Abstract
Description
Technical Field
[0001] The present invention belongs to the field of bioengineering. The present invention specifically relates to an RNA capping enzyme and a mutant thereof, a nucleic acid sequence encoding the capping enzyme and / or its mutant, an expression construct or a host cell comprising the nucleic acid sequence. The present invention also relates to a composition comprising the capping enzyme and a method and a reaction system for capping RNA using the capping enzyme, its mutant or its composition. Background Art
[0002] After transcription in eukaryotic organisms, mRNA needs to be capped at the 5' end. The cap structure is mainly an inverted N7-methylated guanosine added to the RNA head, which is connected to the first nucleotide of RNA through a triphosphate bond. The cap structure plays an important role in the stability, nuclear export and translation of mRNA, and can reduce its immunogenicity in cells. In mRNA therapy and mRNA vaccine technology, capping of exogenous mRNA is very important for its effective expression in mammalian cells.
[0003] Capping enzyme is an effective enzyme that can catalyze the formation of a cap structure, thereby improving the capping effect of mRNA. Capping enzyme plays a very important role in the preparation of mRNA vaccines, and can improve the effectiveness and safety of vaccines. At present, the main capping enzyme used in industrial production processes such as mRNA vaccines is the Vaccinia Capping Enzyme, but it is composed of two subunits, D1 and D12, with low expression and purification efficiency and high price, which limits its application in the industrial production of mRNA vaccines. At the same time, most of the previous patents were optimized for poxvirus capping enzymes, such as mutations or the establishment of fusion proteins, and the sequence diversity was relatively single. To this end, exploring and mining more capping enzymes with higher capping efficiency, compact and complete functions, and easier scale and industrial production is of great significance for mRNA vaccines, gene therapy, etc.
[0004] In order to successfully express in host cells, many viruses have their own RNA 5' triphosphatase (RT), guanylate transferase (GT) and RNA guanine-N7 methyltransferase (RNMT). These enzymes are different in source, mechanism of action and sequence, and their activities are also uneven. Therefore, it is urgent to obtain a variety of capping enzymes from natural sequences in this field, and to obtain capping enzymes with higher capping activity, more stable capping effect, higher capping rate, and more conducive to large-scale and industrial production through mutation and screening. For this reason, the technical solution of the present invention is proposed. Summary of the invention

[0162] 1. Definition
[0163] Capping enzyme: mRNA in eukaryotic organisms is modified after transcription to form a special structure at the 5' end, namely the cap structure, which plays an important role in the stability, transport and translation of mRNA, and can reduce its immunogenicity in cells. Capping enzyme is an effective enzyme that catalyzes the formation of the cap structure. It has the activities of RNA 5' triphosphatase (TPase / RT), guanylate transferase (GTase / GT) and RNA guanine-N7 methyltransferase (MTase / RNMT), and can connect the 7-methylguanine cap structure (m7Gppp) to the 5' end of RNA.
[0164] Isolated or purified: The terms "isolated" or "purified" mean a polypeptide or polynucleotide that is removed from at least one component with which it is naturally associated. For example, a polypeptide can be at least 1% pure, e.g., at least 5% pure, at least 10% pure, at least 20% pure, at least 40% pure, at least 60% pure, at least 80% pure, at least 90% pure, or at least 95% pure, as determined by SDS-PAGE, while a polynucleotide can be at least 1% pure, e.g., at least 5% pure, at least 10% pure, at least 20% pure, at least 40% pure, at least 60% pure, at least 80% pure, at least 90% pure, or at least 95% pure, as determined by agarose electrophoresis.
[0165] Sequence identity: The parameter "sequence identity" describes the relatedness between two amino acid sequences or between two nucleotide sequences.
