[0002] Silicosis is a disease caused by long-term inhalation of free silicon dioxide (SiO 2 ) is a disease caused by dust with diffuse fibrosis of lung tissue as the main manifestation. It is also one of the occupational diseases with the highest incidence rate and the main cause of labor loss of workers in dusty environments. Its main pathological changes are proliferation of pulmonary interstitial cells and excessive deposition of extracellular matrix leading to pulmonary fibrosis. Silicosis has a long history in my country. In the Shang and Zhou dynasties, there were people engaged in ceramics, mining and smelting. In the Song Dynasty, "Tan Yuan" recorded that "the quarrymen in Jiagu Mountain were injured by the stones, and many died of lung scorch." Later doctors had other theories on the cause of the disease, such as "metal and stone are hot" and "metal and stone are dry and toxic." For patients with silicosis fibrosis, they can take drugs such as acetylcysteine tablets, pirfenidone tablets and nintedanib for treatment, but Western medicine treatment has the defects of high price, large side effects and treating symptoms but not the root cause. Therefore, it is urgent to develop traditional Chinese medicine compound preparations for the treatment of silicosis fibrosis. Summary of the invention

[0043] Animal Experimentation:

[0044] (1) Experimental solvent:

[0045] 3% sodium pentobarbital solution: Weigh 3 g of sodium pentobarbital powder using a balance, pour the sodium pentobarbital powder into a 200 mL volumetric flask, add 100 mL of normal saline, dissolve and mix to obtain a 3 g / mL sodium pentobarbital solution, and store in a refrigerator at 4°C for later use.

[0046] (2) Experimental procedures:

[0047] The Wistar rats in the above experimental animal groups were killed by overdose anesthesia with 3% sodium pentobarbital solution, and the killed Wistar rats were fixed on the operating table, and the fur from the neck to the xiphoid process was wiped with a cotton ball soaked in physiological saline. A longitudinal incision was made from the neck to the sternal manubrium, and the layers were peeled away until the trachea and lungs were exposed. A T-shaped incision was made in the trachea, and a cannula was intubated and fixed with a thin thread. 5 mL of physiological saline was drawn with a 5 mL syringe and the lungs were lavaged through the cannula. Lavage was performed three times until the alveoli turned white. The alveolar lavage fluid (15 mL) was collected three times and placed in a 15 mL centrifuge tube and mixed, and filtered with a double-layer sterile gauze to obtain bronchoalveolar lavage fluid (BALF). The BALF was placed in a centrifuge tube and stored in an ice box.

[0048] The lungs, livers and kidneys of Wistar rats were completely separated, the surrounding connective tissues were carefully removed, the thin wires were cut and the cannulas were recovered, the lungs were flushed with saline, the lungs were photographed, the color, gloss, texture, elasticity, lung congestion and swelling of both lungs were observed with the naked eye, and the gross images of the animal samples were preserved. The right lower lobe of the lung was taken for the observation of lung tissue pathology, and the remaining lobes of the lung were placed in different centrifuge tubes and stored in a -80°C refrigerator for Western blot analysis.

[0049] Figure 1 The lung tissue samples of Wistar rats in the model control group, silicosis model group, Shenfu Tang Jiajian preparation prevention group and Shenfu Tang Jiajian preparation treatment group were collected by Figure 1 It can be seen that the surface of the lung tissue of the model control group was smooth, and no obvious granular nodules were found; there were granular nodules on the surface of the lung tissue of the silicosis model 1-week group, silicosis model 2-week group, silicosis model 4-week group and silicosis model 8-week group, and the granular nodules became more obvious with the extension of the feeding time; granular nodules were visible on the surface of the lung tissue of the Shenfu Tang Jiajian preparation prevention group, but the range of granular nodules was smaller than that of the silicosis model 8-week group; compared with the silicosis model 8-week group, although there were granular nodules on the surface of the lung tissue of the Shenfu Tang Jiajian preparation treatment group, the range of granular nodules was smaller than that of the silicosis model 8-week group. Compared with the Shenfu Tang Jiajian preparation high-dose treatment group, the Shenfu Tang Jiajian preparation medium-dose treatment group and the Shenfu Tang Jiajian preparation low-dose treatment group, the lungs of the Shenfu Tang Jiajian preparation high-dose treatment group were pink, soft in texture, and the granular nodules were significantly smaller than those of the Shenfu Tang Jiajian preparation low-dose treatment group and the Shenfu Tang Jiajian preparation medium-dose treatment group. These experimental results indicate that the fibrosis symptoms of rats were alleviated after administration of the modified Shenfu Decoction, and Shenfu Decoction has the potential to prevent and treat silicotic fibrosis.

