Abstract
Description
[Technical field]
[0001] (CROSS REFERENCE TO RELATED APPLICATIONS) This application claims priority to USSN 62 / 955,073, filed December 30, 2019, USSN 62 / 955,062, filed December 30, 2019, USSN 62 / 968,695, filed January 31, 2020, and USSN 62 / 968,724, filed January 31, 2020, the contents of each of which are incorporated herein by reference in their entirety. [Background technology]
[0002] c-KIT (also known as KIT, CD117, and stem cell factor receptor) is a 145 kDa transmembrane tyrosine kinase protein that acts as a type III receptor. The c-KIT proto-oncogene, located on chromosome 4q11-21, encodes the c-KIT receptor, whose ligands are stem cell factor (SCF), hematopoietic stem cell factor, kit ligand, and mast cell growth factor). The receptor has tyrosine protein kinase activity, and binding of the ligand SCF leads to the autophosphorylation of c-KIT and its association with substrates such as phosphatidylinositol 3-kinase (PI3K). Tyrosine phosphorylation by protein tyrosine kinases is particularly important in cell signaling and can mediate signals for key intracellular processes such as proliferation, survival, differentiation, apoptosis, adhesion, invasion, and migration. Deficiency of c-KIT is the cause of focal albinism, an autosomal dominant developmental abnormality of pigmentation characterized by congenital patches of white skin and hair lacking melanocytes. Gain-of-function mutations in the c-KIT gene and expression of constitutively phosphorylated c-KIT are found in most gastrointestinal stromal tumors (GISTs) and mastocytosis. In addition, nearly all gonadal seminomas / dysgerminomas exhibit c-KIT membrane staining, and several reports have clarified that a proportion (10–25%) harbor c-KIT gene mutations. c-KIT deficiency has also been associated with testicular tumors, including germ cell tumors (GCTs) and testicular germ cell tumors (TGCTs). C-KIT mutations have also been associated with a subgroup of cutaneous or acral melanomas.
[0003] Oncogenic genomic alterations in PDGFRα kinase or overexpression of PDGFRα kinase have been shown to be causative in human cancers. Missense mutations in PDGFRα kinase have also been shown to be causative in a subgroup of GISTs. PDGFRα mutations are the inducing factor in oncogenesis in approximately 8–10% of GISTs. The dominant mutation in PDGFRα is exon 18 D842V, but other exon 18 mutations including D846Y, N848K, and Y849K, as well as exon 18 insertion-deletion mutations (indels) including RD841-842KI, DI842-843-IM, and HDSN845-848P, have also been reported. In addition, rare mutations in exons 12 and 14 of PDGFRα have also been reported. PDGFRα exon 18 deletion mutations ΔD842-H845 and ΔI843-D846 have been reported in GISTs. PDGRFα amplification or mutations have also been reported in human tissues with malignant peripheral nerve sheath tumors (MPNSTs). PDGFRα amplification has been reported in multiple cutaneous lesions of undifferentiated pleomorphic sarcoma and in intimal sarcomas. PDGFRα amplification has been linked to a subset of lung cancer patients. 4q12, which contains the PDGFRα locus, is amplified in 3-7% of lung adenocarcinomas and 8-10% of lung squamous cell carcinomas. PDGFRα amplification is universal in pediatric and adult high-grade astrocytomas and identifies a poor prognostic group in IDH1-mutated glioblastomas. PDGFRα amplification was frequent in pediatric (29.3%) and adult (20.9%) tumors. PDGFRα amplification was associated with increased grade and, notably, poor prognosis in IDH1-mutated de novo GBM. It has been reported that the PDGFRα locus in this PDGFRα-amplified glioma has been shown to exhibit an intragenic deletion rearrangement of exons 8 and 9 of PDGFRα. This intragenic deletion is ubiquitous and is present in 40% of glioblastoma multiforme (GBM) cases that exhibit PDGFRα amplification. Tumors with this rearrangement exhibit histological characteristics of oligodendroglioma, and the intragenic deletion of PDGFRα exons 8 and 9 shows elevated constitutive tyrosine kinase activity. The FIP1L1-PDGFRA fusion protein is oncogenic in a subgroup of patients with hypereosinophilic syndrome. The FIP1L1-PDGFRα fusion has also been identified in eosinophilic acute myeloid leukemia, and lymphoblastic T-cell lymphoma.
