FORMULATION COMPRISING ANTIBODY-DRUG CONJUGATE TARGETING CLAUDIN18.2, METHOD FOR PREPARING THE SAME, AND USE THEREOFTECHNICAL FIELD

[0001] The present invention relates to the filed of the pharmaceutical formulation of antibody-drug conjugate (ADC) targeting Claudin18.2 (CLDN18.2) . More specifically, the present invention relates to a pharmaceutical formulation, in particular a lyophilized formulation, a liquid formulation and a reconstituted liquid formulation, comprising an antibody-drug conjugate targeting Claudin18.2, a method for preparing the pharmaceutical formulation and a therapeutic and / or prophylactic use of the pharmaceutical formulation.BACKGROUND

[0002] Claudins are a family of proteins, which are important components constituting the tight junctions between cells. They establish an intercellular barrier that controls the intercellular flow of molecules, the barrier can control the flow of molecules between cells. Claudins family proteins have tetraspanin domains, and their N-terminus and C-terminus are included in the cytoplasm. Different Claudins proteins are expressed in different tissues, and their functional changes are related to the development of cancer in various tissues. For example, Claudin-1 is expressed in colon cancer and has prognostic value, Claudin-18 is highly expressed in gastric cancer and pancreatic cancer, and Claudin-10 is highly expressed in hepatocellular carcinoma. Claudins, as cell membrane surface proteins, are useful targets for various therapeutic strategies.

[0003] Isoform 2 of Claudin-18 (Claudin 18.2 or CLDN18.2) is a highly selective cell lineage marker. Its expression in normal tissues is strictly confined to differentiated epithelial cells of the gastric mucosa, but its expression was absent from the gastric stem cell zone. CLDN18.2 is expressed in a considerable portion of primary gastric cancer, and the expression level thereof is retained in gastric metastatic cancer tissues. In addition to gastric cancer, the expression of CLDN18.2 is also found in pancreatic cancer. CLDN18.2 is an ideal target molecule for treating these cancers (Singh, P., Toom, S. &Huang, Y. Anti-CLDN18.2 antibody as new targeted therapy for advanced gastric cancer. J Hematol Oncol 10, 105 (2017) . https:  / / doi. org / 10.1186 / s13045-017-0473-4) .

[0004] With regard to gastric cancer, in 2014, there were about 410, 000 new cases and 290, 000 deathsin China, accounting for almost half of the new cases and deaths worldwide, and they are still showing an increasing trend. However, there is a large unmet need for clinical tumor treatment, so it is extremely necessary to develop drugs targeting Claudin18.2.

[0005] Despite the clinical success has been achieved by therapeutic antibodies, naked MAbs targeting cell surface tumor antigens rarely provide sufficient efficacy alone. To increase the low activity of Mab, new strategies focus on conjugating them to toxic molecules. Plant and bacterial toxins and chemotherapeutic small molecules can be good candidates because they are very potent and active in very small amounts.

[0006] Due to technological advances over the past several years, the field of antibody-drug conjugates (ADCs) for the treatment of cancer has recently experienced an increasing development activity by pharmaceutical companies in order to solve the problems that the ADCs initially presented in terms of immunogenicity, affinity, specificity, undesirable toxicity, yield, half-life, etc.

[0007] Despite some advances, there is still a need for additional therapeutic strategies for the treatment of tumors as well as for components used in such therapeutic strategies, in particular for anti-Claudin18.2 antibodies with high affinity, good specificity, and / or low risk of immunogenicity, and for ADC molecules with higher activity, low toxicity, prolonged half-life, good specificity or affinity, and / or good half-life or druggability.

[0008] There is a need for a novel pharmaceutical formulation comprising an antibody-drug conjugate targeting Claudin18.2 that is sufficiently stable and suitable for administration. The pharmaceutical formulation, in particular the lyophilized formulation, has good stability, remains stable during storage and transportation, and the lyophilized formulation is readily to be administrated upon reconstitution.

[0010] The present invention provides a pharmaceutical formulation comprising an antibody-drug conjugate targeting Claudin18.2 (ADC) .

