Abstract
Description
[Technical field]
[0001] This application claims priority to U.S. Provisional Application No. 63 / 130,010, filed December 23, 2020, and U.S. Provisional Application No. 63 / 245,288, filed September 17, 2021.
[0002] The present disclosure relates to variants or derivatives of 4-amino-3-(4-phenoxyphenyl)-1,3-dihydro-2H-imidazo[4,5-c]pyridin-2-one, and salts thereof, for use as agonists and antagonists. The present disclosure further relates to pharmaceutical compositions, methods of manufacture, and methods of treatment. [Background technology]
[0003] Small molecule drugs have proven useful for a variety of diseases and conditions. Some act as antagonists by blocking or attenuating a biological response, some act as agonists by activating a biological response, and some can function as both.
[0004] While many small molecule drugs are known, new and diverse therapeutic agents are needed. For example, according to the National Center for Complementary and Integrative Health, multiple sclerosis ("MS") affects approximately 2,500,000 people worldwide, including approximately 400,000 in the United States. Although there are several drugs available for the treatment of MS, not everyone responds well to the available drug therapies. Examples of other diseases and conditions for which there is a need include autoimmune disorders such as lupus, pemphigus vulgaris, myasthenia gravis, Sjogren's syndrome, dry eye, multiple sclerosis, Wegener's granulomatosis, autoimmune hemolytic anemia, idiopathic thrombocytopenic purpura, granulomatosis with polyangiitis, and rheumatoid arthritis.
[0005] The present disclosure provides compounds that are tyrosine kinase inhibitors, particularly Bruton's tyrosine kinase ("BTK") inhibitors, and thus are useful in the treatment of diseases such as cancer, autoimmune diseases, inflammatory diseases, and thromboembolic diseases. Also provided are pharmaceutical compositions containing such compounds and methods for producing such compounds.
[0006] BTK is a member of the Tec family of non-receptor tyrosine kinases and is essential for downstream B cell signaling from the B cell receptor. BTK is expressed in B cells and other hematopoietic cells such as monocytes, macrophages, and mast cells. BTK functions in various aspects of B cell function, maintaining the B cell repertoire (see Non-Patent Document 1). B cells play a role in other autoimmune diseases such as rheumatoid arthritis (see Non-Patent Document 2, Non-Patent Document 3, and Non-Patent Document 4), systemic lupus erythematosus, and cancer (see Non-Patent Document 5, Non-Patent Document 6, and Non-Patent Document 7).
[0007] BTK inhibitors may also have potential for the treatment of allergic diseases (Non-Patent Document 4). It has been noted that irreversible inhibitors suppress passive cutaneous anaphylaxis (PCA) induced by IgE-antigen complexes in mice. These findings are consistent with those observed in BTK mutant mast cells and knockout mice, suggesting that BTK inhibitors may be useful in the treatment of asthma, an IgE-dependent allergic disease of the airways.
[0008] Therefore, compounds that inhibit BTK are believed to be useful in treating diseases such as autoimmune diseases, inflammatory diseases, and cancer.
[0009] It has been surprisingly discovered that certain 4-amino-3-(4-phenoxyphenyl)-1,3-dihydro-2H-imidazo[4,5-c]pyridin-2-one derivatives function as agonists, antagonists, or both, of multiple receptors involved in many diseases and disorders, including those listed above. [0100] Compounds of Formula Ia and Formula Ib were synthesized as shown in the scheme above (designated Compound 13 and Compound 14, respectively) and as described in more detail below.
[0101] Step 1: Under nitrogen atmosphere, a reactor was charged with DMF (21.5 mL). 2,4-Dichloro-3-nitropyridine (SM1) (4.3 g, 22.28 mmol, 1.0 equiv) was added at 20±5° C. After stirring for 5 min, triethylamine (4.75 mL, 33.81 mmol, 1.5 equiv) was added and the mixture was cooled to 0±5° C. HOBt (301 mg, 2.23 mmol, 0.1 equiv) was added followed by a solution of tert-butyl (3R,5S)-3-amino-5-hydroxy-piperidine-1-carboxylate (4.8 g, 22.28 mmol, 1.0 equiv) in DMF (64.5 mL) added dropwise. The temperature was maintained between −10° C. and 0° C. during the addition of the reagents. The reaction mixture was stirred at 0±5° C. for 2 h and then at 20±5° C. for 4 h. HPLC analysis confirmed the production of tert-butyl (3R,5S)-3-[[2-(benzotriazol-1-yloxy)-3-nitro-4-pyridyl]amino]-5-hydroxy-piperidine-1-carboxylate (110).
