At a minimum, such references including numerical parameters will include variations using mathematical and industrial principles accepted in the art (e.g., rounding, measurement or other systematic errors, manufacturing tolerances, etc.) that will not change the least significant digits.

[0077] In the context of two or more nucleic acid or polypeptide sequences (e.g., Vβ17 antibodies and polynucleotides encoding them, Vβ17 polynucleotides encoding them), the terms "same" or "percent identity" refer to two or more sequences or subsequences that are the same or have a specified percentage of amino acid residues or nucleotides that are the same when compared and aligned for maximum correspondence, as measured using one of the following sequence comparison algorithms or by visual inspection.

[0078] For sequence comparison, usually one sequence is used as a reference sequence to which a test sequence is compared. When using a sequence comparison algorithm, the test sequence and the reference sequence are input into a computer, subsequence coordinates are specified (if necessary), and program parameters for the sequence algorithm are specified. Then, the sequence comparison algorithm calculates the sequence identity percentage for the test sequence relative to the reference sequence based on the specified program parameters.

[0079] Optimal alignment of sequences for comparison can be performed, for example, by the local homology algorithm of Smith & Waterman (Adv. Appl. Math. 2:482 (1981), by using the homology alignment algorithm of Needleman & Wunsch, J Mol. Biol., 48:443 (1970), by searching the similarity method of Pearson & Lipman (Proc. Nat'l. Acad. Sci. USA 85:2444 (1988)), by computerized implementations of these algorithms (GAP, BESTFIT, FASTA, and TFASTA in the Wisconsin Genetics Software Package (Genetics Computer Group, 575 Science Dr., Madison, WI), or by visual inspection (see generally, Current Protocols in Molecular Biology, FM Ausubel et al., eds., Current Protocols, a joint venture between Greene Publishing Associates, Inc. and John Wiley & Sons, Inc. (1995 supplemented edition) (Ausubel)).

[0080] Examples of algorithms suitable for determining percent sequence identity and sequence similarity are the BLAST and BLAST 2.0 algorithms, which are described in Altschul et al., (1990) J. Mol. Biol., 215: 403-410 and Altschul et al., (1997) Nucleic Acids Res. 25: 3389-3402, respectively. Software for performing BLAST analyses is publicly available through the National Center for Biotechnology Information. The algorithm involves first identifying high scoring sequence pairs (HSPs) by identifying short words of length W in the query sequence that match or satisfy some positive-valued threshold score T when aligned with a word of the same length in a database sequence. T is referred to as the neighborhood word score threshold (Altschul et al., supra). These initial neighborhood word hits act as seeds for initiating searches to find longer HSPs containing them. The word hits are then extended in both directions along each sequence for as far as the cumulative alignment score can be increased.

[0081] For nucleotide sequences, the cumulative score is calculated using the parameters M (reward score for a pair of matching residues; always > 0) and N (penalty score for mismatching residues; always < 0). For amino acid sequences, a scoring matrix is used to calculate the cumulative score. Extension of the word hits in each direction is stopped when: the cumulative alignment score falls by the amount X from its maximum achieved value; the cumulative score goes to zero or lower due to the accumulation of one or more negative-scoring residue alignments; or the end of either sequence is reached. The BLAST algorithm parameters W, T, and X determine the sensitivity and speed of the alignment. The BLASTN program (for nucleotide sequences) uses as defaults a word length (W) of 11, an expectation (E) of 10, M=5, N=-4, and a comparison of both chains. For amino acid sequences, the BLASTP program uses as defaults a word length (W) of 3, an expectation (E) of 10, and the BLOSUM62 scoring matrix (see Henikoff and Henikoff, Proc. Natl. Acad. Sci. USA 89:10915 (1989)).

[0082] In addition to calculating the percentage of sequence identity, the BLAST algorithm also performs a statistical analysis of the similarity between two sequences (see, e.g., Karlin and Altschul, Proc. Nat'l. Acad. Sci. USA 90: 5873-5787 (1993)). One measure of similarity provided by the BLAST algorithm is the smallest sum probability (P(N)), which provides an indication of the probability that a match between two nucleotide or amino acid sequences would occur by chance. For example, a nucleic acid is considered similar to a reference sequence if the smallest sum probability in a comparison of the test nucleic acid to the reference nucleic acid is less than about 0.1, more preferably less than about 0.01, and most preferably less than about 0.001.

