Abstract
Description
Skin beauty method, multiple component skin penetration method, and multiple component skin penetration system
[0001] The present invention relates to a skin cosmetic method for cosmetically treating skin, a multiple component skin permeation method, and a multiple component skin permeation system.
[0002] A skin beauty composition containing an acetic acid extract of onion bulbs and baking soda, which improves wrinkles and has an excellent feel when used, and a skin beauty method (excluding medical procedures) involving application of the composition have been proposed (see Patent Document 1). In this skin beauty method, the acetic acid extract of onion bulbs alone is known to provide wrinkle improvement, moisturizing effects, and skin beautifying effects (improved skin texture), but it has been found that the inclusion of baking soda in the composition also provides more excellent skin beauty effects (particularly wrinkle improvement) in a shorter period of time. However, the beauty industry is seeking further benefits, including a youthful appearance effect. That is, in addition to wrinkle improvement, moisturizing effects, and skin beautifying effects, a lifting effect that improves facial sagging and wrinkles is also desired. However, the above-mentioned beauty method does not provide a lifting effect.
[0003] Patent No. 6973740
[0004] In view of the above-mentioned problems, an object of the present invention is to provide a sufficient youthful appearance effect and improve user satisfaction.
[0005] This invention is characterized by a skin beauty method (excluding medical procedures) comprising the steps of applying a first composition containing one or more selected from a sirtuin gene activator, a growth factor agent, a proteoglycan, and a cannabinoid, and applying a second composition containing one or more other than the first composition selected from the sirtuin gene activator, the growth factor agent, the proteoglycan, and the cannabinoid.
[0006] The present invention can provide a skin beauty method, a multiple component skin penetration method, and a multiple component skin penetration system that can provide a sufficient youthful appearance effect.

[0008] This invention relates to a first composition containing a first active ingredient and a second composition containing a second active ingredient, which are applied to the skin separately or in combination. One of the first active ingredient and the second active ingredient is a growth factor agent such as a human stem cell culture agent, and the other is a sirtuin gene activator such as NMN. Furthermore, as a method of application, the first penetrating agent containing the first active ingredient and the second penetrating agent containing the second active ingredient can be adjusted individually or collectively with an adjuster such as purified water to form a spray liquid (application liquid), which can then be applied. Application methods include application using a penetration device that promotes penetration into the skin, application with the fingers, or application using a pack. The penetration device can be any suitable device, such as a spray device that sprays, an iontophoresis device that applies a weak negative current to the skin to allow negatively ionized cosmetic ingredients to penetrate the skin, an electroporation device that applies an electric pulse to the skin to temporarily create microscopic holes in the cell membrane and allow cosmetic ingredients to penetrate to the dermis, or a roller-type penetration promoter with multiple microscopic needles attached to the periphery of a cylindrical roller. Note that the term "skin" used here refers to all skin on the body, including the face, neck, décolleté, limbs, chest, abdomen, back, pubic area, and head. An embodiment of the present invention will now be described with reference to the accompanying drawings.
[0009] 1 is a diagram showing the system configuration of a multiple-component skin penetration system 1. The multiple-component skin penetration system 1 is composed of a first penetrating agent 2, a second penetrating agent 3, an adjusting agent 4, and a spray device 5.
[0010] The first penetrating agent 2 is an agent containing a first active ingredient to be penetrated into the skin, and is composed of a liquid or powder. The second penetrating agent 3 is an agent containing a second active ingredient to be penetrated into the skin, and is composed of a liquid or powder.
[0011] In this example, one of the first penetrating agent 2 (first active ingredient) and the second penetrating agent 3 (second active ingredient) is a sirtuin gene activator such as NMN (NMN agent), and the other is a growth factor agent such as a human stem cell culture agent.
[0012] The sirtuin gene activator may be an NAD+ precursor, an NAD+ analog, NAD (NAD+), niacinamide, or a combination thereof, and may be in the form of a powder or liquid. The NAD+ precursor may be nicotinamide mononucleotide (hereinafter referred to as NMN). The NAD+ analog may be deazaflavin or 5-deazaflavin. The 5-deazaflavin may be 5-deazaflavin (TND1128) or 5-deazaflavin (F420), or both. The sirtuin gene activator may be in the form of a powder or liquid, and in the case of a powder, it may be powdered by microgranulation or freeze-drying.
[0013] NMN is a powder or liquid form of nicotinamide mononucleotide (NMN) that is found in breast milk, edamame, broccoli, avocado, tomato, and other organisms. Liquid NMN is extracted from these organisms. Powdered NMN is formed by granulation, such as microgranulation.
[0014] The growth factor agent can be a growth factor, exosome, stem cell culture supernatant, or a combination thereof, and can be in the form of a powder or liquid. The growth factor can be EGF (epidermal growth factor), FGF (fibroblast growth factor), PDGF (platelet-derived growth factor), IGF-1 (insulin-like growth factor), HGF (hepatocyte growth factor), KGF (epidermal growth factor), TGF-β (transforming growth factor), or a combination thereof. The FGF can be any FGF family member, and can be any one or more of FGF1 to FGF23, preferably any one or more of FGF1 to FGF14, more preferably any one or more of FGF1 to FGF10, and even more preferably FGF1 or FGF2. The growth factor agent can be in the form of a liquid or powder. In the case of a powder, the liquid growth factor agent can be freeze-dried and powdered.
[0015] The growth factor agent is derived from human stem cell culture supernatant, which can be obtained by culturing human stem cells of an appropriate origin, such as umbilical cord, umbilical cord blood, fat, or dental pulp, and can be in the form of a liquid or powder.
[0016] In culturing human stem cells, subculture is preferably limited to a certain number of generations, preferably up to five generations. Furthermore, subculture is preferably performed while the growth curve during culture is linear. This prevents contamination with senescent cells and prevents the introduction of growth factors from senescent cells.
