Abstract
Description
Technical Field
[0001] The present invention relates to the technical field of chemical medicine, and in particular to a class of 3-aminoindazole compounds and applications thereof. Background Art
[0002] Fibroblast growth factor receptors (FGFRs) belong to the receptor tyrosine kinase family (RTKs), which contains four receptor subtypes: FGFR1, FGFR2, FGFR3 and FGFR4. The structure of FGFRs mainly consists of three parts: an extracellular region composed of three immunoglobulin-like domains, a single transmembrane region and an intracellular region containing a kinase domain. The activation mechanism of FGFR4 is the same as that of other receptor tyrosine kinases. The ligand binds tightly to the complex formed by the receptor and co-receptor, induces receptor dimerization, phosphorylates tyrosine residues in the intracellular region of the receptor, and activates receptor kinase activity. Activated FGFR4 can phosphorylate downstream target proteins to cause signal cascade reactions. Among them, the FGFR4 subtype is mainly distributed in the liver, lung, lymph and breast tissues. FGFR4 and its specific ligand fibroblast factor FGF19 play important physiological functions in the liver, lung and breast tissues. Studies have shown that FGF19 / FGFR4 is upregulated in 30-50% of liver cancers, and this upregulation is closely related to the poor prognosis of liver cancer, making it an effective new target for the treatment of liver cancer and other related tumors.
[0003] FGFR4 has become a hot target in anti-tumor research today, attracting the attention and R&D boom of many scientific research institutions and pharmaceutical companies. At present, several selective FGFR4 inhibitors have been reported. In 2015, Hagel et al. developed the first selective inhibitor BLU554 based on the difference between the key cysteine at position 552 of FGFR4 and other members and entered clinical research (NCT02508467), showing good therapeutic effects in patients with liver cancer and cholangiocarcinoma. The FGFR4 selective inhibitor H3B-6527 developed by H3 Biomedicine has also entered clinical phase I (NCT03424577). Novartis has developed a class of reversible covalent inhibitors of selective FGFR4, among which the representative compound FGF401 has entered clinical phase I / II (NCT02325739) for the treatment of liver cancer with overexpression of FGFR4. Although many companies at home and abroad have also studied FGFR4 selective inhibitors, no FGFR4 selective inhibitors have been approved for marketing, and more effective FGFR4 selective inhibitors are urgently needed in clinical practice. In addition, due to the mutation of gatekeeping amino acids in the FGFR4 protein kinase region, existing clinical drugs have target-dependent drug resistance problems, and therefore there is an urgent need to study new FGFR4 inhibitors. Summary of the invention
[0004] In view of the above problems, the present invention provides a new class of 3-aminoindazole compounds, which can inhibit the activity of FGFR4 protein kinase with high selectivity and high activity, thereby inhibiting the proliferation of various tumor cells.

DETAILED DESCRIPTION
[0048] The experimental methods in the following examples of the present invention, where no specific conditions are specified, are usually carried out under conventional conditions or under conditions recommended by the manufacturers. The various commonly used chemical reagents used in the examples are all commercially available products.
[0049] Unless otherwise defined, all technical and scientific terms used in the present invention have the same meaning as those commonly understood by those skilled in the art of the present invention. The terms used in the specification of the present invention are only for the purpose of describing specific embodiments and are not intended to limit the present invention.
[0050] The terms "including" and "having" and any variations thereof of the present invention are intended to cover non-exclusive inclusions. For example, a process, method, device, product or equipment comprising a series of steps is not limited to the listed steps or modules, but may optionally include steps not listed, or may optionally include other steps inherent to these processes, methods, products or equipment.
[0051] In the compounds described in the present invention, when any variable (such as R 12 If a (, etc.) compound occurs more than once in any component, its definition at each occurrence is independent of its definition at every other occurrence. Likewise, combinations of substituents and variables are permitted so long as such combinations result in stable compounds. A line drawn from a substituent into the ring system indicates that the indicated bond may be attached to any substitutable ring atom. If the ring system is polycyclic, this means that such bonds are attached only to any suitable carbon atom of an adjacent ring. It is to be understood that one of ordinary skill in the art can select substituents and substitution patterns for the compounds of the invention to provide compounds that are chemically stable and that can be readily synthesized from readily available raw materials by techniques in the art and the methods set forth below. If a substituent is itself substituted with more than one group, it is to be understood that these groups may be on the same carbon atom or on different carbon atoms so long as the structure is stable.
