[0004] The present disclosure relates to the field of medical and non-medical needs for increasing muscle mass and / or muscle function.
[0005] BACKGROUND OF THE DISCLOSURE
[0006] The proportion of elderly people (65 years and older) is expected to represent one third of the French population by 2050. This population is very heterogeneous and is characterized, at the extremes, by individuals in good health, who are active and in good nutritional condition, with little sarcopenia (age-related loss of mass and muscle function) and generally living at home. Conversely, there is (at the same age) an elderly population that is undernourished and highly sarcopenic, generally dependent and living in institutions and who are in a state of chronic undernutrition (Buckinx et al., Burden of frailty in the elderly population: perspectives for a public health challenge. Archives of public health = Archives beiges de sante publique. 2015;73(l): 19). At the same time, in some Western countries, overweight or obese individuals represent more than half of the population. In addition to an increased susceptibility to diabetes or cardiovascular pathologies, part of this population may also present increased muscle wasting which may reduce the autonomy of these individuals and, above all, limit the room for treatment flexibility when treating obesity with low-calorie diets (Barazzoni et al., Sarcopenic Obesity: Time to Meet the Challenge. Obesity facts. 2018;ll(4):294-305). In addition, loss of muscle mass and function can also punctually affect individuals over the course of their lives, depending on their background and experiences. This is particularly the case for "burn victims", immobilized people, cancer patients or people who have undergone intestinal resection or who have intestinal malabsorptions.
[0007] Presently, enhancing muscle mass or muscle function may be obtained at least by practicing physical exercise. Indeed, practicing physical exercise cannot be performed by subjects such as immobilized subjects (e.g. subjects confined to bed following a bone fracture) or the elderly affected with some physical weakness.
[0008] A need for enhancing muscle mass also exists in subjects affected with sarcopenia in pathological situations, such as in cancer subjects affected with severe sarcopenia. How to better prevent sarcopenia in these subjects prevent and treat sarcopenia has become one of the frontiers of global geriatric research. Many molecules have been identified as therapeutic targets, bringing a booming of drug investigation targeting sarcopenia. It may be cited the exploration of pharmacotherapies designed or repurposed for treating sarcopenia, such as (i) drugs targeting myostatin signalling, including myostatin inhibitors, activin receptor antagonists, follistatin fusion proteins and gene therapy, (ii) drugs targeting the renin-angiotensin system, such as ACE inhibitors, angiotensin II type I receptor antagonists, Mitochondrial Assembly Receptor (MASR) agonists, (iii) Testosterone and selective androgen receptor modulators, such as testosterone and (iv) Ghrelin and its mimetics.
[0009] There remains a need for further substances or compositions able to enhance muscle mass or enhance muscle function both (i) for providing to healthy subjects in which preventing sarcopenia or increasing muscle mass or muscle function is desirable and (ii) for preventing or treating sarcopenia in subjects affected with a disease.
[0010] SUMMARY OF THE DISCLOSURE
[0011] The present disclosure relates to a furan fatty acid, or a furan fatty acid-containing compound, for its use for preventing and / or treating a loss in muscle mass in a human or animal subject.
[0012] The present disclosure also relates to a furan fatty acid, or a furan fatty acid-containing compound, for its use for preventing and / or treating a loss in muscle mass caused by a disease affecting a human or animal subject. For the sake of clarity, the said use aims at preventing and / or treating a loss in muscle mass and thus is not aimed at preventing and / or treating the disease per se which, notably, causes a loss in muscle mass.