[0017] The human stem cell culture agent can be a supernatant obtained when stem cells collected from a human are cultured in a culture medium, or a powder obtained by freeze-drying this supernatant using a freeze-drying method, and can be a human stem cell culture supernatant. The human stem cell culture supernatant can be a human umbilical cord stem cell culture supernatant obtained from human umbilical cord, a human umbilical cord blood stem cell culture supernatant obtained from human umbilical cord blood, a human adipose stem cell culture supernatant obtained from human fat, a human bone marrow stem cell culture supernatant obtained from human bone marrow fluid, or a supernatant obtained by culturing other human cells, or a supernatant powder obtained by freeze-drying these supernatants.
[0018] The first penetrating agent 2 and the second penetrating agent 3 are preferably each 100% sirtuin gene activator or 100% growth factor agent, but may each contain other components. Note that all % notations in this specification are expressed in mass percent.
[0019] The volume of the sirtuin gene activator may be 10 ml or more, preferably 50 ml or more, more preferably 100 ml or more, even more preferably 150 ml or more, and even more preferably 200 ml or more. The volume of the sirtuin gene activator may be 1000 ml or less, preferably 800 ml or less, more preferably 600 ml or less, and even more preferably 500 ml or less.
[0020] The amount of growth factor agent can be 0.1 mg or more, preferably 0.5 mg or more, and more preferably 1 mg or more, and can be 20 mg or less, preferably 15 mg or less, and more preferably 10 mg or less.
[0021] The adjusting agent 4 can be purified water or physiological saline. The purified water can be ordinary purified water or sterilized purified water obtained by sterilizing purified water. The physiological saline can be any appropriate type, such as injectable saline, which is purified water sterilized by filtration or distillation and further has pyrogens removed.
[0022] The amount of the adjuster 4 can be 0.5 ml or more, preferably 1 ml or more, and more preferably 2 ml or more. The amount of the adjuster 4 is preferably 20 ml or less, more preferably 15 ml or less, even more preferably 10 ml or less, and is suitably 5 ml or less.
[0023] The ratio of the growth factor agent, sirtuin gene activator, and regulator 4 can be set within the range of the amounts described above. Therefore, the above amounts are sufficient when applying mainly to the face or scalp, but the amounts can be increased within the above ranges when applying to the whole body.
[0024] The first penetrant 2 is adjusted with the adjuster 4 and used as a first spray liquid (first application liquid). Similarly, the second penetrant 3 is adjusted with the adjuster 4 and used as a second spray liquid (second application liquid). Note that the first penetrant 2 and the second penetrant 3 can also be mixed and adjusted with the adjuster 4 to be used as a single integrated spray liquid (integrated application liquid).
[0025] The spray liquid (first spray liquid, second spray liquid, or integrated spray liquid) prepared in this manner can have a viscosity of 200 mPa·s or less, preferably 100 mPa·s or less, more preferably 50 mPa·s or less, still more preferably 10 mPa·s or less, even more preferably 5 Pa·s or less, more preferably 3 mPa·s or less, still more preferably 2 mPa·s or less, and preferably 1 mPa·s or less. This prevents clogging inside the spray nozzle 12a of the spray device 5, which will be described later, when spraying.
[0026] The spray device 5 is a device that sprays pressurized liquid onto a target (human skin in this embodiment) through a nozzle, and will be described in detail below.
[0027] The spray device 5 has a main body 30 for delivering air, a conduit 20 connected at one end to the main body 30, and a handpiece 10 connected at the other end of the conduit 20 and operated by the practitioner.
[0028] The handpiece 10 is a longitudinal unit that can be held by the practitioner's hand. It has a spray cover 11 at its tip and the other end of the conduit 20 connected to its rear end. A liquid input section 15 is provided distally from the center of the top surface of the handpiece 10, into which the spray liquid, prepared by adjusting the first penetrant 2 and the second penetrant 3 with the adjuster 4, is input. The liquid input section 15 has a liquid input hole 15b in the lid of the top surface, and a liquid storage section 15a inside the handpiece 10. The liquid storage section 15a is a sterile chamber, and the spray liquid is input into the liquid storage section 15a through the liquid input hole 15b using a syringe or the like. A setting section 19 is provided distally from the center of the top surface of the handpiece 10, and includes a display section 19a (see FIG. 2) that displays the current spray setting, a switch button 19b (see FIG. 2) that switches the spray setting of the spray liquid, an operation button 19c (see FIG. 2) that switches between starting and stopping the spray of the spray liquid, and a control section 19d that executes various controls. A dial-type spray amount adjustment unit 18 is provided on the right side (front of Figure 1) of the handpiece 10 to adjust the amount of spray liquid per spray, and a manual spray button 16 is provided on the left side (back of Figure 1) of the handpiece 10 to set the spray setting and perform one spray when pressed during manual spraying.
[0029] The conduit 20 has a pipe 20a (see FIG. 2) for supplying gas (air) to the handpiece 10 and an electric wire 20b (see FIG. 2) for supplying power to the handpiece 10. The conduit 20 can be bent freely, allowing the practitioner to freely move the handpiece 10 for treatment while leaving the main body 30 in place.
[0030] The main body 30 has an operation start button 30a on the front for switching between starting and stopping operation, and a power cord (not shown) on the back for connecting to a household power source, and one end of the conduit 20 is connected to the side.
[0031] 2 is a block diagram showing the configuration of the multiple-component skin penetration system 1. A spray tip 12 is provided at the distal end of the handpiece 10, and a transparent, approximately cylindrical spray cover 11 is attached to the outer periphery of the spray tip 12. The spray cover 11 is cylindrical, with its tip cut perpendicular to the axis of the cylinder, and two gas release spaces 11a are provided at the tip, one on each side. That is, the tip is mostly a circular ring-shaped surface on a flat surface, and at least a portion of it is recessed to form the gas release space 11a. In the illustrated example, the gas release space 11a is formed by a recessed notch groove formed by partially switching the end portion, which is the open portion of the spray cover 11.