[0102] Step 2: The next step was carried out immediately in one pot by charging triethylamine (4.75 mL, 33.81 mmol, 1.5 equiv) and 1-(4-methoxyphenyl)-N-[(4-methoxyphenyl)methyl]methanamine (5.7 g, 22.28 mmol, 1.0 equiv). The reaction solution was warmed to 40-45 °C and stirred at this temperature for 62 h. HPLC analysis confirmed that less than 1% intermediate remained. After cooling to 10-20 °C, EtOAc (129 mL) and water (129 mL) were added to the solution and stirred at 10-20 °C for 30 min. The organic phase was separated and washed with aqueous CaCl2 (5%, 43 mL x 5). The organic phase was concentrated under reduced pressure below 35° C. to give a solution of tert-butyl (3R,5S)-3-[[2-[bis[(4-methoxyphenyl)methyl]amino]-3-nitro-4-pyridyl]amino]-5-hydroxy-piperidine-1-carboxylate (111) (94.3% purity) in EtOAc (brown solution), which was used directly in the next step.
[0103] Step 3: tert-Butyl (3R,5S)-3-[[2-[bis[(4-methoxyphenyl)methyl]amino]-3-nitro-4-pyridyl]amino]-5-hydroxy-piperidine-1-carboxylate (111) (14.7 g, 24.76 mmol, 1 equiv.) (theoretical amount) was dissolved in AcOEt (73.5 mL) and charged to a reactor. The reactor was inerted under vacuum and filled with nitrogen 5 times, wet 10% Pd / C (2.64 g, 20 wt.%) was added, the reactor was inerted under vacuum and filled with nitrogen 5 times, then the reactor was inerted under vacuum and filled with hydrogen 3 times. The reaction was kept under hydrogen pressure at 20±5° C. and stirred for 22 h. HPLC analysis confirmed the disappearance of starting material. The suspension was filtered and the cake was washed with EtOAc (73.5 mL×5). The filtrate was concentrated under reduced pressure to give tert-butyl (3R,5S)-3-[[3-amino-2-[bis[(4-methoxyphenyl)methyl]amino]-4-pyridyl]amino]-5-hydroxy-piperidine-1-carboxylate (112) as a yellow foamy solid (12 g, yield=87%, purity 91.1%), which was used in the next step without further purification.
[0104] Step 4: A reactor was charged with THF (60 mL), tert-butyl (3R,5S)-3-[[2-[bis[(4-methoxyphenyl)methyl]amino]-3-nitro-4-pyridyl]amino]-5-hydroxy-piperidine-1-carboxylate (112) (12.0 g, 20.21 mmol, 1.0 equiv.) and triethylamine (4.4 mL, 40.42 mmol, 2.0 equiv.). The solution was stirred at 20±5°C for 10 min, then triphosgene (1.07 mL, 6.46 mmol, 0.32 equiv.) was added in three portions and the resulting solution was stirred at 20-25°C for 1.5 h. [0119] Synthesis of (R)-5-acrylamido-4-(4-amino-2-oxo-3-(4-phenoxyphenyl)-2,3-dihydro-1H-imidazo[4,5-c]pyridin-1-yl)pentanoic acid [ka]
[0120] The compound of formula IV (referred to above as compound (6)) was prepared as shown in the scheme above and as described in more detail below. The starting material, 4-amino-3-(4-phenoxyphenyl)-1-[(3R)-3-piperidyl]imidazo[4,5-c]pyridin-2-one (1), was prepared according to the procedure described in Example 2 of U.S. Pat. No. 9,688,676. 4-amino-3-(4-phenoxyphenyl)-1-[(3R)-3-piperidyl]imidazo[4,5-c]pyridin-2-one (1) was then converted to the oxalate salt by adding DCM to compound (1). The solution was exchanged with EtOH and the resulting solution was reacted with oxalic acid. After filtration, the crude solid was slurried in MTBE, filtered, and dried to obtain the oxalate salt of compound (1).