[0004] Such broad-spectrum c-KIT inhibitors and their formulations would be of high therapeutic value in the treatment of patients with refractory GIST and other disorders. There is a need for oral formulations that provide patients with a significantly more stable product. Mutations, deletions, rearrangements and amplifications of the PDGFRα gene have been associated with many solid and hematological cancers. Given the complex function of the PDGFRα gene and the potential utility of PDGFRα inhibitors in the treatment of various solid and hematological cancers, there is a need for oral formulations of the inhibitor with good therapeutic performance. [0063] In general, solid forms, such as crystalline or amorphous forms, may be referred to herein as being characterized by graphical data, "as depicted in" or "substantially depicted in" FIG. 1. Such data include, for example, powder X-ray diffractograms and solid-state NMR spectra. As is well known in the art, graphical data can potentially be interpreted in a manner that is not necessarily consistent with reference to numerical values or peak positions alone. The data provide additional technical information to further define each solid form (so-called "fingerprint"), which cannot necessarily be described. In any case, those skilled in the art will be aware that the graphical representation of such data may be subject to small variations in peak relative intensity and peak position due to certain factors, such as, but not limited to, variations in instrument response and variations in sample concentration and purity. Crystals are composed of atoms arranged periodically in 3D space, while in amorphous materials atoms are randomly distributed in 3D space. As a result, the X-ray diffraction pattern of a crystalline material will show narrow peaks of high intensity due to the fact that the X-rays are scattered in only certain directions (due to the periodic arrangement of the atoms). In contrast, the X-ray diffraction pattern of an amorphous material will generally show broad peaks of low intensity (halo pattern) because the X-rays are scattered in many different directions, resulting in large ridges distributed over a wide range (2 theta).
[0063]
[0064] All ranges recited herein include endpoints, including those reciting a range "between" two values. The terms "substantially" and "about" are to be construed as modifying a term or value so as not to make it absolute. This includes, at a minimum, the degree of expected experimental error, technique error, and instrument error for a given technique used to measure the value.
[0064]
[0065] The present invention relates to an amorphous solid dispersion of the amorphous form of the compound of formula (I), optionally in a polymer matrix and the amorphous form of the compound of formula (I). The amorphous nature of the solid dispersion and the amount of crystalline drug can be determined by powder X-ray diffraction (PXRD), scanning electron microscope (SEM) analysis, differential scanning calorimetry (DSC), or any other standard quantitative measurement. For example, a material is X-ray amorphous and no sharp peaks are observed in its PXRD pattern, but rather is described as a halo-like pattern. A sharp peak in the pattern indicates the presence of crystalline material. The amorphous form of the compound of formula (I) can be characterized by an X-ray diffraction pattern substantially as shown in FIG. 1.
[0065]
[0066] The amorphous drug may exist within the solid amorphous dispersion as a pure phase, as a solid solution of drug homogeneously distributed throughout the polymer, or any combination of these states or states in between. The dispersion is preferably substantially homogeneous, such that the amorphous drug is dispersed as homogeneously as possible throughout the polymer. As used herein, "substantially homogeneous" means that a relatively small proportion of the drug is present in relatively pure amorphous domains within the solid dispersion.