[0032] A spray nozzle 12a for spraying the spray liquid is provided at the center of the spray tip 12, which is the tip of the handpiece 10, and a gas discharge section 14 is provided at the back of the spray nozzle 12a. A spray gas chamber 14b is provided within this gas discharge section 14, and this spray gas chamber 14b is connected to the spray nozzle 12a. An air port 14a at the tip of the spray gas chamber 14b and the spray nozzle 12a are arranged opposite each other. A spray liquid chamber 13 is provided on the outer periphery between the air port 14a and the spray nozzle 12a, and this spray liquid chamber 13 is connected to the spray nozzle 12a. The spray liquid chamber 13 is a sterile chamber and is connected to a liquid storage section 15a that stores the spray liquid. In addition, the release speed of the spray liquid and gas released from the gas release section 14 through the spray nozzle 12a can be 100 km / h or more, preferably 500 km / h or more, and preferably 1,000 km / h or more. The upper limit of this release speed can be 5,000 km / h or less, preferably 3,000 km / h or less, and preferably 2,000 km / h or less. By releasing compressed gas at high speed in a short period of time, the spray liquid can be released in nano-sized form. By spraying (applying) the spray liquid in nano-sized form, it can penetrate deeper and deeper into the skin.
[0033] The outer periphery and the spray tip side of the spray tip 12 having the spray nozzle 12a are covered by the spray cover 11. Therefore, the spray liquid and gas sprayed from the spray nozzle 12a are confined within the spray cover 11 and sprayed (sprayed) forcefully onto the skin to be sprayed, and excess gas (including the spray liquid) is discharged to the outside through the gas release space 11a.
[0034] A solenoid 17 is provided inside the handpiece 10 downstream of the gas discharge unit 14. This solenoid 17 is provided midway along a pipe 20a that supplies gas to the nebulizing gas chamber 14b of the gas discharge unit 14, and switches between allowing and blocking the passage of gas through the pipe 20a. This solenoid 17 and the control unit 19d that controls it function as a spray control unit.
[0035] A control unit 19d (see FIG. 1) is provided inside the handpiece 10. In response to operation of the switch button 19b, the control unit 19d switches the spray setting of the spray liquid between one of the following: manual spraying, which performs a single spray when the manual spray button 16 is pressed; first intermittent spraying, which performs continuous, instantaneous, short-term spraying (e.g., 0.1 seconds) at a first slow time interval (e.g., 1.5 seconds); second intermittent spraying, which performs continuous, short-term spraying (e.g., 0.1 seconds) at a second time interval (e.g., 1.0 seconds) shorter than the first intermittent spraying; and third intermittent spraying, which performs continuous, short-term spraying (e.g., 0.1 seconds) at a third time interval (e.g., 0.5 seconds) even shorter than the second intermittent spraying. The instantaneous, short-term spraying can be spraying for 1 second or less, preferably spraying for 0.5 seconds or less, more preferably spraying for 0.3 seconds or less, even more preferably spraying for 0.2 seconds or less, and preferably spraying for 0.1 seconds or less. The lower limit of the instantaneous short-term spraying time can be set to 0.001 seconds or more, preferably 0.01 seconds or more, and more preferably 0.05 seconds or more.
[0036] Furthermore, when the manual spray button 16 is pressed when the spray setting is manual spray (or when the timing for spraying in the first intermittent spray, second intermittent spray, or third intermittent spray arrives), the control unit 19d turns on the solenoid 17 for a predetermined instantaneous time (for example, 0.1 seconds) during spraying to open the pipe 20a and allow gas to pass, and then turns off the solenoid 17 to close the pipe 20a and prevent gas from passing through. By controlling spraying in this manner, the moment the solenoid 17 is turned off, negative pressure is generated in the spray liquid chamber 13, and the spray liquid in the liquid storage unit 15a is supplied to the spray liquid chamber 13. The next time the solenoid 17 is turned on, gas is supplied instantaneously, and the spray liquid is sprayed together with the gas from the spray nozzle 12a.
[0037] The control unit 19d also performs an operation to display the current spray setting and an error code, etc., on the display unit 19a when an error occurs. The control unit 19d performs these operations after the operation is turned ON by the operation button 19c, and stops and does not perform each operation when the operation is turned OFF by the operation button 19c. When the solenoid 17 is OFF, the gas supplied from the pipe 20a flows into an exhaust path (not shown).
[0038] The manual spray button 16, solenoid 17, display unit 19a, switch button 19b, operation button 19c, and control unit 19d are supplied with power from the main body 30 via an electric wire 20b.
[0039] A spray amount adjuster 18 is provided downstream of the solenoid 17 within the handpiece 10. This spray amount adjuster 18 acts on the pipe 20a to adjust the amount of gas supplied when the solenoid 17 is turned on. This adjusts the amount of gas and spray liquid ejected in one spray. The spray amount adjuster 18 may also be configured to adjust the spray amount by changing the opening size of the spray nozzle 12a. In this case, the spray range and spray intensity can also be adjusted.
[0040] The main body 30 is provided with a control unit 31, a power supply unit 32, and a gas supply unit 33. The control unit 31 controls the supply of gas by the gas supply unit 33. Specifically, when the operation start button 30a (see FIG. 1) is pressed to turn the power on, the gas supply unit 33 is driven to start supplying gas and also starts supplying power from the electric wire 20b to the unit inside the handpiece 10. When the operation start button 30a (see FIG. 1) is pressed again to turn the power off, the gas supply and power supply are stopped.
[0041] The power supply unit 32 supplies power to each component. While the power supply unit 33 is turned on, it drives an internal pump to continuously supply gas to the nebulizing gas chamber 14b of the handpiece 10 through the pipe 20a. In this embodiment, ambient air is continuously supplied as the gas, but it is also possible to supply an appropriate gas, such as oxygen, hydrogen, or an appropriate mixed gas, by attaching a gas cylinder (not shown).
[0042] 3 is a flowchart showing the operation of the multiple component skin penetration system 1. When the operation start button 30a (see FIG. 1) is pressed to turn on the power, the control unit 19d receives input of the first spray liquid from the liquid input hole 15b (step S1). The received first spray liquid is supplied from the liquid storage unit 15a to the spray liquid chamber 13.
[0043] The control unit 19d accepts the switching of the spray setting by the switching button 19b (step S2).
