Description

[Technical field]

[0001] The present invention relates to antibodies and antigen-binding fragments of antibodies that specifically bind and neutralize Chikungunya virus (CHIKV), which can be engineered to provide prophylactic and therapeutic solutions for the prevention and treatment of CHIKV disease. The present invention also relates to pharmaceutical compositions comprising the antibodies that neutralize CHIKV and uses of the antibodies for the prevention and treatment of CHIKV disease. [Background technology]

[0002] CHIKV is a reemerging mosquito-borne pathogen. Although CHIKV is endemic in Africa, India and Southeast Asia, it also infects millions of people beyond these regions, causing unpredictable outbreaks with high attack rates (Non-Patent Document 1). Mutations in the envelope glycoprotein 1 (E1) of CHIKV allow the virus to be transmitted by Aedes albopictus in addition to Aedes aegypti, causing a severe epidemic in 2005 that spread to Reunion, India and Indonesia, followed by outbreaks that began in travelers in Italy, France and China (Non-Patent Document 2; Non-Patent Document 3; Non-Patent Document 4; Non-Patent Document 5; Non-Patent Document 6). Based on the expanded geographic range of Aedes albopictus, the virus is expected to spread to new areas, and Europe and the Americas are now at risk of CHIKV outbreaks.

[0003] CHIKV is an enveloped, positive-stranded RNA virus in the Alphavirus genus of the Togaviridae family. It is a member of the Semliki Forest virus complex and is closely related to Ross River virus and Onyong-nyong virus (ONNV); it can also be called an arbovirus (arthropod-borne virus) because it is transmitted by arthropods, i.e., mosquitoes.

[0004] CHIKV enters cells by receptor-mediated internalization and a type II membrane fusion event triggered by low pH in early endosomes. CHIKV disease is characterized by acute, post-acute and chronic polyarthritis / polyarthralgia phases, the latter of which is usually symmetric, often results in incapacity and can last for months or years. Other symptoms such as fever, rash, myalgia and / or fatigue are also present during the acute phase. Recent outbreaks have been associated with atypical and severe clinical forms of CHIKV disease that appear to be limited to very young and elderly patients with comorbidities, and some even death.

[0005] Currently, there is no specific preventive or therapeutic treatment for CHIKV disease. CHIKV is usually treated symptomatically with bed rest, fluids, and simple medications such as painkillers and / or nonsteroidal anti-inflammatory drugs (NSAIDs) to relieve symptoms of fever and pain. Vaccine candidates against CHIKV were first proposed 45 years ago, but many vaccine candidates tested to date have not been successful in eliciting protective antibodies or showing significant safety results. [0007] There is still a need for treatments that show improved therapeutic efficacy against CHIKV, including the use of specific monoclonal antibodies that target CHIKV.There is a need in the art for antibodies that neutralize CHIKV that are suitable for prophylactic and therapeutic use.In particular, such antibodies should adequately neutralize different strains of CHIK virus with high target binding affinity, show suitable pharmacokinetic parameters, show suitable half-life when administered, and allow efficient production in large scale, while retaining their binding to FcγRIIIa associated with effector function. [Means for solving the problem]

[0008] As disclosed herein, the inventors of the present application have been able to select and engineer specific CHIKV neutralizing antibodies, improve their pharmacokinetics associated with exposure, and maintain their binding to FcγRIIIa associated with effector functions, making them compatible with the development of therapeutics to prevent and treat CHIKV disease, addressing the need in the art for effective therapies against CHIKV.

[0009] The antibodies of the present invention have high binding affinity (in nanomolar range) to different CHIKV strains. Therefore, they show broad and highly potent neutralizing activity against different CHIKV strains. Furthermore, the antibodies of the present invention have improved binding to human FcRn receptor, and at the same time retain the binding to FcγRIIIa, making them compatible with increased half-life, and at the same time maintain their binding to FcγRIIIa associated with effector function.

DETAILED DESCRIPTION OF THE PREFERRED EMBODIMENTS

[0057] definition An "antibody" is a natural or conventional antibody in which two heavy chains are linked to each other by disulfide bonds and each heavy chain is linked to a light chain by a disulfide bond. In mammals, antibodies are classified into five major classes or isotypes, IgA, IgD, IgE, IgG, and IgM. They are classified according to the heavy chains they contain, alpha, delta, epsilon, gamma, or mu. They differ in the sequence and number of constant domains, hinge structure, and valency of the antibody. There are two types of light chains, lambda (l) and kappa (k), with kappa light chains being the more common of the two. Although they are relatively different in protein sequence, they share similar structure and function.

[0058] Five major heavy chain classes (or isotypes) determine the functional activity of antibody molecules: IgM, IgD, IgG, IgA and IgE. Each chain contains a distinct sequence domain. IgG is the most abundant antibody in normal human serum, accounting for 70-85% of the total immunoglobulin pool. It is a monomer with a molecular weight of about 150 KDa, is the primary antibody of the secondary immune response, and has the longest half-life of the five immunoglobulin classes. IgG is classified into four human subclasses (IgG1, IgG2, IgG3 and IgE), each of which contains a different heavy chain. IgG1 and IgG4 are composed of IgG1, IgG2 and IgG3. They are highly homologous and differ mainly in the hinge region and the extent to which they activate the host's immune system. IgG1 and IgG4 contain two interchain disulfide bonds in the hinge region, IgG2 has four and IgG3 has eleven.

[0059] The light chain contains two domains or regions, one variable domain (VL) and one constant domain (CL). The heavy chain contains four domains, one variable domain (VH) and three constant domains (CH1, CH2 and CH3, collectively referred to as CH). The variable regions of both the light chain (VL) and the heavy chain (VH) determine the binding recognition and specificity to the antigen. The constant region domains of the light chain (CL) and the heavy chain (CH) confer important biological properties such as antibody chain association, secretion, mobility across the placenta, complement binding, and binding to Fc receptors (FcR). The Fv fragment is the N-terminal portion of the Fab fragment of an immunoglobulin and consists of one light chain and one variable portion of a heavy chain. The specificity of an antibody resides in the structural complementarity between the antibody binding site and the antigenic determinant. The antibody binding site is made up of residues mainly from the hypervariable or complementarity determining regions (CDRs). Occasionally, residues from non-hypervariable or framework regions (FR) influence the overall domain structure and therefore the binding site. "Complementarity determining region or CDR" refers to the amino acid sequences that together define the binding affinity and specificity of the natural Fv region of an original immunoglobulin binding site. The light and heavy chains of an immunoglobulin each have three CDRs, called CDR1-L, CDR2-L, CDR3-L (for the light chain complementarity determining region) or CDRL1, CDRL2, CDRL3, and CDR1-H, CDR2-H, CDR3-H (for the heavy chain complementarity determining region) or CDRH1, CDRH2, CDRH3, respectively. The antigen-binding site of a conventional antibody therefore contains six CDRs, including a set of CDRs from each of the heavy and light chain V regions.

[0060] "Framework region" (FR) refers to the amino acid sequences interposed between the CDRs, i.e., those portions of the variable regions of the light and heavy chains of immunoglobulins that are relatively conserved among different immunoglobulins in a single species. The light and heavy chains of immunoglobulins each have four FRs, designated FR1-L, FR2-L, FR3-L, FR4-L, and FR1-H, FR2-H, FR3-H, FR4-H, respectively.

[0061] As used herein, a "human framework region" is a framework region that is substantially identical (about 85% or more, particularly 90%, 95%, 97%, 99% or 100%) to the framework region of a naturally occurring human antibody.

[0062] In one embodiment, the definition of CDR / FR in an immunoglobulin light or heavy chain should be determined based on the IMGT definition (Lefranc, MP et al., 2003, Dev Comp Immunol. 27(1):55-77; www.imgt.org). The CDR sequences mainly dealt with in the present invention are given according to the nomenclature of IMGT.

[0063] The term "antibody" as used herein refers to conventional or full-length antibodies (i.e., antibodies comprising two heavy chains and two light chains), single domain antibodies, and fragments of conventional and single domain antibodies. The term "antibody" as used herein includes, but is not limited to, chimeric antibodies, humanized antibodies, human antibodies, and multispecific antibodies (such as bispecific antibodies and trispecific antibodies). The term "antibody" refers to both antibodies including the signal peptide (or propeptide, if present), and the mature form resulting from secretion and proteolytic processing of the chain(s).

[0064] As used herein, antibody or immunoglobulin also includes the more recently described "single domain antibodies," whose complementarity determining regions are part of a single domain polypeptide. Single domain antibodies are antibodies that are a part of the antibody. Examples of single domain antibodies include heavy chain antibodies, antibodies that naturally lack light chains, single domain antibodies derived from normal four-chain antibodies, and engineered single domain antibodies. Single domain antibodies can also be derived from any species, including but not limited to mouse, human, camel, llama, goat, rabbit, and cow. Single domain antibodies can also be naturally occurring single domain antibodies known as heavy chain antibodies that lack light chains. In particular, camelid species, such as camels, dromedaries, llamas, alpacas, and guanacos, produce heavy chain antibodies that naturally lack light chains. Camelid heavy chain antibodies also lack the CH1 domain.

[0065] The variable heavy chains of these single domain antibodies, devoid of light chains, are known in the art as "VHH" or "nanobodies". Like regular VH domains, VHH contain four FRs and three CDRs. Nanobodies have advantages over regular antibodies: they are about 10 times smaller than IgG molecules, and as a result, properly folded, functional nanobodies can be produced by in vitro expression while achieving high yields. Furthermore, nanobodies are very stable and resistant to the action of proteases. The properties and production of nanobodies have been reviewed by Harmsen and De Haard HJ (Appl. Microbiol. Biotechnol. 2007 Nov;77(1):13-22).