[0097] spray dried dispersion
[0098] As described herein, the dispersion of an active agent and a pharma- ceutically acceptable polymer is made by a spray drying process. As used herein, the term "spray dried dispersion" or "spray dried powder dispersion" refers to the product of a spray drying process, the product comprising a dispersion of at least one active agent and at least one excipient, such as a polymer. In some embodiments, the polymer is selected from the group consisting of polyvinylpyrrolidone, polyethylene oxide, polyethylene glycol, poly(vinylpyrrolidone-co-vinyl acetate), polyoxyethylene-polyoxypropylene block copolymers, graft copolymers (composed of polyethylene glycol, polyvinylcaprolactam and polyvinyl acetate), polymethacrylates, polyoxyethylene alkyl ethers, polyoxyethylene castor oil, polycaprolactam, polylactic acid, polyglycolic acid, poly(lactic-glycolic) acid, lipids, sucrose, glycerol ... cellulose, pullulan, dextran, maltodextrin, hyaluronic acid, polysialic acid, chondroitin sulfate, heparin, fucoidan, polysulfate pentosan, spirulan, hydroxypropyl methylcellulose acetate succinate, hydroxypropyl methylcellulose propionate succinate, hydroxypropyl methylcellulose phthalate, cellulose acetate phthalate, cellulose acetate trimellitate, methylcellulose acetate phthalate, hydroxypropyl cellulose acetate phthalate, cellulose acetate terephthalate, cellulose acetate isophthalate, carboxymethylethylcellulose, hydroxypropyl methylcellulose, hydroxypropyl methylcellulose acetate phthalate, hydroxypropyl methylcellulose propionate phthalate, hydroxypropyl methylcellulose acetate trimellitate, hydroxypropyl methylcellulose propionate trimellitate, cellulose acetate succinate, methylcellulose acetate succinate, The pharma- ceutically acceptable polymer is selected from the group consisting of dextran, dextran acetate, dextran tropionate, dextran succinate, dextran acetate propionate, dextran acetate succinate, dextran tropionate succinate, dextran acetate propionate succinate, poly(methacrylic acid-co-methyl methacrylate) 1:1, poly(methacrylic acid-co-methyl methacrylate) 1:2, poly(methacrylic acid-co-ethyl acrylate) 1:1, hydroxyethyl cellulose, methyl cellulose and hydroxypropyl cellulose, polymethacrylic acid-ethyl acrylate, polymethacrylic acid-methyl methacrylate, polymethyl methacrylate-ethyl acrylate, polytrimethylammonioethyl methacrylate, methyl methacrylate-ethyl acrylate and poly(butyl methacrylate-co-(2-dimethylaminoethyl) methacrylate-co-methyl methacrylate), and mixtures thereof.
[0098]
[0099] In the spray drying process, the active agent and one or more polymers are dissolved in a common solvent. Here, "common" means that the solvent, which can be a mixture of compounds, dissolves both the active agent and the polymer(s). After both the active agent and the polymer are dissolved, the solvent is rapidly removed by evaporation in the spray drying apparatus, resulting in the formation of a substantially uniform solid dispersion. In such a dispersion, the active agent is dispersed as uniformly as possible throughout the polymer, and can be considered as a solid solution of the active agent dispersed in the polymer(s).
[0099]
[0100] The solvent is removed by the spray drying process. The term "spray drying" is conventionally used and broadly refers to a process that involves breaking up a liquid mixture into small droplets (atomization) and rapidly removing the solvent from the mixture in a spray drying apparatus where there is a strong driving force for evaporation of the solvent from the droplets. The spray drying process and spray drying equipment are generally described in Perry's Chemical Engineers' Handbook, pages 20-54 to 20-57 (Sixth Edition 1984). For more information on the spray drying process and equipment, see Marshall, "Atomization and Spray-Drying," 50 Chem. Eng. Prog. Monogr. Series 2 (1954) , and Masters, Spray Drying Handbook (Fourth Edition 1985). Additionally, additional process and spray drying techniques and equipment are generally described in US8.343.550 and US7,780,988, the contents of which are incorporated herein by reference in their entirety for all purposes. A strong driving force for solvent evaporation is generally provided by maintaining the partial pressure of the solvent in the spray drying apparatus well below the vapor pressure of the solvent at the temperature of the drying droplets. This is accomplished by (1) maintaining the pressure in the spray drying apparatus at a partial vacuum (e.g., 0.01 to 0.50 atm), or (2) mixing the droplets with a warm drying gas, or (3) both (1) and (2). In addition, a portion of the heat required for solvent evaporation may be provided by heating the spray solution.