[0044] When the activation button 19c is pressed, the control unit 19d performs a first spray liquid spraying step of spraying in accordance with the spray setting (step S3). That is, if the spray setting is manual spraying, the control unit 19d performs spraying at the timing when the manual spray button 16 is pressed, if the spray setting is first intermittent spraying, the control unit 19d automatically and continuously performs intermittent spraying at the first time interval, if the spray setting is second intermittent spraying, the control unit 19d automatically and continuously performs intermittent spraying at the second time interval, and if the spray setting is third intermittent spraying, the control unit 19d automatically and continuously performs intermittent spraying at the third time interval.
[0045] When the spray setting is changed (step S4: YES), the control unit 19d accepts the change in the spray setting (step S2), and executes spraying with the changed spray setting (step S3).
[0046] When spraying this spray liquid (first spray liquid, second spray liquid, etc.), the tip of spray cover 11 is brought into contact with the skin to perform spraying. When the circular ring-shaped tip of spray cover 11 is brought into full contact with the skin to perform spraying, excess gas is released from gas release space 11a at the tip along with the spray liquid. As a result, much of the spray liquid, which has been turned into a fine mist, penetrates into the skin due to the force of release, and excess gas is released from gas release space 11a, preventing the skin from being overly pressed by the gas pressure.
[0047] In other words, if the gas release space 11a were not present and spraying were performed with the entire tip of the spray cover 11 in contact with the skin, the released gas and spray liquid would press against the skin, and the large amount of gas inside the spray cover 11 would press against the skin, leaving it depressed. This would apply unnecessary pressure to the skin, which could actually stagnate blood flow and hinder penetration of the spray liquid. In contrast, by releasing excess gas from the gas release space 11a, pressure is not applied to the skin, and the skin returns to its original state immediately after the gas and spray liquid are sprayed onto the skin. This allows the penetrated spray liquid to dissolve into the bloodstream and lymphatic system without stress, and a mild, moderate stimulation is applied to the skin, promoting bloodstream and lymphatic flow and allowing the penetrated spray liquid to penetrate deeper.
[0048] If the process is not terminated even after the spraying of the first spray liquid is completed (step S5: NO), a second spray liquid spraying step is performed to receive the second spray liquid (step S6). The received second spray liquid is supplied from the liquid storage section 15a to the spray liquid chamber 13. After this, the process from step S2 onwards is repeated, and the second spray liquid is sprayed.
[0049] With the above configuration and operation, the active ingredient can be sprayed in a fine mist. Since only a small amount of spray liquid is used in one spray (instantaneous spray for 0.1 seconds, etc.), even if the active ingredient is valuable and expensive, it can be sprayed in small amounts many times, so it can be sprayed on the entire face by spraying one area at a time, and it is also possible to spray on the neck and décolleté. It can also be sprayed on the head, and it can be sprayed on the entire head or multiple times on areas of the head where concentrated penetration is desired.
[0050] When a growth factor agent such as human stem cell culture medium is used as the penetrating agent (first penetrating agent or second penetrating agent), spraying it onto the skin of the face, neck, décolleté, etc. can enhance firmness, provide a lifting effect, open up the eyes, reduce wrinkles, slim the contours, and make the skin appear fresh and rejuvenated. In particular, when a human umbilical cord stem cell culture supernatant or a growth factor agent derived from a human umbilical cord stem cell culture supernatant is used, these effects are more pronounced.
[0051] Furthermore, when human stem cell culture medium is sprayed onto injured or burned areas of the skin, the wound or burn heals faster than usual, and leaves a clean wound without any scars after healing. These effects are particularly pronounced when human umbilical cord stem cell culture supernatant or a growth factor agent derived from human umbilical cord stem cell culture supernatant is used.
[0052] Furthermore, when a growth factor agent such as human stem cell culture medium is used as the penetrating agent (first penetrating agent or second penetrating agent), spraying it onto the scalp can provide hair growth and regrowth effects, such as hair growth, increased hair volume, hairline advancement, increased hair firmness, stronger hair strength, thicker hair, and turning gray hair back into black hair, as well as a youthful appearance. In particular, when a human umbilical cord stem cell culture supernatant or a growth factor agent derived from a human umbilical cord stem cell culture supernatant is used, these effects are more pronounced.
[0053] Furthermore, when a sirtuin gene activator such as NMN is used as the penetrating agent (first penetrating agent or second penetrating agent), spraying it onto the skin of the face, neck, décolleté, etc. can increase firmness, provide a lifting effect, open up the eyes, reduce wrinkles, slim the contours, and make the skin look fresh and rejuvenated.
[0054] Furthermore, when a sirtuin gene activator such as NMN is sprayed onto an injured or burned area of the skin, the wound or burn will heal faster than usual, and will leave a clean wound with no scars even after healing.
[0055] Furthermore, when a sirtuin gene activator such as NMN is used as the penetrating agent (first penetrating agent or second penetrating agent), spraying it onto the head can produce effects such as hair growth, increased hair volume, advancement of the hairline, increased hair firmness, stronger hair strength, thicker hair, and turning gray hair back to black hair.
[0056] Furthermore, when a growth factor agent such as human stem cell culture medium is used as one of the first and second penetrating agents, and a sirtuin gene activator such as NMN is used as the other, the above-mentioned effects can be obtained more significantly than when either agent is used alone, and even greater effects can be obtained by combining the two rather than using twice the amount of one agent during the treatment.
[0057] In particular, when the first spray solution using the first penetrating agent and the second spray solution using the second penetrating agent are sprayed separately and in sequence, the active ingredients can be penetrated into the skin one by one, and each ingredient penetrates sequentially through the bloodstream and lymphatic system, resulting in a synergistic effect. In particular, when this treatment is performed on the face, significant changes in face contours and eye size can be achieved, and some people may even suspect cosmetic surgery.
[0058] Furthermore, since the first spray liquid and the second spray liquid are sprayed by the spray device 5, a fixed amount of spray can be sprayed at a constant intensity each time. Therefore, the same effect can be obtained regardless of the skill of the practitioner, as long as the practitioner sprays the same area in the same order.