[0052] As used herein, the term "alkyl" is intended to include branched and straight chain saturated aliphatic hydrocarbon groups having a specific number of carbon atoms. For example, the definition of "C1-C6" in "C1-C6 alkyl" includes groups having 1, 2, 3, 4, 5 or 6 carbon atoms in a straight or branched arrangement. For example, "C1-C6 alkyl" specifically includes methyl, ethyl, n-propyl, isopropyl, n-butyl, tert-butyl, isobutyl, pentyl, and hexyl.
[0053] The term "cycloalkyl" refers to a monocyclic saturated aliphatic hydrocarbon group having a specific number of carbon atoms. For example, "cycloalkyl" includes cyclopropyl, cyclobutyl, cyclopentyl or cyclohexyl, etc.
[0054] The term "alkoxy" refers to a group having an -O-alkyl structure, such as -OCH3, -OCH2CH3, -OCH2CH2CH3, -O-CH2CH(CH3)2, -OCH2CH2CH2CH3, -O-CH(CH3)2, and the like.
[0055] The term "alkylamino" refers to a group having an -N-(alkyl)2 or -NH-alkyl structure, such as -NHCH3, -NHCH2CH3, -NHCH2CH2CH3, -NHCH2CH(CH3)2, -NHCH2CH2CH2CH3, -NHCH(CH3)2, -N(CH3)2, -N(CH2CH)2, -N(CH2CH)2, -N(CH2CH2CH3)2, etc.
[0056] The term "heterocycloalkyl" or "heterocyclyl" is a saturated or partially unsaturated monocyclic, fused, spiro or bridged cyclic substituent, wherein one or more ring atoms are selected from N, O or S(O)m (wherein m is an integer from 0 to 2) heteroatoms, and the remaining ring atoms are carbon, for example: morpholinyl, piperidinyl, tetrahydropyrrolyl, oxetanyl, piperazinyl, pyrrolidinyl, dihydroimidazolyl, dihydroisoxazolyl, dihydroisothiazolyl, dihydrooxadiazolyl, dihydrooxazolyl, dihydropyrazinyl, dihydropyrazolyl, dihydropyridinyl, dihydropyrimidinyl, dihydropyrrolyl, dihydrotetrazolyl, dihydrothiadiazolyl, dihydrothiazolyl, dihydrothiophenyl, dihydrotriazolyl, dihydroazetidinyl, tetrahydrofuranyl, tetrahydrothiophenyl, and the like, and N-oxides thereof. The attachment of the heterocyclic substituent may be achieved through carbon atoms or through heteroatoms.
[0057] As will be appreciated by those skilled in the art, "halo" or "halo" as used herein refers to chlorine, fluorine, bromine and iodine.
[0058] Unless otherwise defined, alkyl, cycloalkyl, aryl, heteroaryl and heterocycloalkyl substituents may be unsubstituted or substituted. For example, C1-C6 alkyl may be substituted with one, two or three substituents selected from OH, halogen, alkoxy, dialkylamino or heterocyclic radicals such as morpholinyl, piperidinyl, etc.
[0059] The present invention includes free forms of compounds of formula I-IV, as well as pharmaceutically acceptable salts and stereoisomers thereof. The term "free form" refers to amine compounds in non-salt form. The pharmaceutically acceptable salts included include not only the exemplary salts of the specific compounds described herein, but also typical pharmaceutically acceptable salts of the free forms of all compounds of formula I-IV. The free form of the specific salt of the compound can be isolated using techniques known in the art. For example, the free form can be regenerated by treating the salt with an appropriate dilute aqueous base solution, such as a dilute aqueous solution of NaOH, a dilute aqueous solution of potassium carbonate, a dilute aqueous ammonia, and a dilute aqueous solution of sodium bicarbonate. The free form is somewhat different from its respective salt form in certain physical properties, such as solubility in polar solvents, but for the purpose of the invention, such acid salts and base salts are equivalent to their respective free forms in other pharmaceutical aspects.
