Description

Technical Field

[0001] The invention belongs to the technical field of microorganisms, and in particular relates to a Burkholderia forestii strain and application thereof. Background Art

[0002] Crop bacterial wilt caused by infection with Ralstonia solanacearum species complex (RSSC) is a major disease worldwide. RSSC can infect more than 450 plant species from more than 50 families, especially Solanaceae crops such as tomatoes, potatoes, eggplants, and tobacco, causing serious economic losses. The yield loss caused by bacterial wilt generally exceeds 50%, or even a total crop failure, and the economic losses caused are difficult to estimate.

[0003] The dominant strain causing tomato bacterial wilt in Guangdong Province is RSSC evolution type I, namely Ralstonia pseudosolanacearum, with six sequence variants, namely 13, 14, 17, 18, 34, and 44. In recent years, the evolution of pathogenicity of tomato bacterial wilt pathogen populations has accelerated, and new sequence variants and highly pathogenic strains have continued to emerge, resulting in the gradual loss of resistance of crop varieties. There is a lack of effective control agents on the market, and the research and development of prevention and control technologies is insufficient. In production, the phenomenon of reduced production or total crop failure due to bacterial wilt is very common, causing huge economic losses to producers.

[0004] At present, the prevention and control of tomato bacterial wilt still mainly relies on chemical pesticides. However, there are not many options for effective pesticides. Farmers often use a mixture of several fungicides. During the peak of the disease, they spray chemical pesticides almost every week, resulting in a large amount of financial and material waste and environmental pollution. In addition, the long-term and large-scale use of chemical pesticides is prone to the risk of drug resistance. Therefore, in order to deal with the chemical pesticide pollution caused by the prevention and control of tomato bacterial wilt, it is necessary to find a suitable biological control method as an effective way to prevent and control crop bacterial wilt. However, the biological resources that can be used for biological control of tomato bacterial wilt are still very limited and need to be further explored. Summary of the invention

[0047] Example 1 Identification of strain BB417

[0048] 1. Isolation of strain BB417

[0049] BB417 was isolated from tomato rhizosphere soil. The specific isolation steps are as follows: weigh 1.0 g of the collected rhizosphere soil sample and add it to a 100 mL Erlenmeyer flask containing 9 mL of sterilized ddH2O, place it in an oscillating incubator, oscillate at 30°C and 160 r / min for 30 min, and then let it stand for 30 s to prepare a soil bacterial suspension; draw 1 mL of the soil bacterial suspension and add it to a 15 mL sterile test tube containing 9 mL of sterilized ddH2O, then invert and mix to prepare 10 -2 Soil bacteria suspension; and so on, get 10 -3 , 10 -4 and 10 -5 Soil bacterial suspension. -3 , 10 -4 and 10 -5 150 μL of soil bacteria suspension was evenly spread on a 15 cm LB medium plate with sterilized glass beads, dried, and inverted in a 30°C incubator for 1 to 3 days to obtain 219 single colonies. The single colony numbered BB417 was selected and streaked on the LB medium plate for purification to obtain a single colony, which was repeated 3 times.

[0050] 2. Screening of strain BB417

[0051] The single colonies obtained by separation and purification were inoculated into each 96-well plate containing 10 μL sterilized ddH2O to prepare bacterial suspension; 5 μL of bacterial suspension was slowly added to the filter paper in the 15 cm plate (60 to 80 filter paper in each plate, and the bacterial suspension added to each filter paper was a different single colony), and then dried and incubated inverted in a 30°C incubator for 36 to 48 hours; bacteria with obvious transparent inhibition zones around the colonies and an inhibition zone diameter of more than 1.0 cm were inoculated into the liquid culture medium, and photographed and recorded ( Figure 1 ), cultured at 30℃ with shaking at 160r / min, and then stored in a -80℃ refrigerator with 50% glycerol for later use.

[0052] 3. 16S rDNA of strain BB417

[0053] The target band of strain BB417 was amplified by PCR using 16S rDNA primers, and the target fragments were recovered, cloned and sequenced. The results showed that the size of BB41716S rDNA fragments was 1428bp. The phylogenetic tree was constructed using MEGA X software, and strain BB417 and Burkholderia strains were clustered in the same branch. The 16S rDNA sequence of BB417 (SEQ ID No. 1) is:

[0054]

[0055] 4. Morphological identification

[0056] The cells of strain BB417 are red after Gram staining and are Gram-negative bacteria. The colonies on the LB plate are round, milky white, with a raised center and smooth edges. Figure 1 ).

[0057] 5. Physiological and biochemical characteristics

[0058] BB417 is motile, and the results of nitrate reduction, citrate utilization, arginine hydrolysis, urease utilization, malonate utilization, casein hydrolysis and esterase tests are positive; the results of denitrification, VP test, methyl red test and amylase test are negative; BB417 can utilize maltose, dulcitol, cellobiose, rhamnose, sorbitol, mannitol and sucrose, but cannot utilize lactose and erythritol.