[0083] Another indication that two nucleic acid sequences or polypeptides are substantially identical is that the polypeptide encoded by the first nucleic acid immunologically cross-reacts with the polypeptide encoded by the second nucleic acid, as described below. Thus, a polypeptide is typically substantially identical to a second polypeptide, e.g., where the two peptides differ only by conservative substitutions. Another indication that two nucleic acid sequences are substantially identical is that the two molecules hybridize to each other under stringent conditions.

[0084] As used herein, the term "polynucleotide", which is synonymously referred to as "nucleic acid molecule", "nucleotide" or "nucleic acid", refers to any polyribonucleotide or polydeoxyribonucleotide, which may be unmodified RNA or DNA or modified RNA or DNA. "Polynucleotide" includes, but is not limited to, single-stranded and double-stranded DNA, DNA that is a mixture of single-stranded and double-stranded regions, single-stranded and double-stranded RNA, and RNA that is a mixture of single-stranded and double-stranded regions, and hybrid molecules comprising DNA and RNA that may be single-stranded or more typically double-stranded or a mixture of single-stranded and double-stranded regions. In addition, "polynucleotide" refers to a triple-stranded region comprising RNA or DNA or both RNA and DNA. The term polynucleotide also includes DNA or RNA containing one or more modified bases, and DNA or RNA having a main chain modified for stability or other reasons. "Modified" bases include, for example, tritylated bases and rare bases such as inosine. A variety of modifications can be made to DNA and RNA; thus, "polynucleotide" includes chemically, enzymatically, or metabolically modified forms of polynucleotides that commonly occur in nature, as well as chemical forms of DNA and RNA that are unique to viruses and cells."Polynucleotide" also includes relatively short nucleic acid chains, commonly referred to as oligonucleotides.

[0085] As used herein, the term "vector" is a replicon into which another nucleic acid segment may be operably inserted so as to bring about the replication or expression of that segment.

[0086] As used herein, the term "host cell" refers to a cell comprising a nucleic acid molecule as provided herein. A "host cell" can be any type of cell, for example, a primary cell, a cell in culture, or a cell from a cell line. In one embodiment, a "host cell" is a cell transfected with a nucleic acid molecule as provided herein. In another embodiment, a "host cell" is a progeny or potential progeny of such a transfected cell. The progeny of a cell may or may not be identical to the parent cell, for example, due to mutations or environmental influences that may occur in the progeny or due to the integration of the nucleic acid molecule into the host cell genome.

[0087] As used herein, the term "expression" refers to the biosynthesis of a gene product. The term encompasses the transcription of a gene into RNA. The term also encompasses the translation of RNA into one or more polypeptides, and also encompasses all naturally occurring post-transcriptional and post-translational modifications. The expressed antibody may be in the cytoplasm of a host cell, in an extracellular environment such as a growth medium for a cell culture, or anchored to a cell membrane.

[0088] As used herein, the terms "peptide", "polypeptide" or "protein" may refer to molecules composed of amino acids and may be recognized as proteins by those skilled in the art. Conventional single-letter or three-letter codes for amino acid residues are used herein. The terms "peptide", "polypeptide" and "protein" are used interchangeably herein to refer to polymers of amino acids of any length. The polymer may be linear or branched, it may contain modified amino acids, and it may be interspersed with non-amino acids. The term also encompasses amino acid polymers that have been modified naturally or by intervention; natural or interventional modifications are, for example, disulfide bond formation, glycosylation, lipidation, acetylation, phosphorylation, or any other manipulation or modification, such as conjugation with a labeling component. The definition also includes, for example, polypeptides containing one or more amino acid analogs (including, for example, non-natural amino acids, etc.) and other modifications known in the art.

[0089] The peptide sequences described herein are written according to the usual convention, with the N-terminal region of the peptide on the left and the C-terminal region on the right. Although isomeric forms of amino acids are known, unless otherwise expressly indicated, it is the L form of the amino acid represented.

[0090] Antibody

[0091] Provided herein are Vβ17 antibodies or antigen-binding fragments thereof, nucleic acids encoding the antibodies and expression vectors, recombinant cells containing the vectors, and compositions comprising the antibodies. Also provided are methods for preparing these antibodies, and methods for using these antibodies to treat diseases. The antibodies disclosed herein have one or more desired functional properties, including but not limited to high affinity binding to Vβ17 or high specificity for Vβ17. In certain embodiments, the antibodies disclosed herein have the ability to treat or prevent a disease or condition when administered to a subject alone or in combination with other therapies.