[0059] The following describes an example of the multiple-component skin penetration method using the above-described multiple-component skin penetration system 1. The first penetrating agent 2, second penetrating agent 3, and adjusting agent 4 used in this example are as follows.
[0060] First penetrating agent 2: NMN <Details> β-nicotinamide mononucleotide Particle size: 16 mesh or less Bulk specific gravity: 0.6 g / ml or more Purity: 99% or more (composition ratio: 100%) Processing: microgranulation processing Shape: white crystalline powder Amount: 500 mg Amount used in this example: 1 / 3 (= 166.67 mg)
[0061] Second permeant 3: Human umbilical cord stem cell culture supernatant <Details> Human mesenchymal stem cells derived from umbilical cord matrix (hMSC-UC) Expanded cell culture Passage 2 (third culture) Storage method: cryopreservation Volume: 1 ml Amount of material in this example: total volume
[0062] Adjusting agent 4: Purified water - 400,000 times more pure than tap water - Amount: 400 ml - Amount used in this example: (1) 3 ml used to mix with 166.67 g of NMN powder as the first penetrating agent 2... After mixing, about 3.17 ml of second spray liquid (2) 2 ml used to mix with 1 ml of human umbilical cord stem cell culture supernatant as the second penetrating agent 3... After mixing, 3 ml of first spray liquid
[0063] 0. Preparation First, the subjects' skin was prepped and cleansed, and then a lymphatic massage was performed on the neck, décolleté, and face as a preliminary step.
[0064] 1. Spraying of the first spray liquid (NMN) The first spray liquid was sprayed onto the décolleté, neck, and face using the spray device 5. Spraying was performed on each area in turn with the tip of the spray cover 11 in contact with the skin. The total number of sprays was approximately 1,000.
[0065] 2. Spraying of the second spray liquid (human umbilical cord stem cell culture supernatant) The second spray liquid was sprayed onto the décolleté, neck, and face using the spray device 5. Spraying was performed on each area in turn with the tip of the spray cover 11 in contact with the skin. The total number of sprays was approximately 1,000.
[0066] The above treatment was performed on two patients and the following results were obtained:
[0067]
[0068] In particular, changes in facial contours are easy to see, so an example of the changes in subject 1 will be described in Figure 4. Figure 4(A) is a photograph of the stage after the completion of the above-mentioned "0. Preparation," i.e., the stage after the lymphatic drainage massage. Figure 4(B) is a photograph of the state after spraying NMN and then human umbilical cord stem cell culture supernatant. Figure 4(C) is a photograph showing the facial contours of Figure 4(A) before spraying superimposed on the facial contours of Figure 4(B) after spraying. Thus, from the state after lymphatic drainage (Before), it is clear that significant effects can be obtained by simply performing this treatment (After) once, in which NMN and human stem cell culture medium are applied sequentially.
[0069] Furthermore, the subject's friend suggested the subject had gone for surgery the following day, and a person she met a week after the procedure also suggested the subject had gone for surgery. This shows that a single application of this treatment, which involves sequentially applying NMN and human stem cell culture medium, can produce significant results, and that these effects are sustained.
[0070] In Example 2, only the order of spraying was changed from Example 1. That is, the first penetrating agent 2 and the second penetrating agent 3 were swapped as follows: (1) 2 ml was used to mix with 1 ml of human umbilical cord stem cell culture supernatant as the first penetrating agent 2...after mixing, 3 ml of the first spray liquid; (2) 3 ml was used to mix with 166.67 g of NMN powder as the second penetrating agent 3...after mixing, approximately 3.17 ml of the second spray liquid. Other than this, the procedure was the same as in Example 1, with massage, spraying of the first spray liquid, and spraying of the second spray liquid being performed in that order. The above treatment was performed on two people, and the following effects were obtained.
[0071]
[0072] Furthermore, because subject 3 had atopic dermatitis, the first spray liquid (human umbilical cord stem cell culture supernatant) and the second spray liquid (NMN) were also sprayed on the atopic area. As a result, improvement in the atopic symptoms was observed, and improvements in the moisture, firmness, and luster of the skin in the atopic area were also observed.
[0073] Next, the same formulation and treatment as in Example 1 were used to spray onto the scalp, which is a part of the skin, and the hair growth effect was confirmed. At this time, the preparatory massage included a massage of the back of the neck and the scalp. The other spray liquids and treatment order were the same as in Example 1. A series of treatments was counted as one session, and a total of four treatments were performed to measure the effect. The interval between each session ranged from one week to one month. As a result of performing the treatment on two people, both of them showed a clear hair growth effect (scalp skin beauty effect).
[0074] Next, the same formulation and treatment as in Example 2 were used to spray onto the scalp, and the hair growth and regrowth effects were confirmed. At this time, the preparatory massage included a massage of the back of the neck and scalp. The other spray liquids and treatment order were the same as in Example 2. A series of treatments was counted as one session, and a total of four treatments were performed to measure the effects. The intervals between each session ranged from one week to one month. As a result of performing the treatment on two people, both of them showed a clear hair growth effect (scalp skin beauty effect).
[0075] Examples 5-10: Treatments were also performed in various ways to measure their effects. The pre-massage, spraying of the first penetrating agent 2, and spraying of the second penetrating agent 3 were performed using the same formulations and methods as in Examples 3 and 4 described above. Additionally, an example was added in which holes were made in the skin surface with a roller (see penetration enhancer 105 described below) before spraying with the spray device 5, and an example was added in which a proteoglycan-containing shampoo was used as a regular shampoo in addition to the treatment. In each example, a series of treatments was considered one session, and a total of four treatments were performed, with intervals between each session ranging from one week to one month, and measurements were taken. The results obtained were as shown in Table 3 below.
[0076] In all cases, hair growth and thickening effects were confirmed. In particular, Examples 5 to 8, in which both human umbilical cord stem cell culture supernatant and NMN were sprayed in sequence, were highly effective, with Examples 7 and 8, in which a roller was also added, being particularly effective. Furthermore, Examples 9 and 10, in which a proteoglycan-containing shampoo was used as a regular shampoo, human umbilical cord stem cell culture supernatant was sprayed as the first penetrating agent, and no second penetrating agent was sprayed, also achieved an increase in downy hair.
