Abstract
Description
Composition for improving sleep comprising natural extract complex and use thereof
[0001] The present invention relates to a composition for preventing or treating sleep disorders, comprising a mixture of Perilla frutescens and peppermint (Mentha piperita) as an active ingredient.
[0002] This invention was supported by the Gyeonggi Regional Cooperation Research Center, Gyeonggi Regional Cooperation Research Center (GRRC) Project: Research Center for Technology to Address Unsolved Diseases of the Aging Society (Project Identification Number: GRRCAjou2023-B01) (Research Period: July 1, 2023 - June 30, 2029, Implementing Organization: Ajou University Industry-Academic Cooperation Foundation).
[0003] Sleep disorders are a common problem faced by modern people. Sleep disorders can be defined as a lack of healthy sleep, difficulty maintaining daytime alertness despite adequate sleep, or a disruption in sleep rhythms that causes difficulty falling asleep or staying awake. Chronic sleep deprivation can increase the risk of developing high blood pressure, diabetes, dementia, and obesity. Furthermore, it weakens the immune system, making people more susceptible to infectious diseases like the common cold. It can also increase the risk of memory impairment, stroke, accelerated skin aging, increased risk of heart disease, and bone loss.
[0004] According to statistics from the National Health Insurance Service, 1,098,819 patients received hospital treatment for sleep disorders in 2022. This represents a whopping 30% increase over the four-year period from 855,025 in 2018. With the number of people suffering from sleep disorders exceeding one million, the number of patients is increasing every year.
[0005] To treat these sleep disorders, chemical hypnotics are used, which reversibly suppress the central nervous system to induce and maintain sleep. Conventional hypnotics have central depressant effects similar to anesthetics, producing sedation in small doses and sleep in moderate doses. However, larger doses can cause coma, paralysis, and respiratory depression. Barbiturates have low safety and can lead to side effects such as tolerance and dependence, as well as serious problems such as memory impairment, delirium, hangovers, dizziness, and headaches.
[0006] Accordingly, the need for natural sleep enhancers that are highly effective and free of drug side effects is increasing, and the desire to improve the quality of sleep not only among insomniacs but also among the general public is increasing, leading to an increase in demand for sleep-enhancing compositions.
[0007] The inventors of the present invention have made great efforts to develop a sleeping drug that is safe in vivo without causing the problems of the above-mentioned side effects, and as a result, they have confirmed that a mixture of natural plants, such as chaga and peppermint, not only increases the duration of sleep but also has the effect of significantly shortening the time it takes to fall asleep, thereby completing the present invention.
[0008] Accordingly, the purpose of the present invention is to provide a pharmaceutical composition, health functional food composition, food additive, cosmetic composition, quasi-drug composition or feed composition for preventing or treating sleep disorders, which comprises a mixture of a Perilla frutescens extract and a peppermint (Mentha piperita) extract as an active ingredient.
[0009] To achieve the above purpose, the present invention provides a pharmaceutical composition for preventing or treating sleep disorders, comprising a mixture of a Perilla frutescens extract and a peppermint (Mentha piperita) extract as an active ingredient.

[0061] To facilitate understanding of the present invention, preferred examples are presented below. However, the following examples are provided solely to facilitate understanding of the present invention, and the scope of the present invention is not limited by the examples.
[0062]
[0063] Example 1. Preparation of tea tree and peppermint extracts
[0064] Perilla frutescens var. acuta kudo) and peppermint (Mentha piperita) were extracted with hot water at 80°C and then sterilized at 121°C for 20 minutes to prepare hot water extracts of Perilla frutescens and peppermint. The obtained extracts were filtered and concentrated under reduced pressure, and then powdered by freeze-drying for use.
[0065]
[0066] Example 2. Establishment of a pentobarbital-induced sleep model
[0067] To confirm the sleep disorder-improving effect of the natural extract complex according to the present invention, ICR mice were purchased from Orient Bio Co., Ltd. The mice were then divided into five groups: a group administered with a PF (Perilla frutescens) hot water extract, a group administered with a MP (Peppermint) hot water extract, a group administered with a PF+MP (Perilla frutescens + Peppermint extract complex), a control group, and a group administered with DZP (diazepam). Each substance was administered orally once. The control group mice were administered saline.
[0068] Thirty minutes after administering the test substance, mice were intraperitoneally administered 45 mg / kg of pentobarbital, a tranquilizer purchased from Hanlim Pharmaceutical Co., Ltd., to induce sleep. Latency time and total sleep duration were then measured. Shorter latency times and longer durations indicate greater sleep-enhancing efficacy.
[0069]
[0070] Example 3. Confirmation of the efficacy of reducing latency and increasing total sleep time (duration) by concentration of natural extract complex.
[0071] In order to confirm the sleep disorder improvement effect according to the concentration of the natural extract complex according to the present invention, the sleep onset time (latency) and total sleep time (duration) were measured by varying the concentration of the PF+MP (peppermint extract complex) administration group in the pentobarbital-induced sleep model constructed as in Example 2, and the results are shown in Fig. 2.
[0072] As shown in Fig. 2, compared to the control group administered saline, the time to fall asleep was significantly reduced in all groups administered the tea tree + peppermint extract complex, and the total sleep time was significantly increased.
[0073]
[0074] Example 4. Synergistic effect of natural extract complex on reducing latency and increasing total sleep time compared to single extract.
[0075] In order to confirm the synergistic effect of the natural extract complex compared to the single extract, the concentrations of each administration group were set to 5 mg / kg, 10 mg / kg, and 20 mg / kg, and then the sleep onset time (latency) and total sleep time (duration) of the administration group of the PF+MP complex were evaluated compared to the PF and MP administration groups, respectively. The results are shown in Figures 3 to 5.
[0076] As shown in Figures 3 to 5, when the tea tree + peppermint extract complex was administered, the time to fall asleep was significantly reduced compared to when a single extract was administered, and the total sleep time was significantly increased, confirming the synergistic effect of the tea tree + peppermint extract complex on improving sleep.
[0077]
[0078] Example 5. Synergistic effects of natural extract compounds on reducing latency and increasing total sleep time (duration) by compound ratio.
[0079] In order to confirm the synergistic effect according to the compound ratio of the natural extract complex, the concentrations of each administration group were set to 5 mg / kg and 10 mg / kg, and then compared with the administration groups of the chazzi hot water extract (PF) and peppermint hot water extract (MP), respectively, to evaluate the sleep onset time (latency) and total sleep time (duration) of the chazzi + peppermint extract complex (PF + MP) administration group with different compound ratios, and the results are shown in Figures 6 and 7.
[0080] As shown in Figures 6 and 7, even when various ratios of the tea tree + peppermint extract complex were administered, the time to fall asleep was significantly reduced compared to when a single extract was administered, and the total sleep time was significantly increased, confirming that the tea tree + peppermint extract complex showed a synergistic effect even when the ratios of the complex were different.
[0081]
[0082] In summary, administration of the present invention's tea tree and peppermint extract complex resulted in a decrease in falling asleep time and an increase in sleep time compared to the single extract administration group, specifically confirming that the tea tree and peppermint extract mixture exhibits a sleep-improving effect. These results confirmed that the present invention's tea tree and peppermint extract complex has a sleep-improving effect.
[0083] A mixture of a Perilla frutescens extract and a peppermint (Mentha piperita) extract according to one embodiment of the present invention showed the efficacy of reducing the time it takes to fall asleep and increasing the sleep time in an animal model, and therefore, as a natural product, it can be usefully used as a composition for improving sleep with few side effects and is effective, and thus has industrial applicability.