[0121] Step 1: Under argon atmosphere, a flask was charged with CHCl and the oxalate salt of 4-amino-3-(4-phenoxyphenyl)-1-[(3R)-3-piperidyl]imidazo[4,5-c]pyridin-2-one (1.3 g, 2.66 mmol, 1 eq.). To this suspension was then added DMAP (487 mg, 3.98 mmol, 1.5 eq.), DIEA (1.35 mL, 7.97 mmol, 3 eq.) and BocO (2.90 g, 13.28 mmol, 5 eq.). The starting material dissolved and the reaction became clear. The solution was stirred at room temperature for 20 h. The reaction was hydrolyzed with water and extracted with CHCl. The organic phase was separated, dried over MgSO, filtered and concentrated under reduced pressure to give the crude product. The crude product was purified by flash chromatography (SiO2, Hep / AcOEt (1 / 1)) to give tert-butyl (3R)-3-[4-[bis(tert-butoxycarbonyl)amino]-2-oxo-3-(4-phenoxyphenyl)imidazo[4,5-c]pyridin-1-yl]piperidine-1-carboxylate (2) (1.86 g, yield=91.2%).
[0122] Step 2: To a solution of tert-butyl (3R)-3-[4-[bis(tert-butoxycarbonyl)amino]-2-oxo-3-(4-phenoxyphenyl)imidazo[4,5-c]pyridin-1-yl]piperidine-1-carboxylate (2) (1 g, 1.42 mmol, 1 equiv.) in MeCN / HO / CCl (15 mL / 20 mL / 15 mL) was added NaIO (1.22 g, 5.7 mmol, 4 equiv.) and RuCl.xHO (32 mg, 142 μmol, 0.1 equiv.) in sequence. The reaction was a light brown mixture. After stirring at room temperature for 1 h, additional NaIO (152 mg, 0.7 mmol, 0.25 equiv.) was added. The reaction was stirred at room temperature for 24 h, at which point it turned dark brown. After hydrolysis with aqueous Na2S2O5 and stirring for 30 min, the solution turned pale yellow, which was extracted with AcOEt, the organic phase was separated, dried over MgSO4, filtered and concentrated under reduced pressure to give the crude product. The crude product was purified by flash chromatography (SiO2, Hep / AcOEt (1 / 1)) to give tert-butyl (3R)-3-[4-[bis(tert-butoxycarbonyl)amino]-2-oxo-3-(4-phenoxyphenyl)imidazo[4,5-c]pyridin-1-yl]piperidine-1-carboxylate (starting material) (450 mg, yield=45%), and tert-butyl (5R)-5-[4-[bis(tert-butoxycarbonyl)amino]-2-oxo-3-(4-phenoxyphenyl)imidazo[4,5-c]pyridin-1-yl]-2-oxo-piperidine-1-carboxylate (desired product) (3) (200 mg, yield=20%).
[0123] Step 2: Electrochemical process - Using an Electrasyn 2.0 (IKA) equipped with a 20 mL vial setup, to a solution of tert-butyl (3R)-3-[4-[bis(tert-butoxycarbonyl)amino]-2-oxo-3-(4-phenoxyphenyl)imidazo[4,5-c]pyridin-1-yl]piperidine-1-carboxylate (2) (300 mg, 0.43 mmol, 1 equiv.) in MeCN (12 mL) was added quinuclidine (2 equiv., 0.86 mmol, 95 mg) as a mediator, 1,1,1,3,3,3-hexafluoropropan-2-ol ("HFIP") (11 equiv., 4.70 mmol, 790 mg), and tetramethylammonium tetrafluoroborate (2.05 equiv., 0.88 mmol, 141 mg) as an electrolyte. Vial cap set: (working electrode) RVC-Reticulated Vitreous Carbon Foam was used as the anode and Ni Foam was used as the cathode. The electrochemical reaction was carried out in constant current mode; a constant current of 8 mA without polarity switching. The total charge was 16 Faradays / mol at room temperature. The reaction was monitored by LC / MS. After the reaction was poured into ice water, a 1M solution of sodium thiosulfate was added. The aqueous phase was washed with ethyl acetate. The organic phase was separated, dried over sodium sulfate, filtered, and concentrated under reduced pressure to obtain the crude product. Purification by flash chromatography on silica gel, eluting with ethyl acetate / pentane (70 / 30) gave the expected product tert-butyl (5R)-5-[4-[bis(tert-butoxycarbonyl)amino]-2-oxo-3-(4-phenoxyphenyl)imidazo[4,5-c]pyridin-1-yl]-2-oxo-piperidine-1-carboxylate (3) (150 mg, yield=49%).