[0077] The present invention is not limited to the above-described embodiment, but can be embodied in various other forms.
[0078] For example, before spraying the first spray liquid, holes may be made in the skin surface using a roller (see penetration promoter 105 described below) with a cylindrical outer periphery on which multiple fine needles of a predetermined length are arranged, followed by spraying the first spray liquid and then the second spray liquid. In this case, the penetration of the spray liquid into the skin can be improved, resulting in better results. In this case, the needles are preferably made of titanium. The predetermined length can be 0.1 mm to 1 mm, preferably 0.1 to 0.5 mm, and preferably 0.25 mm.
[0079] Furthermore, performing a lymphatic massage as a preliminary preparation can improve blood and lymph flow, resulting in better results (the effect of the massage plus the effect of the spray), but spraying the first spray liquid and the second spray liquid may be performed without such preliminary preparation. Even in this case, a clear change can be seen before and after the spraying.
[0080] Furthermore, although the first spray liquid and the second spray liquid are sprayed 1,000 times and 1,000 times, respectively, the number of sprays is not limited to this, and can be any appropriate number, such as 500 times each, 300 times each, or conversely, increased to 1,500 times each. The number of sprays depends on the area of skin to be sprayed, but can be 10 or more times for one type of spray liquid, preferably 50 or more times, more preferably 100 or more times, even more preferably 300 or more times, and preferably 500 or more times. Furthermore, the upper limit of the number of sprays for one type of spray liquid can be 3,000 or less times, preferably 2,000 or less times, more preferably 1,500 or less times, and preferably 1,000 or less times.
[0081] The area covered by the inside of the tip of the circular spray cover 11 can cover 40% or more of the target skin area, preferably 50% or more, more preferably 60% or more, even more preferably 70% or more, even more preferably 80% or more, and preferably 90% or more. Although spraying the same area only once is sufficient, spraying two or more times can allow more active ingredients to penetrate. It is particularly preferable to spray two or more times on areas where intensive care is desired, such as fine lines around the eyes, nasolabial folds, and forehead wrinkles. This increases the penetration of the active ingredients into the areas where intensive care is desired, resulting in more favorable effects.
[0082] Furthermore, for the spray liquid using the first penetrant 2 and the second penetrant 3, either the two are mixed with the adjuster 4 and then poured into the liquid storage section 15a (simultaneous addition of all components), or the first penetrant 2 and adjuster 4 are mixed and poured into the liquid storage section 15a as the first spray liquid and sprayed, and then within a predetermined time, the second penetrant 3 and adjuster 4 are mixed and poured into the liquid storage section 15a as the second spray liquid (sequential addition of components).
[0083] In the sequential component addition, the predetermined time from the completion of spraying of the first spray liquid to the start of spraying of the second spray liquid can be within one week, preferably within 48 hours, more preferably within 24 hours, more preferably within 12 hours, even more preferably within 6 hours, even more preferably within 3 hours, even more preferably within 1 hour, even more preferably within 30 minutes, and preferably within 10 minutes. By adding the components sequentially in this manner, the components of the first penetrant 2 and the second penetrant 3 are sufficiently penetrated, and the effects of both components can be obtained simultaneously, resulting in a synergistic effect. In particular, by shortening the predetermined time between the spraying of the first spray liquid (first penetrant 2 + adjuster 4) and the spraying of the second spray liquid (second penetrant 3 + adjuster 4), the synergistic effect of the two components can be enhanced. Furthermore, adding the components sequentially allows each component to penetrate more thoroughly than mixing them all together and adding them simultaneously.
[0084] When all ingredients are added at once, the overall spray time can be shortened compared to sequential ingredient addition. For example, with sequential ingredient addition, it is necessary to spray the first spray liquid 1,000 times on each part of the face (and the neck and décolleté may also be added), and then spray the second spray liquid again 1,000 times on each part of the face (and the neck and décolleté may also be added), which takes a total of 2,000 sprays, but with simultaneous ingredient addition, it is possible to spray each part of the face (and the neck and décolleté may also be added) only 1,000 times in total, thereby cutting the time in half.
[0085] When all ingredients are added simultaneously, a third penetrant (an agent consisting of a third active ingredient) may be further mixed with the first penetrant 2 and the second penetrant 3, and then all ingredients may be added simultaneously and sprayed. The third penetrant may be any suitable substance, for example, a cannabinoid (third active ingredient) such as CBD (cannabidiol), CBN (cannabinol), or CBG (cannabigenol). If this cannabinoid is used, it is preferable that THC (tetrahydrocannabinol) has been removed.
[0086] The third penetrating agent may also be a proteoglycan. In this case, application to the scalp in particular can improve effects on hair (e.g., hair growth, increased hair volume, hairline advancement, increased hair firmness, stronger hair strength, thicker hair, and gray hair turning black).
[0087] In the case of sequential component addition, after spraying the first spray liquid (first penetrant 2 + regulator 4) and the second spray liquid (second penetrant 3 + regulator 4), the third spray liquid (third penetrant + regulator 4) can be sprayed within a predetermined time. Furthermore, it is also possible to spray the third spray liquid (fourth penetrant + regulator 4) within a predetermined time.
[0088] In addition, while the first active ingredient is NMN (a sirtuin gene activator) and the second active ingredient is human stem cell culture medium (a growth factor agent), which are applied (sprayed) in that order, the order may be reversed, with the first active ingredient being human stem cell culture medium (a growth factor agent) and the second active ingredient being NMN (a sirtuin gene activator), which are applied (sprayed) in that order. In this case, too, the active ingredients can be penetrated sequentially to exert their respective synergistic effects.
[0089] Alternatively, one of the first and second active ingredients may be a human stem cell culture medium, and the other may be a proteoglycan, and applied (sprayed) in that order. In this case, the effects of each ingredient can be fully exerted, resulting in a synergistic effect. In this case, applying the ingredients to the scalp can achieve a high level of hair growth and hair care effects.
