 50% by the expression of klotho. In humans, the expression of klotho is inversely proportional to the progression of Alzheimer's disease, and a report that klotho protein reduces the amount of inflammatory cytokines in the blood of Alzheimer's patients has been submitted. According to reports of multiple research results, Klotho has the effect of enhancing the function of the retinal pigment epithelium (RPE) and protecting RPE cells from oxidative stress. It is known that in the retina of (KL- / -) mice with Klotho gene removed, the degeneration of photoreceptors and the reduction of pigment production in RPE cells are shown. Klotho expression is also confirmed in cultured RPE cells, and experiments confirm that the expressed klotho protein increases the synthesis of L-3,4-dihydroxyphenylalanine (L-3,4-dihydorxyphenylalanine, L-DOPA) and inhibits the generation of vascular endothelial growth factor (VEGF) in the basement membrane. In addition, it is confirmed through research results that klotho has the function of protecting RPE cells from oxidative stress by reducing the generation of reactive oxygen species (ROS) in RPE cells. As a result of experiments on patients with age-related macular degeneration (AMD), the expression of klotho in aqueous humor of the eyeball is reduced, and it is confirmed that this reduction effect is related to oxidative stress and inflammation.

[0006] As a result of continuing to study compounds that induce klotho expression, the present inventors have experimentally confirmed that the novel compound developed by the present inventors has excellent stability and excellent protective effect against oxidative stress (oxidative toxicity) in retinal pigment epithelial cells (RPE), thereby completing the present invention. [0127] Experimental Example 1. Evaluation of the stability of compounds

[0128] Each KS compound of Example 1 was dissolved in 10 mM dimethyl sulfoxide (DMSO) and used, and tolbutamide used as a standard substance was dissolved in methanol at a concentration of 10 mM and used.

[0129] In a glass test tube (13×100 mm), 0.1 M potassium dihydrogen phosphate (KH2PO4) (147.5 μl), 0.1 M magnesium chloride (MgCl2) (25 μl), and 50 μl of 0.5 mg / ml human liver microsomes (mixed-gender pool) purchased from Corning (Corning Life Science) were mixed with each KS compound solution to a final concentration of 1 μM, and pre-incubated in a shaking water bath (37°C, 40 rpm) for 3 minutes, and then incubated for 30 minutes with or without the addition of 25 μl of 10 mM reduced nicotinamide adenine dinucleotide phosphate (NADPH).

[0130] After taking 50μl of the sample before and after the reaction, add 2ml of acetonitrile to stop the reaction and vortex for 1 minute. After centrifuging (4000rpm, 4°C) the mixed solution for 20 minutes, transfer each supernatant to a new glass test tube and dry it for 2 hours using a centrifugal concentrator. After confirming that it is completely dry and adding 200μl of 40% acetonitrile to each glass test tube, vortex for 1 minute to resuspend. After centrifuging (4000rpm, 4°C) the resuspended solution for 10 minutes, take 150μl of the supernatant for use in HPLC-UV analysis. Agilient 1200series was used for HPLC-UV analysis, and Agilent 6410 was used for MS / MS analysis. The conditions used in the analysis are as follows.

[0131] Table 1

[0132] Conditions used for HPLC analysis

[0133]

[0134] Table 2 Conditions used for LC-MS / MS analysis

[0135]

[0136] Figure 1 The results of the KS compounds incubated with human liver microsomes were quantitatively confirmed by MS / MS analysis. Microsome enzymes work in a coenzyme NADPH(+)-dependent manner, and the compounds are metabolized only in reactions with the addition of NADPH(+). The KS compounds showed higher stability than tolbutamide used as a control, and KS104 was found to have the best microsomal stability.

[0137] Experimental Example 2. Comparison of the Effects of Klotho Gene Expression

[0138] The KS compounds of Example 1 were treated with human renal proximal tubular epithelial cells (RPTEC) at different concentrations, and messenger RNA (mRNA) was collected from the cells after 6 hours and the expression of the messenger RNA of the klotho gene was confirmed by polymerase chain reaction. Renal proximal tubular epithelial cells (RPTEC) were purchased from Lonza and cultured at 37°C and 5% CO2 for use in the experiment. After each KS compound was placed in the cell culture medium and exposed for 6 hours, ribonucleic acid (RNA) was isolated from the cells and converted into complementary deoxyribonucleic acid (cDNA) using reverse transcriptase and klotho was amplified by polymerase chain reaction to confirm the expression level. Dimethyl sulfoxide was added instead of the compound to be used as a control group.