[0066] As used herein, an "isolated antibody" refers to an antibody that is primarily free of other antibodies having different antigenic specificities; for example, an isolated antibody, or fragment thereof, or antigen-binding fragment thereof, that binds to CHIKV does not primarily include antibodies that specifically bind to antigens other than CHIKV.

[0067] As used herein, "blocking antibody" or "neutralizing antibody" or "antibody that neutralizes CHIKV activity" or "antibody that exhibits / exercises neutralizing activity against CHIKV" or "antibody that neutralizes CHIKV" or "anti-CHIKV antibody" refers to an antibody whose binding to CHIKV results in the inhibition of at least one biological activity of CHIKV. For example, the antibody can neutralize CHIKV strains by blocking the attachment of CHIKV to cells, thereby preventing the infection of said cells by CHIKV.

[0068] The term "monoclonal antibody" or "mAb" as used herein refers to an antibody molecule of a single amino acid composition directed against a particular antigen and is not to be construed as requiring production of the antibody by any particular method. A monoclonal antibody may be produced by a single clone of B cells or hybridoma, but may also be produced recombinantly, i.e., by protein engineering.

[0069] The term "chimeric antibody" in its broadest sense refers to an engineered antibody that contains one or more regions from one antibody and one or more regions from one or more other antibody(ies). In particular, a chimeric antibody comprises the VH and VL domains of an antibody derived from a non-human animal combined with the CH and CL domains of another antibody, in particular a human antibody. As a non-human animal, any animal can be used, for example, mouse, rat, hamster, rabbit, etc. A chimeric antibody can also refer to a multispecific antibody having specificity for at least two different antigens. In one embodiment, a chimeric antibody has variable domains of mouse origin and constant domains of human origin.

[0070] The term "humanized antibody" refers to an antibody that is originally of non-human origin, either wholly or partially, and that has been modified by replacing certain amino acids, particularly those in the framework regions of the heavy and light chains, to avoid or minimize immune responses in humans. The constant domains of a humanized antibody are most often human CH and CL domains. In one embodiment, a humanized antibody has constant domains of human origin.

[0071] A "fragment" of a (conventional) antibody comprises a portion of a complete antibody, in particular the antigen-binding or variable region of the complete antibody. Examples of antibody fragments include Fv, Fab, F(ab')2, Fab', dsFv, (dsFv)2, scFv, sc(Fv)2, diabodies, bispecific antibodies and multispecific antibodies formed from antibody fragments. A fragment of a conventional antibody can also be a heavy chain antibody or a single domain antibody such as a VHH.

[0072] The term "Fab" refers to an antibody fragment having a molecular weight of about 50,000 and antigen-binding activity, in which about half of the N-terminal side of the H chain and the entire L chain are bound together through disulfide bonds, among fragments obtained by treating IgG with a protease called papain.

[0073] The term "F(ab')2" refers to an antibody fragment obtained by treating IgG with a protease called pepsin, which is slightly longer than Fab linked through the disulfide bonds in the hinge region, has a molecular weight of about 100,000, and has antigen-binding activity.

[0074] The term "Fab'" refers to an antibody fragment having a molecular weight of about 50,000 and antigen-binding activity, which is obtained by cleaving the disulfide bond in the hinge region of F(ab')2.

[0075] "Fc region" or "Fc domain" is defined as the carboxyl-terminal portion of the heavy chain of an antibody, containing protein sequences common to all immunoglobulins as well as determinants unique to each distinct class of immunoglobulin. As an example, the heavy chain of human IgG1 contains the CH1, hinge, CH2 and CH3 regions; the hinge and portions of the CH2 and CH3 regions constitute the Fc region. As shown in FIG. 1, the numbering of amino acid residues in the Fc region for the purposes of the present invention is according to the EU index as set forth in Kabat (Kabat et al., Sequences of Proteins of Immunological Interest, 5th ed., Public Health Service, National Institutes of Health, Bethesda, Md. (1991)). As a result, the phrase "the position of the amino acid is given by the EU index" refers to this numbering of the Fc region as shown in FIG. 1, as set forth in Kabat et al., 1991, supra.

[0076] The domains of the Fc region are central in determining the biological functions of immunoglobulins, and these biological functions are called "effector functions". These Fc domain-mediated activities are mediated by immunological effector cells, including B lymphocytes, natural killer cells, macrophages, basophils, neutrophils and mast cells, or various complement components. These effector functions include the activation of receptors on the surface of the effector cells by binding of the Fc domain of an antibody to said receptor (or "Fc receptor") or to a complement component(s). Antibody-dependent cellular cytotoxicity (ADCC), antibody-dependent cellular phagocytosis (ADCP) and complement-dependent cytotoxicity (CDC) activities belong to these effector functions and involve the binding of the Fc domain to Fc receptors such as FcγRI (CD64), FcγRII, FcγRIII or complement components such as C1q of effector cells. Among the various human immunoglobulin classes, human IgG1 and IgG3 mediated ADCC is more effective than IgG2 and IgG4. The term "Fc receptor" includes, but is not limited to, FcγRI (CD64), FcγRIIA and FcγRIIB (CD32), FcγRIIIA (CD16a) and FcγRIIIB (CD16b), Fcα receptors (FcαRI or CD89) and Fcε receptors (FcεRI and FcεRII (CD23). Several amino acid substitutions have been reported in the literature. This has been reported to be associated with reduced effector functions in different human IgG isotypes (see Table 2 in Strohl 2009, Current Opinion in Biotechnology 20:685-691).

[0077] Single-chain Fv ("scFv") polypeptides are covalently linked VH::VL heterodimers that are usually expressed from gene fusions containing VH- and VL-encoding genes linked by a peptide-encoding linker. Human scFv fragments can contain CDRs that are held in the proper conformation, particularly by using recombinant gene techniques. Divalent and multivalent antibody fragments can either form spontaneously by association of monovalent scFvs or can be generated by coupling monovalent scFvs by a peptide linker, such as a bivalent sc(Fv)2. A "dsFv" is a VH::VL heterodimer stabilized by a disulfide bond. "(dsFv)2" refers to two dsFvs coupled by a peptide linker.

[0078] The term "bispecific antibody" or "BsAb" refers to an antibody that combines the antigen-binding sites of two antibodies in a single molecule. Thus, BsAbs can bind two different antigens simultaneously. Genetic engineering is used with increasing frequency to design, modify, and produce antibodies or antibody derivatives with a desired set of binding properties and effector functions, for example as described in EP2050764A1.

[0079] The term "multispecific antibody" refers to an antibody that combines the antigen-binding sites of two or more antibodies within a single molecule.

[0080] The term "diabody" refers to a small antibody fragment having two antigen-binding sites, which comprises a variable domain of a heavy chain (VH) connected to a variable domain of a light chain (VL) on the same polypeptide chain (VH-VL). By using a linker that is too short to allow pairing between the two domains on the same chain, these domains are forced to pair with complementary domains on another chain to generate two antigen-binding sites.

[0081] The term "hybridoma" refers to cells obtained by fusing B cells produced by immunizing a non-human mammal with an antigen with myeloma cells derived from a mouse or the like, which produce the desired monoclonal antibody having antigen specificity.

[0082] As used herein, "specifically binds" or "specifically binds to" or "binds to" and the like means that an antibody or antigen-binding fragment thereof forms a complex with an antigen that is relatively stable under physiological conditions. Specific binding is at least about 1×10 -8 M or less equilibrium dissociation constant (K D ) (e.g., smaller K D represents tighter binding). Methods for determining whether two molecules specifically bind are well known in the art and include, for example, equilibrium dialysis, surface plasmon resonance, etc. As described herein, antibodies have been characterized for their specific binding to CHIKV and / or CHIKV antigens, for example, using surface plasmon resonance, e.g., BIACORE™.

[0083] As used herein, "CHIKV antigen" refers to the specific native antigen of the antibodies described herein, i.e., protein E2 of CHIKV, which is used, for example, in surface plasmon resonance binding experiments to measure in vitro the binding affinity of the anti-CHIKV antibodies described in the Materials and Methods herein, and may refer to "pE2-E1 protein" or "pE2-E1 target" or "p62-E1" or "his-tagged CHIKV E2 " or "CHIKV target pE2-E1" or "pE2-E1 antigen of CHIKV" refers to a recombinant protein comprising the envelope protein E1 of CHIKV and the specific antigen E2 of CHIKV.

[0084] As used herein, an "acidic environment" refers to an environment with a pH of less than 7; it is understood that binding experiments performed at, for example, pH 6 will yield binding data in an acidic environment.

[0085] A sequence that is "at least 80% identical to a reference sequence" is a sequence that has 80% or more, in particular 85%, 90%, 91%, 92%, 93%, 94%, 95%, 96%, 97%, 98% or 99% sequence identity over its entire length to the entire length of the reference sequence.