[0090] Furthermore, the method of application is preferably spraying as described above, but is not limited to this, and other methods can also be used, such as applying it to the skin by hand, or soaking it in a pack or the like and placing it on the skin. In this case, the penetration of the active ingredient can be increased by making multiple small holes in the skin using a fine needle. In this case, a certain degree of effect can also be obtained, and by applying the active ingredients in order, a synergistic effect of each active ingredient can be exerted.
[0091] This first embodiment can also be provided as a composition kit (beauty kit, skin topical kit). This composition kit can be a set of the first penetrant 2 and the second penetrant 3, a set of the first penetrant 2, the second penetrant 3, and the adjuster 4, a set of the first penetrant and the adjuster 4, a set of the second penetrant and the adjuster 4, or a set of the first penetrant 2, the second penetrant 3, the adjuster 4, and the spray device 5 (penetration enhancer). In this case, a user can purchase these and use them by adjusting the first penetrant 2 with the adjuster 4 and applying it to the skin using the spray device 5, and then adjusting the second penetrant 3 with the adjuster 4 and applying it to the skin using the spray device 5.
[0092] Similarly, the above-described Examples 4 to 10 can be provided as a composition kit for hair growth and restoration, i.e., a hair growth and restoration kit. This hair growth and restoration kit can be a set of the first penetrant 2 and the second penetrant 3, a set of the first penetrant 2, the second penetrant 3, and the regulator 4, a set of the first penetrant and the regulator 4, a set of the second penetrant and the regulator 4, or a set of the first penetrant 2, the second penetrant 3, the regulator 4, and the spray device 5 (penetration enhancer). In this case, a user can purchase these and use them by adjusting the first penetrant 2 with the regulator 4 and applying it to the skin using the spray device 5, and then adjusting the second penetrant 3 with the regulator 4 and applying it to the skin using the spray device 5.
[0093] 5 is a structural diagram showing another example of a composition kit. The composition kit 101 includes a first skin topical product 120 containing a first penetrating agent 102 in a first container 125, a second skin topical product 130 containing a second penetrating agent 103 in a second container 135, a liquid container 140 containing a liquid adjusting agent 104 in a liquid container 145, and a penetration enhancer 105 that enhances the penetration of the cosmetic ingredient into the skin.
[0094] The first penetrating agent 102 may be the same as the first penetrating agent 2 described above, or may contain other components added thereto, and the second penetrating agent 103 may be the same as the second penetrating agent 3 described above, or may contain other components added thereto. The other components to be contained may be any appropriate component, such as niacinamide, collagen, hyaluronic acid, elastin, retinol (vitamin A), vitamin C, ceramide, amino acid, neil-1, fullerene, honey extract, or a combination thereof.
[0095] Niacinamide, also known as nicotinamide, D-melanosine, or wrinkle niacin, is a type of vitamin B. Niacinamide can be present in an amount of 0.01% or more, preferably 0.05% or more, more preferably 0.1% or more, more preferably 0.5% or more, and even more preferably 1% or more. Niacinamide can be present in an amount of 70% or less, preferably 50% or less, more preferably 30% or less, more preferably 20% or less, even more preferably 10% or less, more preferably 5% or less, and preferably 3.5% or less.
[0096] The first penetrant 102 and the second penetrant 103 are preferably powders that dissolve in liquids such as water, purified water, or saline solution, which allows the first penetrant 102 and the second penetrant 103 to be stored in a stable state and can be used as a liquid when needed.
[0097] The first and second skin topical products 120 and 130 are preferably prepared by sterilizing the first and second containers 125 and 135, respectively, and then storing the powdered first and second penetrating agents 102 and 103 in the containers in a clean room, and then sealing the containers with the lids 123 and 133. This allows the first and second skin topical products 120 and 130 to be provided without containing any preservative ingredients.
[0098] The penetration enhancer 105 is a needle roller-type beauty tool in which a cylindrical roller 152 is rotatably supported on one end of a rod-shaped handle 153, and multiple needles 151 protrude from the surface of the roller 152. More than half of the base of the needles 151 is embedded in the roller 152, with the tips protruding from the surface of the roller 152. The protruding length of the needles 151 can be at least 0.1 mm, preferably at least 0.2 mm, and at most 1.0 mm, preferably at most 0.7 mm, more preferably at most 0.5 mm, even more preferably at most 0.4 mm, and even more preferably at most 0.3 mm. For home use, a length of 0.25 mm is preferred. The needles 151 can be made of a metal material, preferably titanium. Using titanium for the needles 151 enhances antibacterial properties. Furthermore, multiple needles 151 are arranged at equal intervals around the circumferential periphery of the roller 152. The number of needles 151 can be set to a minimum of 200, preferably 300, and more preferably 400 or more. The number of needles 151 can be set to a maximum of 1,000, preferably 900, and more preferably 800. In this embodiment, six rows of 60 needles 151 are arranged in a circle at equal intervals around the circumference, for a total of 540 needles. In this embodiment, the penetration enhancer 105 is a needle roller-type cosmetic device, but this is not limiting, and the above-described spray device 5 (see FIG. 1 ) may also be used as the penetration enhancer 105.
[0099] The first container 125 and the second container 135 are each composed of a main body 124, 134 made of a glass bottle and a lid 123, 133 that seals the upper opening of the main body 124, 134, and the first penetrant 102 or the second penetrant 103 is contained in the internal space of the main body 124, 134, respectively.
[0100] The size of the main body 124, 134 is such that it has a capacity sufficient to dissolve the powdered first penetrant 102 or second penetrant 103 in the liquid 104. The lower limit of this capacity can be 1 ml or more, preferably 2 ml or more, more preferably 3 ml or more, and even more preferably 4 ml or more. The upper limit of this capacity is preferably 20 ml or less, more preferably 15 ml or less, even more preferably 10 ml or less, and even more preferably 7 ml or less. In this embodiment, the capacity is 4 ml, and 3 ml of the liquid 104 is poured in to dissolve the first penetrant 102 or second penetrant 103.