[0011] In a first aspect, the present invention provides a pharmaceutical formulation comprising (i) an antibody-drug conjugate targeting Claudin18.2 or a pharmaceutically acceptable salt or a solvate thereof; (ii) a buffer, (iii) a stabilizer, and / or (iv) a surfactant; wherein the pH of the formulation is about 5.5-7.5, for example, about 6.0-6.5, e.g., 5.6, 5.7, 5.8, 5.9, 6.0, 6.1, 6.2, 6.3, 6.4, 6.5, 6.6, 6.7, 6.8, 6.9, 7.0 or 7.2, preferably, about 6.5.

[0110] The term “CLAUDIN” or “CLDN” used herein is the most important skeleton protein that determines the structures of tight junctions between cells, and it is involved in adhesion junctions and plays an important role in the metastasis and invasion of tumor cells. Claudin proteins are widely present in mammalian  epithelial and endothelial cells, primarily on the lateral surface of epithelial cells and on the plasma membrane of basal cells. Different Claudin proteins have their respective specific expression in different tissues, wherein the Claudin18 (CLDN18) gene is located at 3q22.3, has a molecular weight of 24 kDa, comprises 261 amino acid residues, and is a member of the Claudins superfamily. Its protein structure comprises 2 extracellular loops and 4 transmembrane region. Two subtypes of human CLDN18 or Claudin18 protein are Claudin18.1 or CLDN18.1 (UniProt ID: P56856-1) and Claudin18.2 or CLDN18.2 (UniProt ID: P56856-2) , respectively. In the primary structural sequences of the two proteins, only the amino acid residues at some positions from the N-terminus signal peptide to the extracellular loop 1 (Loops) structure are different. Especially at the extracellular loop 1, CLDN18.1 and CLDN18.2 differ by only 8 amino acids. The intergeneric and inerspecies sequence homology of the two subtypes of CLDN18 protein is also very high. The sequences of the extracellular loop 1 of CLDN18.2 are completely identical in different species such as human, mouse and rhesus monkey, while the homology of human and mouse CLDN18.2 proteins reaches 84%, indicating that the sequence of CLDN18.2 protein is extremely conserved (O. Tureci. et al., Gene, 481: 83-92, 2011) . CLDN18.2 or any variants and isoforms thereof may be isolated from cells or tissues in which they are naturally expressed, or recombinantly produced using techniques well known in the art and / or those described herein. In an embodiment, the CLDN18.2 as described herein is a human CLDN18.2.

[0384] Strong endocytosis is one of important properties fo ADC drugs. When the ADC binds to the antigen on the surface of the cell membrane, the ADC-antigen complex enters the cell through endocytosis and kills the target cell. Therefore, the endocytosis efficiency of ADCs is one of the important indexs to determine the tumor inhibition effect.

[0385] To test the endocytosis efficiency of antibodies after being conjugated to small molecule compounds, the endocytosis of different IEX019 molecules on DANG-hCLDNA18.2 cells were tested by flow cytometry. After digestion of DANG-hCLDNA18.2 cells, the cell density was adjusted. The cells were plated onto a 96-well plate at 1*105 cells / well, then were centrifuged at 500g for 3min. The supernatants were discarded. 100 μl of tested molecule was taken to resuspend the cells (the molecular concentration is 50 nM) and 5 replicates were set up for each sample (i.e., endocytosis time: 0h, 1h, 2h, 3h, 4h) . The mixtures were placed on ice and incubated for 1 h. After 1 h, the mixtures were centrifuged at 500g for 3min. The supernatants were discarded. 200 μl of FACS buffer (1%FBS, 1X PBS) was added to each well to rinse twice. One set of samples was taken and transferred to a new 96-well plate and incubated at 37 ℃ for 4 h. The rest of the samples continued to be incubated on ice. The previous operations were repeated. The samples were incubated at 37℃ for 3h, 2h, 1h, and 0h, respectively. After the specific incubation time was completed, the mixtures were centrifuged at 500 g for 3 min. The supernatants were discarded. 100 μl of anti-hFC-PE antibody (SouthernBiotech) diluted at 1: 400 was added to each well. The mixtures were incubated on ice in the dark for 30 min. After the completion of the secondary antibody incubation, the mixtures were centrifuged at 500g for 3 min. The supernatants were discarded. 200 μl of FACS buffer was added to each well to rinse twice. Finally, the cells were resuspended with 100 μl PBS for detection.