[0092] As used herein, the term "antibody" is used in a broad sense and includes immunoglobulins or antibody molecules, including human antibodies, humanized antibodies, composite antibodies and chimeric antibodies and monoclonal or polyclonal antibody fragments. Generally speaking, an antibody is a protein or peptide chain that exhibits binding specificity for a specific antigen. The structure of an antibody is well known. According to the amino acid sequence of the heavy chain constant domain, immunoglobulins can be designated as five major classes (i.e., IgA, IgD, IgE, IgG, and IgM). IgA and IgG are further subclassified into isotypes IgA1, IgA2, IgG1, IgG2, IgG3, and IgG4. Therefore, the antibodies provided herein may be any of the five main species or corresponding subclasses. In a specific embodiment, the antibodies provided herein are IgG1, IgG2, IgG3, or IgG4. Based on the amino acid sequence of its constant domain, the antibody light chain of a vertebrate species can be designated as one of two completely different types, i.e., K and λ. Therefore, in certain embodiments, the antibodies provided herein may include a K light chain constant domain. In certain embodiments, the antibodies provided herein may also include a λ light chain constant domain. According to specific embodiments, the antibodies provided herein include a heavy chain constant region and / or a light chain constant region from a mouse antibody or a human antibody. In specific embodiments, the constant region is a human constant region.

[0093] In addition to the heavy and light chain constant domains, antibodies also contain an antigen binding region consisting of a light chain variable region (VL) and a heavy chain variable region (VH), wherein each variable region contains three domains (i.e., complementarity determining region 1 (CDR1), CDR2, and CDR3). "CDR" refers to one of the three hypervariable regions (HCDR1, HCDR2, or HCDR3) within the non-framework region of the immunoglobulin (Ig or antibody) VH β-sheet framework, or one of the three hypervariable regions (LCDR1, LCDR2, or LCDR3) within the non-framework region of the antibody VL β-sheet framework. Thus, CDRs are variable region sequences interspersed within framework region sequences. CDR regions are well known to those skilled in the art and have been defined, for example, by Kabat as the most hypervariable region within an antibody variable (V) domain (Kabat et al., J. Biol. Chem. 252: 6609-6616 (1977); Kabat, Adv. Prot. Chem. 32: 1-75 (1978)). Chothia also structurally defines CDR region sequences as those residues that are not part of the conserved β-sheet framework and are therefore able to adapt to different conformations (Chothia and Lesk, J. Mol. Biol., 196: 901-917 (1987)). Both terms are recognized in the art. CDR region sequences are also defined by AbM, Contact and IMGT. Exemplary CDR region sequences are shown herein, for example, in the table provided below. The positions of CDRs within canonical antibody variable regions have been determined by comparing many structures (Al-Lazikani et al., J. Mol. Biol., 273: 927-948 (1997); Morea et al., Methods 20: 267-279 (2000)). Because the number of residues within the hypervariable region varies among different antibodies, additional residues relative to the canonical position are usually numbered with a, b, c, etc. next to the residue number in the canonical variable region numbering scheme (Al-Lazikani et al., supra, 1997). Such nomenclature is also well known to those skilled in the art.

[0094] The light chain variable region CDR1 domain is interchangeably referred to herein as LCDR1 or VL CDR1. The light chain variable region CDR2 domain is interchangeably referred to herein as LCDR2 or VL CDR2. The light chain variable region CDR3 domain is interchangeably referred to herein as LCDR3 or VL CDR3. The heavy chain variable region CDR1 domain is interchangeably referred to herein as HCDR1 or VH CDR1. The heavy chain variable region CDR2 domain is interchangeably referred to herein as HCDR2 or VH CDR2. The heavy chain variable region CDR1 domain is interchangeably referred to herein as HCDR3 or VH CDR3.

[0095] As used herein, the term "hypervariable region" (such as VH or VL) when used herein refers to a region in an antibody variable region that is hypervariable in sequence and / or forms structurally defined loops. Typically, an antibody comprises six hypervariable regions; three in VH (HCDR1, HCDR2, HCDR3) and three in VL (LCDR1, LCDR2, LCDR3). Many hypervariable region descriptions are in use and are included herein. The "Kabat" CDR is based on sequence variability and is the most commonly used (see, e.g., Kabat et al., Sequences of Proteins of Immunological Interest, 5th Edition, Public Health Service, National Institutes of Health, Bethesda, MD. (1991)). "Chothia" in contrast refers to the position of the structural loops (see, e.g., Chothia and Lesk, J. Mol. Biol., 196:901-917 (1987)). When numbered using the Kabat numbering convention, the ends of the Chothia CDR-HCDR1 loop vary between H32 and H34, depending on the length of the loop (this is because the Kabat numbering scheme places the insertion at H35A and H35B; if both 35A and 35B are absent, the loop ends at 32; if only 35A is present, the loop ends at 33; if both 35A and 35B are present, the loop ends at 34). The "AbM" hypervariable regions represent a compromise between the Kabat CDR and Chothia structural loops and are used by Oxford Molecular's AbM antibody modeling software (see, e.g., Martin, Antibody Engineering, Vol. 2, Chapter 3, Springer Verlag). The "Contact" hypervariable regions are based on analysis of available complex crystal structures.

