Abstract
Description
TECHNICAL FIELD
[0001] The present application relates to the field of natural pharmaceutical chemistry and the field of cosmetics, in particular to a stable composition formed by coating dihydroartemisinin with fatty alcohol polyoxyethylene ether carboxylic acid or fatty alcohol polyoxyethylene ether carboxylic acid salt. BACKGROUND
[0002] In today's fast-paced era, people are always hoping that anything can be quickly, accurately and effectively solved under the background of work and life pressure, and they do not want to invest a lot of time and effort to harvest the desired effect or other negative effects. In this spirit of pursuit, consumers' demand for functional personal care products is becoming more and more vigorous. Gradually, this demand has extended from stay-on products such as color cosmetics and skin care products to rinse-off products such as personal cleansing and care products. Therefore, cosmetics that take into account both efficacy and safety are increasingly favored by consumers. Among the many functional raw materials, natural sources of functional raw materials have more advantages than a variety of traditional non-natural sources of functional active substances.
[0003] Dihydroartemisinin (DHA) is an extremely important derivative of artemisinin, which has higher bioavailability and easier metabolism than artemisinin, greatly improving its pharmaceutical value. According to research in the medical field, DHA has good effects in terms of anti-malaria, anti-tumor, anti-virus, anti-inflammatory, anti-other parasites, anti-fibrosis, blood pressure reduction, and immune regulation. Existing research has found that dihydroartemisinin not only has the above medical effects, but also has a good inhibitory effect on Malassezia, and can be used as a safe and efficient anti-dandruff ingredient to replace traditional anti-dandruff agents.
[0004] Chinese patent CN109939105A discloses the use of artemisinin and its derivatives in the preparation of drugs and cosmetics for improving the barrier function of the stratum corneum and preventing and treating skin inflammation.
[0005] Chinese patent CN116019734A discloses the anti-dandruff application of dihydroartemisinin. The patent research found that dihydroartemisinin has a clear inhibition zone at a concentration of 5mM and 10mM, and the inhibition zone of 10mM is greater than the positive control OCT, indicating that dihydroartemisinin has a good inhibitory effect on Malassezia.
[0006] However, DHA is poorly soluble in water and other aqueous media, and contains a special peroxide group in the molecule, which is not stable to heat, and is easily decomposed by moisture, heat and reducing substances. In order to improve the water solubility and stability of DHA and reduce its toxicity, the common method in the medical field at present is to encapsulate DHA into drug carriers. For example, Chiara Righeschi et al. prepared two kinds of DHA-loaded liposomes using egg yolk lecithin and cholesterol, which can significantly improve the water solubility of DHA and reduce its toxicity to normal cells. Chen Fangwei et al. used hydroxypropyl-β-cyclodextrin (HP-β-CD) as a carrier to encapsulate DHA, and the feasibility of encapsulation was revealed by molecular simulation. The results showed that the encapsulation of DHA by HP-β-CD was not only feasible but also could significantly improve the water solubility of DHA. The above cases are all from the field of pharmacy, and there is no new report in the field of daily chemical industry. It is of great significance to expand the application field of DHA if it can be applied to cosmetic formulations and stably exert its effect.
[0007] It is unexpectedly found in the present application that the fatty alcohol polyoxyethylene ether carboxylic acid or fatty alcohol polyoxyethylene ether carboxylic acid salt can solubilize DHA, thereby greatly improving the solubility of DHA in aqueous system, and the combination of the two has a stable appearance under high and low temperature and normal temperature conditions. Therefore, the present application provides a stable composition formed by wrapping dihydroartemisinin with fatty alcohol polyoxyethylene ether carboxylic acid or fatty alcohol polyoxyethylene ether carboxylic acid salt. [0021] The present application provides a stable composition. The composition is formed by a combination of fatty alcohol polyoxyethylene ether carboxylic acid / fatty alcohol polyoxyethylene ether carboxylic acid salt and dihydroartemisinin (DHA), and the composition has a stable appearance and can be used in efficacy-type daily-use products. The fatty alcohol polyoxyethylene ether carboxylic acid and DHA can form a stable white gel-like liquid, and the fatty alcohol polyoxyethylene ether carboxylic acid salt and DHA can form a stable colorless transparent liquid. When the aqueous solution of the fatty alcohol polyoxyethylene ether carboxylic acid is adjusted to a pH greater than 7 with a sodium hydroxide solution, the solution is colorless and transparent.
[0022] When the vesicles or micelles formed by the fatty alcohol polyoxyethylene ether carboxylic acid and the fatty alcohol polyoxyethylene ether carboxylic acid salt in water can encapsulate hydrophobic ingredients, they have a solubilizing effect. At the same time, during cleaning dilution or use, the vesicles or micelles open, and the solubilized ingredients can be slowly released, so they also have a sustained-release effect on the active ingredients.