[0050] The lungs were rinsed with physiological saline and then dried with filter paper. The wet weight of the lungs was measured using an electronic balance and the lung coefficient was calculated: lung coefficient = (wet weight of lungs / body weight of Wistar rats) × 100%. Figure 2This is a statistical chart showing the changes in lung coefficient indicators of Wistar rats in the model control group, silicosis model group, Shenfu Decoction plus or minus preparation prevention group and Shenfu Decoction plus or minus preparation treatment group. Compared with the model control group, the lung coefficient of Wistar rats in the silicosis model group was significantly increased. After treatment with Shenfu Decoction plus or minus preparation, the lung coefficient of Wistar rats in the Shenfu Decoction plus or minus preparation treatment group was reduced, and was dose-dependent.

[0051] To further investigate the pathological morphology of the lung tissue of Wistar rats in each experimental animal group, the right lower lobe of the lung was immersed in a pre-prepared 4% paraformaldehyde fixative, fixed in a 4°C refrigerator for 48 hours, embedded in paraffin, and sliced (the slice thickness was 4 μm). The lung tissue sections of Wistar rats were stained with hematoxylin-eosin (HE) for observation. Figure 3 As shown, Figure 3 HE staining of lung tissues of Wistar rats in the model control group, silicosis model group, Shenfu Tang Jiajian preparation prevention group, and Shenfu Tang Jiajian preparation treatment group. Figure 3 It can be seen that the alveolar tissue structure of Wistar rats in the 8-week model control group was normal, and no obvious inflammatory cell infiltration was observed. The silicotic silicotic nodules of Wistar rats in the 8-week silicosis model group were obvious, the alveolar tissue septa were thickened, and inflammatory cell infiltration was observed. Silicotic silicotic nodules were present in the prevention group of Shenfu Decoction and the treatment group of Shenfu Decoction, and the alveolar septa were thickened and inflammatory cells were infiltrated. However, the fibrosis area was smaller than that in the 8-week silicosis model group. Compared with the prevention group of Shenfu Decoction and the low-dose treatment group of Shenfu Decoction, the lung tissue of Wistar rats in the medium-dose treatment group of Shenfu Decoction and the high-dose treatment group of Shenfu Decoction showed reduced inflammatory cell infiltration and reduced silicotic nodule area.

[0052] The liver and kidney were immersed in a pre-prepared 4% paraformaldehyde fixative, fixed in a 4°C refrigerator for 48 hours, embedded in paraffin, and sliced (slice thickness was 4 μm). The liver and kidney tissue slices of Wistar rats were stained with HE for observation. Figure 4 HE staining images of liver tissue and kidney tissue of Wistar rats in the model control 8-week group, silicosis model 8-week group, Shenfu Tang Jiajian preparation prevention group and Shenfu Tang Jiajian preparation treatment group. Figure 4 The first and third rows of the HE results of liver tissue are Figure 4 It can be seen that the liver cells of Wistar rats in the 8-week model control group, the prevention group of Shenfu Decoction plus or minus preparation, and the treatment group of Shenfu Decoction plus or minus preparation were neatly arranged and radiated in cords, without necrosis and degeneration, the liver lobule structure was not destroyed and was intact and clear, and no obvious abnormal proliferation of connective tissue was found in the portal area. Figure 4The second and fourth rows are the HE results of renal tissue. The results show that the glomerular structure of Wistar rats in the model control 8-week group, the prevention group of Shenfu Decoction plus or minus preparation, and the treatment group of Shenfu Decoction plus or minus preparation were clear, the renal collecting ducts were arranged tightly and regularly, the renal tubular epithelial cells were arranged regularly and the basement membrane was intact, and no inflammatory cell infiltration was observed. The above results indicate that Shenfu Decoction plus or minus preparation is non-toxic to Wistar rats.

[0053] Take 1 mL of bronchoalveolar lavage fluid (BALF) and dilute it 20 times with normal saline. Take 10 μL of the diluted BALF and add it to the bovine Bowman cell counting plate. Under an optical microscope, count the total number of white blood cells in the four large squares at the four corners of the bovine Bowman cell counting plate. The cell counting formula is: number of cells / mL = (total number of white blood cells / 4) × dilution factor × 10 4 mL. Figure 5 Figure 1 is a statistical chart of the number of white blood cells in bronchoalveolar lavage fluid of Wistar rats in the model control group, silicosis model group, Shenfu Tang Jiajian preparation prevention group, and Shenfu Tang Jiajian preparation treatment group. Figure 5 It can be seen that compared with the model control group, the total white blood cell count in the bronchoalveolar lavage fluid of Wistar rats in the silica-induced silicosis model group was significantly increased, and different doses of Shenfu Decoction plus minus preparation treatment groups could significantly reduce the total white blood cell count in the BALF of Wistar rats in the silica-induced silicosis model group. Among the Shenfu Decoction plus minus preparation treatment groups, the Shenfu Decoction plus minus preparation medium dose treatment group and the Shenfu Decoction plus minus preparation high dose treatment group could better reduce the total white blood cell count in the BALF.