[0096] Recently, a universal numbering system has been developed and widely adopted, the ImMunoGeneTics (IMGT) Information (Lafranc et al., Dev. Comp. Immunol. 27(1):55-77 (2003)). IMGT is an integrated information system dedicated to the study of immunoglobulins (IG), T cell receptors (TR) and major histocompatibility complex (MHC) of humans and other vertebrates. In this article, CDRs are referenced both by amino acid sequence and by position within the light or heavy chain. Since the "position" of CDRs within the immunoglobulin variable domain structure is conserved between species and is present in structures called loops, CDRs and framework residues are easily identified by using a numbering system that aligns variable domain sequences based on structural features. This information can be used to transplant and replace CDR residues from an immunoglobulin from one species into an acceptor framework, usually from a human antibody. Honegger and Plückthun developed an additional numbering system (AHon), J. Mol. Biol., 309:657-670 (2001). The correspondence between numbering systems, including, for example, Kabat numbering and the IMGT unique numbering system, is well known to those skilled in the art (see, for example, Kabat, supra; Chothia and Lesk, supra; Martin, supra; Lefranc et al., supra). The exemplary system shown herein combines Kabat and Chothia.

[0098] Hypervariable regions may include "extended hypervariable regions" as follows: 24-36 or 24-34 (LCDR1), 46-56 or 50-56 (LCDR2), and 89-97 or 89-96 (LCDR3) in VL, and 26-35 or 26-35A (HCDR1), 50-65 or 49-65 (HCDR2), and 93-102, 94-102 or 95-102 (HCDR3) in VH. CDR sequences reflecting each of the above numbering schemes are provided herein, included in Tables 1 to 6.

[0099] The term "constant region" or "constant domain" refers to the carboxyl terminal portion of the light chain and the heavy chain, which is not directly involved in the binding of the antibody to the antigen, but exhibits various effector functions, such as interactions with Fc receptors. These terms refer to the portion of the immunoglobulin molecule that has a more conserved amino acid sequence relative to the rest of the immunoglobulin (the variable region containing the antigen binding site). The constant region may include the CH1, CH2 and CH3 regions of the heavy chain and the CL region of the light chain.

[0100] The term "framework" or "FR" residues are those variable region residues that flank the CDRs. FR residues are found, for example, in chimeric, humanized, human domain antibodies, diabodies, linear antibodies, and bispecific antibodies. FR residues are those variable domain residues other than hypervariable region residues or CDR residues.

[0101] As used herein, the term "isolated antibody" refers to an antibody that is substantially free of other antibodies with different antigenic specificities (e.g., an isolated antibody that specifically binds to Vβ17 is substantially free of antibodies that do not bind to Vβ17). In addition, an isolated antibody may be substantially free of other cellular material and / or chemicals.

[0102] As used herein, the term "monoclonal antibody" refers to an antibody obtained from a group of substantially homogeneous antibodies, i.e., the individual antibodies constituting the group are identical except for possible naturally occurring mutations that may be present in small amounts. The monoclonal antibodies provided herein can be prepared by hybridoma methods, phage display techniques, single lymphocyte gene cloning techniques, or by recombinant DNA methods. For example, a monoclonal antibody can be produced by a hybridoma comprising a B cell obtained from a transgenic non-human animal such as a transgenic mouse or rat, the transgenic non-human animal having a genome comprising a human heavy chain transgene and a light chain transgene.

[0103] As used herein, the term "antigen binding fragment" refers to an antibody fragment, such as, for example, a diabody, Fab, Fab', F(ab')2, an Fv fragment, a disulfide-stabilized Fv fragment (dsFv), (dsFv)2, a bispecific dsFv (dsFv-dsFv'), a disulfide-stabilized diabody (dsdiabody), a single-chain antibody molecule (scFv), a single domain antibody (sdAb), a scFv dimer (a bivalent bivalent antibody), a multispecific antibody formed from a portion of an antibody comprising one or more CDRs, a camelized single domain antibody, a nanobody, a domain antibody, a bivalent domain antibody, or any other antibody fragment that binds to an antigen but does not contain a complete antibody structure. An antigen binding fragment is capable of binding to the same antigen as the antigen to which the parent antibody or parent antibody fragment binds. According to specific embodiments, the antigen binding fragment comprises a light chain variable region, a light chain constant region, and an Fd segment of the heavy chain. According to other specific embodiments, the antigen binding fragment comprises Fab and F(ab').