[0059] 6. Whole genome analysis

[0060] The strain BB417 contains three circular chromosomes, with chromosome genome sizes of 3420068bp, 3626782bp and 1244966bp, respectively, and G+C contents of 67.06%, 66.8% and 66.59%, respectively. There are 7361 coding genes with a total length of 7246470bp, including 68 tRNAs, 17 sRNAs and 4 ncRNAs. The BB417 genome also contains 2 prophages, 2 Gls( Figure 2 ). The results of multi-gene phylogenetic tree analysis and cluster analysis showed that strain BB417 and Burkholderia arboris (R-24201) were clustered into one branch ( Figure 3 ). This indicates that strain BB417 is Burkholderia arboris.

[0061] Example 2 Antibacterial effect of strain BB417 on tomato Ralstonia solanacearum strains

[0062] Activate BB417 strains and strains Ssf-4, RS129, RS180, RS397, RS404, TS-2 of Ralstonia pseudosolanalis, the pathogen of tomato bacterial wilt (the above strains were isolated and preserved by the Institute of Protection of Guangdong Academy of Agricultural Sciences) and GMI1000 (provided by Professor Lu Yongjun of Sun Yat-sen University). After culturing Ralstonia solanacearum for 2 days and biocontrol bacteria for 1 day at 30°C, pick a single colony into 5mL LB and shake at 30°C and 160r / min to obtain bacterial solution for use. Prepare 9cm plates containing bacteria using Ralstonia solanacearum strains as indicator bacteria. Put 3 filter paper pieces on each plate, draw 5μL BB417 bacterial solution and slowly add it to the filter paper pieces in the 9cm plates, repeat 3 times; after drying, invert and culture in a 30°C incubator for 48h; measure and record the diameter of the inhibition zone using the cross method (Table 1), and take photos and record ( Figure 4 ).

[0063] From Table 1 and Figure 4 It can be seen that BB417 has certain antibacterial activity against tomato Ralstonia solanacearum strains of different sequence variants. There are differences in the antibacterial effects of BB417 on 8 strains of Ralstonia solanacearum of 6 sequence variants, and the diameters of the inhibition zones are 1.41-2.00 cm respectively.

[0066] Example 3 Biological functions of strain 183

[0067] Determination of IAA production capacity of strain BB417: IAA detection medium shake test: Add 5mL of king culture medium to each test tube, add 100μL of BB417 bacterial solution with OD600=1, add 100μL of sterile water to the indoleacetic acid (IAA) detection medium culture medium as a blank control, and culture on a shaker at 30℃ and 180r / min for two days. After centrifugation of the bacterial solution, take 200μL of supernatant and 200uL of IAA colorimetric solution in a 1.5mL centrifuge tube, use the shaken 100μL of sterile water as a control, and observe the color change after leaving at room temperature for 20min. If the liquid in the test tube turns red, it indicates that IAA is produced.

[0068] Determination of the ability of strain BB417 to secrete siderophore: The strain was inoculated onto CAS detection medium using the single-point method. After culturing at 30°C for 3 days, the presence of a yellow halo around the colony was observed. If so, the bacteria were able to produce siderophore.

[0069] Detection of nitrogen fixation and phosphorus solubilization ability of strain BB417: Use the streak method to inoculate BB417 onto Ashuber's nitrogen fixation medium. After culturing at 30°C for 2 days, observe whether a transparent circle is formed around it. Use the three-point method to inoculate the BB417 bacterial solution onto the phosphorus solubilization medium to observe whether a transparent circle is formed around it.

[0070] Determination of enzyme production capacity of strain BB417: The activities of amylase, cellulase, β, 1-3 glucan hydrolase and protease of strain BB417 were determined according to the methods of Hu Rong et al., Liu Wan, Chen Yunyun, Wang Chao et al.

[0071] The results showed that BB417 had protease hydrolysis activity, the ability to solubilize phosphate, fix nitrogen and produce IAA, no cellulase β and 1,3 glucanase hydrolysis activity, and the ability to produce iron carriers ( Figure 5 ).

[0072] Example 4 Strain BB417 has the ability to promote growth of tomato plants

[0073] 1. Preparation of bacterial culture of strain BB417

[0074] Pick a single colony of activated BB417 on the LB plate and inoculate it into a test tube containing 5 mL of LB liquid medium, and culture it in a 30°C incubator at 180 r / min for 24 h. For further cultivation, inoculate the obtained BB417 seed liquid into a conical flask containing 150 mL of LB liquid medium at a 1% inoculum amount, and continue to culture it for 24 h under the same culture conditions.

[0075] 2. Vaccination and investigation

[0076] When the tomato plant had two leaves and one heart, the tomato seedlings with the same growth were selected for growth promotion ability determination. Each tomato seedling was irrigated with 40 mL of the concentration of 1×10 8 cfu / mL biocontrol liquid, and 40mL diluted LB liquid medium was used as the control. 15 seedlings were treated in each treatment, and 3 replicates were set. Each treatment was randomly arranged. After 15 days, the tomato plant height, root length, stem diameter, fresh weight and root weight were measured and photographed and recorded ( Figure 6 ) and calculated the growth-promoting effect (GPE%).