[0054] The remaining 14 mL of BALF was further placed in a 4°C centrifuge and centrifuged at 12000 g for 10 min to obtain a cell pellet. The cell pellet was resuspended with 0.5 mL of precooled (4°C) physiological saline and fully blown to obtain a cell suspension. 5 μL of the cell suspension was aspirated and placed on a clean glass slide for cell smear. After smearing, a fixed area was circled with a crayon. After it was slightly dried, it was fixed with 4% paraformaldehyde fixative for 20 min, and then Wright-Giemsa staining was performed: according to the Wright-Giemsa method According to the instructions of the Wright-Giemsa stain reagent, add 0.5mL of Wright-Giemsa stain A onto the smear and 20s later, add 1mL of Wright-Giemsa stain B onto the Wright-Giemsa stain A. Use an ear bulb to blow gently to mix the Wright-Giemsa stain A and Wright-Giemsa stain B. After staining for 1 minute, rinse with PBS buffer (PH=7), then rinse with tap water, and place in a fume hood to air dry. Observe the number of macrophages on the air-dried slide under an optical microscope.

[0055] Figure 6Figure 2 is the Wright-Giemsa staining of macrophages in the model control group for 8 weeks, the silicosis model group, the prevention group of Shenfu Decoction plus minus preparation, and the treatment group of Shenfu Decoction plus minus preparation. Figure 6 It can be seen that compared with the model control 8-week group, the macrophages of Wistar rats in the silicosis model group increased significantly with the extension of feeding time. Compared with the silicosis model group, the number of macrophages of Wistar rats in the prevention group of Shenfu Decoction plus or minus preparation decreased, and the number of macrophages of Wistar rats in the treatment groups of different doses of Shenfu Decoction plus or minus preparation decreased significantly.

[0056] Figure 7 Expression of α-SMA in lung tissue of Wistar rats in the model control group for 8 weeks, silicosis model group for 8 weeks, Shenfu Tang plus minus preparation prevention group, and Shenfu Tang plus minus preparation treatment group. Figure 7 It can be seen that α-smooth muscle actin (α-SMA) was positively expressed in large quantities in the silicotic nodules and interstitial fibrosis areas of the lung tissue of Wistar rats in the 8-week silicosis model group, and the expression intensity and area were particularly obvious, while the positive expression range and intensity of the model control 8-week group, the Shenfu Decoction prevention group and the Shenfu Decoction treatment group were significantly weaker than those in the 8-week silicosis model group.

[0057] The lung tissue stored in the centrifuge tube was cut into 80 mg small pieces, 600uL tissue cell lysis solution and 6uL protease inhibitor were added, homogenized, and then lysed on ice for 40 minutes to obtain a lysate. The lysate was centrifuged at 12,000×g for 10 minutes at 4°C, and the supernatant (i.e., α-SMA protein) was taken. The α-SMA protein was diluted with ultrapure water to obtain a diluent. The concentration of the diluent was 10ug / uL. The diluent was mixed with the loading buffer. The volume ratio of the diluent to the loading buffer was 4:1. It was heated at 100°C for 10 minutes, cooled, and subjected to Western blot analysis. β-actin was used as an internal reference. The results are as follows: Figure 8 As shown, Figure 8 Western blot and α-SMA protein statistics of Wistar rats in the model control 8-week group, silicosis model 8-week group, Shenfu Tang Jiajian preparation prevention group and Shenfu Tang Jiajian preparation treatment group. Figure 8It can be seen that in Western blot, the expression of α-SMA protein in the lung tissue of Wistar rats in the 8-week silicosis model group was significantly higher than that in the 8-week model control group. Compared with the 8-week silicosis model group, the expression of α-SMA protein in the lung tissue of Wistar rats in the prevention group and the treatment group of Shenfu Tang Jiajian preparation was significantly decreased, among which, the expression of α-SMA protein in the high-dose treatment group of Shenfu Tang Jiajian preparation was close to that in the 8-week model control group, and α-SMA protein is a marker protein of myofibroblasts, which represents the degree of pulmonary fibrosis to some extent. The decrease of α-SMA protein indicates that the degree of fibrosis is improved. Therefore, the high-dose treatment group of Shenfu Tang Jiajian preparation can significantly alleviate silicosis fibrosis in rats, indicating that Shenfu Tang Jiajian preparation can effectively treat pulmonary fibrosis.