[0166] For the purposes of the present invention, the degree of sequence identity between two amino acid sequences can be determined using the Clustal Omega algorithm (Sievers F, et al. (2011). Molecular Systems Biology 7:539) as implemented in the online align tool (https: / / www.uniprot.org / align) of the UniProt website, preferably version 1.2.4 or higher. The optional parameter used is iteration 0. The percent identity is obtained using the Percent Identity Matrix output by the program, which is calculated as follows:
[0167] (same residues x 100) / (alignment length - total number of gaps in the alignment)
[0168] For the present invention, the degree of sequence identity between two nucleotide sequences can be determined using the Clustal Omega algorithm (Sievers F, et al. (2011). Molecular Systems Biology 7:539) as implemented in the online align tool (https: / / www.ebi.ac.uk / Tools / msa / clustalo / ) of the EBI website, preferably version 1.2.4 or higher. Default parameters are used. The percent identity is obtained using the Percent Identity Matrix output by the program, which is calculated as follows:
[0169] (same deoxyribonucleotides x 100) / (alignment length - total number of gaps in the alignment)
[0170] For the present invention, the sequence identity degree between two amino acid sequences can be measured using the Needle program as in the EMBOSS software package (EMBOSS:The European Molecular Biology Open Software Suite, Rice et al., 2000, Trends Genet. 16:276-277), preferably the Needleman-Wunsch algorithm (Needleman and Wunsch, 1970, J. Mol. Biol. 48:443-453) performed in version 3.0.0 or higher. The optional parameters used are gap penalty (gap penalty) 10, gap extension penalty (gap extension penalty) 0.5 and EBLOSUM62 (EMBOSS version of BLOSUM62) substitution matrix. Use the output result (obtained using the -nobrief option) marked as "highest identity (longest identity)" using Needle as the identity percentage, and calculate as follows:
[0171] (same residues x 100) / (alignment length - total number of gaps in the alignment)
[0172] For the present invention, the sequence identity degree between two nucleotide sequences can be measured using the Needle program as in the EMBOSS software package (EMBOSS:The European Molecular Biology Open Software Suite, Rice etc., 2000, see above), preferably the Needleman-Wunsch algorithm (Needleman and Wunsch, 1970, see above) performed in version 3.0.0 or higher. The optional parameters used are gap penalty 10, gap extension penalty 0.5 and EDNAFULL (EMBOSS version of NCBI NUC4.4) substitution matrix. Use the output result (obtained using the -nobrief option) marked as "highest identity" using Needle as identity percentage, and calculate as follows:
[0173] (same deoxyribonucleotides x 100) / (alignment length - total number of gaps in the alignment)
[0174] Coding sequence: The term "coding sequence" means a polynucleotide that directly specifies the amino acid sequence of a polypeptide. The boundaries of the coding sequence are usually determined by an open reading frame, which usually begins with the ATG start codon or alternative start codons such as GTG and TTG, and ends with a stop codon such as TAA, TAG and TGA. The coding sequence can be a DNA, cDNA, synthetic or recombinant polynucleotide.
[0175] Nucleic acid construct: The term "nucleic acid construct" means a single-stranded or double-stranded nucleic acid molecule that is isolated from a naturally occurring gene, or that is modified in a manner that does not otherwise exist in nature to contain a segment of nucleic acid, or that is synthetic. When the nucleic acid construct contains the regulatory sequences required for expression of the coding sequence of the present invention, the term nucleic acid construct is synonymous with the term "expression cassette".
[0176] Control sequence: The term "control sequence" is meant to include all components that are necessary or advantageous for the expression of a polynucleotide encoding a polypeptide. Each control sequence may be native or foreign to the polynucleotide encoding the polypeptide, or each control sequence may be native or foreign to each other. These control sequences include, but are not limited to, a leader sequence, a polyadenylation sequence, a propeptide sequence, a promoter, a signal peptide sequence, and a transcription terminator. At a minimum, a control sequence includes a promoter and termination signals for transcription and translation. The control sequence may be provided together with a linker for introducing specific restriction sites that facilitate the connection of the control sequence to the coding region of the polynucleotide encoding the polypeptide.
[0177] Operably linked: The term "operably linked" means a configuration in which a control sequence is placed at an appropriate position relative to the coding sequence of a polynucleotide such that the control sequence directs the expression of the coding sequence.