[0104] As used herein, the term "single-chain antibody" refers to a conventional single-chain antibody in the art, which comprises a heavy chain variable region and a light chain variable region connected by a short peptide of about 15 to about 20 amino acids. As used herein, the term "single-domain antibody" refers to a conventional single-domain antibody in the art, which comprises a heavy chain variable region and a heavy chain constant region or only a heavy chain variable region.

[0105] As used herein, the term "human antibody" refers to an antibody produced by a human or an antibody having an amino acid sequence corresponding to an antibody produced by a human prepared using any technique known in the art. [0106] As used herein, the term "humanized antibody" refers to a non-human antibody that has been modified to increase sequence homology with a human antibody, such that the antigen-binding properties of the antibody are retained but its antigenicity in the human body is reduced.

[0107] As used herein, the term "chimeric antibody" refers to an antibody whose amino acid sequence of an immunoglobulin molecule is derived from two or more species. The variable regions of both the light and heavy chains often correspond to the variable regions of an antibody with desired specificity, affinity and capacity derived from one mammalian species (e.g., mouse, rat, rabbit, etc.), while the constant region corresponds to a sequence in an antibody derived from another mammalian species (e.g., human) to avoid eliciting an immune response in that species.

[0108] As used herein, the term "multi-specific antibody" refers to an antibody comprising multiple immunoglobulin variable domain sequences, wherein the first immunoglobulin variable domain sequence in the multiple immunoglobulin variable domain sequences has binding specificity to the first epitope, and the second immunoglobulin variable domain sequence in the multiple immunoglobulin variable domain sequences has binding specificity to the second epitope. In one embodiment, the first epitope and the second epitope do not overlap or do not overlap substantially. In one embodiment, the first epitope and the second epitope are on different antigens such as different proteins (or different subunits of multimeric proteins). In one embodiment, the multi-specific antibody comprises the third, fourth or fifth immunoglobulin variable domain. In one embodiment, the multi-specific antibody is a bispecific antibody molecule, a tri-specific antibody molecule or a tetra-specific antibody molecule.

[0109] As used herein, the term "bispecific antibody" refers to a multispecific antibody that binds no more than two epitopes or two antigens, such as two epitopes or antigens of different genetic targets (e.g., Vβ17, CD69 or CD25). Bispecific antibodies are characterized by having a first immunoglobulin variable domain sequence with binding specificity to a first epitope (e.g., an epitope on a Vβ17 antigen) and a second immunoglobulin variable domain sequence with binding specificity to a second epitope. In one embodiment, the first epitope and the second epitope are on different antigens, such as different proteins (or different subunits of a multimeric protein). [0112] The term "KD" refers to the dissociation constant obtained from the ratio of Kd to Ka (i.e., Kd / Ka) and expressed as a molar concentration (M). According to the present disclosure, the KD value of an antibody can be determined using methods in the art. For example, the KD of an antibody can be determined using surface plasmon resonance, such as by using a biosensor system (e.g., System), or by using bio-layer interferometry technology (such as Octet RED96 system). The smaller the value of antibody KD, the higher the affinity of the antibody for binding to the target antigen.

[0152] The full-length bispecific antibodies provided herein can be generated, for example, using Fab arm exchange (or half-molecule exchange) between two monospecific bivalent antibodies by the following manner: a substitution is introduced at the heavy chain CH3 interface in each half molecule to promote the formation of heterodimers of two antibody half molecules with different specificities in an in vitro cell-free environment or using co-expression. The Fab arm exchange reaction is the result of a disulfide bond isomerization reaction and a CH3 domain dissociation-association. The heavy chain disulfide bonds in the hinge region of the parent monospecific antibody are reduced. The resulting free cysteine of one of the parent monospecific antibodies forms an inter-heavy chain disulfide bond with a cysteine residue of a second parent monospecific antibody molecule, while the CH3 domain of the parent antibody is released and re-formed by dissociation-association. The CH3 domain of the Fab arm can be engineered to promote heterodimerization rather than homodimerization. The resulting product is a bispecific antibody with two Fab arms or half molecules, each of which binds to a different epitope, for example, an epitope on a first anti-target and an epitope on a second target. Other methods of making multispecific antibodies are known and contemplated.

[0153] As used herein, "homodimerization" refers to the interaction of two heavy chains having the same CH3 amino acid sequence. As used herein, "homodimer" refers to an antibody having two heavy chains having the same CH3 amino acid sequence.