[0086] The percentage of "sequence identity" can be determined by comparing two optimally aligned sequences across a comparison window, where for optimal alignment of the two sequences, the portion of the polynucleotide or polypeptide sequence in the comparison window may contain additions or deletions (i.e., gaps) compared to the reference sequence (which does not contain additions or deletions). The percentage is calculated by determining the number of positions where the same nucleic acid base or amino acid residue occurs in both sequences to obtain the number of matched positions, and dividing the number of matched positions by the total number of positions in the comparison window and multiplying the result by 100 to obtain the percentage of sequence identity. Optimal alignment of sequences for comparison is performed by global pairwise alignment, for example, using the algorithm of Needleman and Wunsch J. Mol. Biol. 48:443 (1970). The percentage of sequence identity can be easily determined, for example, using the program Needle with the BLOSUM62 matrix and the following parameters: gap-open=10, gap-extend=0.5.

[0087] A "conservative amino acid substitution" is one in which an amino acid residue is replaced by another amino acid residue having a side chain R group with similar chemical properties (e.g., charge or hydrophobicity). In general, conservative amino acid substitutions do not substantially change the functional properties of a protein. Examples of groups of amino acids with side chains with similar chemical properties include: 1) aliphatic side chains: glycine, alanine, valine, leucine, and isoleucine; 2) aliphatic hydroxyl side chains: serine and threonine; 3) amide-containing side chains: asparagine and glutamine; 4) aromatic side chains: phenylalanine, tyrosine, and tryptophan; 5) basic side chains: lysine, arginine, and histidine; 6) acidic side chains: aspartic acid and glutamic acid; and 7) sulfur-containing side chains: cysteine and methionine. Conservative amino acids substitution groups are: valine-leucine-isoleucine, phenylalanine-tyrosine-tryptophan, lysine-arginine, alanine-valine, glutamate-aspartate, and asparagine glutamine.

[0088] The term "antigen-binding portion" of an antibody, "antigen-binding fragment" of an antibody, etc., as used herein, includes any naturally occurring, enzymatically obtainable, synthetic, or genetically engineered polypeptide or glycoprotein that specifically binds to an antigen to form a complex. The term "antigen-binding portion" of an antibody, or "antibody fragment" as used herein, refers to one or more fragments of an antibody that retain the ability to bind to CHIKV and / or CHIKV antigens. Such antigen-binding portion typically includes the CDRs of an antibody.

[0089] "Antibody-dependent cell-mediated cytotoxicity" or "ADCC" is a process in which antibodies secreted and bound to Fc receptors (FcRs) present on certain cytotoxic cells (e.g., natural killer (NK) cells, neutrophils, and macrophages) mediate the production of these cytotoxic effectors. ADCC refers to a form of cytotoxicity that allows the specific binding of an antibody-binding cell to a target cell carrying an antigen and subsequently killing the target cell. In other words, ADCC is a mechanism of cell-mediated immunity in which effector cells of the immune system, mainly natural killer cells, actively lyse target cells to which specific antibodies are bound. ADCC is one of the mechanisms by which antibodies, as part of the humoral immune response, can limit and contain infection. To estimate the ADCC activity of a molecule of interest, we also considered in vitro ADCC assays such as those described in U.S. Pat. No. 5,500,362 or 5,821,337.

[0090] The expression "amino acid sequence(s) which differ from sequence X or Y by one or two amino acid substitution(s)" means that said sequence differs from sequence X or Y by at most two amino acid substitutions, i.e. by no more than one or two amino acid substitutions.

[0091] For example, the phrase "a sequence X which differs from a sequence Y by an amino acid substitution Z and optionally one or two additional amino acid substitution(s)" means that said sequence X is: - only the amino acid substitution Z, or - an amino acid substitution Z and one or two amino acid substitutions different from amino acid substitution Z means that it is different from array Y.

[0092] The phrase "SEQ ID NO:X with one amino acid substitution at position Y" means a sequence that differs from SEQ ID NO:X by one amino acid substitution at position Y of SEQ ID NO:X.

[0093] As used herein, "CHIKV" refers to Chikungunya virus, an enveloped, positive-stranded RNA virus in the Alphavirus genus of the Togaviridae family, described in the introduction above. Also included within CHIKV are different representative infectious strains ("CHIKV strains"), including, but not limited to, the Eastern / Central / Southern African (ECSA) genotype, such as the LR2006OPY1[LR] strain having the sequence set forth in NCBI accession number DQ443544.2 dated October 24, 2006; the Western African genotype, such as the NI64IbH35 strain having the sequence set forth in NCBI accession number HM045786.1 dated December 28, 2010; and the Asian genotype, such as the RSU1 strain having the sequence set forth in NCBI accession number HM045797.1 dated December 28, 2010, and the 99659[2014 Caribbean] strain having the sequence set forth in NCBI accession number KJ451624 dated September 11, 2014. Other strains belonging to different genotypes have also been identified, for example the S27 strain, which has, as another example, the sequence designated by NCBI accession number AF369024.2 dated January 14, 2014, and the UniProtKB / Swiss-Prot reference Q8JUX5 dated September 16, 2015: and the SL15649, which has, as another example, the sequence designated by NCBI accession number GU189061 dated December 14, 2011. Other examples of referenced CHIKV strains can be found in the Viral Pathogens Database for their genomes at: https: / / www.viprbrc.org / brc / vipr_genome_search.spg?method=doQuickTextSearch&decorator=toga&pageTo=1&selectionContext=1476362322448 or for related proteins at: https: / / www.viprbrc.org / brc / vipr_protein_search.spg?method=doQuickTextSearch&decorator=toga&pageTo=1&selectionContext=1476362669763 Available in.

[0094] Like other alphavirus genomes, the CHIKV genome contains two envelope glycoproteins. It encodes the proteins, E2 and E1, which are derived from a larger polyprotein precursor (capsid / E3 / E2 / 6K / E1; designated NCBI accession number: NC_004162.2 dated June 27, 2012) and are embedded in the viral membrane. Mature virions contain three major structural proteins: the nucleocapsid protein and two glycoproteins, E1 and E2, where E2 functions in attachment to cells and E1 is involved in viral fusion. A third glycoprotein, E3, is associated with mature virions in some alphaviruses but not others, and the 6K protein, a membrane-bound peptide resulting from cleavage of the polyprotein precursor releasing E2 and E1, is incorporated at low levels into the particle. The organization of alphavirus surface glycoproteins in particles has been defined using cryo-electron microscopy (Cryo-EM), while the atomic structures of the CHIKV glycoproteins have recently been elucidated by X-ray crystallography for both the mature particle and the immature precursor polyprotein. 240 copies of each of the three glycoproteins (E3 / E2 / E1) combine on the mature virus to form 80 spikes that constitute an icosahedral protein shell around the viral membrane (Voss et al., 2003). JE et al. 2010, Nature 468:709-712). The folding, surface transport and function of these glycoproteins depend on their proper interaction with each other. E1 consists of three b-sheet domains, designated I, II and III; E2 contains three immunoglobulin-like domains (A, B and C, with A at the N-terminus). In the complex, domain B is at the membrane distal end and abuts E3, domain C is closest to the viral membrane and domain A is in the center (Fox JM et al. 2015, Cell 163:1095-1107 and WO2015010125). Sequences and information for the E1, E2 and E3 proteins of CHIKV are provided, by way of non-limiting example, in PDB entries 2xFB and 2xFC (last updated on November 24, 2010), PDB entries 3N40, 3N41, 3N42, 3N43 and 3N44 (last updated on December 1, 2010), respectively.

[0095] Antibodies mainly used in this invention For therapeutic purposes, it is desirable to generate mAbs that are better suited to their desired pharmaceutical properties, in particular by improving their binding to the antigen(s) they target, their stability, pharmacokinetics and pharmacodynamics, and their function.

[0096] First, the anti-CHIKV antibodies are based on fully human parent antibodies, mAb1 and mAb2, respectively, which have high binding affinity (in the nanomolar range) to different CHIKV strains, especially to their respective protein E2, and therefore exert broad and highly potent neutralizing activity against various CHIKV strains.

[0097] mAb1: - array: [ka] a heavy chain variable domain consisting of (framework regions comprising CDRH1, CDRH2 and CDRH3); - array: [ka] a light chain variable domain consisting of (framework regions including CDRL1, CDRL2 and CDRL3) Including, mAb2: - array: [ka] a heavy chain variable domain consisting of: - array: [ka] and the framework regions include CDRL1, CDRL2 and CDRL3.

[0098] In a first aspect, the present invention provides modified antibodies of mAb1 and mAb2 that bind to CHIKV and have improved binding to the FcRn receptor in an acidic environment because they contain at least one amino acid substitution in their Fc domains, such mutations result in an increase in the serum half-life of such modified antibodies when administered to a patient. Examples of such substitutions include, but are not limited to, modifications at positions 250 (e.g., E or Q); 250 and 428 (e.g., L or F); 252 (e.g., L / Y / F / W or T), 254 (e.g., S or T) and 256 (e.g., S / R / Q / E / D or T); or modifications at positions 428 and / or 433 (e.g., H / L / R / S / P / Q or K) and / or 434 (e.g., A, W, H, F or Y); or modifications at positions 250 and / or 428; or modifications at positions 307 or 308 (e.g., 308F, V308F) and 434, wherein said amino acid positions are given using the EU index numbering (Figure 1).

[0099] The inventors have determined that, as shown in FIG. 4 and Example 2, binding to the human and mouse FcRn receptors at pH 6 is enhanced when substitutions selected from the group below are introduced into the Fc region of the mAb1 or mAb2 antibodies: i. an alanine at position 434, or ii. alanine at positions 307, 380 and 434, respectively; or iii. a glutamine at position 250 and a leucine at position 428, respectively; or iv. a leucine at position 428 and a serine at position 434, respectively; or v. A tyrosine at position 252, a threonine at position 254 and a glutamic acid at position 256.