[0178] Expression: The term "expression" includes any step involved in the production of the polypeptide including, but not limited to, transcription, post-transcriptional modification, translation, post-translational modification, and secretion.
[0179] Expression vector: The term "expression vector" means a linear or circular DNA molecule comprising a polynucleotide encoding a polypeptide operably linked to additional nucleotides that provide for its expression.
[0180] Host cell: The term "host cell" means any cell type that is susceptible to transformation, transfection, transduction, etc. using a nucleic acid construct comprising a polynucleotide of the present invention. [0196] In one aspect, the present invention provides a nucleic acid construct comprising the nucleic acid sequence of the present invention. In one embodiment, the nucleic acid construct of the present invention may further comprise elements or regulatory sequences required for expressing the nucleic acid sequence.
[0197] In one embodiment, nucleic acid construct of the present invention can be constructed as follows: the nucleotide sequence is operably connected to one or more (several) regulating and controlling sequences, and the regulating and controlling sequences instruct the expression of the nucleotide sequence under the conditions compatible with the regulating and controlling sequences in a suitable host cell. Depending on the type of nucleic acid construct, it may be desirable or necessary to operate the nucleotide sequence before the nucleotide sequence is inserted into a plasmid or other vector. The technology of using recombinant DNA methods to modify the nucleotide sequence is known in the art.
[0198] In one embodiment, the regulatory sequence may be a suitable promoter sequence, which is a nucleotide sequence recognized by a host cell for expressing a polynucleotide encoding a capping enzyme. The promoter sequence contains a transcriptional regulatory sequence that mediates the expression of the capping enzyme. The promoter may be any nucleotide sequence that shows transcriptional activity in the selected host cell, including mutant, truncated and hybrid promoters, and may be obtained from a gene encoding an extracellular or intracellular capping enzyme that is homologous or heterologous to the host cell.

Other useful promoters for yeast host cells include, for example, a xylose-inducible promoter (regulated by the XylR transcription factor derived from Bacillus licheniformis) and promoters described by Romanos et al., 1992, Yeast 8:423-488 and Chen et al., 2020, Nature Microbiology 5:1349-1360.
[0202] The promoters used to direct the expression of the nucleic acid construct of the present invention in a mammalian host include, but are not limited to, a strong mammalian expression promoter derived from human cytomegalovirus (CMV), a strong mammalian expression promoter derived from human elongation factor 1α (EF1α), a mammalian expression promoter derived from simian vacuolating virus 40 (SV40), a mammalian promoter derived from phosphoglycerate kinase gene (PGK1), and the like.
[0203] In one embodiment, the regulatory sequence may be a suitable transcription terminator sequence, which is a sequence recognized by the host cell to terminate transcription. The terminator sequence is operably connected to the 3' end of the nucleotide sequence encoding the capping enzyme. Any terminator that is functional in the selected host cell can be used in the present invention. For filamentous fungal host cells, preferred terminators can be obtained from the genes of the following enzymes: Aspergillus nidulans anthranilate synthase, Aspergillus niger glucoamylase, Aspergillus niger α-glucosidase, Aspergillus oryzae TAKA amylase and Fusarium oxysporum trypsin-like protease. For yeast host cells, preferred terminators can be obtained from the genes of the following enzymes: Saccharomyces cerevisiae enolase, Saccharomyces cerevisiae cytochrome C (CYC1) and Saccharomyces cerevisiae glyceraldehyde-3-phosphate dehydrogenase. For yeast host cells, other useful terminators are described by Romanos et al., 1992 and Chen et al., 2020.
[0204] In one embodiment, the regulatory sequence may be a suitable leader sequence, which is an mRNA non-translated region important for the translation of the host cell. The leader sequence may be operably linked to the 5'-end of the nucleotide sequence encoding the capping enzyme. Any leader sequence that is functional in the selected host cell may be used in the present invention. Preferred leaders for filamentous fungal host cells are obtained from the genes of the following enzymes: Aspergillus oryzae TAKA amylase and Aspergillus nidulans triose phosphate isomerase. Suitable leaders for yeast host cells are obtained from the genes of the following enzymes: Saccharomyces cerevisiae enolase (ENO-1), Saccharomyces cerevisiae 3-phosphoglycerate kinase, Saccharomyces cerevisiae alpha factor, and Saccharomyces cerevisiae alcohol dehydrogenase / glyceraldehyde-3-phosphate dehydrogenase (ADH2 / GAP).
