Abstract

This invention relates to the field of food biotechnology and fermentation microbiology, specifically a method for obtaining lactic acid bacteria (LAB) isolates as starter culture candidates. The method is carried out by preparing a wheat-based fermentation substrate and adding gelatin protein as a nitrogen source to form a fermentation medium of wheat, gelatin, and fructose. Indigenous microorganisms in the substrate are inoculated into the fermentation medium and incubated under conditions that support the growth of lactic acid bacteria until lactic acid bacteria isolates are obtained. Fermentation is carried out at a temperature of 30–37°C for 12–72 hours, with a gelatin concentration of 10% (w / v) and a wheat to water ratio of 1:1 to 1:5 (w / v). The fermentation medium, a combination of wheat, gelatin, and fructose, functions as a selective medium for growing and selecting lactic acid bacteria before further isolation on standard microbiological media.This method produces lactic acid bacteria isolates that can be used as starter culture candidates in food fermentation processes.