Total proteins were lysed in RIPA buffer (20mM Tris, 150mM NaCl, ImM EDTA, 1% NP40, pH 7.5 + complete cocktail protease inhibitors + Pierce phosphatase inhibitors tablet). 40 pg of cell extracts were loaded and blotted onto a nitrocellulose membrane. The membranes were washed, blocked with 5% nonfat milk and incubated in primary antibody overnight at 4°C. The membranes were then washed in TBS-Tween (lOmM Tris, 140mM NaCl, and 0.2% Tween-20), incubated with the appropriate secondary antibody coupled to horseradish peroxidase, and washed again in TBS-Tween. [0223] B. RESULTS Example 1 : In vitro effects of FuFAs on muscle metabolism
[0224] 1.1 FuFA-F2 supplementation has little influence on C2C12 myoblast differentiation
[0225] For in vitro studies, the C2C12 cell line is classically used because it recapitulates the different phases observed in vivo: proliferation, withdrawal from the cell cycle, and fusion into myotubes. After a proliferation phase, the C2C12 myoblasts reach confluence and their terminal differentiation is induced by lowering the serum level in the culture medium. C2C12 cells were grown for 5 days in a differentiation medium, with or without FuFA-F2 (1, 10, and 50 pM) (Figure 1A). In humans, the levels of FuFAs in plasma are around 1 pM (Puchta et al., 1988, Liebigs Ann Chem, Vol. 1 : 25-28; Wahl et al., 1994, Hrc-JH High Res Chrom, Vol. 17(5) : 308- 311; Xu et al., 2020, Journal of Lipid Research, Vol. 61(4) : 560-569). The results show that FuFA-F2 had no significant effects on myoblast fusion index except for the highest dose of 50 pM, where a slight increase in differentiation was shown (61% with FuFA vs 57% for control, p<0.05) (Figure 1A). Consistent with this observation, FuFA-F2 had no effect on the expression of myogenin, a key myogenic factor for differentiation (Figure IB).
[0226] 1.2. FuFA-F2 supplementation increases protein accretion in C2C12 myotubes
[0227] Next were employed C2C12 myotubes that had been differentiated from myoblasts in order to test the influence of FuFA-F2 at this stage (Figure 2A). It was observed that whatever the dose, 3 days of FuFA-F2 supplementation significantly increase total protein content in C2C12 myo tubes (Figure 2B).
[0228] 1.3 : FuFA-F2 supplementation decreases autophagy in C2C12 myotubes
[0229] It is shown that FuFA-F2 increased protein accretion in myotubes. Protein homeostasis is determined by the dynamic balance between protein synthesis and degradation. Protein synthesis was evaluated by measuring the phosphorylation of Akt (Ser473), mTOR (Ser2448) and 4E-BP1 (Thr37 / 46), which are major regulators of translation and protein synthesis. Unfortunately, basal level of Akt and 4E-BP1 phosphorylation was undetectable with C2C12 myotubes in the present experiments (Figure 3A). In addition, it was found no difference in mTOR phosphorylation with or without FuFA-F2 (Figure 3A-B) perhaps because a rapid down regulation of this pathway occurred in high glucose medium.
[0230] AMP activated protein kinase (AMPK) is a crucial cellular energy sensor protein and is activated when intracellular ATP level decreases to regulate autophagy initiation (Hardie et al., 2012, Nat Rev Mol Cell Bio, Vol. 13 (4) : 251-262). Activation level of AMPK as assessed by phosphorylation at Thrl72 was significantly decreased by FuFA-F2 whatever the dose (Figure 3A-B). The autophagy-related signaling was further evaluated by measuring the MTORC1 mediated phosphorylation by ULK1 at Ser757 which disrupts the interaction between ULK1 and AMPK. Surprisingly ULK1 phosphorylation was decreased for the highest dose of FuFA-F2, however the observation that total Ulk level was also decreased was not in favor of autophagy activation (Figure 3C-D). Therefore, the autophagic flux was assessed by measuring the conversion of LC3-I to LC3-II, through lipidation. It was found that the ratio of LC3-II / LC3-I was decreased by FuFA-F2 whatever the dose (Figure 3C-D) suggesting that FuFA-F2 inhibited the autophagy process in C2C12 myotubes.
