Description

[Technical field]

[0001] Related Applications This application claims the benefit of U.S. Provisional Patent Application No. 63 / 173,611, filed April 12, 2021, the contents of which are incorporated by reference in their entirety herein.

[0002] FIELD OF THEINVENTION The present invention relates to compositions and methods for efficiently detecting pH changes using solvatochromic indicators. In a specific embodiment, the present invention also relates to efficiently detecting infection at a surgical site based on visual detection of pH changes. [Background technology]

[0003] background Surgical site infections are one of the most common causes of hospital-acquired or nosocomial infections. These infections are generally caused by gram-positive bacterial species such as Staphylococcus aureus and Streptococcus pyogenes, and gram-negative bacteria such as Pseudomonas aeruginosa and Escherichia coli. Clinical symptoms may include localized redness, pain, wound drainage, delayed healing, swelling, and fever.

[0004] Millions of surgical procedures are performed in inpatient hospitals, outpatient affiliated hospitals, or free-standing settings. Surgical site infections can occur in up to 5% of these surgical procedures, and infections are associated with significant morbidity and mortality. Moreover, it often leads to increased hospital stays and additional costs due to prolonged antibiotic therapy.

[0005] Core guidelines for surgical procedures often include recommendations for preventing surgical site infections that are often generalized across surgical procedures. Among these recommendations, administration of preoperative antimicrobials before skin incision, thorough cleaning of the skin before and after surgery, and wound monitoring are the most commonly used methods. Additionally, hospitals around the world have adjusted their surgical protocols in an attempt to reduce the likelihood of surgical site infections. However, protocols for checking incisions for obvious signs of infection have remained unchanged, despite the fact that it is very difficult to proceed once signs of infection have occurred, and therefore why surgical site infections are so dangerous. If a surgical site infection is detected early, the wound can be treated with antibiotics as opposed to the need for additional surgery. These concerns apply broadly to all types of wound sites, especially in developing countries where medical care can be difficult to obtain.

[0006] For this purpose, there is a need for a method for efficiently detecting bacterial infection in both clinical and non-clinical environments. The disclosed compositions, methods and uses provide for efficiently detecting pH changes in medical, biomedical, veterinary or clinical environments, or any other environments that need to be kept free of bacteria.Detecting pH changes can be applied to visually detect infection in wound sites, such as surgical sites.Disclosed herein is a novel method for detecting pH changes, and therefore bacterial infection, in any environment.

[0007] Additionally, there is a need for inexpensive, easily accessible tools to diagnose infection in developing countries, where people may not be able to see a doctor, and the use of simple, widely applicable compositions to indicate infection would allow a person to know that medical attention is needed. [0104] Having now generally described the present disclosure, the same will be more readily understood by reference to the following examples, which are provided by way of illustration and are not intended to be limiting of the disclosure, unless otherwise specified. EXAMPLES

[0105] Example 1: Determine viable materials that change color with pH The bulb of at least one sugar beet plant is cut into pieces of approximately 200 g. The pieces are added to 400 milliliters (ml) of distilled water in a beaker and boiled at 400° C. A polyester cotton blend filament material is cut into 2 meter long strings and stirred into the boiling mixture and boiled for at least 30 minutes with frequent stirring. After boiling, the filaments are removed and dried on a non-absorbent surface. This process is repeated with various types of materials including yarn, polyester, cotton, vicryl, and monocryl.

[0106] After drying, each of the filament materials is treated with a solution that is either pH 5, pH 7 (neutral, control), or pH 9 to determine the intensity of the visual response during the solvatochromic reaction. The solution appears red at pH 5 and purple at pH 9. See FIG. 1A. The material treated with the pH 9 solution showed a color response that clearly distinguished between pH 9 and pH 5. The polyester cotton blend showed the most robust color response. See FIG. 1B and C. The yarn (75% viscose and 25% polyester) also showed a robust color change at pH 9. The color change response of the sugar beet extract is further tested by adding the extract to a cotton swab and testing against pH 5 and pH 9 solutions with the addition of water and 91% isopropyl alcohol as well. See FIG. 2. The addition of 91% isopropyl alcohol does not affect the color change response and aids in the solubility of the extract containing solution. The addition of extra water may reduce the color change response. See FIG. 2.

[0107] Example 2: Preparation of sugar beet extract A sugar beet extract is made by cutting 250 grams of sugar beet bulbs into small cubes. The sugar beet cubes are added to 600 ml of distilled water and boiled at 400° C. for at least 45 minutes. The mixture is removed from the heat, cooled, and then blended in an emulsion blender. Alternatively, the mixture can be sonicated or subjected to an equivalent treatment. The mixture is then returned to the heat and boiled for at least 30 minutes. The mixture is then removed from the heat and cooled. 100 ml of 91% isopropyl alcohol is added and the mixture is poured into a strainer to remove any remaining solid material. The filtration step can be repeated as necessary until all solid material is removed.

