Description

[Technical field]

[0001] The present disclosure relates to the large-scale manufacture of pharmaceutical compounds and novel intermediates for use in the manufacture thereof. [Background technology]

[0002] International Patent Application WO 2005 / 023633 discloses morpholinopyrimidines useful for treating cancer, their preparation methods, and pharmaceutical compositions thereof. In particular, WO 2005 / 023633 discloses the compound 4-{4-[(3R)-3-methylmorpholin-4-yl]-6-[1-((R)-S-methylsulfonimidoyl)cyclopropyl]pyrimidin-2-yl}-1H-pyrrolo[2,3-b]pyridine (hereinafter referred to as compound of formula (I)) as experimental example 2.02 on page 60. The structure of compound of formula (I) is shown below. [ka]

[0003] The synthetic route for compounds of formula (I) is described on pages 51-57, 66 and 67 of WO 2005 / 023391 and is summarized in Scheme 1 below. [ka] [Prior art documents] [Patent documents]

[0004] [Patent Document 1] International Publication No. 2011154737 Brochure Summary of the Invention [Problem to be solved by the invention]

[0005] There are several potential disadvantages to the route to compounds of formula (I) shown in Scheme 1. For example, out of a total of 15 steps in Scheme 1 (the starting compound (R)-3-methylmorpholine is commercially available, but there are five synthetic steps to obtain this compound), the longest linear sequence is made up of 14 steps, which is non-convergent and may not be desirable on an industrial scale. In addition, the cyclopropanation step is extremely challenging, requiring a continuous stirred tank process with difficult workup, only moderate yields, and many impurities formed. The introduction of reactive sulfoximines early in the scheme also poses significant problems for subsequent cyclopropanation and Suzuki coupling. The scheme also requires the early introduction of expensive chiral morpholines. In addition, the use of rhodium in the sulfoximine formation step can be expensive. Taken together or individually, these potential disadvantages may make the route shown in Scheme 1 less attractive for use on an industrial scale at reasonable cost.

[0006] The compound of formula (I) is being developed as an active pharmaceutical compound for treating cancer. Therefore, suitable methods for the safe, cost-effective, efficient and environmentally friendly manufacture of the compound of formula (I) are desirable. In view of the potential disadvantages highlighted above, it is desirable to develop a more efficient, economical and shorter route, i.e., one with fewer steps. [Means for solving the problem]

[0007] Applicants have now discovered a route to synthesize the cyclopropyl key intermediate of formula (II), depicted in Scheme 2 below, where LG1 is a leaving group. [ka] DETAILED DESCRIPTION OF THE PREFERRED EMBODIMENTS

[0130] Where 1H NMR spectra are given, they were recorded on a Bruker DRX500 (500 MHz), Bruker 400 (400 MHz). Either the central peak of chloroform-d (CDCl3; δH 7.27 ppm) or dimethylsulfoxide-d6 (d6-DMSO; δH 2.50 ppm), or the internal standard of tetramethylsilane (TMS; δH 0.00 ppm) was used as a reference.