[0231] 1.4. FuFA-F2 supplementation increase mTOR phosphorylation
[0232] The observation that basal level of Akt phosphorylation was undetectable with C2C12 myotubes in the present experiments cultivated with a classical high glucose medium suggested a rapid down regulation of this pathway masking a potential activation. A similar hypothesis may also be done for mTOR activation. To avoid this possibility, C2C12 myotubes were treated as before and then cultured them for 2 hours in low-glucose medium before adding again the different treatments as indicated for 30 minutes (Figure 4A). In this condition, it was shown that insulin, used as positive control, induced a strong increase in Akt phosphorylation at Ser473 (Figure 4B- C). Compared to low glucose medium, the normal high-glucose medium and FuFA-F2 induced a similar increase of Akt phosphorylation (Figure 4B-C). In addition, treatment with FuFA-F2 did not alter insulin-induced phosphorylation of Akt (Figure 4B-C). This indicates that FuFA- F2 treatment does not appear to regulate Akt phosphorylation. Furthermore, in FuFA-F2 treated myoblasts it was observed a significant increase in mTOR phosphorylation at Ser2448 while no increase was detected in the presence of insulin alone (Figure 4B-C). This data set suggests that FuFA-F2 stimulates mTOR phosphorylation and protein synthesis.
[0233] To determine whether mTOR phosphorylation is induced only by 30 min of incubation in the presence of FuFA-F2 or whether it is an effect partly related to the 3-day treatment with FuFA- F2, the experiment was repeated without pretreatment (Figure 4D). In this condition it was still observed that only insulin is able to induce a strong increase in Akt phosphorylation (Figure 4E- F). However, in contrast to the previous experiment a rapid effect of FuFA-F2 on mTOR phosphorylation was not observed (Figure 4E-F). This observation suggests that the activation of mTOR by FuFA-F2 was related to the 3-day treatment. 1.5. FuFA-F2 supplementation modulates the metabolic and contractile characteristics of C2C12 myotubes
[0234] It was further explored whether this increase in myotubes anabolism is associated with a change in the metabolic and contractile characteristics of muscle fibers. To address this issue, the influence of FuFA-F2 on mitochondrial activity in C2C12 myotubes was tested. It was found that 3 days of FuFA-F2 treatment of C2C12 myotubes had no effect on citrate synthase activity (Figure 5A), commonly used as a quantitative marker of intact mitochondria. However, at the 10 pM dose, FuFA-F2 tended to increase the Complex IV activity (+60 %, p<0.07) (Figure 5B), a key enzymatic activity of the mitochondrial respiratory chain. To examine whether this trend of increased of mitochondrial respiratory chain activity with 10 M FuFA-F2 was associated with a change in the contractile type of myosin heavy chain (MyHC), the expression of slow and fast MyHC by western-blot was studied. It was found that at the 10 pM dose, FuFA-F2 significantly increased the expression of slow MyHC which are characteristic of high mitochondrial activity (Figure 5C-D). Surprisingly, a decrease of fast MyHC was observed for the highest dose of FuFA-F2 (Figure 5C-D).
[0235] Example 2 : In vivo effects of FuFAs on muscle metabolism
[0236] 2.1. FuFA-F2 supplementation in C57B16 mice increase muscle mass
[0237] To investigate in vivo the potential impact a furan-containing compound under the form of a triacylgycerol of FuFA-F2 on muscle characteristics, C57B16 mice were supplemented by oral gavage with vehicle or FuFA-F2 (20mg / kg) for 3 and a half weeks (Fig. 6A). It was observed that mice receiving FuFA-F2 ate more and had a significantly higher body weight than animals receiving the vehicle alone (Fig. 6B-C). In addition, analysis of the body composition using an EchoMRI revealed that the mice supplemented with FuFA-F2 had a greater lean mass than the control mice (Fig. 6D). However, fat mass was similar between the two groups (Fig. 6E). In agreement with these observations, the weight of the quadriceps and gastrocnemius, the largest muscles of the hindlimbs, is significantly increased in mice receiving FuFA-F2 (Fig. 6F). These data demonstrated that 3 weeks of FuFA-F2 supplementation was sufficient to stimulate muscle anabolism in mice.