[0100] Furthermore, the inventors have shown that the binding of mAb1 and mAb2 to their CHIKV target pE2-E1 is not affected by the above substitutions when introduced into their Fc regions, but enhances binding to human and mouse FcRn, as described in Example 3 and Figure 5. As shown in Example 2 and Figure 4, substitutions in the Fc regions of mAb1 or mAb2 show the strongest binding to human and mouse FcRn when either tyrosine at position 252, threonine at position 254 and glutamic acid at position 256, respectively, or leucine at position 428 and serine at position 434, respectively, are introduced into their respective Fc regions.

[0101] As known in the art, the Fc region is essential in determining the biological functions of immunoglobulins, referred to as "effector functions". ADCC is a part of cell-mediated immunity, where effector cells of the immune system (mainly natural killer cells) lyse target cells to which specific antibodies are bound. Therefore, ADCC is one of the mechanisms that limit and contain infection. Cell-mediated activity involves the binding of the Fc domain to Fc receptors, such as FcγRI (CD64), FcγRII, and FcγRIII, on effector cells.

[0102] As primarily addressed in the present invention, where monoclonal antibodies are highly specific for CHIKV strains and subject to therapeutic treatment, their ability to activate ADCC is an important parameter to measure, since ADCC appears to be related to the activity of anti-CHIKV antibodies in controlling infection. As shown in Example 4 and Figure 6, the inventors have demonstrated that the binding of mAb1 and mAb2 to FcγRIIIa is retained when substitutions selected from the following group are introduced into their respective Fc regions: i. an alanine at position 434, or ii. alanine at positions 307, 380 and 434, respectively; or iii. a glutamine at position 250 and a leucine at position 428, respectively; or iv. A leucine at position 428 and a serine at position 434, respectively.

[0103] In contrast, binding to FcγRIIIa was reduced when mAb1 and mAb2 were substituted with tyrosine at position 252, threonine at position 254, and glutamic acid at position 256, respectively.

[0104] Thus, the inventors have identified antibodies with improved binding to the human FcRn receptor, whilst retaining binding to FcγRIIIa, making the antibodies of the invention compatible with increased half-life whilst maintaining their effector functions.

[0105] Thus, in a first aspect, the present invention relates to an isolated monoclonal antibody that binds to CHIKV and comprises three heavy chain complementarity determining regions (CDRH) and three light chain complementarity determining regions (CDRL), wherein: i. the CDRH has the amino acid sequence of SEQ ID NO:5, 6, and 7, and the CDRL has the amino acid sequence of SEQ ID NO:8, GNT, and SEQ ID NO:10; or ii. the CDRH has the amino acid sequence of SEQ ID NO:11, 12, and 13, and the CDRL has the amino acid sequence of SEQ ID NO:14, GTS, and SEQ ID NO:16; or iii. the CDRH and CDRLH have an amino acid sequence that differs from the sequence of i. or ii. by one or two amino acid substitutions; The antibody comprises: i. an alanine at position 434, or ii. alanine at positions 307, 380 and 434, respectively; or iii. a glutamine at position 250 and a leucine at position 428, respectively; or iv. a leucine at position 428 and a serine at position 434, respectively; or v. tyrosine at position 252, threonine at position 254 and glutamic acid at position 256, respectively; and further comprising an Fc region comprising at least one residue selected from the group consisting of: Here, the amino acid positions are given by the EU index.

[0106] Fc regions containing such substitutions are shown in Figures 1 and 2 (SEQ ID NO: 17; SEQ ID NOs: 59 to 63).

[0107] In one embodiment, the anti-CHIKV antibody comprises at least an Fc region comprising an alanine at position 434. In another embodiment, the anti-CHIKV antibody comprises an Fc region comprising an alanine at positions 307, 380, and 434. In another embodiment, the anti-CHIKV antibody comprises an Fc region comprising a glutamine at position 250 and a leucine at position 428. In another embodiment, the anti-CHIKV antibody comprises an Fc region comprising a leucine at position 428 and a serine at position 434. In another embodiment, the anti-CHIKV antibody comprises an Fc region comprising a tyrosine at position 252, a threonine at position 254, and a glutamic acid at position 256.

[0108] The antibodies primarily of interest in the present invention are derived from mAb1 or mAb2 and contain three heavy chain complementarity determining regions (CDRHs) and three light chain complementarity determining regions (CDRLs), which are: i. the amino acid sequences of SEQ ID NO:5, 6 and 7, and the amino acid sequences of SEQ ID NO:8, GNT and SEQ ID NO:10, respectively; or ii. the amino acid sequences of SEQ ID NO: 11, 12 and 13, and the amino acid sequences of SEQ ID NO: 14, GTS and SEQ ID NO: 16, respectively; or iii. an amino acid sequence which differs from the sequence in i. or ii. by one or two amino acid substitutions; Each of them has:

[0109] In a further embodiment, the antibody comprises CDRs having amino acid sequences which differ by one or two amino acid substitutions from the sequences of SEQ ID NOs: 5, 6 and 7, and from the amino acid sequences of SEQ ID NOs: 8, GNT and SEQ ID NO: 10, respectively, or from the sequences of SEQ ID NOs: 11, 12 and 13, and from the sequences of SEQ ID NO: 14, GTS and SEQ ID NO: 16, respectively.

[0126] The inventors have shown that the binding of mAb1 and mAb2 to their CHIKV target pE2-E1 is not affected when they contain an Fc region containing at least one residue as described above, as described in Example 3 and Figure 5. The inventors have also shown that these antibodies containing an Fc region containing at least one residue as described above all show enhanced binding to human and mouse FcRn (Example 2 and Figure 4), which has a favorable effect on their respective half-lives and is consequently beneficial for anti-CHIKV therapy; they also show the highest binding to FcRn in human and mouse when either a tyrosine at position 252, a threonine at position 254 and a glutamic acid at position 256, respectively, or a leucine at position 428 and a serine at position 434, respectively, are introduced into their respective Fc regions.

[0127] Thus, in an exemplary embodiment, the variant antibody comprises three heavy chain complementarity determining regions (CDRH) having the amino acid sequences of SEQ ID NO:5, 6 and 7, respectively, and three light chain complementarity determining regions (CDRL) having the amino acid sequences of SEQ ID NO:8, GNT and SEQ ID NO:10, respectively, and at least an Fc region comprising a leucine at position 428 and a serine at position 434, respectively, or at least a tyrosine at position 252, a threonine at position 254 and a glutamic acid at position 256, respectively, where said amino acid positions are given by the EU index.

[0128] Furthermore, as shown in Example 4 and FIG. 6, the present inventors demonstrated that the binding of mAb1 and mAb2 to FcγRIIIa corresponds to the following groups: i. an alanine at position 434, or ii. alanine at positions 307, 380 and 434, respectively; or iii. a glutamine at position 250 and a leucine at position 428, respectively; or iv. Leucine at position 428 and serine at position 434, respectively were shown to be retained when introduced into their respective Fc regions.

[0129] In contrast, binding to FcγRIIIa was reduced when mAb1 and mAb2 were substituted with tyrosine at position 252, threonine at position 254, and glutamic acid at position 256, respectively.

[0130] Thus, the inventors have identified at least one antibody that retains binding to FcγRIIIa while binding to its target is unaffected and has improved binding to the human FcRn receptor, while maintaining their effector functions, which, in terms of its increased half-life, makes it compatible with the development of therapeutics to prevent and treat CHIKV disease.

[0158] In one embodiment, the antibody is a combination of heavy and light chains or heavy and light chains encoded by the nucleotide sequences set forth in Table 3 below.

[0159] [Table 3]

[0160] As already mentioned, for therapeutic purposes, it is highly desirable to generate mAbs that are reliable in terms of stability, pharmacokinetics and pharmacodynamics, as well as their functionality, while retaining their binding affinity to their specific targets.

[0161] In a second embodiment, we identified hot spots that can be used to improve homogeneity and mitigate the disadvantages of chemistry, production and control (CMC). Such analysis focused on undesirable solvent-exposed motifs such as oxidation, deamination, isomerization, acidic cleavage, glycosylation and additional free cysteine residues that are identified either in silico or experimentally to potentially result in degradation products or heterogeneity of antibody formulations. As an example, deamination of asparagine and glutamine residues can occur depending on factors such as pH and surface exposure. Asparagine residues are particularly prone to deamination when present primarily in the sequence Asn-Gly, and less so in other dipeptide sequences such as Asn-Ala. Therefore, such deaminations are important for the development of novel antibody preparations. When a amino acid site, particularly Asn-Gly, is present in an antibody or polypeptide described herein, it is desirable to remove the site, typically by a conservative substitution to remove one of the involved residues.

[0162] As shown in Example 1 and Figure 3, the inventors have identified at least three amino acids in CDRH3 of mAb2 (SEQ ID NO: 13) that may have adverse consequences themselves when used for therapeutic purposes or because they result in unfavorable motifs based on the previous criteria. As shown in Figure 3, the inventors generate at least three variant antibodies of mAb2 that maintain their binding affinity to the pE2-E1 antigen of CHIKV by single substitutions at positions 8, 12 and 13, respectively, of SEQ ID NO: 13 to suppress the undesirable amino acids or motifs. Thus, the inventors have generated variants of mAb2 that have higher stability, homogeneity to CMC criteria, with less product-related impurities while retaining their target affinity when produced in bioreactors.