[0394] In the synthesis process of ADC drugs, small molecule compounds may be linkeded to antibodies through cleavable linkers, and after entering into the cell membrane via endocytosis, the linkers are cleaved, and the small molecules are released to kill the target cells. After the death of the target cell, the small molecule compound is released from the target cell into the intercellular space to further kill the non-target cells within a certain range, and such effect is known as the bystander killing effect. Since cells within tumors may vary greatly in target expression levels (tumor heterogeneity) , the bystander killing effect is important  for effectively killing tumor cells and inhibiting tumor growth.

[0395] Non-targeting cell DAN-G and target cell DANG-hCLDN18.2 were used in the present invention to test the bystander killing effect of IEX019 molecules.

[0396] After digesting the used cells with EDTA / Trypsin, the density was adjusted. Six-well cell culture plates were prepared, and 7.5×104 cells of DANG cells and DANG-hCLDN18.2 cells were added. The two kinds of cells were co-incubated. 200 μl of tested samples (IgG (SEQ ID NO: 13, SEQ ID NO: 14) , IEX019-02, IEX019-05, IEX019-06, Exatecan (Macklin, E881532) were added to each well at a final concentration of 50 nM with 3 replicates for each sample. The cell culture plate was placed in an incubator at 37 ℃ for 5 days. After 5 days, the culture supernatants were discarded. After rinsed with PBS, Trypsin-EDTA was added to digest the cells. All digested cells were collected and transferred to a 96-well plate. According to the antibody incubation protocol of flow cytometry technology, the cells were incubated with primary antibody (IEX019-01, 100 nM) and secondary antibody (anti-hFc-PE, Southern Biotech) at 4℃ for 1 h and 0.5 h, respectively. After rinsed with PBS, the live / dead Violet dye (Thermo, L34964) was diluted at 1:1, 000 and added to each well at 100 μl for incubation at 4℃ for 20 min. After rinsed with PBS, cells were resuspended with 100 μl PBS for detection. The populations in each sample were distinguished using the live / dead dye, wherein the populations with IEX019-01-negative (i.e., hCLDN18.2-negative) were DAN-G cells, and cells with IEX019-01-positive (i.e., hCLDN18.2-positive) were DAN-G-hCLDN18.2. The number of two kinds of cells in each sample was counted, and the relative cell viability of each kind of cells was calculated according to the following equation, and the curve was fitted by Graph Pad Prism 8.0.

[0437] The purpose of the present experiment was to develop a long-term stable IEX019-02 formulation. The development of formula of the formulation consisted of two steps: 1) pH buffer solution screening (store at 40 ℃ stress condition for 4 weeks) . 2) auxiliary material screening, including liquid samples were placed at 25 ℃ and 40 ℃ stress condition for 4 weeks, freeze-thaw for 5 cycles, and 3 days of oscillation (150 rpm, 25℃) . Lyophilized samples were placed at 40℃ for 2, 4, and 12 weeks. Analytical methods include DSC, appearance, pH, molecule concentration of ADCs, insoluble particles (HIAC) , SE-HPLC, iCIEF, Caliper-R / NR, DAR value, free drug, reconstitution time, and binding activity.

[0455] 2.9 Purity of reduced / non-reduced capillary gel electrophoresis Caliper-NR / R

[0456] The non-reduced Caliper-SDS was analyzed on a SCIEX / PA800 Plus instrument equipped with a PDA detector (diode array detector) . Samples were diluted to 2.0 mg / mL. 50 μL of the sample was placed in a 1.5 mL centrifuge tube, 5 μL of NEM (N-ethylmaleimide) was added to the centrifuge tube, and 45 μL of SDS sample buffer solution (PB-CA (citrate phosphate buffer solution) containing 0.8%SDS) was added. The samples were mixed well and centrifuged, and placed at 70℃ for 10 minutes. Capillary right-end injection method (effective capillary separation length was 20.2 cm) was used. The detection wavelength was 220 nm. After 20 seconds of continuous injection using a forward voltage of -5 kV, the separation was performed with a forward voltage of +15 kV for 35 minutes.