[0238] 2.2. FuFA-F2 supplementation promotes more oxidative muscle metabolism in mice
[0239] To determine if this increase in muscle mass is accompanied by changes in the contractile and metabolic characteristics of the muscle fibers, the expression of myosin heavy chains and mitochondrial activity in the quadriceps was measured. The results did not indicate any significant difference in the expression of the different myosin heavy chains between mice treated or not with FuFA. However, a trend towards an increase in MyHCIIa expression at the expense of MyHCIIb with FuFA supplementation was observed (Fig. 7A). Citrate synthase (CS), a marker of mitochondrial activity, was not modified (Fig. 7B). It was found a significant increase of the complex IV activity of mitochondrial respiratory chain complexes in FuFA group (Fig. 7C). These set of data indicates that FuFA-F2 supplementation promotes more oxidative muscle metabolism in mice.
[0240] Summary of the results shown in the examples 1 and 2
[0241] Several studies suggest that FuFAs could have beneficial health effects. However, only a few studies have been performed to date.
[0242] In the present study, it was tested for the first time the influence of FuFA-F2 extracted from Hevea brasiliensis latex (Liengprayoon et al., 2011, Phytochemistry, Vol. 72(14-15) : 1902- 1913) on skeletal muscle phenotype. To address this potential effect of FuFA-F2 cultured skeletal muscle cells were used. Mouse C2C12 skeletal muscle cell lines are commonly used to study muscle differentiation, muscle hypertrophy and atrophy. In this work the use of 3 different doses of FuFA-F2 (1, 10 and 50 pM) was chosen. The lowest dose of IpM is comparable to the concentration found in human plasma (Puchta et al., 1988, Liebigs Ann Chem (1) : 25-28; Wahl et al., 1994, Hrc J High Res Chrom, Vol. 17(5) : 308-311; Xu et al., 2020, Journal of Lipid Research, Vol. 61(4) : 560-569).
[0243] First, the influence of FuFA-F2 on the myoblasts differentiation was tested. It was found that FuFA-F2 had no effects on myoblast fusion index except for the highest dose of 50 pM, where a slight increase in differentiation was shown. Then, the effect of FuFA-F2 supplementation on C2C12 myotubes was studied. It was shown that 3 days of FuFA-F2 treatment significantly increase total protein content in C2C12 myotubes whatever the dose. This observation indicates clearly that FuFA-F2 increased protein accretion in myotubes. To understand how FuFA-F2 induces an increase in total protein content and muscle anabolism protein homeostasis was investigated. The dynamic balance between protein synthesis and degradation determines protein homeostasis. It was first explored the synthesis pathways but in a classical hyperglycemic medium it was not possible to detect Akt phosphorylation and no difference on mTOR activation with FuFA-F2 treatment was seen. It was hypothesized that the high glucose medium could mask a possible effect by inducing a rapid down regulation of Akt / mTOR pathway. To avoid this possibility, C2C12 myotubes were cultured for 2 hours in low-glucose medium before adding the different treatments for 30 minutes. Under this condition it was observed that 3 days of FuFA- F2 treatment induced a strong increase in mTOR phosphorylation. This observation indicates that FuFA-F2 stimulates protein synthesis. However, in these experiments any influence of FuFA-F2 on Akt phosphorylation was observed. Although the classical pathway of mTOR regulation involves Akt, other mechanisms of activation of this pathway have already been shown (Memmott et al., 2009, Cellular Signalling, Vol. 21(5) : 656-664). In particular, several studies indicate that the LKB1 / AMPK pathway inhibits mTORCl (Carretero et al., 2007, Oncogene, Vol. 26(11) : 1616-1625; Gwinn et al., 2008, Molecular Cell, Vol. 30(2) : 214-226; Shaw et al., 2004, Cancer Cell, Vol. 6(1) : 91-99). Thus, it can be hypothesized that AMPK inhibition induced by FuFA-F2 observed in this study is responsible for the stimulation of the mTOR pathway in C2C12 myotubes. It was next investigated protein degradation. Autophagy is a major intracellular degradation process that is essential for the clearance of altered proteins and organelles. In addition, autophagy has been shown to be critical for skeletal muscle homeostasis (Masiero et al., 2009, Cell Metabolims, Vol. 10(6) : 507-515). This process involves the formation of double-membraned structures known as autophagosomes. LC3-II is a standard marker for autophagosomes, so we assessed autophagy by measuring the LC3-II / LC3-I ratio. It was found that FuFA-F2 decreased significantly this ratio whatever the dose indicating that this fatty acid inhibited the autophagy process in C2C12 myotubes. It was also found that FuFA-F2 decreased AMPK activation, a regulator of autophagy initiation (Hardie et al., 2012, Nat Rev Mol Cell Bio, Vol. 13(4) : 251-262). Thus, FuFA-F2 increases the anabolism of C2C12 myotubes by increasing protein synthesis on the one hand and inhibiting autophagy on the other. In addition, it was found herein that this stimulation of anabolism induced by FuFA-F2 was also accompanied by a change in the metabolic and contractile characteristics of muscle fibers for the intermediate dose of 10 |1M. By stimulating the mitochondrial respiratory chain and inducing the expression of slow myosin heavy chain, FuFA-F2 supplementation could promote the utilization of glucose by the muscle. In a pathological context, this may delay or limit insulin resistance.
