Description

Technical Field

[0001] The present application relates to the technical field of organic compound synthesis, and more specifically, to a preparation method of a fully substituted 5,5'-bioxazole compound and its application. Background Art

[0002] It is highly desirable to develop efficient atomic and step-economic strategies to synthesize biaryl heterocyclic skeletons that are widely present in the field of bioactive molecules and optoelectronic materials. In recent years, people have developed transition metal-catalyzed methods to expand the synthetic molecular library of bioxazole compounds. Most of the classical synthetic methods are based on the coupling reaction of heteroaryl halides and organometallic reagents. This type of method can prepare a variety of heterodiaryls and exhibits high levels of activity for the coupling of heteroaryl chlorides and large sterically hindered aryls and heteroaryl halides (J.Am.Chem.Soc., 2007, 129, 3358-3366). However, the synthesis of bioxazole compounds catalyzed by transition metals has the following challenges:

[0003] 1) The substrates often need to be prepared in advance, and the reaction produces stoichiometric byproducts that are environmentally unfriendly, resulting in poor atom economy;

[0004] 2) When electron-poor aromatic rings are selected as substrates for aromatic coupling to obtain azole compounds, their substrate compatibility is poor and their chemical selectivity is poor and difficult to control;

[0005] 3) Using stoichiometric strong base as an additive pollutes the environment and does not conform to the concept of green chemistry.

[0006] In addition, people have also developed strategies to use existing heterocyclic skeletons to achieve oxidative coupling through CH bond activation to synthesize heterocyclic compounds, which has better step economy and atom economy. For example, in 2011, Ofial reported a method for selective CC coupling of benzoazoles and azoles by breaking two CH bonds at the C2 position, synthesizing a class of asymmetric 2,2'-bisheteroaryl compounds that have not been widely explored (Angew. Chem. Int. Ed. 2011, 50, 2178-2182); in 2012, You Jinsong's research group used two structurally similar non-benzo-fused cyclic azoles to selectively perform oxidative cross-coupling (Chem. Eur. J. 2012, 18, 6158-6162.), but there are still limitations, that is, the substrate starts with a heterocyclic compound, only one cross-dehydrogenation coupling reaction is achieved, the functional group compatibility is poor, and it is difficult to carry out later modification.

[0007] Although many natural products with important biological activities contain directly connected azole compounds, the 2,2'-connection of azole compounds is already a rare skeleton structure in nature. The 5,5'-connection of azole compounds is even less reported. The synthesis of the rare 5,5'-bioxazole compounds is obtained by direct carbon-carbon coupling of two molecules of oxazole compounds (Chin. Chem. Lett., 2021, 32, 425-428). In 2014, Zhao Yu's research group only reported one case of using oxalate and isocyanoacetate to synthesize 5,5'-bioxazole substituted with methyl formate at the 4th position under silver catalysis (Angew. Chem. Int. Ed., 2014, 53, 5435-5439). It should be pointed out that the one-step synthesis of fully substituted 5,5'-bioxazoles through a simple raw material direct multiple dehydrogenation strategy has not been reported.

[0008] On the other hand, enamines are versatile reaction intermediates for the synthesis of nitrogen-containing skeletons due to their intrinsic nucleophilicity. They can participate in a series of selective functionalization reactions. The directed CH functionalization of olefins by enamines has become an effective synthetic strategy for the preparation of various polysubstituted amines and olefin derivatives. In 2015, Wang Jian's research group reported a ruthenium-catalyzed [4+2] cycloaddition reaction of enamides and alkynes, and published a mild and economical scheme for the construction of polysubstituted pyridines (J.Am.Chem.Soc.2015,137,9489-9496), where enamines act as four-atom synthons.

[0009] In 2019, Teck-Peng Loh reported a practical method for synthesizing N-acyl enamine amides by iron-catalyzed oxidative coupling of enamides with formamide (ACS Catal. 2019, 9, 8128-8135). The products can be further converted into important biologically active molecules such as pyrimidine-4-ones and 4-hydroxypyridin-2-ones. Here, enamines serve as C1 synthons.

[0010] Application Contents

[0011] In order to overcome at least one problem existing in the above-mentioned prior art, the present application provides a method for preparing a fully substituted 5,5'-bioxazole compound: with the assistance of an easily available and traceless weakly coordinating directing group, 1,1-multiple dehydrogenation difunctionalization of olefins under metal catalysis is achieved to construct a polycyclic aromatic heterocyclic molecular skeleton.

[0012] In order to solve the above technical problems, the technical solution adopted in this application is:

The reaction equation is as follows:

[0014]

[0015] Wherein Ar is an o-, m- or p-substituted aromatic compound, and R is an alkyl group or a phenyl group.