[0060] Pharmaceutically acceptable salts of the present invention can be synthesized from compounds of the present invention containing a basic or acidic moiety by conventional chemical methods. Typically, salts of basic compounds are prepared by ion exchange chromatography or by reacting a free base with a stoichiometric amount or excess of an inorganic or organic acid in the desired salt form in an appropriate solvent or combination of solvents. Similarly, salts of acidic compounds are formed by reacting with an appropriate inorganic or organic base.
[0061] Therefore, the pharmaceutically acceptable salts of the compounds of the present invention include conventional non-toxic salts of the compounds of the present invention formed by reacting the alkaline compounds of the present invention with inorganic or organic acids. For example, conventional non-toxic salts include salts derived from inorganic acids such as hydrochloric acid, hydrobromic acid, sulfuric acid, aminosulfonic acid, phosphoric acid, nitric acid, etc., and also include salts prepared from organic acids such as acetic acid, propionic acid, succinic acid, glycolic acid, stearic acid, lactic acid, malic acid, tartaric acid, citric acid, ascorbic acid, pamoic acid, maleic acid, hydroxymaleic acid, phenylacetic acid, glutamic acid, benzoic acid, salicylic acid, p-aminobenzenesulfonic acid, 2-acetoxy-benzoic acid, fumaric acid, toluenesulfonic acid, methanesulfonic acid, ethanedisulfonic acid, oxalic acid, isethionic acid, trifluoroacetic acid, etc.
[0062] If the compound of the present invention is acidic, then suitable "pharmaceutically acceptable salts" refer to salts prepared by pharmaceutically acceptable non-toxic bases including inorganic bases and organic bases. Salts derived from inorganic bases include aluminum salts, ammonium salts, calcium salts, copper salts, iron salts, ferrous salts, lithium salts, magnesium salts, manganic salts, manganous salts, potassium salts, sodium salts, zinc salts, etc. Ammonium salts, calcium salts, magnesium salts, potassium salts and sodium salts are particularly preferred. Salts derived from pharmaceutically acceptable organic non-toxic bases, including salts of primary, secondary and tertiary amines, substituted amines including naturally occurring substituted amines, cyclic amines and basic ion exchange resins such as arginine, betaine, caffeine, choline, N,N'-dibenzylethylenediamine, diethylamine, 2-diethylaminoethanol, 2-dimethylaminoethanol, aminoethanol, ethanolamine, ethylenediamine, N-ethylmorpholine, N-ethylpiperidine, glucosamine, glucosamine, histidine, hydroxocobalamin, isopropylamine, lysine, methylglucosamine, morpholine, piperazine, piperidine, guaiac, polyamine resins, procaine, purine, theobromine, triethylamine, trimethylamine, tripropylamine, tromethamine and the like.
[0063] Berg et al., "Pharmaceutical Salts," J. Pharm. Sci. '1977: 66: 1-19, describes in more detail the preparation of the above-mentioned pharmaceutically acceptable salts and other typical pharmaceutically acceptable salts.
[0064] Since deprotonated acidic moieties such as carboxyl groups in the compounds can be anionic under physiological conditions, and this charge can then be balanced by the internal protonated or alkylated basic moieties such as quaternary nitrogen atoms that carry cations, it should be noted that the compounds of the present invention are potential internal salts or zwitterions.
[0065] In some embodiments, the present invention provides a method for treating hyperproliferative diseases or symptoms such as tumors in humans or other mammals using compounds having formula I-IV and pharmaceutically acceptable salts thereof.
[0066] In some embodiments, the compounds of the present invention and pharmaceutically acceptable salts thereof can be used to treat or control hyperproliferative diseases such as liver cancer, bile duct cancer, breast cancer, gastric cancer, and prostate cancer.
[0067] Drug metabolites and prodrugs
[0068] The metabolites of the compounds involved in the present invention and their pharmaceutically acceptable salts, as well as prodrugs that can be converted into the structures of the compounds involved in the present invention and their pharmaceutically acceptable salts in vivo, are also included in the claims of the present invention.
