In the invention, the spinel nano nickel ferrite is added into the chitosan solution with fixed content of glucose oxidase to improve the transmission of electronic, and is used for electrochemical gaging of the glucose. The preparation method of the modified glassy carbon electrode as glucose sensor is as follows: the spinel nano nickel ferrite is dispersed into the chitosan solution with certain concentration; the mixed solution is evenly dispersed by ultrasound; the glucose oxidase with certain amount is added into the mixed solution; the mixed solution is dispersed evenly by ultrasound once again to obtain a modifier of the nickel ferrite, the chitosan and the glucose oxidase.