[0124] Step 3: To a solution of tert-butyl (5R)-5-[4-[bis(tert-butoxycarbonyl)amino]-2-oxo-3-(4-phenoxyphenyl)imidazo[4,5-c]pyridin-1-yl]-2-oxo-piperidine-1-carboxylate (3) (820 mg, 1.15 mmol, 1 equiv.) in THF / H2O (30 mL / 30 mL) was added LiOH (471 mg, 19.4 mmol, 17 equiv.). The solution was stirred at room temperature for 3 h. The reaction mixture was hydrolyzed with 1N aqueous HCl until pH=1-2 and then extracted with AcOEt. The organic phase was separated, dried over MgSO4, filtered and concentrated under reduced pressure to give the desired compound: (4R)-4-[4-[bis(tert-butoxycarbonyl)amino]-2-oxo-3-(4-phenoxyphenyl)imidazo[4,5-c]pyridin-1-yl]-5-(tert-butoxycarbonylamino)pentanoic acid (4) (780 mg, yield=92.8%).
[0125] Step 4: Under an argon atmosphere, a flask was charged with CHCl (5 mL), (4R)-4-[4-[bis(tert-butoxycarbonyl)amino]-2-oxo-3-(4-phenoxyphenyl)imidazo[4,5-c]pyridin-1-yl]-5-(tert-butoxycarbonylamino)pentanoic acid (720 mg, 0.98 mmol, 1 equiv.) (4), and TFA (2 mL). The mixture was stirred at room temperature for 6 h. After evaporating CHCl and TFA and drying under high vacuum for 10 h, (4R)-5-amino-4-[4-amino-2-oxo-3-(4-phenoxyphenyl)imidazo[4,5-c]pyridin-1-yl]pentanoic acid (5) (410 mg, 96%) was obtained, which was used directly in the next step without further purification.
[0126] Step 5: To a suspension of (4R)-5-amino-4-[4-amino-2-oxo-3-(4-phenoxyphenyl)imidazo[4,5-c]pyridin-1-yl]pentanoic acid (5) (425 mg, 0.98 mmol, 1 equiv.) and CHCl (6 mL) was added DMF (0.5 mL) to solubilize the starting material. The reaction solution was then cooled to 0° C. and charged with DIEA (343 μL, 1.96 mmol, 2 equiv.) and a solution of acryloyl chloride (41 μL, 0.49 mmol, 0.5 equiv.) in CHCl (2 mL) was added dropwise. The solution was allowed to warm to room temperature and stirred for 2 h. The reaction was concentrated under reduced pressure and purified directly by preparative HPLC followed by supercritical HPLC to give (4R)-4-[4-amino-2-oxo-3-(4-phenoxyphenyl)imidazo[4,5-c]pyridin-1-yl]-5-(prop-2-enoylamino)pentanoic acid (compound of formula IV, also referred to above as (6)) (180 mg, yield=38%, purity 98%). 1H NMR (500 MHz, DMSO-d6) δ ppm 2.02 (m, 1 H), 2.17 (m, 2 H), 2.28 (m, 1 H), 3.65 (m, 2 H), 4.36 (m, 1 H), 4.78 (s, 2 H), 5.53 (dd, J=8 , 2Hz , 1 H), 6.02 (dd, J=17 , 2 Hz, 1 H), 6.09 (dd, J=17 , 8 Hz ,1 H), 6.65 (m, 1 H), 7.15 (d, J=8 Hz, 4 H), 7.21 (t, J=7 Hz, 1 H), 7.45 (m, 4 H), 7.71 (d, J=5 Hz, 1 H), 8.28 (t, J=6 Hz, 1H) LC / MS: 488.2 (M+H) + / 486.1 (MH) - .