[0244] To investigate in vivo the potential impact of FuFA-F2 on muscle characteristics, C57B16 mice were supplemented by oral gavage with vehicle or FuFA-F2. Example 3 : In vivo effects of FuFA-F2 incorporated in a High Fat and High Sucrose (HFHS) diet on muscle mass

[0247] Forty six-weeks old male C57BL / 6J mice (Charles River, L’ Arbresle, France), weighing about 21 g, were housed (5 per cage) under conditions of constant temperature (20-22°C), humidity (45-50%) and a standard dark cycle (20.00-08.00 hours). The mice were randomized, according to their initial weight, into four groups of ten animals and fed for 12 weeks with one of the four following semi-purified diets: 1) control diet, 2) HFHS diet, 3) HFHS + FuFA-F2 (0.048%, 40mg / kg body weight / day), 4) HFHS + FuFA-F2 (0.127%, 105mg / kg body weight / day). In this example, “FuFA-F2” is the triacylglycerol form of FuFA-F2.
[0248] The control diet contains 5% lipids as a mixture of rapeseed oil, high oleic sunflower oil, sunflower oil and linseed oil (oil mixture of Carrefour), the high fat high sucrose (HFHS) diets contain 25% lipids (5% of a mixture of rapeseed oil, high oleic sunflower oil, sunflower oil and linseed oil and 20% of lard) and 30% sucrose. [0258] In conclusion, the inventors have shown for the first time that supplementation in FuFA-F2 increases protein content in C2C12 myotubes . This anabolic effect is explained by the fact that treatment of myotubes with FuFA-F2: i) decreases autophagy by inhibiting autophagosomes formation as attested by the decrease of LC3II / LC3I ratio; ii) increases protein synthesis as shown by the stimulation of mTOR phosphorylation. In addition, the inventors have found that FuFA-F2 modulates the metabolic and contractile characteristics of C2C12 myotubes.
[0259] In addition, the inventors have found that oral gavage with FuFA-F2 for 3 weeks in C57B16 mice increased muscle mass and promoted more oxidative muscle metabolism in mice. Altogether, these data demonstrated that FuFA-F2 extracted from Hevea brasiliensis latex stimulates muscle anabolism both in vitro and in vivo, and promotes a more oxidative metabolism. These results highlight that FuFA-F2 supplementation could have health benefits by promoting skeletal muscle anabolism.
[0260] Further, the examples also show that feeding subjects with various amounts FuFA-containing compounds specified in the present disclosure cause an increase of muscle mass, including in a dose-dependent manner. A furan fatty acid for its use according to claim 2, which is selected from the furan fatty acids wherein,- n is 2, m is 8, R1 means a methyl group, R2 means a hydrogen atom and R3 means a methyl group, or- n is 4, m is 8, R1 means a hydrogen atom, R2 means a hydrogen atom and R3 means a methyl group, or- n is 4, m is 8, R1 means a methyl group, R2 means a hydrogen atom and R3 means a methyl group, or- n is 2, m is 10, R1 means a methyl group, R2 means a hydrogen atom and R3 means a methyl group, or- n is 4, m is 10, R1 means a hydrogen atom, R2 means a hydrogen atom and R3 means a methyl group, or- n is 4, m is 10, R1 means a methyl group, R2 means a hydrogen atom and R3 means a methyl group, or- n is 4, m is 10, R1 means a hydrogen atom, R2 means a hydrogen atom and R3 means a methyl group, or- n is 4, m is 8, R1 means a hydrogen atom, R2 means a hydrogen atom and R3 means a hydrogen atom, or- n is 4, m is 12, R1 means a hydrogen atom, R2 means a hydrogen atom and R3 means a methyl group , or- n is 4, m is 12, R1 means a methyl group, R2 means a hydrogen atom and R3 means a methyl group.