[0459] Gradiently diluted references, controls, and test samples were added to a 96-well high-binding plate, and the products were bound to the antigen coated on the plate. After washing, peroxidase-labeled goat anti-human IgG Fcγ detection antibody (Jackson, 109-035-098#) was added to the plate. The detection antibody can bind to the captured ADC product. After the final washing, TMB (3, 3', 5, 5'-tetramethylbenzidine) substrate solution was added to the plate. TMB can react specifically with peroxide in the presence of peroxidase to generate single color signal, and the signal intensity is proportional to the amount of ADC products bound on the plate. After the color reaction was terminated, the absorbance value of 450 nm-620 nm was read on the microplate reader.

[0463] A greater than 5.2 ml of water for injection was taken by a 10 ml syringe, and the air was excluded so that the final volume of water for injection in the syringe was 5.2 ml. The syringe was inserted through the center of the stopper. The timer was pressed after adding the water for injection along the wall of the bottle. The aluminum cap of the bottle mouth was grasped with a hand. The wrist was rotated slightly clockwise. If no particles werre observed within 10 minutes, the timer was pressed again, and the time was the reconstitution time.

[0464] 2.12 DSC

[0465] DSC is a thermal analysis instrument that measures the heat difference between a test sample and a reference when heated. This experiment was measured using MicroCal PEAQ DSC (Malvern, SYS12907) . Prior to the analysis, the protein sample was first diluted to 1 mg / mL using its reference buffer solution. 400 μL of each reference buffer solution was added to the odd-numbered wells of the 96-well plate, and 400 μL of samples were added to the even-numbered wells of the same plate. The experimental parameters were set so that the scan temperature rised from 10 ℃ to 95 ℃ at a rate of 90 ℃ / h. Thermalgram analysis was performed using MicroCal PEAQ DSC automated data analysis software.

[0466] 2.13 Detection of glass transition temperature (Tg’ )

[0467] The glass transition temperature (Tg') of the protein formula was determined by TA 2000 DSC. 20 μL of sample was placed in an aluminum sample tray, and the sample tray to which the sample was added and an empty sample tray (reference) were placed in the DSC instrument. The corresponding parameters (see the table below) , including the heating / cooling rate, the target temperature of heating / cooling, and the holding time of the platform after reaching the target temperature, were set in the control panel. Once completing the setup, the data sampling began. The mDSC pattern was recorded during this process and the glass transition temperature (Tg') was obtained by Universal Analysis.

[0468] 2.14 Collapse temperature (Tc)

[0469] The collapse temperature (Tc) was measured by Lyostat5 LED lyophilization microscopy. When testing a sample, the sample holder was placed on the temperature control block, the side door of the sample cell was closed to ensure that the sample holder was in the center of the temperature control block. A piece of quartz glass with 16 mm diameter was placed on the sample holder with a vacuum pen and was brought to contact with the temperature control block. Next, a 70 μm thick metal gasket was clamped with tweezers and placed on the quartz slide, keeping the metal gasket coaxial with the center of the quartz sheet. A certain volume (typically 2 μL) of the test sample was taken with a pipette and dropped in the center of the quartz sheet. A 13 mm diameter coverslip was carefully placed on the metal gasket with a vacuum pen to ensure that the edge of the sample was moved above the light penetration hole of the temperature control block for easy visualization of the sample. The lid was rotated back on the platform, and the nitrogen outlet  pipe was fixed on the lid to avoid frosting. Then, the corresponding parameters (see the table below) , including the heating / cooling rate, the target temperature of heating / cooling, the holding time of the platform after reaching the target temperature, and the frequency of image collection, were set in the control panel. Once completing the setup, the data sampling began. The sublimation front moved from the outer edge of the sample to the middle during the lyophilization. The front of the sublimation front was in a frozen state, and the rear thereof was in a dry strate. When the sample temperature was higher than the collapse temperature (Tc) , collapse occured. The photos in the experiment were carefully reviewed, starting from the photos of the cooler temperatures. When a tiny void appeared in the collected photo, it was a microstructure collapse, and the corresponding temperature was deemed as the collapse temperature.