[0108] Example 3: Preparation of turmeric extract An extract of turmeric is made by cutting 500 grams of turmeric root into small cubes. The turmeric cubes are added to 1200 ml of distilled water and boiled at 400°C for at least 45 minutes. The filtration step can be repeated as necessary until all solid material has been removed.

[0109] Example 4: Preparation of Solvatochromic Mixtures Sugar beet extract and turmeric extract are prepared as described in Examples 2 and 3, respectively. A solvatochromic mixture containing approximately 10% isopropyl alcohol is prepared by mixing equal amounts of sugar beet extract and turmeric extract. The solvatochromic mixture can be used in a similar manner as the sugar beet extract described in Example 1.

[0110] Example 5: Solvatochromic mixtures contain antibacterial properties To determine whether the solvatochromic mixtures of Examples 1 and 2 have antibacterial properties, sugar beet bulb extracts were exposed to E. coli in culture. Petri dishes containing LB agar or similar are prepared using standard procedures. The petri dishes are inoculated with E. coli using standard laboratory procedures. Filament material is treated with the solvatochromic mixtures of Examples 1, 2, 3, or 4 and boiled together to sterilize. After a short cooling period, the filament material is placed on one half of the petri plate, the other half being a positive growth control. The plate is incubated at 37° C. for 12 hours. After incubation, the positive growth control side of the petri plate shows normal growth, while the side treated with the solvatochromic mixture shows no growth, confirming the antibacterial properties of the solvatochromic mixture. See FIG. 3.

[0111] Example 6: Development of materials containing solvatochromic mixtures Any form of sterile filament can be used to create a material containing a solvatochromic indicator that is useful for diagnosing bacterial infection in a biomedical environment or any other environment where bacterial growth is a problem, such as the food service industry.

[0112] For example, any material suitable for sutures can be used to make sutures containing a solvatochromic mixture. The filament material is added to a tube containing 35 ml of the solvatochromic mixture of Example 4. The material is immersed in the solvatochromic mixture at room temperature for 48 hours. After 48 hours, the material is removed from the solvatochromic mixture and allowed to dry on a non-absorbent surface.

[0113] Example 7: Thin film coating of treated filaments Materials made with the solvatochromic mixture in Example 5 can be treated with a thin film coating to confine the solvatochromic mixture while allowing interaction with the external environment. For example, a thin film coating of Parylene C (FIG. 4A) has been shown to work well with the solvatochromic mixture of the present disclosure, confine the solvatochromic mixture while being permeable enough to allow detection of pH in the external environment. Briefly, any deposition process can be used to apply the thin film coating. Here, the suture treated with the solvatochromic mixture is coated using a VSi Parylene C deposition process (FIG. 4B, VSi Parylene, Broomfield Colorado, 80021). Parylene C is deposited using a liquid-promoted adhesion protocol and subsequent coating as shown in FIG. 4B.

[0114] Parylene C treated sutures are sewn into a material that mimics skin. Three sets of sutures are set up, one treated with pH 5 solution, one treated with pH 9 solution, and one control. See Figure 5A-C. A solution of appropriate pH is added to the sutures as time zero, the control is untreated. See Figure 5D-F. After 10 minutes, a color change reaction can be seen in response to the pH 9 solution, while the pH 5 solution does not cause a color change (compare Figure 5G and 5I). Note that Parylene C still allows absorption of the pH 9 solution that has diffused underneath the artificial membrane. This demonstrates that the thin film coating does not affect uptake into the suture and subsequent color change (see subtle color change in the control stitch, Figure 5H).

[0115] Example 8: Identification of additional solvatochromic indicators useful for diagnosing bacterial infections Any material with solvatochromic properties that allow for a visible color change reaction in response to a basic pH environment can be used to create a solvatochromic indicator for use in the methods disclosed herein. For example, purple cabbage (Brassica oleracea, which has purple or red leaves), also known as red cabbage or blaukraut, has suitable properties and has been shown to cause a strong color change in response to a pH 9 environment. See Figure 6A.

[0116] 250 grams of purple cabbage leaves are cut into small strips and added to 600 ml of 400° C. distilled water and boiled for at least 45 minutes. The strips are removed from the heat and allowed to cool for 10 minutes. After the cooling period, an emulsion blender is used to disrupt the cell membranes. Sonication or any equivalent procedure can also be used to create the purple cabbage leaf extract. The mixture is then returned to the heat and boiled for an additional 30 minutes. After cooling for at least 10 minutes, 100 ml of 91% isopropyl alcohol is added to the solution. The mixture is poured into a strainer to remove any remaining solid material. The filtration step can be repeated as necessary until all solid material has been removed. The cabbage extract can be used alone or in combination with the sugar beet extract. See Figures 6A and 6B.