[0205] In one embodiment, the regulatory sequence may be a polyadenylation sequence, which is a sequence operably connected to the 3' end of the nucleotide sequence, and when transcribed, the host cell recognizes it as a signal for adding polyadenosine residues to the transcribed mRNA. Any polyadenylation sequence that is functional in the selected host cell can be used in the present invention. For filamentous fungal host cells, preferred polyadenylation sequences are obtained from the genes of the following enzymes: Aspergillus oryzae TAKA amylase, Aspergillus niger glucoamylase, Aspergillus nidulans anthranilate synthase, Fusarium oxysporum trypsin-like protease and Aspergillus niger α-glucosidase. For yeast host cells, useful polyadenylation sequences are described by Guo and Sherman, 1995, Molecular Cellular Biology 15: 5983-5990. For mammalian host cells, useful polyadenylation sequences are described by Qin et al., 2023, Nature Communications 14: 1500.
[0206] In one embodiment, the regulatory sequence may be a signal peptide coding region, which encodes a signal peptide connected to the N-terminus of the capping enzyme and directs the capping enzyme to enter the cell secretory pathway. The 5' end of the coding sequence of the polynucleotide may inherently contain a signal peptide coding sequence, which is naturally connected in the translation reading frame together with the coding sequence segment encoding the capping enzyme. Alternatively, the 5' end of the coding sequence may contain a signal peptide coding sequence that is exogenous to the coding sequence. An exogenous signal peptide coding sequence may be necessary when the coding sequence does not naturally contain a signal peptide coding sequence. Alternatively, an exogenous signal peptide coding sequence may simply replace the natural signal peptide coding sequence to enhance the secretion of the capping enzyme. However, any signal peptide coding sequence that directs the expressed capping enzyme to enter the secretory pathway of the selected host cell may be used. Signal peptide coding sequences useful for bacterial host cells are those obtained from the genes for Bacillus sp. NCIB 11837 maltogenic amylase, Bacillus licheniformis subtilisin, Bacillus licheniformis β-lactamase, Bacillus stearothermophilus α-amylase, Bacillus stearothermophilus neutral proteinase (nprT, nprS, nprM), and Bacillus subtilis prsA. Additional signal peptides are described by Simonen and Palva, 1993, Microbiological Reviews 57: 109-137. Signal peptide coding sequences useful for filamentous fungal host cells are those obtained from the genes for Aspergillus niger neutral amylase, Aspergillus niger glucoamylase, Aspergillus oryzae TAKA amylase, Humicola insolens cellulase, Humicola insolens endoglucanase V, Humicola lanuginosa lipase, and Rhizomucor manhei aspartic proteinase. Useful signal peptides for yeast host cells are obtained from the genes for Saccharomyces cerevisiae alpha factor and Saccharomyces cerevisiae invertase. Other useful signal peptide coding sequences are described by Romanos et al., 1992, supra.
[0207] In one embodiment, the regulatory sequence may be a propeptide coding sequence encoding a propolypeptide located at the N-terminus of the capping enzyme. The resulting polypeptide is referred to as a proenzyme or propolypeptide (or in some cases a zymogen). The propolypeptide is generally inactive and can be converted from the propolypeptide to an active polypeptide by catalytic or autocatalytic cleavage of the propeptide. The propeptide coding sequence can be obtained from the genes for Bacillus subtilis alkaline protease (aprE), Bacillus subtilis neutral protease (nprT), Myceliophthora thermophila laccase (WO 95 / 33836), Rhizomucor manhei aspartic protease, and Saccharomyces cerevisiae alpha factor.
[0208] In one embodiment, when both the signal peptide and propeptide sequences are present at the N-terminus of the capping enzyme, the propeptide sequence is placed next to the capping enzyme N-terminus and the signal peptide sequence is placed next to the amino terminus of the propeptide sequence.