[0471] 3.1 Purpose of the experiment

[0472] The purpose of this experiment is to screen the optimal pH of buffer system for IEX019-02 at a concentration of 40 mg / mL.

[0473] 3.2 Experimental protocol

[0474] As shown in Table 10, three buffer systems were studied at a concentration of 40 mg / mL IEX019-02 in 8 groups of experiments.

[0475] Under high temperature conditions (40℃) , the stabilities of 3 different pH buffer systems in 8 groups of experiments were evaluated for 4 weeks. The test items performed in the study included DSC, appearance, pH, molecule concentration of ADC, SE-HPLC, Caliper-NR / R, free drug, DAR value, and iCIEF.

[0477] 3.3 Sample preparation

[0478] IEX019-02 stock solution (10 mg / mL ADC, 20 mM histidine solution, 8% (w / v) sucrose, pH 6.0) was concentrated in ultrafiltration tubes, and then was exchanged into systems of 20 mM citric acid buffer solution (pH 5.0, 6.0) , 20 mM succinic acid buffer solution (pH 5.0, 5.5, 6.0) and 20 mM histidine buffer solution (pH 5.5, 6.0, 6.5) , respectively. The concentration of IEX019-02 was adjusted to approximately 40 mg / mL. The solutions were filtered with a 0.22 μm PES filter in a biosafety cabinet to obtain the samples F1-F8 as shown in Table 10. The samples were then filled into 2 mL vials with a filling volume of 1 mL / vial. After filling, the vials were stoppered and capped immediately. The samples were placed under different storage conditions according to Table 10, and at least 1 vial was placed at each sampling point for each formula.

[0480] The results of pH buffer solution screening for the formulae of IEX019-02 formulation were summarized in Table 11-20.

[0482] As shown in Table 11 and FIG. 8, the Tm onset values of samples in 8 kinds of pH buffer systems (F1-F8) all over than 58℃, which indicated that the molecules had good conformational stability.

[0484] 3.4.2 Appearance

[0485] At T0, the samples were slightly yellow, slightly opalescence liquid with a small amount of protein particles in the 8 pH buffer systems (F1-F8) . After being placed at 40℃ for 1 or 2 weeks, a large number of visible particles and turbidity were observed for F1~F2 (citric acid buffer system, pH 5.0 and pH 6.0) , F6 (histidine / hydrochloride histidine buffer system, pH 5.5) and F3~F4 (succinic acid / sodium succinate buffer system, pH 5.0 and pH 5.5) . A small amount of particles were observed for F5 (succinic acid / sodium succinate buffer system, pH 6.0) and F7~F8 (histidine / hydrochloride histidine buffer system, pH 6.3 and 6.5) and the samples were clear. After being placed at 40℃ for 4 weeks, a large number of particles were presented in all samples, but the amounts of particles for F1~F4 were relatively larger.

[0494] As shown in Table 14, compared with T0, the SE-HPLC monomers in the 8 pH buffer systems (F1-F8) decreased significantly after being placed at 40℃ for 1, 2 and 4 weeks. At the end of week 1, the SE-HPLC monomer decreased by 1.7%~23.1%, and the SE-HPLC monomer in F6~F8 (histidine buffer system, pH 5.5, pH 6.3 and pH 6.5) decreased by 1.7%-2.6%. At the end of week 2, the SE-HPLC monomer decreased by 8.2%~30.2%, and the SE-HPLC monomer in F6~F8 (histidine buffer system, pH 5.5, pH 6.3 and pH 6.5) decreased by 8.2%-10.5%. At the end of week 4, the SE-HPLC monomer decreased by 9.4%~56.2%, and the SE-HPLC monomer in F6~F8 (histidine buffer system, pH 5.5, pH 6.3 and pH 6.5) decreased by 9.4%-10.8%which was less than that of other pH buffer systems. Therefore, histidine buffer solution was preferred.

