Abstract
Description
TECHNICAL FIELD
[0001] The present application belongs to the technical field of biological medicine, and relates to a benzimidazole derivative as a novel HDAC inhibitor, a pharmaceutical composition containing the same, a pharmaceutical preparation, and a medical use thereof. BACKGROUND
[0002] Malignant tumor is the second leading cause of death worldwide, and its incidence and mortality continue to rise, becoming a major public health challenge. In recent years, research has found that the occurrence and development of cancer are closely related to abnormal epigenetic regulation. Epigenetics regulates gene expression through reversible mechanisms such as DNA methylation and histone modification, without changing the DNA sequence. Among them, histone deacetylases (HDACs) affect various life processes, including gene transcription, DNA damage repair, apoptosis, cycle regulation, metabolism and angiogenesis, through deacetylation of histones and non-histones. The mutation and abnormal expression of HDACs can lead to the occurrence of various diseases, especially tumors, and can also cause other nervous system diseases, autoimmune diseases, inflammatory diseases and heart diseases, making HDACs an important target for cancer and neurodegenerative disease treatment. So far, 18 HDAC subtypes have been identified in the epigenetic modification family of HDACs, and they are divided into four classes according to their homology with yeast proteins: Class I (HDAC1, 2, 3 and 8), Class II (IIa: HDAC4, 5, 7, 9; IIb: 6 and 10), Class III (SIRT 1-7), and Class IV (HDAC11). Among them, Class I, II and IV are zinc ion (Zn 2+ ) dependent enzymes, and Class III is an NAD + dependent enzyme.
[0003] The development of anti-tumor drugs targeting HDACs has become a hot spot in the field of epigenetic therapy. The pharmacophore model of the structure of HDAC inhibitors (HDACIs) is mainly divided into three parts: the recognition group (Cap) interacting with the surface of the receptor, the chelating group (ZBG) acting with Zn 2+ in the bottom of the active pocket, and the hydrophobic long chain (Linker) connecting the two parts. According to the structural characteristics, HDACIs can be divided into hydroxamic acids, benzamides, cyclic peptides and short-chain fatty acids, etc. So far, five HDAC inhibitors (SAHA, FK-228, PXD-101, LBH-589 and Chidamide) have been approved for marketing worldwide, mainly for the treatment of hematological malignancies. However, HDAC inhibitors face challenges in the treatment of solid tumors, and the research of anti-tumor drugs targeting HDACs has both profound scientific significance and urgent clinical needs.