[0209] In one embodiment, it is also desirable to add a regulatory sequence that allows the expression of the capping enzyme to be regulated relative to the growth of the host cell. Examples of regulatory systems are those systems that cause gene expression to respond to chemical or physical stimuli, including those systems that are turned on or off by the presence of regulatory compounds. Regulatory systems in prokaryotic systems include lac, tac, and trp operator systems. In yeast, the ADH2 system or the GAL1 system and the XYL system can be used. In filamentous fungi, Aspergillus niger glucoamylase promoter, Aspergillus oryzae TAKA α-amylase promoter, and Aspergillus oryzae glucoamylase promoter can be used as regulatory sequences. In mammalian cells, constitutive promoters including EF1α, CMV, and SV40 and other induction systems can be used for regulation. Other examples of regulatory sequences are those that allow gene amplification. In eukaryotic systems, these regulatory sequences include dihydrofolate reductase genes amplified in the presence of methotrexate, and metallothionein genes amplified with heavy metals. In these cases, the polynucleotide encoding the capping enzyme is operably connected to the regulatory sequence.
[0210] In one embodiment, the nucleic acid construct of the present invention may further comprise one or more (several) convenient restriction sites to allow insertion or substitution of the isolated polynucleotide encoding the capping enzyme at these sites. In one embodiment, the nucleic acid sequence may be expressed by inserting a nucleotide sequence or nucleic acid construct comprising the sequence into an appropriate vector for expression. In the process of preparing the nucleic acid construct, the coding sequence is placed in the vector so that the coding sequence is operably linked to an appropriate regulatory sequence for expression.
[0211] In one embodiment, the vector for expression can be any vector (for example, plasmid or virus), which can easily carry out the recombinant DNA step, and can produce the expression of the nucleic acid sequence. The selection of the vector will usually depend on the compatibility of the vector with the host cell into which the vector will be introduced. The vector can be a linear or closed circular plasmid. In one embodiment, the vector can be an autonomously replicating vector, that is, a vector existing as an extrachromosomal entity (entity), and its replication is independent of chromosomal replication, for example, a plasmid, an extrachromosomal element, a minichromosome (minichromosome) or an artificial chromosome. In one embodiment, the vector can contain any feature for ensuring self-replication. In one embodiment, the vector can be a vector that is integrated into the genome and replicated together with the chromosome into which the vector is integrated when introduced into the host cell. In one embodiment, a single vector or plasmid or two or more vectors or plasmids can be used, which contain the complete DNA (total DNA) to be introduced into the host cell genome together, or a transposon (transposon) can be used.
[0212] In one embodiment, the vector preferably contains one or more (several) selective markers, which allow simple selection of cells transformed, transfected, transduced, etc. Selectable markers are genes whose products provide biocide or virus resistance, resistance to heavy metals, prototrophy to auxotrophs, etc. In one embodiment, examples of bacterial selective markers are dal genes from Bacillus subtilis or Bacillus licheniformis, or markers that confer antibiotic resistance. In one embodiment, the resistance gene is selected from the group consisting of ampicillin resistance gene, kanamycin resistance gene, chloramphenicol resistance gene, spectinomycin resistance gene, tetracycline resistance gene, bleomycin resistance gene, streptomycin resistance gene, hygromycin resistance gene, gentamicin resistance gene, apramycin sulfate resistance gene, erythromycin resistance gene, nourseoinhibitor resistance gene, geneticin resistance gene, and puromycin resistance gene. In one embodiment, the resistance gene is selected from the group consisting of an ampicillin resistance gene, a kanamycin resistance gene, a chloramphenicol resistance gene, or a tetracycline resistance gene. Suitable markers for yeast host cells are ADE2, HIS3, LEU2, LYS2, MET3, TRP1, and URA3. Selectable markers for filamentous fungal host cells include, but are not limited to, amdS (acetamidase), argB (ornithine carbamoyltransferase), bar (phosphinothricin acetyltransferase), hph (hygromycin phosphotransferase), niaD (nitrate reductase), pyrG (orotidine-5'-phosphate decarboxylase), sC (sulfate adenylyltransferase), and trpC (anthranilate synthase), and their equivalents. Fluorescent markers such as mCherry are included for mammalian cells. Preferred for use in Aspergillus cells are the amdS and pyrG genes of Aspergillus nidulans or Aspergillus oryzae and the bar gene of Streptomyces hygroscopicus.