[0154] As used herein, "heterodimerization" refers to the interaction of two heavy chains with different CH3 amino acid sequences. As used herein, "heterodimer" refers to an antibody with two heavy chains containing different CH3 amino acid sequences.

[0155] The "knob-in-hole" strategy (see, e.g., PCT Publication WO2006 / 028936) can be used to generate full-length bispecific antibodies. In short, selected amino acids that form the interface of the CH3 domains in human IgG can be mutated at positions that affect the interaction of the CH3 domains, thereby promoting heterodimer formation. Amino acids with small side chains (knobs) are introduced into the heavy chain of an antibody that specifically binds a first antigen, and amino acids with large side chains (knobs) are introduced into the heavy chain of an antibody that specifically binds a second antigen. Upon co-expression of the two antibodies, heterodimers are formed due to the preferential interaction of the heavy chain with the "hole" with the heavy chain with the "knob". Exemplary CH3 substitution pairs that form a knob-in-hole structure (expressed as modified position in the first CH3 domain of the first heavy chain / modified position in the second CH3 domain of the second heavy chain) are: T366Y / F405A, T366W / F405W, F405W / Y407A, T394W / Y407T, T394S / Y407A, T366W / T394S, F405W / T394S, and T366W / T366S_L368A_Y407V.

[0156] Other strategies may also be used, such as using electrostatic interactions to promote heavy chain heterodimerization by replacing positively charged residues on one CH3 surface and negatively charged residues on a second CH3 surface, such as U.S. Patent Publication No. US2010 / 0015133, U.S. Patent Publication No. US2009 / 0182127, U.S. Patent Publication No. US2010 / 028637; or U.S. Patent Publication No. US2011 / 0123532. In other strategies, heterodimerization can be promoted by the following substitutions (represented as modified position in the first CH3 domain of the first heavy chain / modified position in the second CH3 domain of the second heavy chain): L351Y_F405AY407V / T394W, T366I_K392M_T394W / F405A_Y407V, T366L_K392M_T394W / F405A_Y407V, L351Y_Y407A / T366A_K409F, L351Y_Y407A / T366V K409F Y407A / T366A_K409F or T350V_L351Y_F405AY407V / T350V_T366L_K392L_T394W, such as US Patent Publication No. US2012 / 0149876 or US Patent Publication No. US2013 / 0195849.

[0157] In addition to the above methods, the bispecific antibodies provided herein can also be generated in an in vitro cell-free environment by the following means: introducing asymmetric mutations in the CH3 region of two monospecific homodimeric antibodies, and forming bispecific heterodimeric antibodies from two parental monospecific homodimeric antibodies under reducing conditions, thereby publishing antibodies according to international patents. In the method, the first monospecific bivalent antibody and the second monospecific bivalent antibody are engineered to have certain substitutions at the CH3 domain that promote the stability of the heterodimer; these antibodies are incubated together under reducing conditions sufficient to isomerize the disulfide bonds of the cysteine in the hinge region; thereby generating bispecific antibodies by exchanging Fab arms. The incubation conditions can optionally be restored to non-reducing conditions. Exemplary reducing agents that can be used are 2-mercaptoethylamine (2-MEA), dithiothreitol (DTT), dithioerythritol (DTE), glutathione, tris (2-carboxyethyl) phosphine (TCEP), L-cysteine and β-mercaptoethanol, preferably a reducing agent selected from the group consisting of 2-mercaptoethylamine, dithiothreitol and tris (2-carboxyethyl) phosphine. For example, the following conditions can be used: in the presence of at least 25mM 2-MEA or in the presence of at least 0.5mM dithiothreitol, at a pH of 5 to 8, for example, at a pH of 7.0 or at a pH of 7.4, at a temperature of at least 20°C, incubated for at least 90 minutes.

[0167] In some embodiments described herein, the immune effector properties of antibodies provided herein can be enhanced or silenced via Fc modification by techniques known to those skilled in the art. For example, Fc effector functions such as C1q binding, complement dependent cytotoxicity (CDC), antibody dependent cell-mediated cytotoxicity (ADCC), antibody dependent cell-mediated phagocytosis (ADCP), downregulation of cell surface receptors (e.g., B cell receptor; BCR), etc., can be provided and / or controlled by modifying residues in Fc that contribute to these activities.

[0168] "Antibody-dependent cell-mediated cytotoxicity" or "ADCC" refers to a cell-mediated reaction in which nonspecific cytotoxic cells expressing Fc receptors (FcRs), such as natural killer (NK) cells, neutrophils, and macrophages, recognize bound antibody on a target cell and subsequently cause lysis of the target cell.

