Provided herein are triazacyclododecanesulfonamide ("TCD")-based protein secretion inhibitors, such as Sec61 inhibitors, methods for their preparation, related pharmaceutical compositions, and methods for their use. For example, provided herein are compounds of Formula (I) and pharmaceutically acceptable salts and compositions comprising them. The compounds disclosed herein can be used, for example, in the treatment of diseases involving inflammation and / or cancer.