[0101] Next, Examples 11 to 18 in which the composition kit 101 was used by a user at home will be described with reference to Table 4. In Examples 11 to 18, the first penetrating agent 102 and the second penetrating agent 103 were each dissolved in 3 ml of purified water (liquid 104) before use.
[0102] In Examples 11 to 14, a needle roller-type beauty device was used as the penetration enhancer 105. After perforating the facial skin with the needle roller-type beauty device, the first penetrating agent and the second penetrating agent were applied. This series of steps was repeated five times, and the effects were measured with intervals ranging from one week to one month between each of the five measurements.
[0103] In addition, in Examples 15 to 18, a spray device 5 was used as the penetration enhancer 105, and after the first penetrant was applied (sprayed) using this spray device 5, the second penetrant was applied (sprayed) using the spray device 5. In Examples 15 to 18, the effects of a single implementation were measured.
[0104] Furthermore, since Examples 11 to 18 were for home use, no prior massage was performed, etc. Furthermore, the first and second osmotic agents in Examples 11 to 18 were prepared by dissolving 1 mg of human umbilical cord stem cell culture supernatant (freeze-dried powder) in 3 ml of purified water (liquid 104), 150 mg of NMN (powder) in 3 ml of purified water (liquid 104), or 1 mg of proteoglycan (freeze-dried powder) in 3 ml of purified water (liquid 104), respectively.
[0105]
[0106] As shown in Table 4, the effect was observed in all cases. Figure 6 is an explanatory diagram using photographs to explain the before and after of Example 16. In this example, the subject was a 76-year-old woman.
[0107] Figure 6(A) is a photograph of the state before the start, Figure 6(B) is a photograph of the state after NMN (powder) dissolved in purified water has been sprayed using sprayer 5, and Figure 6(C) is a photograph of the state after human umbilical cord stem cell culture supernatant (freeze-dried powder) dissolved in purified water has been sprayed using sprayer 5. In this example, spraying of NMN (powder) was carried out over a period of approximately 15 minutes, followed by an interval of approximately 10 minutes for photography and preparation for the next step, followed by spraying of human umbilical cord stem cell culture supernatant (freeze-dried powder) over a period of approximately 15 minutes, for a total time of approximately 40 minutes from start to finish.
[0108] Significant effects were observed in lifting, eye opening, wrinkle reduction, cheek position, skin firmness, skin luster, and whitening. Figure 6 compares the skin from above the mouth to the nose, which is the most obvious example. It is clearly evident that wrinkles are fewer and shallower from before treatment in Figure 6(A) to after NMN treatment in Figure 6(B). Furthermore, compared to Figure 6(B), which shows the improved condition after spraying NMN (powder) with the spray device 5, Figure 6(C), which shows the condition after spraying human umbilical cord stem cell culture supernatant (freeze-dried powder) with the spray device 5, shows further improvement. Thus, sequential application of the first penetrant 102 and the second penetrant 103 results in two stages of improvement, resulting in a highly youthful appearance.
[0109] Next, Examples 19 to 57 will be described with reference to Tables 5 and 6. These Examples were carried out using various combinations of sirtuin gene activators and growth factor agents to measure the effects.
[0110]
[0111]
[0112] The sirtuin gene activator uses one or more of NAD+, NMN, 5-deazaflavin, and niacinamide, all of which are used in powder form by dissolving them in purified water as a solvent.
[0113] The growth factor agents used are one or more of EGE, EGF, PDGF, exosomes, and human stem cell culture supernatant, all of which are in powder form and dissolved in purified water as a solvent. FGF1 is used as the FGF. The human stem cell culture supernatant is a human umbilical cord stem cell culture supernatant (freeze-dried powder) derived from the umbilical cord. Furthermore, the growth factor agents used, EGE, EGF, PDGF, exosomes, and human stem cell culture supernatant, are all freeze-dried powders and dissolved in purified water as a solvent.
[0114] In Tables 5 and 6, the numerical values in each example indicate volume, and the symbol "-" indicates that the component was not included. Specifically, the numerical values for growth factor agents (0.2, 0.5, 1, 2, 5) all indicate volume in ml. The numerical values for sirtuin gene activators (50, 100, 150, 200, 250, 300, 500) all indicate volume in mg. The numerical values for solvent (purified water) (2, 3) all indicate volume in ml.
[0115] In Examples 19 to 57, no prior massage or the like was performed, and the procedure was as follows: the first penetrant dissolved in a solvent was sprayed onto the entire facial skin and the skin under the chin using the spray device 5 also used in Example 1, and after an interval of about 3 minutes, the second penetrant dissolved in a solvent was sprayed onto the entire facial skin and the skin under the chin using the spray device 5.
[0116] As described in the effects of each example, improvements were observed in all of the following effects: lifting effect, eye opening, wrinkle reduction, cheek position, skin firmness, skin luster, and whitening. The example using NAD+ as a sirtuin gene activator showed the highest effect.
[0117] Furthermore, in Examples 49 to 52, in which a growth factor agent was not used but NAD+ was used as a sirtuin gene activator, dissolved in purified water, and sprayed onto the skin using spray device 5, the effect was lower than in the examples using a growth factor agent, but a certain degree of effect was still obtained. In addition, in Examples 49 to 52, NAD+ and a growth factor agent may be mixed and dissolved in purified water and sprayed using spray device 5. In this case, the growth factor agent can be a growth factor, exosomes, stem cell culture supernatant, or a combination of these, with exosomes being preferred. Even in this case, although the effect is lower than when the growth factor agent and the sirtuin gene activator are applied separately and sequentially, a certain degree of effect can still be obtained. Specifically, when 1 mg of exosomes and 200 mg of NAD+ were mixed and dissolved in 3 ml of purified water and sprayed using spray device 5, significant improvements were observed in eye opening, wrinkle reduction, skin firmness, skin luster, and whitening, as well as improvements in the lifting effect and cheek position.
[0118] The present invention can be used in industries that provide beauty treatments and hair growth treatments.