[0169] The ability of antibodies to induce ADCC can be enhanced by engineering their oligosaccharide components. Most of the glycans in human IgG1 or IgG3 are N-glycosylated at Asn297 in the form of well-known bibranched G0, G0F, G1, G1F, G2 or G2F. Antibodies produced by unengineered CHO cells typically have a glycan fucose content of about at least 85%. Removing core fucose from bibranched complex-type oligosaccharides attached to the Fc region enhances the ADCC of antibodies via improved FcγRIIIa binding without changing antigen binding or CDC activity. Such antibodies can be achieved using different methods reported for the successful expression of relatively highly defucosylated antibodies with bibranched complex-type Fc oligosaccharides, such as controlling the culture osmotic pressure (Konno et al., Cytotechnology 64:249-65, 2012), using the variant CHO cell line Lec13 as a host cell line (Shields et al., J Biol Chem 277:26733-26740, 2002), using the variant CHO cell line EB66 as a host cell line (Olivier et al., MAbs; 2(4), 2010; Epub ahead of print; PMID: 20562582), using the rat hybridoma cell line YB2 / 0 as a host cell line (Shinkawa et al., J Biol Chem 277:26733-26740, 2002), using the variant CHO cell line EB66 as a host cell line (Olivier et al., MAbs; 2(4), 2010; Epub ahead of print; PMID: 20562582), using the rat hybridoma cell line YB2 / 0 as a host cell line (Shinkawa et al., J Biol Chem 278:3466-3473, 2003), introduction of small interfering RNA specific for the α-1,6-fucosyltransferase (FUT8) gene (Mori et al., Biotechnol Bioeng 88:901-908, 2004), or co-expression of β-1,4-N-acetylglucosaminyltransferase III and Golgi α-mannosidase II or the potent α-mannosidase I inhibitor, kifnosine (Ferrara et al., J Biol Chem 281:5032-5036, 2006, Ferrara et al., Biotechnol Bioeng 93:851-861, 2006; Xhou et al., Biotechnol Bioeng 99:652-65, 2008).

[0170] In some embodiments described herein, ADCC initiated by antibodies provided herein can also be enhanced by certain substitutions in antibody Fc. Exemplary substitutions are, for example, substitutions at amino acid positions 256, 290, 298, 312, 356, 330, 333, 334, 360, 378 or 430 (residues are numbered according to the EU index), as described in U.S. Patent No. 6,737,056.

[0370] Example

[0371] Example 1: Antibodies that bind to Vβ17

[0372] 1.1: Identification of Vβ17 monoclonal

[0373] A cell-based approach was used to identify Vβ17-specific monoclones.

[0374] Mice were tolerized with NS0-WT cells during the neonatal period and then boosted three times per week with the NS0 cell line expressing TCRVα10.2*Vβ17.2.

[0375] High titers were evaluated by examining specificity for Vβ17.2 using three variants of the SKW3 cell line [SKW3-1 (Vα10*Vβ17), SKW3-2 (Vα12*Vβ17), SKW3-3 (Vα10*Vβ7 / 9)].

[0376] like Figure 1 As shown, several monoclonal antibodies binding to Vβ17 were identified by flow cytometry using different cell lines and primary cells. Anti-TCR Vβ17-PE Ab, Beckman-Colter, Cat: IM2048 was used as a positive control. Goat anti-mouse IgG PE, Southern Biotech, Cat: 1030-09, E2518-RJ69Y was used as a negative control.

[0377] Example 2: Evaluation of Vβ17 Antibody Properties

[0378] 2.1: Proliferation and activation

[0379] To test the proliferation of Vβ17+ cells, 96-well ELISA plates were coated with 10ug / ml of Vβ17 antibody or control antibody (B17B21) and incubated for 24 hours. PBMCs were thawed and labeled with cell tracer purple to track proliferation. 0.5MPBMC cells / well / 200ul of 10% RPMI medium (without IL-2) were cultured in a CO2 incubator for 24 hours. Cells were transferred from 96 wells to 48-well culture plates containing 200μl of 10% RPMI medium (without IL-2). The proliferation and activation status of CTV dilutions were examined using markers CD25, CD69, and CD71.

[0380] like Figure 2 As shown, single clones from 7 parents showed good agonistic properties. 26 single clones with good agonistic properties were selected for purification.

[0381] Differential proliferation kinetics were observed for 26 purified antibodies. Figure 3 As shown, single clones from parents 28B2, 28C9 and 28D2 showed good proliferation and were used for further assays.