[0188] In one embodiment, the antibody is a heavy and light chain or a combination of heavy and light chains encoded by the nucleotide sequences set forth in Table 4 below.

[0189] [Table 4]

[0190] In a third aspect, the inventors combined the above advantageous aspects. On the one hand, substitutions were made within CDRH3 to suppress amino acids or motifs that are not favorable based on the criteria of chemical, manufacturing and control (CMC) disadvantages, and on the other hand, because they contain at least one amino acid substitution in their Fc domain, they generated variants of mAb2 with improved binding to the FcRn receptor in an acidic environment while retaining binding to FcγRIIIa associated with effector functions.

[0191] As described in Example 2 and Figures 4C and 4D, the inventors have demonstrated that variants of mAb2 in which substitutions in CDRH3 have been introduced fall into the following group: i. a leucine at position 428 and a serine at position 434, respectively; or ii. a tyrosine at position 252, a threonine at position 254, and a glutamic acid at position 256 The present inventors have demonstrated that substitutions selected from the following, when introduced into the Fc region, exhibit enhanced binding to the human and mouse FcRn receptors at pH 6.

[0192] Furthermore, the inventors confirmed that for antibodies stacking the above-described substitutions in their CDRH3 and their Fc regions, as described in Example 3 and Figure 5, their binding to the CHIKV target pE2-E1 is not affected.

[0193] As shown in Example 4 and Figures 6B and 6D, the inventors also confirmed that binding to FcγRIIIa was retained for such antibodies when at least a leucine at position 428 and a serine at position 434, respectively, were introduced into their Fc regions. In contrast, FcγRIIIa binding was weakened when such antibodies were substituted with a tyrosine at position 252, a threonine at position 254, and a glutamic acid at position 256, respectively.

[0194] Therefore, the inventors have confirmed the beneficial effect of substitutions introduced into the respective Fc regions of the antibodies according to the invention that are substituted in CDRH3, suppressing amino acids or motifs that are not advantageous based on the criteria of chemistry, manufacturing and control (CMC) disadvantages.

[0216] Common expression systems include E. coli host cells and plasmid vectors, insect host cells and baculovirus vectors, and mammalian host cells and vectors. Other examples of host cells include, but are not limited to, prokaryotic cells (such as bacteria) and eukaryotic cells (such as yeast cells, mammalian cells, insect cells, plant cells, etc.). Specific examples include E. coli, yeast of the genus Kluyveromyces or Saccharomyces, mammalian cell lines (e.g., Vero cells, CHO cells, 3T3 cells, COS cells, HEK293 cells, etc.), and primary or established mammalian cell cultures (e.g., produced from lymphoblasts, fibroblasts, embryonic cells, epithelial cells, neuronal cells, adipocytes, etc.). Examples include mouse SP2 / 0-Ag14 cells (ATCC CRL1581), mouse P3X63-Ag8.653 cells (ATCC CRL1580), CHO cells defective in the dihydrofolate reductase gene (hereinafter referred to as the "DHFR gene") (Urlaub, G. et al., 1980, Proc Natl Acad Sci USA. 77(7):4216-4220), rat YB2 / 3HL.P2.G11.16Ag.20 cells (ATCC CRL1662, hereinafter referred to as the "YB2 / 0 cells"), etc. YB2 / 0 cells are of interest because the ADCC activity of chimeric or humanized antibodies is enhanced when expressed in these cells.

[0217] In particular, for the expression of a humanized antibody, the expression vector is either a type in which the gene encoding the antibody heavy chain and the gene encoding the antibody light chain are present on separate vectors, or a type in which both genes are present on the same vector (tandem type). In terms of ease of construction of a humanized antibody expression vector, ease of introduction into animal cells, and balance between the expression levels of the antibody H and L chains in animal cells, a tandem type humanized antibody expression vector is commonly used (Shitara, K. et al., 1994, J Immunol Methods. Jan. 3:167(1-2):271-8). Examples of tandem type humanized antibody expression vectors include pKANTEX93 (WO97 / 10354), pEE18, etc.

[0218] The present invention also relates to a method for producing a recombinant host cell expressing an antibody according to the invention, said method comprising the steps of: (i) introducing, in vitro or ex vivo, a recombinant nucleic acid or vector as described above into a competent host cell, (ii) culturing the obtained recombinant host cell in vitro or ex vivo, and (iii) optionally selecting cells which express and / or secrete said antibody.

[0219] Such recombinant host cells can be used for the production of the anti-CHIKV antibodies described herein.

[0220] Thus, the present invention relates to a method for producing a monoclonal antibody according to the present invention, said method comprising the steps of: (i) culturing the cell line described above; (ii) purifying the monoclonal antibody produced; and optionally (iii) formulating said monoclonal antibody into a pharmaceutical composition.

[0221] Method for producing anti-CHIKV antibodies The anti-CHIKV antibodies primarily addressed in the present invention are produced by any technique known in the art, including, but not limited to, any chemical, biological, genetic or enzymatic technique, either alone or in combination.

[0222] Knowing the amino acid sequence of the desired sequence, the skilled artisan can easily produce said antibodies or immunoglobulin chains by standard techniques for producing polypeptides. For example, they can be synthesized using the well-known solid-phase method, in particular using commercially available peptide synthesizers (such as those produced by Applied Biosystems, Foster City, Calif.), following the manufacturer's instructions. Alternatively, antibodies and immunoglobulin chains can be synthesized by recombinant DNA techniques well known in the art. For example, these fragments can be obtained as DNA expression products after incorporation of a DNA sequence encoding the desired (poly)peptide into an expression vector and introduction of such a vector into a suitable eukaryotic or prokaryotic host expressing the desired polypeptide, from which the fragments can then be isolated using well-known techniques.

[0223] In particular, the present invention further relates to a method for producing an antibody comprising the steps of: (i) culturing a host cell transformed according to the present invention; (ii) expressing said antibody or polypeptide; and (iii) recovering the expressed antibody or polypeptide.

[0224] In other words, the present invention: (i) providing a cell expressing an anti-CHIKV antibody; (ii) culturing the cells; (iii) purifying the antibody; and (iv) optionally formulating the antibody into a pharmaceutical composition. The present invention relates to a method for producing an antibody, comprising:

[0225] Methods for producing humanized or chimeric antibodies involve conventional recombinant DNA and gene transfection techniques well known in the art (see Morrison, SL and Oi, VT, 1984, Annu Rev Immunol 2:239-256 and patent documents US 5,202,238; and US 5,204,244).

[0226] In a specific embodiment, the chimeric antibody of the present invention can be produced by obtaining nucleic acid sequences encoding mouse VL and VH domains, constructing a chimeric antibody expression vector as described above, inserting them into an expression vector for animal cells carrying genes encoding the CH and CL of a human antibody, and expressing the coding sequences by introducing the expression vector into the animal cells.

[0227] The antibodies of interest in the present invention are suitably separated from the culture medium by conventional immunoglobulin purification procedures, such as protein A affinity chromatography, ceramic hydroxyapatite chromatography, mixed mode chromatography, size-exclusion chromatography, and the like.

[0228] Fab can be obtained by treating an antibody that specifically reacts with CHIKV with a protease such as papain. Fab can be obtained by inserting DNA sequences encoding both chains of the Fab of an antibody into a vector for expression in prokaryotes or eukaryotes. It can also be produced by introducing the vector into a prokaryotic or eukaryotic cell (as appropriate) to express the Fab.

[0229] F(ab')2 can be obtained by treating an antibody specifically reactive with CHIKV with the protease, pepsin. F(ab')2 can also be produced by linking Fab' through a thioether bond or a disulfide bond as described below.

[0230] Fab' can be obtained by treating F(ab')2 specifically reacting with CHIKV with a reducing agent such as dithiothreitol. Fab' can also be produced by inserting a DNA sequence encoding the Fab' chain of an antibody into a vector for expression in a prokaryotic or eukaryotic expression, and introducing the vector into a prokaryotic or eukaryotic cell (as appropriate) to effect expression.

[0231] scFvs can be produced by constructing DNA encoding scFv fragments using sequences of CDRs or VH and VL domains as described above, inserting the DNA into a prokaryotic or eukaryotic expression vector, and then introducing the expression vector into a prokaryotic or eukaryotic cell (as appropriate) to express the scFv. To generate humanized scFv fragments, a well-known technique called CDR grafting can be used, which involves selecting complementarity determining regions (CDRs) according to the present invention and grafting them into the framework of a human scFv fragment of known three-dimensional structure (see, for example, WO98 / 45322; WO87 / 02671; US5,859,205; US5,585,089; US4,816,567; EP0173494).

[0232] Single chain antibodies or VHHs against CHIKV can be obtained, for example, by a method comprising the steps of: (a) immunizing a mammal belonging to the family Camelidae with CHIKV or a fragment thereof in order to induce antibodies (and in particular heavy chain antibodies) against CHIKV; (b) obtaining a biological sample from the thus immunized Camelidae, comprising heavy chain antibody sequences and / or VHH sequences against CHIKV; and (c) recovering (e.g. isolating) the heavy chain antibody sequences and / or VHH sequences against CHIKV from said biological sample. Suitable single chain antibodies or VHHs can also be obtained, by way of non-limiting example, by screening a library comprising heavy chain antibody sequences and / or VHH sequences of heavy chain antibody sequences and / or VHH sequences competing for binding to pE2-E1 of CHIKV.