5. A furan fatty acid for its use according to claim 2, wherein the furan fatty acid is of formula (I) wherein n is 4, m is 8, R1 means a hydrogen atom, R2 means a hydrogen atom and R3 means a methyl group.

A furan fatty acid-containing compound for its use according to any one of claims 1 to 3, which is of formula (II) belowwherein groups RA, RB and RC represent, independently of each other, (i) a hydroxyl group, (ii) -O-C(=O)-R4 , wherein R4 means a group of formula (III) as defined in claim 2 or (iii) -O- C(=O)-R4 , wherein R4 means a (C6-C22) linear or branched, alkyl chain, a (C6-C22) linear or branched alkenyl group or a (C6-C22) linear or branched alkynyl group, provided that at least one of RA, RB and RC represents -O-C(=O)-R4 , wherein R4 means a group of formula (III) as defined in claim 2.
7. A furan fatty acid-containing compound of formula (II) according to claim 5, wherein RA, RB and RC are identical and each means -O-C(=O)-R4 , wherein R4 means a group of formula (III) as defined in claim 2 wherein n is 4, m is 8, R1 means a hydrogen atom and R3 means a methyl group.
8. A furan fatty acid-containing compound of formula (II) according to claim 5, wherein RA, RB and RC are identical and each means -O-C(=O)-R4 , wherein R4 means a group of formula (III) as defined in claim 2 wherein n is 4, m is 10, R1 means a hydrogen atom and R3 means a methyl group.
9. A furan fatty acid-containing compound of formula (II) according to claim 5, wherein RA, RB and RC are identical and each means -O-C(=O)-R4 , wherein R4 means a group of formula (III) as defined in claim 2 wherein n is 4, m is 8, R1 means a hydrogen atom and R3 means a hydrogen atom.
10. A furan fatty acid, or a furan fatty acid-containing compound, for its use according to any one of claims 1 to 9, wherein the subject is a human subject or an animal subject.

A furan fatty acid, or a furan fatty acid-containing compound, for its use according to any one of claims 1 to 10, wherein the human or animal subject is selected among elderly subjects affected by sarcopenia, overweight and obese subjects affected by sarcopenic obesity and subjected to a food regime, subjects affected by cachexia, notably a cachexia linked to cancer, subjects affected by an inflammatory bowel disease or a chronic obstructive pulmonary disease, immobilized subjects, subjects with severe burns and affected with septic or viral infections such as with a SARS-CoV-2 virus, subjects having a heart failure or a respiratory failure, subjects having cirrhosis, subjects affected with a rheumatoid arthritis,, convalescent subjects, immobilized subjects, and subjects with bowel resections or intestinal malabsorptions.
12. A furan fatty acid, or a furan fatty acid-containing compound, for its use according to any one of claims 1 to 10, which is comprised in an oral composition, and more particularly in an oral composition selected from the group consisting of a food product, a beverage, a pharmaceutical, a nutraceutical, a food additive, a dietary supplement, a dairy product and a living biotherapy product (LBPs).
13. A non-therapeutic use of a furan fatty acid, or of a furan fatty acid-containing compound as defined in any one of claims 1 to 9, for maintaining or increasing muscle mass in a subject in need thereof, in particular in a subject selected from a malnourished subject, an elderly subject, in particular a malnourished elderly subject, and a subject engaged in strenuous exercise.
14. The non-therapeutic use according to claim 13, wherein the furan fatty acid, or the furan fatty acid-containing compound is of formula (I) or (II) as defined in any one of claims 2 to 6.