[0497] As shown in Table 15, the iCIEF main peaks of the 8 pH buffer systems (F1-F8) decreased by 11.6%-21.8%compared with T0 after being placed at 40℃ for 4 weeks. The iCIEF main peak for F7 (histidine buffer, pH 6.3) , F8 (histidine buffer, pH 6.5) , F5 (succinic acid buffer, pH 6.0) decreased by 11.6%-17.1%, which was lower than other pH buffer systems (iCIEF main peak decreased by 18.6%-21.8%) . Therefore, F7, F8 and F5 were superior.

[0500] As shown in Table 16, compared with T0, the purity of Caliper-R for 8 pH buffer systems (F1-F8) decreased by 1.0 %-4.6 %after being placed at 40 ℃ for 4 weeks. The purity of Caliper-R for F5 (succinate buffer system, pH 6.0) and F6~F8 (histidine buffer system, pH 5.5, pH 6.3 and pH 6.5) decreased by 1.0 %, which was lower than other pH buffer systems (the purity of Caliper-R decreased by 1.4 %-4.6 %) . Therefore, F5, F6, F7 and F8 showed better performances.

[0509] As shown in Table 19, compared with T0, the DAR values for 8 pH buffer systems (F1-F8) decreased by 1.84-2.95 after being placed at 40 ℃ for 4 weeks. The DAR value for F8 (histidine buffer system, pH 6.5) decreased by 1.84, which was less than other pH buffer systems (DAR value decreased by 2.19-2.95) . Therefore, F8 performed better.

[0512] Based on the DSC and the stability results at 40℃, the sample with F8 formula (20 mM histidine buffer solution, pH 6.5) had a higher Tm Onset value, and showed a good appearance under stability conditions, and had relatively higher purity of size exclusion chromatography and high stability of charge isomer, lower decrease in drug-to-antibody ratio and less free drug, so F8 formula (20 mM histidine buffer  solution, pH 6.5) was finally selected for the next round of auxiliary materials screening.

[0514] 4.1 Purpose of the experiment

[0515] The purpose of this study is to screen the optimal auxiliary materials for histidine buffer system and a IEX019-02 concentration of 40 mg / mL.

[0516] 4.2 Experimental protocol

[0517] The formula design and sampling plan for the auxiliary materials screening experiment were shown in Tables 20 and 21. Lyophilized samples of 3 sets of formula with 4 kinds of auxiliary materials were incubated under high temperature conditions for 12 weeks, and liquid samples were incubated under high temperature conditions (40℃ and 25℃ for 4 weeks) , freeze-thaw (5 cycles from room temperature to -70℃) and oscillation conditions (150 rpm, oscillation at 25℃ for 3 days) , and the optimal auxiliary materials were selected according to the stability of the three sets of formulae. The test items included appearance, pH value, molecule concentration of ADC, SE-HPLC, iCIEF, Caliper-R / NR, drug-to-antibody ratio, free drug, collapse temperature Tc, glass transition temperature Tg’ , DSC, insoluble particles (HIAC method) , moisture of lyophilized samples and binding activity (ELISA) , etc.

[0520] 4.3 Sample preparation

[0521] According to the composition of the stock solution and experimental design, IEX019-02 stock solution (40 mg / mL ADC, 20 mM histidine solution, 8% (w / v) sucrose, pH 6.5) was taken and exchanged into a solution of 20 mM histidine solution and 8.8%trehalose dihydrate (w / v) (pH 6.5) and a solution of 20 mM histidine buffer solution, 2%sucrose (w / v) , 4%mannitol (w / v) (pH 6.5) by ultrafiltration, respectively. The concentration of IEX019-02 was greater than 40mg / ml at this time. According to Table 20, the IEX019-02 was then diluted to 40 mg / mL by adding the corresponding ultrafiltration buffer, and then 5%polysorbate 80 was added to a concentration of 0.02% (w / v) , to obtain Fe01-Fe03. After the completion of the preparation, the samples for each formula were filtered with a 0.22 μm PES filter and filled into 20 mL and 6 mL vials with a filling volume of 5 mL / vial and 3 mL / vial. After filling, the 20 mL vials were semi-stoppered and lyophilized according to the procedures described in Table 22 immediately, the 6 mL vials were stoppered and capped. The samples were placed under different storage conditions according to Table 21, and at least 1 vial was placed at each sampling point for each formula.