[0382] Differential CD71 upregulation was observed for 26 purified antibodies. Figure 4 As shown, monoclones from parental 28B2, 28C9 and 28D2 showed good CD71 upregulation and were used for further assays.

[0383] 2.2: Vβ17 binding

[0384] The 10 best agonistic mAbs binding to Vβ17 were evaluated using variants of the SKW3 cell line. Figure 5 As shown, 10 purified antibodies belonging to 3 different parents bind well to SKW3-1 and SKW3-2 cell lines, but not to SKW3-3 cells that do not express Vβ17. Purified antibodies from the 14H7 parent did not bind well to the SKW3-2 cell line and were included as a negative control.

[0385] The binding of 10 selected purified monoclonal clones to PBMCs was evaluated. Figure 6 As shown, 10 purified antibodies belonging to 3 different parentages bound well to T cells on PBMCs.

[0386] like Figure 7 As shown, these 10 candidate monoclonals differ in isotype based on their heterozygous parents. Purified monoclonal antibodies from 3 different parents have different isotypes: 28D2 has IgG1, 28B2 has IgG2a, and 28C9 has IgG2b.

[0387] 2.3: VB17 antibody internalization

[0388] Internalization assay was used to screen 10 candidate antibodies. MMAF and antibody were added to 5000 cells / well / 50μl, 50nM concentration per well, Ab control, MMAF control and 3-fold dilution until point 7. MMAF antibody was incubated with cells for 20 minutes and the assay was repeated. The plate was incubated at 37°C for 96 hours and Cell Titre Glo was added to the plate. Luminescence was read in TECANSPARK and viability was calculated using the formula MMAF+Ab / MMAF X 100%. Data were plotted in GraphPad Prism.

[0389] like Figure 8 As shown, monoclones from the 28B2 parent showed good internalization. Monoclones from the parent 28C9, which had very good agonist properties, showed poor internalization. Monoclones from the parents 28D2 and 14H7 showed no internalization.

[0390] Example 3: Epitope binning of Vβ17 monoclonal

[0391] Epitope classification helps to separate the final monoclonals into individual epitope recognition groups and antibodies that recognize epitopes that are different from those of commercial antibodies. The distinction in epitope recognition is supported by the diversity of V gene sequences.

[0392] like Figure 9A-9C As shown, the epitope classification matrix of 10 candidate antibodies using OctetRED96 shows 3 separate specificities. Antibody 1 was loaded onto an anti-mouse IgG Fc capture (AMC) sensor, then allowed to bind to the antigen (TCRVα10.2*Vβ17 huFc), and then they were run on the antibody 2 panel. Purified monoclonal antibodies from 3 different parents were classified into 3 different groups.

[0393] In addition, 10 candidate antibodies were classified differently compared to a commercial positive control antibody as assessed using Octet RED96. Figure 10A-10C As shown, the positive control antibody TCRVb17 PE (Beckman-colter, catalog number: IM2048) was classified differently from the 10 candidate antibodies.

[0394] like Fig.11 As shown, monoclonal clones from the same parent have the same VH sequence. The heavy chain sequences of monoclonal clones belonging to the three different hybrids are different from each other.

[0395] like Fig.12 As shown, monoclonal clones from the same parent have identical VK sequences. The light chain sequences of monoclonal clones belonging to the three different hybrids are different from each other.

[0396] Example 4: Further characterization of other VB17 antibodies

[0397] like Fig.13 As shown, 12 additional antibodies were tested for proliferation and activation curves. These 12 additional antibodies had moderate agonistic properties and good PBMC binding and were used for epitope sorting and cell-based competition assays.

[0398] like Figure 14A-Figure 14C As shown in , monoclonal clones from one hybrid parent 23A9 were classified similarly to the positive control antibody (TCRVb17-PE).

[0399] like Figure 15A-Figure 15C As shown, monoclonals from 23A9 and commercial antibodies were classified as similar to each other but different from monoclonals from other hybrid parents.

[0400] Example 5: Cell-based competition assay for Vβ17 antibodies

[0401] Cells were treated with live / dead dye for 15 minutes at 4°C and then plated in 96-well plates. Cells were treated with different concentrations of humanized Vb17 antibody (B17B633.001) and incubated for 40 minutes at 4°C. Cells were washed and incubated with 20ug / ml purified mouse Vb17 antibody for 40 minutes at 4°C. Cells were stained with secondary antibodies, washed and read on Novocyte. Fig.16 As shown in , most monoclonals did not compete with the commercial antibodies for binding to cells expressing Vβ17.