[0233] Modification of the anti-CHIKV antibody of the present invention A further object of the invention includes function-conservative variants of the antibodies described herein.

[0234] For example, certain amino acids can be substituted in a protein structure by other amino acids without appreciable loss of activity. Because the interacting ability and properties of a protein determine its biologically functional activity, certain amino acid substitutions can be made in a protein sequence, and of course in its DNA coding sequence, to nevertheless obtain a protein with similar properties. It is therefore believed that various changes can be made in antibody sequences, or the corresponding DNA sequence encoding said antibodies, without appreciable loss of their binding activity.

[0235] An amino acid may be a saccharide that is more hydrophobic than another amino acid that has a similar hydropathic index or score. It has been demonstrated in the art that it is possible to substitute amino acids and still result in a protein having similar biological activity, i.e., a biologically functional equivalent protein can be obtained. It is known in the art. It is also possible to use well-established techniques such as alanine scanning techniques to identify all amino acids that can be substituted in an antibody without significant loss of binding to the antigen. Such residues can be qualified as neutral because they are not involved in antigen binding or in maintaining the structure of the antibody. One or more of these neutral positions can be substituted by alanine or by another amino acid without changing the main properties of the antibody.

[0236] Thus, as outlined above, amino acid substitutions are generally based on the relative similarity of the amino acid side chain substituents, e.g., their hydrophobicity, hydrophilicity, charge, size, etc. Exemplary substitutions that take into account several of the aforementioned characteristics are well known to those of skill in the art and include: arginine and lysine; glutamate and aspartate; serine and threonine; glutamine and asparagine; and valine, leucine and isoleucine.

[0237] Another type of amino acid modification of an antibody may be useful by deleting one or more carbohydrate moieties found in the antibody and / or adding one or more glycosylation sites that are not present in the antibody, i.e., to alter the original glycosylation pattern of the antibody. The presence of either the tripeptide sequences asparagine X-serine, where X is any amino acid except proline, and asparagine X-threonine creates a potential glycosylation site. Addition or deletion of glycosylation sites to an antibody is usually accomplished by altering the amino acid sequence such that it contains one or more of the above tripeptide sequences (for N-linked glycosylation sites).

[0238] Another type of covalent modification involves chemically or enzymatically coupling glycosides to antibodies. These procedures are advantageous in that they do not require the production of antibodies in host cells that have the capacity for glycosylation for N- or O-linked glycosylation. Depending on the coupling mode used, sugar(s) can be attached to (a) arginine and histidine, (b) free carboxyl groups, (c) free sulfhydryl groups such as those of cysteine, (d) free hydroxyl groups such as those of serine, threonine, or hydroxyproline, (e) aromatic residues such as those of phenylalanine or tyrosine, or (f) the amide group of glutamine. For example, such methods are described in WO87 / 05330.

[0239] Removal of any carbohydrate moieties present on an antibody can be accomplished chemically or enzymatically. Chemical deglycosylation requires exposure of the antibody to the compound trifluoromethanesulfonic acid, or an equivalent compound. This treatment results in the cleavage of most or all sugars except the linking sugar (N-acetylglucosamine or N-acetylgalactosamine), while leaving the antibody intact. Chemical deglycosylation has been described by Sojahr, H. et al. (1987, Arch Biochem Biophys. 259(1):52-57) and by Edge, A. S. et al. (1981, Anal Biochem. 118(1):131-137). Enzymatic cleavage of carbohydrate moieties on antibodies can be accomplished by the use of various endo- and exoglycosidases described by Thotakura, N. R. et al. (1987, Methods Enzymol 138:350-359).

[0240] Another type of covalent modification of antibodies involves linking the antibody to one of a variety of nonproteinaceous polymers, e.g., polyethylene glycol, polypropylene glycol, or polyoxyalkylenes, in the manner described in U.S. Pat. Nos. 4,640,835; 4,496,689; 4,301,144; 4,670,417; 4,791,192, or 4,179,337.

[0241] Pharmaceutical Compositions The anti-CHIKV antibodies featured in the present invention can also be combined with pharma- ceutically acceptable excipients, and optionally with sustained release matrices, such as biodegradable polymers, to form therapeutic compositions.

[0242] Therefore, the present invention also relates to a pharmaceutical composition comprising an anti-CHIKV antibody of the present invention and a pharma- ceutically acceptable carrier.

[0243] The present invention also relates to an antibody according to the invention for use as a medicament.

[0244] "Pharmaceutically" or "pharmacologically acceptable" refers to molecular entities and compositions that do not produce adverse allergic or other untoward reactions when administered appropriately to a mammal, particularly a human. A pharmaceutically acceptable carrier or excipient refers to a non-toxic solid, semi-solid or liquid filler, diluent, encapsulating material or formulation auxiliary of any type.

[0245] The form of the pharmaceutical composition, the route of administration, the dosage and the administration regime will of course depend on the condition to be treated, the severity of the disease, the age, weight, and sex of the patient, etc.

[0246] The pharmaceutical compositions can be formulated for topical, oral, parenteral, intranasal, intravenous, intramuscular, subcutaneous or intraocular administration, and the like.

[0247] In particular, the pharmaceutical compositions contain pharma- ceutically acceptable vehicles for injectable dosage forms. These can be, in particular, isotonic, sterile, saline solutions (monosodium or disodium phosphate, sodium chloride, potassium chloride, calcium chloride or magnesium chloride, etc. or mixtures of such salts), or dry, in particular lyophilized compositions that, when sterile water or saline is added as the case may be, allow the constitution of an injectable solution.

[0248] The dose used for administration can be adapted as a function of various parameters, in particular as a function of the mode of administration used, the pathology involved or alternatively the desired duration of the treatment.

[0249] To prepare a pharmaceutical composition, an effective amount of the antibody can be dissolved or dispersed in a pharma- ceutically acceptable carrier or aqueous medium.

[0250] In one embodiment, the invention relates to a pharmaceutical composition comprising a prophylactically or therapeutically effective amount of an antibody or antigen-binding fragment thereof that binds to CHIKV as described herein, and a pharma- ceutically acceptable carrier.

[0251] The pharmaceutical forms suitable for injectable use include sterile aqueous solutions or dispersions; and sterile powders for the extemporaneous preparation of sterile injectable solutions or dispersions. In all cases, the form must be sterile and must be fluid to the extent that easy syringability exists. It must be stable under the conditions of manufacture and storage and must be preserved so as to prevent the contaminating action of microorganisms, such as bacteria and fungi.

[0252] The carrier can be, for example, a solvent or dispersion medium containing water, ethanol, polyol (e.g., glycerol, propylene glycol, liquid polyethylene glycol, and the like), or suitable mixtures thereof. Proper fluidity can be maintained, for example, by the use of a coating such as lecithin, by the maintenance of the required particle size in the case of dispersions, and by the use of surfactants, stabilizers, cryoprotectants, or antioxidants. Prevention of the action of biological organisms can be achieved by antibacterial and antifungal agents. In many cases, it will be preferable to include isotonic agents, for example, sugars or sodium chloride.

[0253] Sterile injectable solution is prepared by incorporating the active compound in the required amount in a suitable solvent together with the required several other components listed above, followed by sterilization by filtration.Generally, dispersion is prepared by incorporating various sterilized active ingredients into a sterile vehicle that contains the basic dispersion medium and other components required from those listed above. For the case of sterile powder for preparing sterile injectable solution, general preparation methods include vacuum drying and freeze-drying techniques, which produce powders of active ingredients plus any additional desired components from their solutions that have been previously sterilized and filtered.

[0254] Upon formulation, solutions will be administered in a manner compatible with the dosage form and in such amount as is therapeutically effective. The formulations are easily administered in a variety of dosage forms, such as the types of injectable solutions described above, although drug release capsules and the like can also be used.

[0255] For parenteral administration in aqueous solution, for example, the solution should be appropriately buffered if necessary, and the diluent should first be made isotonic with sufficient saline or glucose. These particular aqueous solutions are particularly suitable for intravenous, intramuscular, subcutaneous and intraperitoneal administration. In this regard, those skilled in the art will know, in light of this disclosure, the sterile aqueous medium that can be used. For example, a single dosage can be dissolved in 1 mL of isotonic NaCl solution and added to 1000 mL of bolus hypodermic fluid or injected into the proposed site of infusion (see, for example, "Remington's Pharmaceutical Sciences", 15th Edition, pages 1035-1038, 1570-1580). Some variation in dosage will necessarily occur depending on the condition of the subject being treated. The person responsible for administration will determine the appropriate dose for the individual subject in any event.

[0256] The antibody may be formulated into a therapeutic mixture and contain from about 0.01 to 100 milligrams, or thereabouts, per dose.

[0257] According to certain embodiments, one or more doses of the anti-CHIKV antibodies described herein can be administered to a subject over a defined time course.

[0258] The method according to this embodiment comprises administering to the subject multiple doses of the antibody against CHIKV sequentially. As used herein, "administering sequentially" means that each dose of the antibody against CHIKV is administered to the subject at different time points, for example, on different days, separated by a predetermined interval (for example, hours, days, weeks or months). The present invention includes a method comprising administering to the patient one initial dose of the antibody against CHIKV, followed by one or more secondary doses of the antibody against CHIKV, and optionally followed by one or more tertiary doses of the antibody against CHIKV.