[0213] In one embodiment, the vector preferably contains elements that allow the vector to be integrated into the host cell genome or the vector to replicate autonomously in the cell independently of the genome. In order to be integrated into the host cell genome, the vector can rely on the sequence of the polynucleotide encoding the capping enzyme or any other vector element for integration into the genome by homologous or non-homologous recombination. Alternatively, the vector may contain additional nucleotide sequences for guiding the precise position of integration into the host cell genome chromosome by homologous recombination. In order to increase the possibility of integration at a precise position, the integration element should preferably contain a sufficient number of nucleic acids, such as 100 to 10,000 base pairs, preferably 400 to 10,000 base pairs, and most preferably 800 to 10,000 base pairs, which have a high degree of identity with the corresponding target sequence to enhance the probability of homologous recombination. The integration element can be any sequence, which is homologous to the target sequence in the host cell genome. In addition, the integration element can be a non-coding or encoding nucleotide sequence. On the other hand, the vector can be integrated into the genome of the host cell by non-homologous recombination.
[0214] In one embodiment, for autonomous replication, the vector may further comprise an origin of replication that enables the vector to replicate autonomously in the host cell. The origin of replication may be any plasmid replicator that mediates autonomous replication, which functions in the cell. The term "origin of replication" or "plasmid replicator" is defined herein as a nucleotide sequence that enables a plasmid or vector to replicate in vivo. Examples of origins of replication used in bacterial host cells are the origins of replication of plasmids pBR322, pUC19, pACYC177, and pACYC184 that allow replication in Escherichia coli, and the origins of replication of plasmids pUB110, pE194, pTA1060, and pAMβ1 that allow replication in Bacillus. Examples of origins of replication used in yeast host cells are the 2 micron origin of replication, ARS1, ARS4, a combination of ARS1 and CEN3, and a combination of ARS4 and CEN6. Examples of origins of replication for use in filamentous fungal host cells are AMA1 and ANS1 (Gems et al., 1991, Gene 98:61-67; Cullen et al., 1987, Nucleic Acids Research 15:9163-9175; WO 00 / 24883). Isolation of the AMA1 gene and construction of a plasmid or vector comprising the gene can be accomplished according to the methods disclosed in WO 00 / 24883.
[0215] In one embodiment, more than one copy of the nucleic acid can be inserted into the host cell to increase the production of the expression product. The increase in nucleic acid copy number can be obtained by the following method: the sequence of at least one additional copy is integrated into the host cell genome, or an amplifiable selective marker gene is included in a polynucleotide, wherein the amplified copy containing the selective marker gene can be selected by culturing cells in the presence of a suitable selection agent (selectable agent), thereby also containing cells of additional copies of the polynucleotide. The method for connecting the above-mentioned elements to construct a nucleic acid construct is well known to those skilled in the art (see, for example, Sambrook et al., 1989, see above).
[0216] In one embodiment, the vector is a plasmid vector. In one embodiment, the plasmid vector is selected from the group consisting of a prokaryotic expression vector, a yeast expression vector, a plant expression vector, an insect expression vector, and a mammalian expression vector.

[0218] In one embodiment, the recombinant host cell comprises one or more isolated polynucleotides encoding a capping enzyme, and the recombinant host cell is advantageously used for the recombinant production of the capping enzyme. The term "host cell" includes any progeny of a parent cell that is different from the parent cell because of mutations that occur during replication. The choice of host cells will largely depend on the gene encoding the capping enzyme and its source.