[0069] Combination medication
[0070] The compounds of formula I-IV can be used in combination with other drugs known to treat or improve similar conditions. When administered in combination, the administration method and dosage of the original drug remain unchanged, and the compounds of formula I-IV are taken simultaneously or subsequently. When the compounds of formula I-IV are taken simultaneously with one or more other drugs, it is preferred to use a pharmaceutical composition containing one or more known drugs and the compounds of formula I-IV. Drug combination also includes taking the compounds of formula I-IV and one or more other known drugs in overlapping time periods. When the compounds of formula I-IV are used in combination with one or more other drugs, the dosage of the compounds of formula I-IV or the known drugs may be lower than when they are used alone.

[0225] The brief steps for stable strain construction are as follows:
[0226] 1. The kinase domain gene of human FGFR4 (NM_002011.5) was fused to the N-terminus of ETV6 (NM_001987.4), and the fusion gene TEL-FGFR4 was cloned into the pMSCV vector. Positive clones were selected and verified by sequencing.
[0227] 2. Using the recombinant plasmid TEL-FGFR4-pMSCV as a template, construct the FGFR4 gatekeeper mutant plasmids TEL-FGFR4V550L-pMSCV and TEL-FGFR4 V550M-pMSCV.
[0228] 3. Transfect 293T cells with the three successfully constructed plasmids and the empty plasmid without the target gene, and collect and preserve the virus-containing supernatant culture medium.
[0229] 4. Use this supernatant medium to infect Ba / F3 cells for three days, change to normal medium containing puromycin for one day of recovery, and then select positive cell lines in the absence of interleukin-3 and with the addition of puromycin.
[0230] 5. The positive stable cell lines screened were verified by western-blot.
[0231] Cell proliferation inhibitory activity study:
[0232] 1. Add 2000 positive stable cell lines obtained by screening into a 96-well cell culture plate, and use the Ba / F3 cell line transfected with an empty plasmid without the target gene as a control.
[0233] 2. After 6-12 hours, add an equal volume of pre-diluted 3-aminoindazole compound (3-fold dilution).
[0234] 3. After 72 h, add Cell Counting Kit-8 (CCK-8) reagent to detect cell viability.
[0235] 4. Calculate IC of small molecule inhibition using GraphPad Prism software 50 .
[0236] The test results are shown in Table 2.

[0239] It can be seen from the data in Table 2 that the 3-aminoindazole compounds of the present invention have strong inhibitory activity on the cell proliferation of BaF3-FGFR4, BaF3-FGFR4V550L and BaF3-FGFR4 V550M stable strains.
[0240] Experimental Example 3: Test of the inhibitory activity of compounds on tumor cell proliferation
[0241] In vitro cell proliferation inhibition was detected by CCK8 assay: Tumor cells (MDA-MB-453 (breast cancer cell line), Hep3B (liver cancer cell line) and H1299 (lung cancer cell line)) from the Maurice Wilkins Center of Excellence at the University of Auckland, New Zealand were stored in the corresponding growth medium and passaged at least two times before recovery. An appropriate number of cells in logarithmic growth phase were inoculated on a 6×7 96-well plate with a volume of 100 μL per well, and then cultured overnight in a 37°C incubator containing 5% CO2. In addition, the test compound was first dissolved in DMSO to make a 10 μM / L stock solution, and then diluted from 1×10 -5 M / L to1×10 -10 M / L dilution was performed in 10 gradients, and 2 μL of compound solutions of different concentrations were taken and mixed evenly with 998 μL of culture medium. Then, 100 μL of the culture medium mixed with the compound was taken and added to the corresponding 96-well plate. 2 μL of DMSO was used as a blank control. After 48 hours of co-incubation, 20 μL of Cell Counting Kit-8 (CCK-8) (5 mg / mL) reagent was added, and after shaking for 10 minutes, the number of stained cells was read using SynergyTMHT (Bio Tek) / OD570. Finally, the data was processed with GraphPad Prism, and the nonlinear regression curve was fitted using the S-type dose-response to obtain the IC 50 Values (Table 3).