[0077] Depend on Figure 6 It can be seen that the plant height, root length, stem diameter, fresh weight and root weight of tomato plants were measured 15 days after the tomato plants were treated with BB417 bacterial solution. The results showed that compared with the control treatment, BB417 had a significant growth-promoting effect on the plant height, fresh weight and root weight of tomato plants, among which the growth-promoting effect on plant height was 22.29%, the growth-promoting effect on fresh weight was 55.07%, and the growth-promoting effect on root weight was 97.14%; however, the growth-promoting effect of BB417 on the root length and stem diameter of tomato plants was not significantly different from that of the control treatment.

[0078] Example 5 Strain BB417 inhibits gene expression of Ralstonia solanacearum

[0079] 1. Co-culture of BB417 and RS550

[0080] Take the bacterial suspension of BB417 and RS550 cultured overnight, dilute with sterile water to an OD value of 0.1, take 500 μL of the dilution respectively and add it to 150 mL of NB liquid culture medium at the same time, and culture for 24 hours. Take 500 μL of RS550 dilution alone and add it to 150 mL of NB liquid culture medium and culture for 24 hours as a control; then use an RNA extraction kit to extract RNA, determine the concentration, and adjust the RNA concentration of the co-cultured strain and the RS550 RNA cultured alone with ddH2O.

[0081] 2. Expression of virulence-related genes in RS550

[0082] The phcA, phcB, esp and pheC genes are pathogenicity-related genes of Ralstonia solanacearum. After knocking out these genes, the pathogenicity of Ralstonia solanacearum will decrease. Figure 7 As shown in the figure, the co-culture of BB417 and RS550 can significantly reduce the expression levels of phcA, phcB and esp3 genes. The co-culture of BB417 and RS550 can reduce the expression level of pheC, but the difference between the expression level of pheC and RS550 cultured alone is not significant ( Figure 7 ).

[0083] Example 6: Effect of strain BB417 on tomato bacterial wilt

[0084] 1. Preparation of Ralstonia solanacearum fermentation broth

[0085] Preparation of suspension of Ralstonia solanacearum strain RS550: Pick up activated single colonies on TZC medium and rejuvenate on TZC slant medium, culture in a 30°C incubator for 24 hours, transfer white colonies to inoculate into a test tube containing 5 mL of TZC liquid medium, and culture in a 30°C incubator at 180 r / min for 24 hours to obtain Ralstonia solanacearum suspension.

[0086] 2. Vaccination and investigation

[0087] Tomatoes were sown in sterilized soil mixed with nutrient soil and garden soil in a ratio of 1:1, and transplanted when the tomatoes had 4 leaves and one heart. One week after planting, 50 mL of BB417 solution (final concentration of 1×10 8 cfu / mL), and 50 mL of 20% thiamethoxam diluted 300 times was used as treatment and tap water as control. After 5 days, each treatment was injected with 50 mL of RS550 bacterial solution (final concentration of 1×10 8cfu / mL), and only irrigation with bacterial wilt solution was used as a positive control. Fifteen tomato seedlings were treated for each treatment, with 3 replicates. The treatments were randomly arranged, the culture temperature was 25-30°C, and the humidity was 60-80%. After 7 days, the disease level was investigated and the control effect was statistically analyzed. Grading of tomato bacterial wilt: Level 0: healthy plants without wilting; Level 1: 1-25% of the leaves wilt; Level 2: 26-50% of the leaves wilt; Level 3: 51-75% of the leaves wilt; Level 4: 76-100% of the leaves wilt. The disease index and biocontrol efficiency of tomato bacterial wilt were calculated using the following formula:

[0088] Disease index = ∑ (number of diseased plants × disease grade) / (total number of plants surveyed × highest grade) × 100

[0089] Biocontrol rate (%) = (control disease index - treatment group disease index) / control disease index × 100%

[0090] The results showed that 7 days after the treatment with Ralstonia solanacearum, most of the tomato plants treated with Ralstonia solanacearum alone were infected, with an average disease rate of 88.89% and a disease index of 82.77; the average disease rate of tomato plants treated with BB417 bacterial solution was 37.78%, with a disease index of 31.10, and the control effect of strain BB417 on tomato bacterial wilt was 62.44%; the average disease rate of tomato plants treated with 20% thiamethoxam 300-fold solution was 40%, with a disease index of 36.77, and the control effect of strain BB417 on tomato bacterial wilt was 55.58% (Table 2). The results of variance analysis showed that the control effect of BB417 bacterial solution on tomato bacterial wilt was higher than that of 20% thiamethoxam, but the difference was not significant (Table 2, Figure 8 ).