[0259] The terms "initial dose", "secondary dose" and "tertiary dose" refer to the sequence of administration of antibodies against CHIKV in chronological order. Thus, an "initial dose" is a dose administered at the beginning of a treatment regimen (also referred to as a "baseline dose"); a "secondary dose" is a dose administered after the initial dose; and a "tertiary dose" is a dose administered after the secondary dose. The initial, secondary and tertiary doses may all contain the same amount of antibodies against CHIKV, but generally may differ from each other in terms of frequency of administration. However, in certain embodiments, the amount of antibodies against CHIKV contained in the initial, secondary and / or tertiary doses may vary from each other during the course of treatment (e.g., adjusted up or down as appropriate). In certain embodiments, two or more doses (e.g., 2, 3, 4, or 5) of antibodies against CHIKV are administered. A dose is administered at the start of a treatment regimen as a "loading dose", followed by doses administered on a less frequent basis (eg, "maintenance doses").

[0273] Diagnostic Use In another aspect, the monoclonal antibodies or antigen-binding fragments featured in the present invention are used to detect the presence or absence of a CHIKV antigen in a sample. In one embodiment, the antibodies described herein are used as components of an assay that includes contacting a sample with an antibody or antigen-binding fragment described herein and detecting binding of the monoclonal antibody or antigen-binding fragment to a CHIKV antigen, where detection of binding indicates the presence of the CHIKV antigen or where absence of detection of binding to the CHIKV antigen indicates the absence of the CHIKV antigen.

[0274] In particular, the monoclonal antibodies or antigen-binding fragments described herein find use both as components of therapeutic agents and as components of diagnostic assays.

[0275] In one embodiment, the antibody is intended for in vitro or ex vivo use. For example, CHIKV can be detected in a biological sample obtained from a subject using the anti-CHIKV antibody described herein in vitro or ex vivo.

[0276] The present invention relates to: i. contacting a biological sample of a subject with an anti-CHIKV antibody, particularly under conditions sufficient for the antibody to form a complex with said biological sample; ii. determining the level of antibody bound to said biological sample; iii. detecting the presence of CHIKV infection by comparing the measured levels of bound antibody with a control, wherein an increased level of bound antibody compared to the control indicates CHIKV infection; The present invention further relates to an in vitro or ex vivo method for detecting the presence of a CHIKV infection in a subject, comprising the steps of:

[0280] In said method, an increased level of antibody bound to said biological sample compared to a control indicates ineffectiveness of said treatment of CHIKV infection.

[0281] Said control is in particular a biological sample of the same type as the biological sample to be analyzed, but obtained from the subject temporally prior to the course of treatment of the CHIKV infection.

[0282] In one embodiment, the anti-CHIKV antibodies or antigen-binding fragments thereof (e.g., E2-binding fragments) described herein can also be labeled with a detectable molecule or substance, such as a fluorescent molecule, a radioactive molecule, or any other label known in the art that provides a signal (either directly or indirectly).

[0283] As used herein, with respect to an antibody according to the invention, the term "labeled" is intended to encompass direct labeling of the antibody by coupling (i.e., physically linking) a detectable substance, such as a radioactive agent or a fluorophore (e.g., fluorescein isothiocyanate (FITC) or phycoerythrin (PE) or indocyanine (Cy5)), to the polypeptide, as well as indirect labeling through reactivity with a substance capable of detection of the polypeptide.

[0284] A "sample" or "biological sample" that can be used in a diagnostic assay for CHIKV according to the present invention includes any tissue or fluid sample that can be obtained from a patient under normal or pathological conditions.

[0289] material and method Analysis and manipulation of monoclonal antibodies: The amino acid sequences of the anti-CHIKV antibodies were analyzed using Antibody Inspector (an in-house developed tool for antibody sequence analysis) coupled with a 3D model / structure analysis tool (Biovia Discovery Studio Suite) to screen for potential problems and disadvantages for development. The disadvantage analysis focuses on motifs where solvent exposure is undesirable, such as oxidation, deamination, isomerization, acidic cleavage, glycosylation, and additional free Cys. All solvent exposure disadvantages are prioritized based on their location (CDR, framework of variable domain, constant domain). Mutations were suggested to minimize the disadvantages found in the sequences.

[0290] Optimized antibody generation: Codon-optimized gene fragments were synthesized and cloned into mammalian expression vectors. Transfections were performed using the Expi293F Expression System (Thermo Fisher Scientific) according to the manufacturer's protocol. Samples of harvested conditioned media were purified by passage through a protein A column and the eluted fractions were buffer exchanged into Gibco PBS, pH 7.4.

[0291] Binding analysis: antigen binding, human and mouse FcRn binding, FcγIIIa binding The binding of recombinant CHIKV E2 antigen was measured by surface plasmon resonance using a Biacore T200 instrument. Anti-tetra-His antibody (Qiagen) was immobilized on a CM5 series S sensor chip at saturation level by the standard amine coupling procedure provided by Biacore. The constructs of recombinant CHIKV E2 antigen, namely His-tagged CHIKV E2 LR2006 (SEQ ID NO: 9) and His-tagged CHIKV E2 SL15649 (SEQ ID NO: 15) recombinant proteins, were based on Voss et al., 2010 (Voss JE et al. 2010, Nature 468: 709-712) and were transiently expressed and purified by HEK293 cells (Pal et al., 2013, PloS Pathog 9, e1003312; Smith et al., 2015, Cell Host & Microbe 18: 86-95). Essentially, these constructs are designed as signal peptide-E3_E2-(G4S)4-E1-His8, but in the mature form, the signal peptide and E3 are removed by cleavage. His-tagged CHIKV E2 LR2006 and His-tagged CHIKV E2 SL15649 recombinant antigens were diluted in HBS-EP+ running buffer and injected for 30 seconds to achieve capture levels between 10 and 30 RU. Test antibodies were serially diluted 3-fold from 30 nM to 1.1 nM. Low affinity binders were serially diluted from 900 nM. Each antibody was injected in duplicate over the captured antigen and control surfaces for 3 minutes at a flow rate of 65 μL / min and allowed to dissociate for 5 or 15 minutes. Surfaces were regenerated with glycine at pH 1.5. Kinetic constants were calculated using a 1:1 binding model in the Biacore T200 evaluation software.

[0292] To measure binding to FcRn, recombinant human or mouse FcRn was directly immobilized onto the chip of a CM5 series S sensor using amine chemistry to achieve a surface density of 1700RU and 800RU, respectively. Test antibodies were diluted to 200 and 50nM in 50mM sodium phosphate, 150mM NaCl, 0.05% surfactant P20, pH 6.0 or pH 7.4. Diluted samples were injected for 3 min, followed by dissociation in buffer at 10μL / min for 5 min in duplicate. The surface was regenerated with borate buffer, pH 8.5.

[0293] Binding to FcγRIIIa was measured using a Biacore 3000 instrument. Anti-HPC4 antibodies were immobilized at saturating levels on a CM5 chip by amine coupling. Two polymorphs of recombinant human FcγRIIIa were compared in the analysis (Val158 and Phe158). Recombinant human HPC4-tagged FcγRIIIa-V158 and FcγRIII-F158 were diluted in HBS-P+ buffer containing 2 mM CaCl2 and injected over Fc2 or Fc4, respectively, for 30 s at 10 μL / min to achieve capture levels of 10–40 RU. Samples were diluted to 900, 300, and 100 nM and injected for 2 min, followed by dissociation in duplicate for 3 min at 30 μL / min in buffer. Surfaces were regenerated with 10 mM EDTA in HBS-EP+ buffer at 20 μL / min for 3 min.

[0294] result EXAMPLES

[0295] Antigen Binding of mAb2 with Substitutions in CDRH3 The results are shown in Figure 3. The nucleotide sequences introduced to eliminate potential deamination and oxidation motifs were The effect of mutations in CDRH3 of mAb2 was measured on binding to the pE2-E1 antigen of CHIKV derived from the LR2006 strain of CHIKV. - for mAb2 and its derived variants mAb10, mAb11 and mAb12, which contain an isoleucine at position 8 of their CDRH3, a glutamine at position 12 of their CDRH3 and an alanine at position 13 of their CDRH3, respectively, as well as for variants containing an alanine at position 8 of their CDRH3 and variants containing different combinations of the two substitutions therein listed above.

[0296] Among the seven mutants generated by eliminating potential deamination and oxidation motifs, only mAb11 (mAb2 N108Q), which contains a glutamine at position 12 of its CDRH3, retained target binding affinity equivalent to the parent mAb. Of note, mAb10 (mAb2 M104I), which contains an isoleucine at position 8 of its CDRH3, and mAb12 (mAb2 G109A), which contains an alanine at position 13 of its CDRH3, showed intermediate profiles; the double mutants lost their target binding affinity, as did the variant containing an alanine at position 8 of its CDRH3 (mAb2 M104A). EXAMPLES

[0297] FcRn binding The results are shown in Figure 4. Binding to human FcRn (Figures 4A and 4C) and mouse (Figures 4B and 4D) was: - mAb1 and mAb3, mAb4, mAb5, mAb6 and mAb7, which respectively comprise in their Fc region an alanine at position 434, an alanine at each of positions 307, 380 and 434, a glutamine at position 250 and a leucine at position 428, a leucine at position 428 and a serine at each of positions 434, as well as a tyrosine at position 252, a threonine at position 254 and a glutamic acid at position 256. - mAb2 and mAb8, mAb9, mAb11, mAb13 and mAb14 which comprise, in their Fc region, a tyrosine at position 252, a threonine at position 254 and a glutamic acid at position 256, a leucine at position 428 and a serine at position 434, respectively, and a glutamine at position 12 of their CDRH3, a tyrosine at position 252, a threonine at position 254 and a glutamic acid at position 256 and a glutamine at position 12 of their CDRH3, a leucine at position 428 and a serine at position 434 and a glutamine at position 12 of their CDRH3, respectively. was measured at pH 6.0.