[0523] 4.4 Results and discussion

[0524] Results of auxiliary materials screening experiment are summarized in Table 23-35.

[0525] 4.4.1 Tc, Tg’ and Tm valus of the samples

[0526] As shown in Table 23, the Tcs of 3 formulae (Fe01-Fe03) were all higher than -21℃. The Tcs of Fe01 and Fe02 were close, and Tc of Fe03 was higher and was -9.2℃. Tg' varied widely, which were -28.3℃, -26.6℃, and -32.8℃, respectively. The Tm onsets of 3 formulae (Fe01-Fe03) were all higher than 67.0℃, and the Tm values were higher than 79℃, indicating that these three formulae had good stability.

[0528] 4.4.2 Results of reconstitution time and moisture

[0529] As shown in Table 24, the reconstitution time of the lyophilized samples of the three formulae (Fe01-Fe03) varied widely, and the reconstitution time of Fe01 was relatively short. There was no significant change in the moisture of the three groups during the stability process, which was less than 0.6 %.

[0532] As shown in Table 25, the liquid samples of the three formulae (Fe01-Fe03) were slightly yellow, slightly opalescence liquid with no visible particle at T0. After 3 days of oscillation, freeze-thawing for 5 times, and being placed at 25℃ for 4 weeks, there were essentially free of visible particles or free of visible particles for the 3 formulae (Fe01-Fe03) . However, visible particles appeared after being placed at 40 ℃for 2 weeks and 4 weeks. The lyophilized samples were slightly yellow complete powder cake before reconstitution, and there were essentially free of visible particles after reconstitution.

[0541] 4.4.6 Insoluble particles (HIAC)

[0542] As shown in Table 28, there was no significant change in the insoluble particles of the liquid samples of the three formulae (Fe01-Fe03) during the five cycles of freeze-thawing compared with T0. However, the insoluble particles of Fe03 increased significantly after oscillation at 25℃ for 3 days, and the remaining samples (Fe01-Fe02) did not change significantly. The 3 formulations (Fe01-Fe03) were placed at a high temperature of 40 ℃ for 2 weeks, and the particles increased significantly. The insoluble particulates of the samples of the 3 formulae (Fe01-Fe03) increased slightly after lyophilization. After lyophilization, the samples were placed at a high temperature of 40℃ for 2 weeks, 4 weeks and 12 weeks, and there was no significant change in the insoluble particles for Fe01, while the insoluble particlesfor Fe02 and Fe03 increased significantly.

[0575] The 20 mg / mL stock solution was divided into 20 mL vials at 5.6 mL / bottle. The 40mg / ml was divided into 20mL vials at 5.6 ml / bottle. The vials were semi-stoppered and then lyophilized, and the lyophilization process is shown in Table 37. The lyophilized DP was placed under different test conditions (5℃ long-term experiment, 25℃ accelerated experiment and 40℃ high-temperature experiment) as described in Table 36 and sampled for detection at the specified time. 3 vials of DP at each time point were placed.

[0578] The results of stability experiments under long-term, accelerated and high-temperature conditions are summarized in Table 38.

[0579] The results showed that: after the samples with a IEX019-02 concentration of 20 mg / mL and 40 mg / mL were investigated at 2-8 ℃ for 12 weeks, the appearance before reconstitution did not change significantly compared with that at T0 before reconstitution, the appearance after reconstitution did not change significantly compared with that of T0, and all of the pH value, ADC concentration, reconstitution time, moisture, drug-to-antibody ratio, purity of size exclusion chromatography, purity of non-reduced capillary electrophoresis, purity of reduced capillary electrophoresis, insoluble particles, charge variants and free drugs did not change significantly, which indicated that the sample was stable under the condition of 2-8℃.However, the sample with a IEX019-02 concentration of 20 mg / mL had a shorter reconstitution time than the sample with a concentration of 40 mg / mL.