[0023] The present application takes advantage of the encapsulation, solubilization, and sustained-release effects of the micelles formed by the fatty alcohol polyoxyethylene ether carboxylic acid / fatty alcohol polyoxyethylene ether carboxylic acid salt, not only increasing the water solubility of DHA, but also improving its stability and reducing its cytotoxicity. Therefore, the composition of the present application greatly expands the application scenarios of DHA.

[0026] Dihydroartemisinin
[0027] Dihydroartemisinin (DHA) is one of the most important derivatives of artemisinin, and has the characteristics of higher bioavailability and easier metabolism compared to artemisinin. According to research in the medical field, DHA has good effects in terms of anti-malaria, anti-tumor, anti-virus, anti-inflammatory, anti-other parasites, anti-fibrosis, antihypertensive, and immune regulation. Existing research has found that dihydroartemisinin not only has the above medical effects, but also has a good inhibitory effect on malassezia, and can be used as a safe and efficient anti-dandruff ingredient of natural origin to replace traditional anti-dandruff agents.
[0028] In some embodiments, the composition of the present application comprises 0.01-1% by weight of dihydroartemisinin. In preferred embodiments, the composition of the present application comprises 0.01-0.6% by weight of dihydroartemisinin. In more preferred embodiments, the composition of the present application comprises 0.1-0.5% by weight of dihydroartemisinin.
[0029] Fatty alcohol polyoxyethylene ether carboxylic acid or salt thereof
[0030] Fatty alcohol polyoxyethylene ether carboxylate (AEC) is a high performance surfactant, which is modified from non-ionic surfactant and has the characteristics of both anionic and non-ionic surfactants. Because of the oxygen ethylene bond in the molecule, it has the properties of non-ionic surfactant, and its ionic characteristics can be changed by adjusting the pH. AEC is resistant to hard water and has good compatibility with cationic surfactants. At the same time, because the terminal hydrophilic group is an acid group, it is mild to the human body. Therefore, fatty alcohol polyoxyethylene ether carboxylate has good water solubility, chemical stability, wetting property, emulsifying property, hard water resistance, and good foam performance, strong washing power and calcium soap dispersing power, and low skin irritation, and can be used as a cosmetic, a washing product, a crude oil demulsifier, a thick oil viscosity reducer, a sewage treatment agent, a softener, an emulsifier, an antistatic agent, etc. Han Jinying studied the aggregation behavior of fatty alcohol polyoxyethylene ether carboxylate (AEC) acid type product in aqueous solution by transmission electron microscopy and dynamic light scattering method, and found that it can form vesicles due to hydrogen bonding in aqueous solution, thus showing a light blue to milky white turbidity state.
[0031] When the aqueous solution of fatty alcohol polyoxyethylene ether carboxylic acid is adjusted to pH greater than 7 with sodium hydroxide solution, the solution is colorless and transparent.
[0032] When the vesicles or micelles formed by fatty alcohol polyoxyethylene ether carboxylic acid and fatty alcohol polyoxyethylene ether carboxylate in water can wrap hydrophobic ingredients, they have a solubilizing effect. At the same time, during the process of cleaning dilution or use, the vesicles or micelles open, and the solubilized ingredients can be slowly released, so they also have a slow-release effect on active ingredients.
[0033] The present application finds that fatty alcohol polyoxyethylene ether carboxylic acid and fatty alcohol polyoxyethylene ether carboxylate can solubilize DHA, thereby greatly improving its solubility in aqueous systems. The combination of the two has a stable appearance under high and low temperature and room temperature conditions, indicating that DHA is wrapped by the micelles of fatty alcohol polyoxyethylene ether carboxylic acid and fatty alcohol polyoxyethylene ether carboxylate, and the stability is improved. The composition has a good inhibitory effect on malassezia, and can replace traditional anti-dandruff agents and be applied to anti-dandruff products.

[0101] From the results of Comparative Example 1, Comparative Example 2 and Example 6, it can be seen that the effective content of surfactant is 15%, and the content of dihydroartemisinin is 0.5%, the composition of sodium lauroamphoacetate, sodium cocamphoacetate and dihydroartemisinin in Comparative Example 1 appears color change at room temperature and high temperature, and the appearance is unstable, while the lauryl alcohol polyether-5 carboxylic acid in Example 6 can form a stable white liquid with dihydroartemisinin, which shows that the vesicular structure formed by lauryl alcohol polyether-5 carboxylic acid in aqueous solution can wrap dihydroartemisinin, thereby increasing its stability. From the results of Comparative Example 3 and Example 12, it can be seen that the effective content of surfactant is 15%, and the content of dihydroartemisinin is 0.5%, the composition of lauryl alcohol polyether sulfosuccinate disodium and dihydroartemisinin in Comparative Example 3 appears color change at room temperature and high temperature, and the appearance is unstable, while the sodium lauryl alcohol polyether-5 carboxylate obtained by neutralizing lauryl alcohol polyether-5 carboxylic acid with sodium hydroxide in Example 12 can form a stable colorless transparent liquid with dihydroartemisinin, which shows that the micelles formed by lauryl alcohol polyether-5 carboxylate in aqueous solution can wrap dihydroartemisinin, thereby increasing its stability.