[0100]
[0101] The drying gas can be virtually any gas, but inert gases such as nitrogen, nitrogen-enriched air, or argon are utilized to minimize the risk of fire or explosion due to ignition of flammable vapors and to minimize undesirable oxidation of the drug, concentration enhancing polymer, or other materials in the dispersion. The temperature of the drying gas at the gas inlet of the apparatus is typically about 60°C to about 300°C. The temperature of the product particles, drying gas, and evaporated solvent at the outlet or distal end of the collection cone typically ranges from about 0°C to about 100°C.
[0101]
[0102] A suitable solvent for the spray drying process can be any organic compound in which the active agent and the polymer are mutually soluble. The solvent should be relatively low toxic and removed from the dispersion to an acceptable level according to the International Committee on Harmonization (ICH) guidelines. Removal of the solvent to this level may require a subsequent processing step such as tray drying or secondary drying. In some embodiments, the solvent is an alcohol, a ketone, an ether, an ester, a halogenated alkane, an amide, a sulfone, an acid, or a nitro compound. In some embodiments, the solvent is methanol, ethanol, n-propanol, isopropanol, or butanol. In some embodiments, the solvent is acetone, methyl ethyl ketone (MEK), or methyl isobutyl ketone (MIBK). In some embodiments, the solvent is methyl acetate, ethyl acetate, or propyl acetate. In some embodiments, the solvent is diethyl ether, tetrahydrofuran (THF), 2-methyl THF, 2,5-dimethyl THF, or 2,2,5,5-tetramethyl THF. In some embodiments, the solvent is acetonitrile, methylene chloride, toluene, 1,1,1-trichloroethane, dimethylacetamide (DMA), nitromethane, acetic acid, or dimethylsulfoxide (DMSO). Mixtures of solvent and water are suitable as long as the polymer and compound of formula (I) are sufficiently soluble to make a spray-drying process practical. In some embodiments, the water:solvent mixture is water:acetone. In some embodiments, the water:solvent mixture is water:THF. In some embodiments, the water:solvent mixture is water:methanol. In some embodiments, the water:solvent mixture is water:ethanol. In some embodiments, the water:solvent mixture is water:methyl ethyl ketone. In some embodiments, the solvent:solvent mixture is methanol:ethyl acetate. In some embodiments, the solvent is a solvent mixture ethanol:ethyl acetate. In some embodiments, the solvent:solvent mixture is methanol:dichloromethane. In some embodiments, the solvent:solvent mixture is ethanol:dichloromethane.
[0102]
[0103] The composition of the solvent-borne feed depends on the desired ratio of drug to polymer in the dispersion and the solubility of the drug and polymer in the solvent. [0104] The average residence time of the particles in the drying chamber should be at least 10 seconds, preferably at least 20 seconds. Typically, after solidification, the formed powder remains in the spray drying chamber for about 5-60 seconds to allow further evaporation of the solvent. The final solvent content of the solid dispersion upon exiting the dryer should be low to reduce the mobility of the drug molecules in the dispersion, thereby improving stability. Generally, the solvent content of the dispersion upon leaving the spray drying chamber should be less than about 10% by weight. In some embodiments, the solvent content of the dispersion upon leaving the spray drying chamber is less than about 9% by weight. In some embodiments, the solvent content of the dispersion upon leaving the spray drying chamber is less than about 8% by weight. In some embodiments, the solvent content of the dispersion upon leaving the spray drying chamber is less than about 7% by weight.