[0298] All mutants showed increased FcRn binding affinity, which is predicted to result in increased half-life and therefore favorable impact on their usefulness in anti-CHIKV therapy. Mutants containing leucine at position 428 and serine at position 434, respectively, and tyrosine at position 252, threonine at position 254, and glutamic acid at position 256, respectively, showed the strongest binding when compared to other mutants. EXAMPLES

[0299] Effect of substitutions in the Fc region on target binding of mAb1 and mAb2 The results are shown in Figure 5. The effect of substitutions in the Fc region was measured for mAb1 and mAb2 on binding to the pE2-E1 antigen of CHIKV from CHIKV strains LR2006 (Figure 5A) and SL15649 (Figure 5B), respectively. Binding was: - mAb1 and mAb3, mAb4, mAb5, mAb6, mAb7, mAb8, mAb9, mAb10, mAb11, mAb12, mAb13, mAb14, mAb15, mAb16, mAb17, mAb18, mAb19, mAb20, mAb21, mAb22, mAb23, mAb24, mAb25, mAb26, mAb27, mAb28, mAb29, mAb30, mAb31, mAb32, mAb33, mAb34, mAb35, mAb36, mAb37, mAb38, mAb39, mAb40, mAb41, mAb42, mAb43, mAb44, mAb45, mAb46, mAb47, mAb48, mAb49, mAb50, mAb51, mAb52, mAb53, mAb54, mAb55, mAb56, mAb57, mAb58, mAb59, mAb60, mAb61, mAb62, mAb63, mAb64, mAb65, mAb66, mAb67, mAb68, mAb69, mAb61 5, mAb6 and mAb7; - mAb2 and mAb8, mAb9, mAb11, mAb13 and mAb14, which comprise a tyrosine at position 252, a threonine at position 254 and a glutamic acid at position 256, a leucine at position 428 and a serine at position 434, respectively, a glutamine at position 12 of their CDRH3, a tyrosine at position 252, a threonine at position 254 and a glutamic acid at position 256, and a glutamine at position 12 of their CDRH3, a leucine at position 428 and a serine at position 434, and a glutamine at position 12 of their CDRH3, respectively. Each of these was measured.

[0300] Substitutions in the Fc region that enhanced binding of mAb1 and mAb2 to FcRn did not affect binding to the targeted pE2-E1 antigen of CHIKV. EXAMPLES

[0301] FcγRIIIa binding The results are shown in Figure 6. The effect of substitutions in the Fc region of mAb1 and mAb2, respectively, was measured on binding to FcγRIIIa, which is expressed by NK and macrophages and can induce antibody-dependent cell-mediated cytotoxicity (ADCC) and cytokine release by macrophages.

[0302] Binding results are shown for mAb1 (Figure 6A) and mAb2 (Figure 6B) at the high affinity receptor for human FcγRIIIa (FcγRIIIaV158) and for mAb1 (Figure 6C) and mAb2 (Figure 6D) at the low affinity receptor for human FcγRIIIa (FcγRIIIaF158), respectively.

[0303] Join: - mAb1 and mAb3, mAb4, mAb5, mAb6 and mAb7, comprising an alanine at position 434, an alanine at positions 307, 380 and 434, respectively, a glutamine at position 250 and a leucine at position 428, a leucine at position 428 and a serine at position 434, respectively, and a tyrosine at position 252, a threonine at position 254 and a glutamic acid at position 256, respectively; - mAb2 and mAb8, mAb9, mAb11, mAb13 and mAb14, which comprise a tyrosine at position 252, a threonine at position 254 and a glutamic acid at position 256, a leucine at position 428 and a serine at position 434, respectively, and a glutamine at position 12, a tyrosine at position 252, a threonine at position 254 and a glutamic acid at position 256, and a glutamine at position 12, a leucine at position 428 and a serine at position 434, respectively, of their CDRH3; Each of these was measured.

[0304] Among the substitutions in the Fc region of mAb1 and mAb2 that enhanced binding to FcRn, tyrosine at position 252, threonine at position 254, and glutamic acid at position 256 led to reduced binding affinity to FcγRIIIa, which may have a negative impact on cell-mediated effector function and anti-CHIKV therapy compared to mAb1 or mAb2 without Fc region substitutions. Other mutations such as alanine at position 434, alanines at positions 307, 380, and 434, respectively, glutamine at position 250 and leucine at position 428, leucine at position 428, and serine at position 434, respectively, retained binding affinity to FcγRIIIa.

[0305] an alanine at position 434, an alanine at positions 307, 380, and 434, respectively, or a glutamine at position 250 and a leucine at position 428, or a leucine at position 428 and mAb1 and mAb2, which contain a tyrosine at position 252, a threonine at position 254, and a serine at position 434, respectively, but do not contain a glutamic acid at position 256, retained FcγRIIIa binding, which is linked to effector function.

[0306] Therefore, mAb1 or mAb2 containing a leucine at position 428 and a serine at position 434 exhibits the best FcRn binding while retaining FcγRIIIa binding linked to effector function.

[0311] The results are shown in Table 8 below and in Figures 7A to 7C. mAb1 and mAb7 showed highly potent activity (EC 3.0 for mAb1) against viruses of all three genotypes of Asian, East Central and Southern African (ESCA) and West African CHIKV lineages, as shown in Figures 7A, 7B and 7C, respectively. 50 values <10 ng / mL for mAb 1 and <1 ng / mL for mAb 7). An anti-lysozyme antibody (mAb CTR) was used as a nonspecific negative control.

[0312] [Table 8] EXAMPLES

[0313] In vivo protection studies in mice In vivo studies using the DBA1 / J mouse model were performed in strict accordance with the recommendations in the Guide for the Care and Use of Laboratory Animals of the National Institutes of Health. Protocols were approved by the Institutional Animal Care and Use Committee at the National Institute of Allergy and Infection Diseases (NIAID). Infection experiments were performed in an A-BSL3 facility with approval from the NIAID Animal Studies Committee.

[0314] DBA / 1J mice aged 7–8 weeks were inoculated with 0.05 ml of CHIKV-LR2006 subcutaneously in the pad of the right hind paw toward the ankle. 5.5 CCID 50 Three days after CHIKV infection, 0.1 ml / mL of CHIKV-LR2006 was administered by subcutaneous inoculation in the footpad, followed by a single dose of recombinant human IgG anti-CHIKV mAb by intraperitoneal route at individual specific doses. Viral titers at the site of injection were monitored 5 days after infection. For prophylaxis studies, 105.5 CCID 50 Recombinant human IgG anti-CHIKV mAb was administered by intraperitoneal injection either 2, 7, or 14 days prior to subcutaneous infection of the footpad with 0.1 ml / 0.1 of CHIKV-LR2006. Viral titers at the site of injection were monitored 3 days after infection.

[0315] In vivo mAb-mediated prophylaxis Mice were treated with a single 250 μg dose (approximately 12.5 mg / kg) of recombinant hIgG anti-CHIKV mAb (mAb1, mAb7) or a control anti-isotype lysozyme mAb (mAb CTR All mice treated with isotype control mAb were pretreated with CCID 50 DBA1 / J-mice showed high virus titers 3 days after injection (μg tissue) (Figure 8). Pretreatment with both mAb7 and mAb1 at different times before infection (days -2 to -14) completely protected DBA1 / J-mice from the virus load at the site of injection, as the virus levels in their respective right hind legs were nearly equivalent to those of uninfected mice from the SHAM-PBS group.

[0316] In vivo mAb therapy after exposure DBA / 1J mice aged 7–8 weeks were given 10 0.05 ml of 100 mg ... 5.5 CCID 50 Each mouse was inoculated with 0.05 ml of 1000 mg / mL of CHIKV-LR. A single 250 μg dose of each individual mAb (mAb1, mAb2, mAb7, mAb11 or mAb14) was administered by intraperitoneal route 3 days after CHIKV infection. Viral titers at the site of injection were monitored 5 days after infection. All tested mAbs showed the same potency to neutralize tissue viral load in the mouse model of CHIKV infection (Figure 9A). mAb7 and mAb14 were further characterized in a dose titration study. For that purpose, DBA1 / J mice were inoculated with 0.05 ml of 1000 mg / mL of CHIKV-LR subcutaneously in the pad of the right hind paw towards the ankle. 5.5 CCID 50Mice were inoculated with 0.1 ml of CHIKV-LR2006 per 100 μg dose. mAb7 or mAb14 were given at 10, 25, 50, 100 and 250 μg doses (approximately 0.5, 1, 2.5, 5, 12.5 mg / kg) by a single intraperitoneal injection at 3 dpi. The primary outcome was the viral titer at 5 dpi at the site of viral challenge in the hind leg. mAb7 reduced the viral titer in the joints in DBA / 1J mice infected with CHIKV in a dose-dependent manner (Figure 9B). A significant reduction was observed at doses from 50 to 250 μg, with the highest effect achieved at the highest dose. mAb14 significantly reduced the viral titer whatever the dose, but no dose effect was observed in the tested conditions. EXAMPLES