[0240] 7. Advantages of the present invention
[0241] The advantages of the present invention are at least embodied in the following aspects:
[0242] 1. The present invention tests the overall Cap0 capping activity, not the individual activities of the three steps of dephosphorylation, guanosine transfer, and methylation. The overall capping efficiency is better than or comparable to that of existing commercial poxvirus capping enzymes;
[0243] 2. The capping enzyme of the present invention has a sequence identity of less than 20% with the existing poxvirus capping enzyme, and is a brand-new capping enzyme with commercial potential;
[0244] 3. In the production and purification process of the capping enzyme, the present invention selects Pichia pastoris as the capping enzyme production and purification system, which is different from the conventional E. coli system. Compared with the prokaryotic expression system, this eukaryotic expression system has the function of post-translational protein processing and modification, and the purified protein is closer to mammalian cells in terms of physiological properties or functions;
[0245] 4. The capping enzyme derived from Marseille virus of the present invention has better activity in different species and has better versatility in cross-species expression than the positive control NP868R;
[0246] 5. The capping enzyme derived from orbivirus, Marseille virus or rotavirus of the present invention has a high activity and a sequence length of less than 800aa, and the shortest active sequence is 622aa, which is significantly shorter than the control capping enzyme sequence of 868aa. It is well known in the art that when the activity is basically equivalent, the delivery vector of the in vivo system has a payload limitation, and the shorter the sequence, the easier it is to be suitable for the delivery of the vector. In addition, the shorter the sequence of the enzyme, the more stable the conformation, and the easier it is to synthesize in vitro;
[0247] 6. The commercial capping enzyme derived from poxvirus needs to express two subunits at the same time to complete Cap0 capping, and the process is cumbersome and requires multiple purifications. The capping enzyme of the present invention can complete Cap0 capping with a single subunit, which greatly simplifies the production steps and reduces the production cost. DETAILED DESCRIPTION
[0248] The present invention will be further described below in conjunction with specific embodiments, but these specific embodiments cannot be understood as limiting the scope of protection of the present invention. Those skilled in the art may make various changes or modifications to these specific embodiments without departing from the scope of the technical solution of the present invention, and the changed and modified embodiments still fall within the scope of protection of the present invention.

After the expression is completed, the cells are collected by centrifugation, suspended in 20mM Tris, 150mM NaCl, 5mM Imidazole, 5% Glycerol, pH8.0 for ultrasonic disruption, and then centrifuged at 12000g, 4 degrees for 10 minutes to collect the supernatant, which is purified by nickel affinity filler and washed with washing buffer (20mM Tris, 300mM NaCl, 20mMImidazole, 5% Glycerol, pH8.0), and finally eluted with elution buffer (20mM Tris, 300mM NaCl, 250mM Imidazole, 5% Glycerol, pH8.0). The protein expression of different cloned cells is tested by SDS. The corresponding high-expressing cells can be amplified and purified according to the above expression method. If necessary, the protein can be purified with molecular sieves, and the final protein concentration is 0.5-1 mg / ml and stored in storage buffer (20mM Tris, 300mM NaCl, 1mM EDTA, 0.1mM TCEP, 20% glycerol, pH7.4). The protein can be stored at -80 degrees for long-term storage.

[0268] 2.4 LC-MS analysis of mRNA capping products
[0269] LC–MS analysis was performed using an Agilent 1290 Infinity II UPLC system coupled to a TOF mass spectrometer (Agilent 6230) with an electrospray source operating in negative ionization mode. MS spectra were collected in the range of m / z 400 to 3000. All samples were separated on an AdvanceBio Oligonucleotides Column (2.1 mm × 150 mm; 2.7 μm particle size, Agilent) at a column temperature of 30 °C at a flow rate of 0.2 mL / min. Mobile phase A consisted of 89.3 mM hexafluoroisopropanol and 9.6 mM triethylamine, and mobile phase B was 100% MeOH. The gradient was maintained at 95% A for 2 min and then increased to 70% A in a linear curve over 18 min. The gradient was changed to 20% A at 20.4 min and then increased linearly to 95% A in 3.6 min. The mass spectrometry results were processed using the Bioconfirm program for deconvolution and quantification.