[0016] Preferably, the inert solvent is any one or more of toluene, tetrahydrofuran, 1,4-dioxane, ethylene glycol dimethyl ether, N,N'-dimethylacetamide, N-methylpyrrolidone, dimethyl sulfoxide, acetonitrile, 1,2-dichloroethane, ethanol, and water.

[0017] Preferably, the metal catalyst is any one of pentamethylcyclopentadienylrhodium chloride dimer, pentamethylcyclopentadienyliridium chloride dimer, palladium acetate, and dichloro(p-methylisopropylphenyl)ruthenium dimer, or a combination thereof.

[0019] Preferably, the oxidant is any one or more of silver acetate, silver carbonate, silver oxide, copper acetate, and potassium persulfate.

[0020] Preferably, the additive is any one of lithium acetate and lithium carbonate or a combination thereof.

[0021] Preferably, the amount of the metal catalyst used is 2 mol% of the amount of the N-(1-aryl vinyl)amide compound represented by formula II.

[0023] The preparation method of the fully substituted 5,5'-bioxazole compound in some preferred embodiments of the present application comprises the following specific steps:

[0024] S1: In a reactor, 1.6 mg of pentamethylcyclopentadiene iridium dichloride dimer, 1.9 mg of silver trifluoromethanesulfonimide, 55.2 mg of silver carbonate, 26.4 mg of lithium acetate, 1.0 mL of solvent toluene, and 32.2 mg of N-(1-arylvinyl)acetamide were added in sequence in air;

[0025] S2: reacting the reaction solution at 140°C for 12 hours;

[0026] S3: After the reaction is completed, the mixture is separated by column chromatography to obtain the target compound.

[0027] The present application also provides the use of the fully substituted 5,5'-bioxazole compound prepared by the above preparation method in luminescent materials.

[0028] The present invention realizes multiple dehydrogenation 1,1 difunctionalization of olefins under the conditions of inert solvents by co-promoting metal catalysts and silver salt oxidants to synthesize fully substituted 5,5'-bioxazole compounds. Specifically, firstly, N-(1-aryl vinyl)amide) is used as an easily available and easily converted raw material, and amide is used as a weakly coordinated traceless directing group to realize the C( sp 2 )-H activation, and then the amide undergoes enol tautomerism and intramolecular oxidative cyclization, thereby constructing carbon-carbon bonds and carbon-hetero bonds to obtain fully substituted 5,5'-bioxazole compound products. This method has high atom economy (the product is obtained by cross-dehydrogenation coupling, and the transformation removes 4 CH bonds and 2 NH bonds) and step economy (no need to pre-halogenate or metallate the raw materials), and the substrate has a wide range of applicability. The easily available N-(1-aryl vinyl) amide is used as a reaction substrate and participates in the construction of nitrogen-containing fused heterocycles as a traceless directing group.

[0101] 2. Application Example 1

[0102] The molecules were dissolved in dichloromethane (1.0×10 -5 mol L -1) in the ultraviolet / visible absorption spectrum (UV) and photoluminescence (PL) emission spectrum ( Figure 3 ). Maximum absorption wavelength (λ max ) appears at 287nm, which is due to the π→π of the conjugated double bond. * Transition; photoluminescence spectrum was obtained by excitation at the maximum absorption wavelength. 1h showed blue PL emission at 509 nm in dichloromethane. Subsequently, the fluorescence emission of molecule 1j in different solvents was tested ( Figure 4 ), such as benzonitrile (ACN), dimethyl sulfoxide (DMSO), N,N-dimethylformamide (DMF), tetrahydrofuran (THF), toluene (Tol). There is a certain interaction between the molecule 1h and the solvent, which causes its fluorescence emission to have different degrees of red shift or blue shift in different solvents. Then, in toluene solvent (1.0×10 -5 mol L -1 ) was tested for 1h and the fluorescence quantum yield (QY) was 83.14% ( Figure 5 ). This indicates that 1h has good application prospects in luminescent materials.

[0103] The present invention realizes multiple dehydrogenation 1,1 difunctionalization of olefins under the conditions of inert solvents by co-promoting metal catalysts and silver salt oxidants to synthesize fully substituted 5,5'-bioxazole compounds.

[0104] In summary, this application reports the first transition metal-catalyzed simple construction of a fully substituted 5,5'-oxazole compound skeleton based on a multiple dehydrogenation cyclization strategy. The substrates for this transformation have a wide range of applications and strong functional group tolerance. The applicant also achieved the modification of drug molecules such as probenecid. Furthermore, we conducted a fluorescence test on the obtained 5,5'-oxazole molecule, and its fluorescence quantum yield reached 83.14%, indicating that this type of new skeleton has great application potential in luminescent materials.