[0139] The information of primers used in the experiments is as follows.

[0140] KL-F: GATAGAGAAAAATGGCTTCCCTCC (sequence 1)

[0141] KL-R: GGTCGGTAAACTGAGACAGAGTGG (sequence 2)

[0142] GAPDH-F: TGACAACTTTGGTATCGTGGAAGG (sequence 3)

[0143] GAPDH-R: AGGGATGATGTTCTGGAGAGCC (sequence 4)

[0144] Figure 2 shows the results of confirming the expression of the klotho gene by polymerase chain reaction. A gel photograph and a graph showing the expression level of the bands shown in the gel photograph by quantifying them using Image J. The upper band in the gel photograph shows the secreted form of klotho, and the lower band shows the membrane form of klotho. The quantitative graph shows the two bands together. The expression effect of KS104 showed the best effect, and it was confirmed that the ability to express the klotho gene was more than twice as high as not only KS101, but also other fluorine-binding compounds.

[0145] Experimental Example 3. Evaluation of cytotoxicity

[0146] The KS compound of Example 1 was treated with different concentrations in cultured human retinal epithelial cells (ARPE) and cultured for 24 hours. Then, the cells were placed in EX-cytox of GOGEN Company to react for 1 hour and the change in absorbance was measured. The lower the absorbance, the more damaged the cells were due to toxicity. Figure 3 As shown, it was confirmed that KS104 had the lowest cytotoxicity.

[0147] Experimental Example 4. Analysis of intracellular reactive oxygen species

[0148] Renal proximal tubular epithelial cells (RPTEC) were treated with the KS compounds of Example 1 to examine how the amount of intracellular reactive oxygen species changes.

[0149] 2.5 μM of each KS compound was added to the cultured RPTEC cells and cultured for 6 hours. Then, Invitrogen's Reactive Oxygen Detection Reagents were added and cultured for 1 hour. The fluorescence change was measured using a spectrophotometer to confirm the amount of reactive oxygen species (ROS) present in the cells.

[0150] Figure 4 This is a graph showing the amount of active oxygen remaining in RPTEC cells. It was confirmed that the amount of active oxygen in cells treated with KS103 compound and KS104 compound was reduced by about 30% compared with cells treated with only a solvent (dimethyl sulfoxide).

[0151] Experimental Example 5: Confirmation of changes in oxidative stress-related gene expression

[0152] After adding 2.5 μM of each compound to relatively aged 7th passage (#7) renal proximal tubule epithelial cells (RPTEC) and culturing for 6 hours, messenger RNA was collected and total gene expression was confirmed using a next-generation sequencer. In order to confirm the changes in gene expression caused by aging, the genes (TXN, AKT1, NR4A3, NLRP3, FOXP3 genes) whose expression increased in young cells were confirmed by comparing the gene expression with that in relatively young 4th passage (#4) RPTEC cells. The expression levels of the relevant genes in young cells were compared with the expression change values caused by treating the compounds of Example 1 (treating the compounds of Example 1 to aged cells). As a result, it was confirmed that when the KS104 compound was treated to aged cells, the expression of the relevant genes was induced more significantly than that in young cells ( Figure 5 , Figure 6 ).

[0153] Experimental Example 6. Protective effect on retinal pigment epithelial cells

[0154] In order to confirm the preventive or therapeutic effect of the KS compound of Example 1 on macular degeneration, the effect of protecting retinal pigment epithelial cells from oxidative stress was confirmed.

[0155] First, human retinal pigment epithelial cells (ARPE) purchased from ATCC in the U.S. were used. 10,000 ARPE cells were placed in each well and cultured for 18 hours, and then the medium was replaced with DMEM F-12 medium containing no serum (fetal bovine serum (FBS)).

[0156] In this case, each KS compound was added at a concentration of 2.5 μM, and buthionine sulfoximine (BSO) was treated at a concentration of 50 mM for 6 hours to induce oxidative stress. In order to measure the viability of these cells, Cyto X (Cell Viability Assay Kit) was added and the absorbance over time was measured at a wavelength of 450 nm.

[0157] As a result, it was confirmed that both KS103 and KS104 compounds inhibited cell apoptosis caused by oxidative stress ( Figure 7 ).
